| 5 | Establishment of Real-Time Quantitative PCR Method for the Determination of Transposon Copy Number in Cronobacter sakazakii显示文摘[Objective] This study aimed to establish a Real-Time quantitative PCR method for the determination of transposon copy number in C.sakazakii.[Method] With single-copy housekeeping gene atpD as the reference gene,recombinant plasmid containing both single-copy housekeeping gene atpD and EZ-TN5 transposon was constructed;based on the established standard curves for real-time quantitative detection of atpD gene and EZ-TN5 transposon,copy number of atpD gene and EZ-TN5 transposon in three C.sakazakii mutants was detected and the ratio was calculated.[Result] Correlation coefficients of the standard curves for real-time quantitative detection of atpD gene and EZ-TN5 transposon were 0.999 and 0.998,respectively;the ratios of copy number of atpD gene and EZ-TN5 transposon in three C.sakazakii mutants were 0.98,1.17 and 0.91,respectively,which indicates that EZ-TN5 transposon in C.sakazakii mutants is a single-copy.[Conclusion] Real-time quantitative PCR method established in this study had high availability and could replace the Southern blot method to detect the copy number of EZ-TN5 transposon in different bacteria. | Fei WANG Xinjun DU Rong ZHANG Guixiang XU Shuo WANG | 2012 | Agricultural Biotechnology2012,1,1: | 0 |
| 6 | Study on Dynamic Characteristics of the Growth and Development of Armeniaca dasycarpa(Ehrh.) Borkh显示文摘[Objective]This study aimed to analyze the dynamics of growth and development of Armeniaca dasycarpa( Ehrh.) Borkh fruit,thus providing the theoretical basis for cultivation and breeding of good-quality and high-yield A. dasycarpa. [Method] With A. dasycarpa cultivar Yechengzixing as the experimental material,P. cerasifera Ehrh cultivar Huangguoyingtaoli and A. vulgaris Lam. cultivars Luopuhongdaike,Wanshujianali and Kuchetuoyong as control,the vertical diameter,horizontal diameter and fresh weight of fruit and embryos were measured every 7 d during the growth and development period. [Result] The result showed that the whole fruit growth period of Yechengzixing,lasted 93 d from the end of flowering to fruit maturity. The dynamics of fruit vertical diameter,horizontal diameter and fresh weight of Yechengzixing presented'rapid-slow-rapid'double S-shaped curves. The whole fruit growth period of Yechengzixing could be divided approximately into three stages: first rapid growth stage,slow growth stage and second rapid growth stage,which lasted 29,42 and 21 d,respectively. Daily increment of fruit vertical diameter and horizontal diameter of Yechengzixing reached the maximum at the first rapid growth stage,while daily increment of fruit fresh weight reached the maximum at the second rapid growth stage. The dynamics of vertical diameter,horizontal diameter and fresh weight of Yechengzixing embryos presented S-shaped curves. Fruit growth and development dynamics of the tested materials presented double S-shaped curves. The whole fruit growth period of Yechengzixing,Huangguoyingtaoli and three apricot cultivars lasted 93,119 and 77- 98 d,respectively. Thus,the duration of fruit growth period of Yechengzixing was 26 d shorter than that of Huangguoyingtaoli and exhibited no significant difference compared with three apricot cultivars. The duration of each growth stage and daily increment of Yechengzixing fruit had no significant difference compared with three apricot cultivars. Moreover,Huangguoyingtaoli had a longer slow growth stage and lower daily increment than Yechengzixing. The results showed that they have similar decomposition process, which occur in two steps. The first step is the loss of water and the second step is the decomposition of anhydrous complexes.But for cis-[Cu(gly)2]· H2O,the tempoerature of losing water is higher than that of trans-isomer. | 8 | Photo-induced Ag modulating carbon dots:Greatly improved fluorescent properties and derived sensing application显示文摘Carbon dots(CDs)have been attracted much attention and widely studied due to their excellent fluorescence(FL)properties,better biocompatibility and outstanding photo/chemical stability.However,the disadvantage of lower quantum yield(QY)still limits its wide application.Herein,we reported a novel and convenient strategy to prepare photo-induced Ag/CDs(p-Ag/CDs)by irradiating the mixed Ag+and hydrophobic CDs(h-CDs)acetone solution with ultraviolet(UV)light.The obtained p-Ag/CDs exhibit a greatly enhanced FL emission together with a blue shift(460 nm)than h-CDs(520 nm).The QY of p-Ag/CDs is measured to be 51.1%,which is 10.4 times higher than that of h-CDs(4.9%),indicating that photo-induced Ag modulation can effectively improve the optical properties of CDs.The mechanisms for the FL enhancement and blue shift of h-CDs are studied in detail.The results prove that the greatly enhanced FL emission is from the generated Ag nanoparticles(AgNPs)by UV light irradiation based on metal-enhanced fluorescence(MEF),and the increased oxygen-contained groups in this process lead to the blue shift in CDs fluorescence.Interestingly,the p-Ag/CDs exhibit higher sensitivity and selectivity for sulfide ions(S2-)detection than that of h-CDs,which have a lower response to S2-.This work not only offers a novel strategy to improve the FL properties of materials but also endows them with new functions and broadens their application fields. | Yuwei Wang Ye Li Guixiang Yang Xiaofeng Yang Chenglu Yan Huaqiao Peng Huiyong Wang Juan Du Baozhan Zheng Yong Guo | 2024 | Chinese Chemical Letters2024,35,1: | 0 |
