| 1 | Soil Organic Matter Dynamics Along a Vertical Vegetation Gradient in the Gongga Mountain on the Tibetan Plateau显示文摘Our knowledge about soil organic matter (SOM) dynamics is limited although this is an important issue in the study of responses of ecosystems to global climate changes. Twelve sampling plots were set up every 200 m from 1 700 to 3 900 m along the vertical vegetation gradient along the east slope of Gongga Mountain. Samples were taken from all 12 plots for SOM content measurement, although only 5 of the 12plots were subjected to radiocarbon measurements. A radiocarbon isotope method and a time-dependent model were used to quantify the SOM dynamics and SOM turnover rates along the vertical vegetation gradient. The results showed that the SOM turnover rate decreased and turnover time increased with soil depth for all vegetation types. The litter layer turnover rates presented a clear trend along the gradient. The litter layer turnover rates decreased with an increase in elevation, except that the litter layer turnover rate of mixed forest was higher than that of evergreen forest. Climatic factors, such as temperature and precipitation,were the main factors influencing the surface soil carbon dynamics. The turnover rates of the subsoil (including the A, B, and C horizons in the soil profiles) along the vertical gradient had no clear trends. The SOM of subalpine shrub and meadow turned over more slowly than that of the forest types in almost all soil horizons. The characteristic of short roots distributing in the upper part of the soil profile leads to different SOM dynamics of shrub and meadow compared with the forest types. Coniferous and mixed forests were susceptible to carbon loss from the young carbon pool, but their long and big roots resulted in high △14C values of the deep soil profiles and increased the input of young carbon to the deep soil. In evergreen forest,the carbon cumulative ability from the B horizon to the C horizon was weak. | 4 | Protective effects of Rheum tanguticum polysaccharide against hydrogen peroxide-induced intestinal epithelial cell injury显示文摘AIM: To describe the effect of Rheum tanguticum polysaccharide (RTP) on hydrogen peroxide-induced human intestinal epithelial cell injury.METHODS: Hydrogen peroxide (100 μmol/L) was introduced to induce human intestinal epithelial cell injury.Cells were pretreated with RTP (30,100,300 μg/mL) for 24 h before exposure to hydrogen peroxide. Cell viability was detected by MTr assay and morphological observation.Acridine orange staining and flow cytometry were performed to assess cell apoptosis. Lactate dehydrogenase (LDH) activity, production of malondialdehyde (MDA) and superoxide dismutase (SOD) activity were measured by spectrophotometry with corresponding assay kits.RESULTS: Following exposure to H2O2, a marked decrease in cell survival and SOD activity, increased production of MDA, LDH leakage and cell apoptosis were found.Pretreatment of the cells with RTP could significantly elevate cell survival, SOD activity and decrease the level of MDA, LDH activity and cell apoptosis.CONCLUSION: RTP may have cytoprotective and antioxidant effects against H2O2-induced intestinal epithelial cell injury by inhibiting cell apoptosis and necrosis. | 5 | X-box-binding protein 1-modified neural stem cells for treatment of Parkinson's disease显示文摘X-box-binding protein 1-transfected neural stem cells were transplanted into the right lateral ventricles of rats with rotenone-induced Parkinson's disease. The survival capacities and differentiation rates of cells expressing the dopaminergic marker tyrosine hydroxylase were higher in X-box-binding protein 1-transfected neural stem cells compared to non-transfected cells. Moreover, dopamine and 3,4-dihydroxyphenylacetic acid levels in the substantia nigra were significantly increased, α-synuclein expression was decreased, and neurological behaviors were significantly ameliorated in rats following transplantation of X-box-binding protein 1-transfected neural stem cells. | 6 | Heme oxygenase-1 alleviates ischemia/reperfusion injury in aged liver显示文摘AIM: To investigate if ischemia/reperfusion (I/R) injury in aged liver could be alleviated by heme oxygenase-1 (HO-1).METHODS: Three groups of SD rats (16 mo old) were studied. Group 1: