| 7 | A pH-responsive biomimetic drug delivery nanosystem for targeted chemo-photothermal therapy of tumors显示文摘Smart drug delivery nanosystem is significant for tumor treatments due to its possibility of temporally,spatially,and dose-controlled release.However,the therapeutic efficacy of drug delivery nanosystem is often compromised in cancer treatment as the enrichment of therapeutic agents in the reticuloendothelial system.Herein,doxorubicin(DOX)loaded biomimetic drug delivery nanosystem with macrophage cell membrane(MCM)camouflaged,MnFe_(2)O_(4)-DOX-MCM nanocube(NC),is developed for cancer treatment with tumor targeting,pH-stimuli drug release,and chemo-photothermal therapeutic effects.The nanosystem shows the capability of immune escape and enhanced cellular uptake of cancer cells due to the MCM decoration.Acid-labile bond between the MnFe2O4 NCs and DOX remains stable at physiological condition and release drugs immediately in response to the endo-/lysosome pH stimuli.Meanwhile,the photothermal effect of the nanosystem destroys tumor tissue,which further promotes chemotherapeutic efficacy.In vivo results demonstrate the tumor homing ability and produce a notable synergistic therapeutic effect of the NCs.Thus,biomimetic pH-responsive drug delivery nanosystem,MnFe_(2)O_(4)-DOX-MCM NCs,is an effective nanoplatform,which might be potential application for cancer synergistic treatment. | Yanmin Ju Zhiyi Wang Zeeshan Ali Hongchen Zhang Yazhou Wang Nuo Xu Hui Yin Fugeng Sheng Yanglong Hou | 2022 | Nano Research2022,15,5: | 1 |
| 8 | Temperature and Tumor Microenvironment Dual Responsive Mesoporous Magnetic Nanospheres for Magnetothermal Effect-Induced Cancer Theranostics显示文摘The development of smart drug delivery systems(SDDSs)based on engineered nanomaterials is important for clinical applications.Nevertheless,controllable administration of chemotherapeutic drugs for deep tumors and the avoidance of side effects caused by off-targeting during delivery remain a great challenge.Herein,a stimulus-responsive system of mesoporous nanospheres(composed of Cu@Fe_(2)C@mSiO_(2))with good magnetothermal effect is introduced into the tumor microenvironment.This system plays an important role in image-guided controllable targeted drug delivery that is independent of tumor depth.Aggregation-induced emission luminogen-based fluorescence imaging and magnetic resonance imaging were utilized since these techniques visualize the delivery process in real time.In addition,the degraded nanocarriers showed high catalytic activity for Fenton and Fenton-like reactions,upregulating the level of hydroxyl radicals(•OH)in cancer cells to realize chemodynamic therapy.The induced•OH led to the overexpression of pho-STAT3,activating the STAT3 signaling pathway,eventually inducing cancer cell apoptosis.Through metabolic monitoring,this SDDS is removed from the body after its degradation in vivo.The synergistically enhanced therapeutic effect was obtained in the chemo-chemodynamic therapy of 4T1 tumor-bearing mice,offering a platform for efficient cancer therapy with a personalized theranostic strategy. | Zhiyi Wang Shuren Wang Xiaoguang Zhang Ziyuan Li Zeeshan Ali Donghai Yu Hongtao Zhang Fugeng Sheng Song Gao Yanglong Hou | 2023 | CCS Chemistry2023,5,2: | 0 |
| 9 | A Thylakoid Membrane Protein Functions Synergistically with GUN5 in Chlorophyll Biosynthesis显示文摘Chlorophyll(Chl)is essential for photosynthetic reactions and chloroplast development.While the enzymatic pathway for Chl biosynthesis is well established,the regulatory mechanism underlying the homeostasis of Chl levels remains largely unknown.In this study,we identified CBD1(Chlorophyll Biosynthetic Defect1),which functions in the regulation of chlorophyll biosynthesis.The CBD1 gene was expressed specifically in green tissues and its protein product was embedded in the thylakoid membrane.Furthermore,CBD1 was precisely co-expressed and functionally correlated with GUN5(Genome Uncoupled 5).Analysis of chlorophyll metabolic intermediates indicated that cbd1 and cbd1gun5 mutants over-accumulatedmagnesium protoporphyrin Ⅸ(Mg-Proto Ⅸ).In addition,the cbd1 mutant thylakoid contained less Mg than the wild type not only as a result of lower Chl content,but also implicating CBD1 in Mg transport.This was supported by the finding that CBD1 complemented a Mg^(2+)uptake-deficient Salmonella strain under low Mg conditions.Taken together,these results indicate that CBD1 functions synergistically with CHLH/GUN5 in Mg-Proto IX processing,and may serve as a Mg-transport protein to maintain Mg homeostasis in the chloroplast. | Chi Zhang Bin Zhang Baicong Mu Xiaojiang Zheng Fugeng Zhao Wenzhi Lan Aigen Fu Sheng Luan | 2020 | Plant Communications2020,1,5: | 0 |
| 10 | A SPX domain vacuolar transporter links phosphate sensing to homeostasis in Arabidopsis显示文摘Excess phosphate(Pi)is stored into the vacuole through Pi transporters so that cytoplasmic Pi levels remain stable in plant cells.We hypothesized that the vacuolar Pi transporters may harbor a Pi-sensing mechanism so that they are activated to deliver Pi into the vacuole only when cytosolic Pi reaches a threshold high level.We tested this hypothesis using Vacuolar Phosphate Transporter 1(VPT1),a SPX domain-containing vacuolar Pi transporter,as a model.Recent studies have defined SPX as a Pi-sensing module that binds inositol polyphosphate signaling molecules(InsPs)produced at high cellular Pi status.We showed here that Pi-deficient conditions or mutation of the SPX domain severely impaired the transport activity of VPT1.We further identified an auto-inhibitory domain in VPT1 that suppresses its transport activity.Taking together the results from detailed structure-function analyses,our study suggests that VPT1 is in the auto-inhibitory state when Pi status is low,whereas at high cellular Pi status InsPs are produced and bind SPX domain to switch on VPT1 activity to deliver Pi into the vacuole.This thus provides an auto-regulatory mechanism for VPT1-mediated Pi sensing and homeostasis in plant cells. | Mingda Luan Fugeng Zhao Guangfang Sun Min Xu Aigen Fu Wenzhi Lan Sheng Luan | 2022 | Molecular Plant2022,15,10: | 0 |
