| 1 | The GhREV transcription factor regulate the development of shoot apical meristem in cotton(Gossypium hirsutum)显示文摘Background:Manual topping is a routine agronomic practice for balancing the vegetative and reproductive growth of cotton(Gossypium hirsutum)in China,but its cost-effectiveness has decreased over time.Therefore,there is an urgent need to replace manual topping with new approaches,such as biological topping.In this study,we examined the function of Gh REV transcription factors(a classⅢhomeodomain-leucine zipper family,HD-ZIPⅢ)in regulating the development of shoot apical meristem(SAM)in cotton with the purpose of providing candidate genes for biological topping of cotton in the future.Results:We cloned four orthologous genes of At REV in cotton,namely Gh REV1,Gh REV2,Gh REV3,and Gh REV4.All the Gh REVs expressed in roots,stem,leaves,and SAM.Compared with Gh REV1 and Gh REV3,the expression level of Gh REV2 and Gh REV4 was higher in the SAM.However,only Gh REV2 had transcriptional activity.Gh REV2 is localized in the nucleus;and silencing it via virus-induced gene silencing(VIGS)produced an abnormal SAM.Two key genes,Gh WUSA10 and Gh STM,which involved in regulating the development of plant SAM,showed about 50%reduction in their transcripts in VIGS-Gh REV2 plants.Conclusion:Gh REV2 positively regulates the development of cotton SAM by regulating Gh WUSA10 and Gh STM potentially. | YANG Doudou AN Jing LI Fangjun ENEJI A.Agrinya TIAN Xiaoli LI Zhaohu | 2020 | Journal of Cotton Research2020,3,1: | 1 |
| 2 | Enhancing maize's nitrogen-fixing potential through ZmSBT3, a gene suppressing mucilage secretion∞显示文摘Maize(Zea mays)requires substantial amounts of nitrogen,posing a challenge for its cultivation.Recent work discovered that some ancient Mexican maize landraces harbored diazotrophic bacteria in mucilage secreted by their aerial roots.To see if this trait is retained in modern maize,we conducted a field study of aerial root mucilage(ARM)in 258 inbred lines.We observed that ARM secretion is common in modern maize,but the amount significantly varies,and only a few lines have retained the nitrogen‐fixing traits found in ancient landraces.The mucilage of the high‐ARM inbred line HN5‐724 had high nitrogen‐fixing enzyme activity and abundant diazotrophic bacteria.Our genome‐wide association study identified 17 candidate genes associated with ARM across three environments.Knockouts of one candidate gene,the subtilase family gene ZmSBT3,confirmed that it negatively regulates ARM secretion.Notably,the ZmSBT3 knockout lines had increased biomass and total nitrogen accumulation under nitrogen‐free culture conditions.High ARM was associated with three ZmSBT3 haplotypes that were gradually lost during maize domestication,being retained in only a few modern inbred lines such as HN5‐724.In summary,our results identify ZmSBT3 as a potential tool for enhancing ARM,and thus nitrogen fixation,in maize. | Jingyang Gao Peijiang Feng Jingli Zhang Chaopei Dong Zhao Wang Mingxiang Chen Zhongliang Yu Bowen Zhao Xin Hou Huijuan Wang Zhaokun Wu Razia Sultana Jemim Haidong Yu Doudou Sun Pei Jing Jiafa Chen Weibin Song Xuecai Zhang Zijian Zhou Jianyu Wu | 2023 | Journal of Integrative Plant Biology2023,65,12: | 0 |
| 3 | Three-dimensional-printed titanium prostheses with bone trabeculae enable mechanical-biological reconstruction after resection of bone tumours显示文摘Reconstruction after resection has always been an urgent problem in the treatment of bone tumours.There are many methods that can be used to reconstruct bone defects;however,there are also many complications,and it is difficult to develop a safe and effective reconstruction plan for the treatment of bone tumours.With the rapid development of digital orthopaedics,three-dimensional printing technology can solve this problem.The three-dimensional printing of personalised prostheses has many advantages.It can be used to print complex structures that are difficult to fabricate using traditional processes and overcome the problems of stress shielding and low biological activity of conventional prostheses.In this study,12 patients with bone tumours were selected as research subjects,and based on individualised reverse-engineering design technology,a three-dimensional model of each prosthesis was designed and installed using medical image data.Ti6Al4V was used as the raw material to prepare the prostheses,which were used to repair bone defects after surgical resection.The operation time was 266.43±21.08 minutes(range 180-390 minutes),and intraoperative blood loss was 857.26±84.28 mL(range 800-2500 mL).One patient had delayed wound healing after surgery,but all patients survived without local tumour recurrence,and no tumour metastasis was found.No aseptic loosening or structural fracture of the prosthesis,and no non-mechanical prosthesis failure caused by infection,tumour recurrence,or progression was observed.The Musculo-Skeletal Tumour Society(MSTS)score of limb function was 22.53±2.09(range 16-26),and ten of the 12 patients scored≥20 and were able to function normally.The results showed that three-dimensional printed prostheses with an individualised design can achieve satisfactory short-term clinical efficacy in the reconstruction of large bone defects after bone tumour resection. | Feifei Pu Wei Wu Doudou Jing Yihan Yu Yizhong Peng Jianxiang Liu Qiang Wu Baichuan Wang Zhicai Zhang Zengwu Shao | 2022 | Biomaterials Translational2022,3,2: | 0 |