control donors received physiological saline 24 h before their livers were harvested; group 2: donors were pretreated with hemih 24 h before their livers were harvested; and group 3: donors received hemin 24 h before their livers were harvested and zinc protoporphyrin (ZnPP,HO-1 inhibitor) was given to recipients at reperfusion. The harvested livers were stored in University of Wisconsin solution (4 ℃) for 6 h, and then transplanted to syngeneic rats. Serum glutamic oxaloacetic transaminase (SGOT),apoptotic cells, and apoptotic gene were measured 3, 6,12, 24, 48 h after reperfusion. We measured the apoptotic index by TUNEL, determined the expression of antiapoptotic Bcl-2 and proapoptotic (caspase-3) gene products by Western blot.RESULTS: After 3, 6, 12, 24, and 48 h of reperfusion, the SGOT levels (584.4±85.8 u/L, 999.2±125.2 u/L, 423.4±161.3u/L, 257.8±95.8 u/L, and 122.4±26.4 u/L) in hemin group were significantly (all P<0.05) lower than those in saline group (1082.2±101.2 u/L, 1775.2±328.3 u/L, 840.4±137.8 u/L,448.6±74.3 u/L, and 306.2±49.3 u/L). Liver HO-1 enzymatic activity correlated with beneficial effects of hemin and deleterious effects of adjunctive ZnPP treatment. Markedly less apoptotic (TUNEL+) liver cells 3, 6, 12, 24, and 48 h after reperfusion (5.16±0.73, 10.2±0.67, 9.28±0.78, 7.14±1.12,and 4.78±0.65) (P<0.05) could be detected in hemin liver grafts, as compared to controls (7.82±1.05, 15.94±1.82,11.67±1.59, 8.28±1.09, and 6.36±0.67). We detected the increased levels of Bcl-2 (1.5-fold) expression and compared with saline controls. These differences were most pronounced at 12 h after transplantation. In contrast, an active form of proapoptotic caspase-3 (p20) protein was found to be 2.9-fold lower at 24 h in hemin-pretreated group, as compared to saline liver transplant controls.CONCLUSION: HO-1 overexpression can provide potent protection against cold I/R injury. The microcrystalline structure of rapidly solidified Cu-Cr-Sn-Zn alloy is smaller grain structure examined by optical metallography. The effects of aging processes on the microstructure and properties of the lead frame alloy were investigated. | 10 | Silencing the gene encoding C/EBP homologous protein lessens acute brain injury following ischemia/reperfusion显示文摘C/EBP homologous protein, an important transcription factor during endoplasmic reticulum stress, participates in cell apoptosis mediated by endoplasmic reticulum stress. Previous studies have shown that C/EBP homologous protein mediates nerve injury during Alzheimer's disease, subarachnoid hemorrhage and spinal cord trauma. In this study, we introduced C/EBP homologous protein short hairpin RNA into the brains of ischemia/reperfusion rat models via injection of lentiviral vector through the left lateral ventricle. Silencing C/EBP homologous protein gene expression significantly reduced cerebral infarction volume, decreased water content and tumor necrosis factor-α and interleukin-1β mRNA expression in brain tissues following infarction, diminished the number of TUNEL-positive cells in the infarct region, decreased caspase-3 protein content and increased Bcl-2 protein content. | 12 | Suppressing interfacial side reactions of zinc metal anode via isolation effect toward high-performance aqueous zinc-ion batteries显示文摘Aqueous zinc(Zn)-ion batteries(AZIBs)are one of the most promising large-scale energy storage devices because of the excellent features of zinc metal anodes,including high theoretical capacity(5,855 mAh·cm^(–3)and 820 mAh·g^(−1)),high safety,and natural abundance.Nevertheless,the large-scale applications of AZIBs are mainly limited by the severe interfacial side reactions of zinc metal anodes,which results in low plating/stripping Coulombic efficiency and poor cycling stability.To address this issue,we report an artificial Ta_(2)O_(5)protective layer on zinc foil(Ta_(2)O_(5)@Zn)for suppressing side reactions during Zn deposition/stripping.The results of density functional theory calculation and experiments indicate that Ta_(2)O_(5)@Zn anode can inhibit the side reactions between the electrolyte and zinc anode through the isolation effect.Benefiting from this advantage,the symmetric cells with Ta_(2)O_(5)@Zn anode delivered an ultralong lifespan of 3,000 h with a low overpotential at 0.25 mA·cm^(−2)for 0.05 mAh·cm^(−2).Furthermore,the full cells consisting of Ta_(2)O_(5)@Zn anode and MnO_(2)or NH_(4)V_(4)O_(10)cathode all present outstanding electrochemical performance,indicating its high reliability in practical applications.This strategy brings new opportunities for the future development of rechargeable AZIBs. | Feng Tao Kaijia Feng Yong Liu Jiangzhuo Ren Yi Xiong Chengbo Li Fengzhang Ren | 2023 | Nano Research2023,16,5: | 2 |
