| 2 | Hepatitis B virus X protein enhances hepatocarcinogenesis by depressing the targeting of NUSAP1 mRNA by miR-18b显示文摘Objective: The aim of this study was to investigate the underlying mechanism whereby HBx modulates the targeting of NUSAP1 by miR-18b to enhance hepatocarcinogenesis.Methods: We employed an integrated approach of bioinformatics analysis and molecular experiments in hepatoma cells, HBV transgenic mice, and clinical liver cancer tissues to investigate the role of HBx-regulated miR-18b in the development of liver cancer.Results: In this study, we report that the HBx-mediated tumor suppressor miR-18b modulates hepatocarcinogenesis during the host-HBV interaction. The expression levels of miR-18b were lower in clinical HBV-positive liver cancer tissues and liver tissues of HBV-transgenic mice. Interestingly, HBx inhibited miR-18b expression by inducing the methylation of CpG islands in its promoter. Accordingly, we tested the hypothesis that HBx enhanced hepatocarcinogenesis by increasing the expression of target genes of miR-18b. Moreover, we identified nucleolar spindle-associated protein 1(NUSAP1) as one of the target genes of miR-18b.NUSAP1 was expressed at high levels in liver cancer tissues. Interestingly, HBx up-regulated NUSAP1 by suppressing miR-18b.Functionally, miR-18b significantly inhibited the proliferation of hepatoma cells by depressing NUSAP1 levels in vivo and in vitro.Conclusions: Thus, we conclude that the targeting of NUSAP1 mRNA by the tumor suppressor miR-18b is controlled by HBxmodulated promoter methylation during the host-virus interaction, leading to hepatocarcinogenesis. | 3 | Germline mutations in hereditary diffuse gastric cancer显示文摘Gastric cancer is one of the leading causes of cancer-related deaths worldwide. Among which, about 1%–3% of gastric cancer patients were characterized by inherited gastric cancer predisposition syndromes, knowing as hereditary diffuse gastric cancer(HDGC). Studies reported that CDH1 germline mutations are the main cause of HDGC. With the help of rapid development of genetic testing technologies and data analysis tools, more and more researchers focus on seeking candidate susceptibility genes for hereditary cancer syndromes. In addition, National Comprehensive Cancer Network(NCCN) guidelines recommend that the patients of HDGC carrying CDH1 mutations should undergo prophylactic gastrectomy or routine endoscopic surveillances. Therefore, genetic counseling plays a key role in helping individuals with pathogenic mutations make appropriate risk management plans. Moreover, experienced and professional genetic counselors as well as a systematic multidisciplinary team(MDT) are also required to facilitate the development of genetic counseling and benefit pathogenic mutation carriers who are in need of regular and standardized risk management solutions. In this review, we provided an overview about the germline mutations of several genes identified in HDGC, suggesting that these genes may potentially act as susceptibility genes for this malignant cancer syndrome. Furthermore, we introduced information for prevention, diagnosis and risk management of HDGC. | 4 | Lymphangiogenesis contributes to exercise-induced physiological cardiac growth显示文摘Background:Promoting cardiac lymphangiogenesis exerts beneficial effects for the heart.Exercise can induce physiological cardiac growth with cardiomyocyte hypertrophy and increased proliferation markers in cardiomyocytes.However,it remains unclear whether and how lymphangiogenesis contributes to exercise-induced physiological cardiac growth.We aimed to investigate the role and mechanism of lymphangiogenesis in exercise-induced physiological cardiac growth.Methods:Adult C57 BL6/J mice were subjected to 3 weeks of swimming exercise to induce physiological cardiac growth.Oral treatment with vascular endothelial growth factor receptor 3(VEGFR3) inhibitor SAR1 3 1 675 was used to investigate whether cardiac lymphangiogenesis was required for