1Simulated effect of soil freeze-thaw process on surface hydrologic and thermal fluxes in frozen ground region of the Northern Hemisphere显示文摘Soil freeze-thaw process is closely related to surface energy budget,hydrological activity,and terrestrial ecosystems.In this study,two numerical experiments(including and excluding soil freeze-thaw process)were designed to examine the effect of soil freeze-thaw process on surface hydrologic and thermal fluxes in frozen ground region in the Northern Hemisphere based on the state-of-the-art Community Earth System Model version 1.0.5.Results show that in response to soil freeze-thaw process,the area averaged soil temperature in the shallow layer(0.0175−0.0451 m)decreases by 0.35℃in the TP(Tibetan Plateau),0.69℃in CES(Central and Eastern Siberia),and 0.6℃in NA(North America)during summer,and increases by 1.93℃in the TP,2.28℃in CES and 1.61℃in NA during winter,respectively.Meanwhile,in response to soil freeze-thaw process,the area averaged soil liquid water content increases in summer and decrease in winter.For surface heat flux components,the ground heat flux is most significantly affected by the freeze-thaw process in both summer and winter,followed by sensible heat flux and latent heat flux in summer.In the TP area,the ground heat flux increases by 2.82 W/m2(28.5%)in summer and decreases by 3.63 W/m2(40%)in winter.Meanwhile,in CES,the ground heat flux increases by 1.89 W/m2(11.3%)in summer and decreases by 1.41 W/m2(18.6%)in winter.The heat fluxes in the Tibetan Plateau are more susceptible to the freeze-thaw process compared with the high-latitude frozen soil regions.Soil freeze-thaw process can induce significant warming in the Tibetan Plateau in winter.Also,this process induces significant cooling in high-latitude regions in summer.The frozen ground can prevent soil liquid water from infiltrating to deep soil layers at the beginning of thawing;however,as the frozen ground thaws continuously,the infiltration of the liquid water increases and the deep soil can store water like a sponge,accompanied by decreasing surface runoff.The influence of the soil freeze-thaw process on surface hydrologic and thermal fluxes varies seasonally and spatially.Di Ma SiQiong Luo DongLin Guo ShiHua Lyu XianHong Meng BoLi Chen LiHui Luo 2021Research in Cold and Arid Regions2021,13,1:4

3PITX2 in pancreatic stellate cells promotes EMT in pancreatic cancer cells via the Wnt/β-catenin pathway显示文摘Since the prognosis of patients with pancreatic cancer is very poor and there is a lack of treatment methods,this study is performed to investigate the function of PITX2 in pancreatic stellate cells(PSCs)in the progression of pancreatic cancer.Scientific hypotheses are proposed according to bioinformatics analysis and tissue microarray analysis.Stable knockdown of PITX2 in PSCs is achieved through lentiviral infection.The relative expressions of PITX2,α-SMA,vimentin,CTNNB1,AXIN1 and LEF1 are measured in wild-type PSCs and PITX2-knockdown PSCs.Proliferative capacity is measured by EdU assay.After coculture with PSCs,the proliferation,invasion and migration capacity of pancreatic cancer cells are tested.EMT and Wnt/β-catenin downstream genes of pancreatic cancer cells are investigated to reveal the potential mechanism.Bioinformatics analysis reveals that the PITX2 gene is highly expressed in stromal cells in pancreatic cancer and is correlated with squamous-type PDAC.Analysis of PDAC tissue microarray further demonstrates that high PITX2 level in stromal cells is correlated with poor prognosis in PDAC.After stable knockdown of PITX2 in PSCs,the relative protein levels ofα-SMA,vimentin,CTNNB1,AXIN1 and LEF1 are decreased,and the proliferative capacity of PSCs is also decreased.After coculture with PSCs,in which PITX2 expression is downregulated,the proliferation,invasion and migration capacities of pancreatic cancer cells are inhibited.Thus,our results show that PITX2-silenced PSCs inhibit the growth,migration and invasion of pancreatic cancer cells via reduced EMT and Wnt/β-catenin signaling.Di Wu Weibo Chen Yang Yang Yi Qin Guangchen Zu Yue Zhang Yong An Donglin Sun Xiaowu Xu Xuemin Chen 2023Acta Biochimica et Biophysica Sinica2023,55,9:1
4Sensory nerves directly promote osteoclastogenesis by secreting peptidyl-prolyl cis-trans isomerase D(Cyp40)显示文摘Given afferent functions,sensory nerves have recently been found to exert efferent effects and directly alter organ physiology.Additionally,several studies have highlighted the indirect but crucial role of sensory nerves in the regulation of the physiological function of osteoclasts.Nonetheless,evidence regarding the direct sensory nerve efferent influence on osteoclasts is lacking.In the current study,we found that high levels of efferent signals were transported directly from the sensory nerves into osteoclasts.Furthermore,sensory hypersensitivity significantly increased osteoclastic bone resorption,and sensory neurons(SNs)directly promoted osteoclastogenesis in an in vitro coculture system.Moreover,we screened a novel neuropeptide,Cyp40,using an isobaric tag for relative and absolute quantitation(iTRAQ).We observed that Cyp40 is the efferent signal from sensory nerves,and it plays a critical role in osteoclastogenesis via the aryl hydrocarbon receptor(AhR)-Ras/Raf-p-Erk-NFATc1 pathway.These findings revealed a novel mechanism regarding the influence of sensory nerves on bone regulation,i.e.,a direct promoting effect on osteoclastogenesis by the secretion of Cyp40.Therefore,inhibiting Cyp40 could serve as a strategy to improve bone quality in osteoporosis and promote bone repair after bone injury.Junqin Li Bin Liu Hao Wu Shuaishuai Zhang Zhuowen Liang Shuo Guo Huijie Jiang Yue Song Xing Lei Yi Gao Pengzhen Cheng Donglin Li Jimeng Wang Yang Liu Di Wang Nazhi Zhan Jing Xu Lin Wang Guozhi Xiao Liu Yang GuoXian Pei 2023Bone Research2023,11,4:0