| 13 | AT_1 receptor in rostral ventrolateral medulla mediating blunted baroreceptor reflex in spontaneously hypertensive rats显示文摘AIM: To determine the role of AT1 receptor in the rostral ventrolateral medulla (RVLM) in mediating the blunted baroreceptor reflex in spontaneously hypertensive rats (SHR). METHODS: Intravenous injections of graded doses of phenylephrine (1, 5, 10, 20, and 40μg/kg) increased the blood pressure to elicit the baroreceptor reflex in both SHR and normotensive Wistar rats anesthetized with urethane. The baroreceptor reflex sensitivity (BRS) was determined before and after microinjection of Ang II, losartan, or AT1 receptor antisense oligodeoxynucleotides into the RVLM. AT1 receptor protein level in the RVLM was measured by Western blotting. RESULTS: The BRS was significantly decreased in SHR compared with normal rats. Bilateral microinjection of AT~ receptor antagonist losartan (250 nmol/h) into the RVLM partly reversed the blunted BRS in SHR, but had no significant effect on the BRS in normal rats. Ang II (1.5 nmol/h) significantly inhibited the BRS in normal rats, which was completely abolished by pretreatment with losartan. However, no significant change in the BRS was observed after microinjection of Ang Ⅱ into the RVLM in SHR. Bilateral microinjection of AT1 receptor antisense oligodeoxynucleotides (ASODN) into the RVLM partially recovered the blunted BRS in SHR after 3 h, but no significant change in the BRS was observed in normal rats. The AT1 receptor protein level significantly decreased after administration of ASODN. | 15 | PC-407 inhibited proliferation and induced apoptosis in human colon cancer SW-1116 cells显示文摘AIM: To study whether PC-407 [4-[5-naphthyl-3- (trifluoromethyl)-1H-pyrazol-1-yl] benzenesulfonamide] inhibits cell viability and induces apoptosis in human colon cancer SW-1116 cells. METHODS: Inhibition of SW-1116 proliferation was measured by MTT assay. Morphological assessment of apoptosis was performed with fluorescence microscope and electron microscope. DNA fragmentation was visualized by agarose gel electrophoresis.The amount of apoptotic cells was measured by flow cytometry. RESULTS: PC-407 inhibited SW-1116 cell proliferation in a concentration-dependent manner after 3 d of treatment, and the IC5o for PC-407 inhibition of cell number was 16.67±0.17μmol/L. After incubation of SW-1116 cells with PC-407 20μmol/L for 24 h, morphological changes of typical apoptosis were observed by AO/EB staining or transmission electron microscopy. Flow cytometry analysis showed that PC-407 induced apoptosis in SW-1116 cells in a time- and concentration-dependant manner. The agarose gel electrophoresis of DNA revealed a 'ladder' pattern 48 h later. | 17 | Complete Genome Sequences of the SARS-CoV: the BJ Group (Isolates BJ01-BJ04)显示文摘Beijing has been one of the epicenters attacked most severely by the SARS-CoV (severe acute respiratory syndrome-associated coronavirus) since the first patient was diagnosed in one of the city's hospitals. We now report complete genome sequences of the BJ Group, including four isolates (Isolates B J01, B J02, B J03,and B J04) of the SARS-CoV. It is remarkable that all members of the BJ Group share a common haplotype, consisting of seven loci that differentiate the group from other isolates published to date. Among 42 substitutions uniquely identifled from the BJ group, 32 are non-synonymous changes at the amino acid level.Rooted phylogenetic trees, proposed on the basis of haplotypes and other sequence variations of SARS-CoV isolates from Canada, USA, Singapore, and China, gave rise to different paradigms but positioned the BJ Group, together with the newly discovered GD01 (GD-Ins29) in the same clade, followed by the H-U Group (from Hong Kong to USA) and the H-T Group (from Hong Kong to Toronto), leaving the SP Group (Singapore) more distant.