exercise-induced physiological cardiac growth by VEGFR3 activation.Furthermore,human dermal lymphatic endothelial cell(LEC)-conditioned medium was collected to culture isolated neonatal rat cardiomyocytes to determine whether and how LECs could influence cardiomyocyte proliferation and hypertrophy.Results:Swimming exercise induced physiological cardiac growth accompanied by a remarkable increase of cardiac lymphangiogenesis as evidenced by increased density of lymphatic vessel endothelial hyaluronic acid receptor 1-positive lymphatic vessels in the heart and upregulated LYVE-1 and Podoplanin expressions levels.VEGFR3 was upregulated in the exercised heart,while VEGFR3 inhibitor SAR131675 attenuated exercise-induced physiological cardiac growth as evidenced by blunted myocardial hypertrophy and reduced proliferation marker Ki67 in cardiomyocytes,which was correlated with reduced lymphatic vessel density and downregulated LYVE-1 and Podoplanin in the heart upon exercise.Furthermore,LEC-conditioned medium promoted both hypertrophy and proliferation of cardiomyocytes and contained higher levels of insulinlike growth factor-1 and the extracellular protein Reelin,while LEC-conditioned medium from LECs treated with SAR131675 blocked these effects.Functional rescue assays further demonstrated that protein kinase B(AKT) activation,as well as reduced CCAAT enhancer-binding protein beta(C/EBPβ) and increased CBP/p300-interacting transactivators with E(glutamic acid)/D(aspartic acid)-rich-carboxylterminal domain 4(CITED4),contributed to the promotive effect of LEC-conditioned medium on cardiomyocyte hypertrophy and proliferation.Conclusion:Our findings reveal that cardiac lymphangiogenesis is required for exercise-induced physiological cardiac growth by VEGFR3 activation,and they indicate that LEC-conditioned medium promotes both physiological hypertrophy and proliferation of cardiomyocytes through AKT activation and the C/EBPβ-CITED4 axis.These results highlight the essential roles of cardiac lymphangiogenesis in exercise-induced physiological cardiac growth. | Yihua Bei Zhenzhen Huang Xing Feng Lin Li Meng Wei Yujiao Zhu Shuqin Liu Chen Chen Mingming Yin Huimin Jiang Junjie Xiao | 2022 | Journal of Sport and Health Science2022,11,4: | 2 |
| 5 | Dual effect of WISP-1 in diverse pathological processes显示文摘Wnt-1 inducible signaling pathway-1(WISP-1), also known as CCN-4, belongs to the connective tissue growth factor(CTGF) family. WISP-1 is primarily expressed in embryonic stem cells and is involved in adult organ development. WISP-1 participates in many cellular processes, including proliferation, differentiation, apoptosis and adhesion. In addition, WISP-1 plays an important role in diverse pathophysiological processes, such as embryonic development, inflammation, injury repairs and cancers. Recent studies showed that WISP-1 was highly correlated with tumor progression and malignant transformation, whereas it played an oncogenic role in colorectal cancer,cholangiocarcinoma, hepatocellular carcinoma and breast cancer. However, interestingly, WISP-1 exerts a tumorsuppressing role in lung and prostate cancers. WISP-1 promotes cell proliferation, adhesion, motility, invasion,metastasis and epithelial-to-mesenchymal transition via particular signaling pathways. | 8 | MAGI1 inhibits migration and invasion via blocking MAPK/ERK signaling pathway in gastric cancer显示文摘Objective: To explore the association of membrane-associated guanylate kinase inverted 1(MAGI1) with gastric cancer(GC) and the related molecular mechanisms.Methods: The reverse transcription-polymerase chain reaction(RT-PCR) and immunohistochemistry(IHC)were utilized to measure the MAGI1 expression level in GC tissues. Quantitative real-time PCR and Western blotting were used to ensure the MAGI1 expression in GC cell lines. Small hairpin RNA(sh RNA) was applied for knockdown of endogenous MAGI1 in GC cells. MTT assay and colony formation assay, scratch wounding migration assay and transwell chamber migration