| 3 | High-throughput screening for amyloid-β binding natural small-molecules based on the combinational use of biolayer interferometry and UHPLC−DAD-Q/TOF-MS/MS显示文摘Discovery of drugs rapidly and effectively is an important aspect for Alzheimer's disease(AD).In this study,a novel high-throughput screening(HTS)method aims at screening the small-molecules with amyloid-β(Aβ)binding affinity from natural medicines,based on the combinational use of biolayer interferometry(BLI)and ultra-high-performance liquid chromatography coupled with diode-array detector and quadrupole/time-of-flight tandem mass spectrometry(UHPLC−DAD-Q/TOF-MS/MS)has been firstly developed.Briefly,the components in natural medicines disassociated from biotinylated Aβwere collected to analyze their potential Aβbinding affinity by UHPLC−DAD-Q/TOF-MS/MS.Here,baicalein was confirmed to exhibit the highest binding affinity with Aβin Scutellaria baicalensis.Moreover,polyporenic acid C(PPAC),dehydrotumulosic acid(DTA),and tumulosic acid(TA)in Kai-Xin-San(KXS)were also identified as potent Aβinhibitors.Further bioactivity validations indicated that these compounds could inhibit Aβfibrillation,improve the viability in Aβ-induced PC-12 cells,and decrease the Aβcontent and improve the behavioral ability in Caenorhabditis elegans.The molecular docking results confirmed that PPAC,DTA,and TA possessed good binding properties with Aβ.Collectively,the present study has provided a novel and effective HTS method for the identification of natural inhibitors on Aβfibrillation,which may accelerate the process on anti-AD drugs discovery and development. | Minsong Guo Fengdan Zhu Wenqiao Qiu Gan Qiao Betty Yuen-Kwan Law Lu Yu Jianming Wu Yong Tang Chonglin Yu Dalian Qin Xiaogang Zhou Anguo Wu | 2022 | Acta Pharmaceutica Sinica B2022，12，4： | 3 |
| 4 | Disk Tree——A Case of Parallel Storage Architecture to Improve Performance in Random Access Pattern显示文摘Parallelism is an important way to improve the performance of a storage device. RAID (Redundant array of independent disk) is an example, which uses multi channels to realize parallelism of disk operation. While,because of the limits of bus and channel speed, the number of parallel channels in RAID cannot be increased as far as we wish. In order to break the bottleneck of bus, this paper presents a new approach of storage architecture, disk tree. Disk tree is a tree structure and uses unit controller as a node, which has one channel to connect with up node and two or three channels to connect with down nodes. Disk is the leaf of this tree. In host, multi storage spaces of multi channels are virtualized to a storage space. | 15 | Statistical analysis on protein-protein interface in crystals: Specific and non-specific interfaces are differentially distributed显示文摘The distribution of contact areas, or fractions of contacting, of protein-protein interfaces in crystals of pure polypeptides contains two components: a major exponential distribution and a minor flatter distribution. Suppose the two components belong to specific and non-specific contacts,respectively, then the probability of a contact with a givena rea, or fraction of contacting, can be estimated.