| 4 | Influence of dissolved organic matter on methylmercury transformation during aerobic composting of municipal sewage sludge under different C/N ratios显示文摘Current knowledge about the transformation of total mercury and methylmercury(Me Hg) in aerobic composting process is limited. In this study, the composition and transformation of mercury and dissovled organic matter(DOM) in aerobic composting process of municipal sewage sludge were were comprehensively characterized, and the differences among the three C/N ratio(20, 26 and 30) were investigated. The main form of mercury in C/N 20 and 26 was organo-chelated Hg(F3, 46%-60%);while the main form of mercury in C/N 30 was mercuric sulfide(F5, 64%-70%). The main component of DOM in C/N 20 and 26 were tyrosine-like substance(C1, 53%-76%) while the main fractions in C/N 30 were tyrosine-like substance(C1, 28%-37%) and fulvic-like substance(C2, 17%-39%). The mercury and DOM varied significantly during the 9 days composting process. Compared to C/N 20 and 26, C/N 30 produced the less Me Hg after aerobic composting process, with values of 658%(C/N 20), 1400%(C/N 26) and 139%(C/N 30) of the initial, respectively. Meanwhile, C/N 30 produced the best compost showed greater degree of DOM molecular condensation and humification. Hg fraction had been altered by DOM, as indicated by a significant correlation between mercury species and DOM components. | 6 | Urine biomarkers discovery by metabolomics and machine learning for Parkinson’s disease diagnoses显示文摘Parkinson’s disease(PD)is a complex neurological disorder that typically worsens with age.A wide range of pathologies makes PD a very heterogeneous condition,and there are currently no reliable diagnostic tests for this disease.The application of metabolomics to the study of PD has the potential to identify disease biomarkers through the systematic evaluation of metabolites.In this study,urine metabolic profiles of 215 urine samples from 104 PD patients and 111 healthy individuals were assessed based on liquid chromatography-mass spectrometry.The urine metabolic profile was first evaluated with partial leastsquares discriminant analysis,and then we integrated the metabolomic data with ensemble machine learning techniques using the voting strategy to achieve better predictive performance.A combination of 8-metabolite predictive panel performed well with an accuracy of over 90.7%.Compared to control subjects,PD patients had higher levels of 3-methoxytyramine,N-acetyl-l-tyrosine,orotic acid,uric acid,vanillic acid,and xanthine,and lower levels of 3,3-dimethylglutaric acid and imidazolelactic acid in their urine.The multi-metabolite prediction model developed in this study can serve as an initial point for future clinical studies. | Xiaoxiao Wang Xinran Hao Jie Yan Ji Xu Dandan Hu Fenfen Ji Ting Zeng Fuyue Wang Bolun Wang Jiacheng Fang Jing Ji Hemi Luan Yanjun Hong Yanhao Zhang Jinyao Chen Min Li Zhu Yang Doudou Zhang Wenlan Liu Xiaodong Cai Zongwei Cai | 2023 | Chinese Chemical Letters2023,34,10: | 0 |

| 9 | Cellular senescence-driven transcriptional reprogramming of the MAFB/NOTCH3 axis activates the PI3K/AKT pathway and promotes osteosarcoma progression显示文摘Osteosarcoma is the most common primary malignancy of bones and primarily occurs in adolescents and young adults.However,a second smaller peak of osteosarcoma incidence was reported in the elderly aged more than 60.Elderly patients with osteosarcoma exhibit different characteristics compared to young patients,which usually results in a poor prognosis.The mechanism underlying osteosarcoma development in elderly patients is intriguing and of significant value in clinical applications.Senescent cells can accelerate tumor progression by metabolic reprogramming.Recent research has shown that methylmalonic acid(MMA)was significantly up-regulated in the serum of older individuals and played a central role in the development of aggressive characteristics.We found that the significant accumulation of MMA in elderly patients imparted proliferative potential to osteosarcoma cells.The expression of MAFB was excessively up-regulated in osteosarcoma specimens and was further enhanced in response to MMA accumulation as the patient aged.Specifically,we first confirmed a novel molecular mechanism between cellular senescence and cancer,in which the MMA-driven transcriptional reprogramming of the MAFB-NOTCH3 axis accelerated osteosarcoma progression via the activation of PI3K-AKT pathways.Moreover,the down-regulation of the MAFB-NOTCH3 axis increased the sensitivity and effect of AKT inhibitors in osteosarcoma through significant inhibition of AKT phosphorylation.In conclusion,we confirmed that MAFB is a novel age-dependent biomarker for osteosarcoma,and targeting the MAFB-NOTCH3 axis in combination with AKT inhibition can serve as a novel therapeutic strategy for elderly patients with osteosarcoma in experimental and clinical trials. | Zhenhao Zhang Doudou Jing Baijun Xuan Zhicai Zhang Wei Wu Zengwu Shao | 2024 | Genes & Diseases2024,11,2: | 0 |