assay, as well as transwell chamber invasion assay were employed respectively to investigate the GC cell proliferation, migration and invasion in MAGI1-knockdown and control GC cells. The potential molecular mechanism mediated by MAGI1 was studied using Western blotting and RT- PCR.Results: RT-PCR and IHC verified MAGI1 was frequently expressed in matched adjacent noncancerous mucosa compared with GC tissues and the expression of MAGI1 was related to clinical pathological parameters. Functional assays indicated that MAGI1 knockdown significantly promoted GC cell migration and invasion. | 11 | Treatment and survival status of patients with EGFR mutation-positive stage Ⅳ lung adenocarcinoma: five-year follow-up results in the Ordos Area of Inner Mongolia, China显示文摘Objective We aimed to determine the epidermal growth factor receptor(EGFR) mutation status and treatment survival of patients with stage Ⅳ lung adenocarcinoma living in the Ordos area of Inner Mongolia, China.Methods EGFR testing and first-line tyrosine kinase inhibitor(TKI) treatment rates of patients with stage Ⅳ lung adenocarcinoma were analyzed from June 2012 to June 2016. Kaplan-Meier survival curves were constructed to compare patients who received different treatment strategies and those harboring different EGFR mutation statuses.Results EGFR testing and mutation rates were 65.60% and 52.90%, respectively, and improved continuously from June 2012 to June 2016. Among patients with EGFR mutations, 38.9% had EGFR 19 del, 48.2% had L858 R, 4.2% had co-existing mutations in exons 19 and 21, and 8.4% had uncommon mutations. The median overall survival(OS) was 29.5, 26.5, and 16.0 months for patients receiving both TKI and chemotherapy, TKI alone, and chemotherapy alone, respectively(P = 0.047). The OS was 26.5 and 30.0 months for patients harboring EGFR 19 del and L858 R mutations, respectively(P = 0.096).Conclusion The high OS rates of stage Ⅳ lung adenocarcinoma patients living in the Ordos area may be attributed to continuous improvements in EGFR testing and first-line TKI treatment rates. | 13 | Integration of flexible,recyclable,and transient gelatin hydrogels toward multifunctional electronics显示文摘Facing the challenges posed by exponentially increasing e-waste,the development of recyclable and tran-sient electronics has paved the way to an environmentally-friendly progression strategy,where electron-ics can disintegrate and/or degrade into eco-friendly end products in a controlled way.Natural polymers possess cost and energy efficiency,easy modification,and fast degradation,all of which are ideal prop-erties for transient electronics.Gelatin is especially attractive due to its unique thermoreversible gelation processes,yet its huge potential as a multifunctional electronic material has not been well-researched due to its limited mechanical strength and low conductivity.Herein,we explored versatile applications of gelatin-based hydrogels through the assistance of multifunctional additives like carbon nanotubes to enhance their electromechanical performances.The optimized gelatin hydrogel displays not only a high conductivity of 0.93 S/m,electromagnetic shielding effectiveness of 39.6 dB,and tensile stress tolerance of 263 kPa,but also shows a negative permittivity phenomenon,which may find versatile applications in novel electronics.As a proof of concept,hydrogels were assembled as wearable sensors to sensitively de-tect static and dynamic pressures and strains generated by solids,liquids,and airflow,as well as diverse body movements.Furthermore,the recyclability,biocompatibility,and degradability of gelatin-based hy-drogels were well studied and analyzed.This work outlines a facile method to design multifunctional transient materials for wearable,sustainable,and eco-friendly electronics. | Rui Yin Chen Zhang Jian Shao Youyou Chen Ao Yin Qiang Feng Shuqin Chen Fei Peng Xing Ma Cheng-Yan Xu Feihua Liu Weiwei Zhao | 2023 | Journal of Materials Science & Technology2023,,14: | 0 |
