1Molecular cloning and expression analysis of Crustin-like gene from Chinese shrimp Fenneropenaeus chinensis显示文摘A new member of antimicrobial protein genes of the Crustin family was cloned from haemocytes of the Chinese shrimp Fenneropenaeus chinensis by 3′and 5′ RACE. The full-length cDNA of Crustin-like gene contains a 390 bp open reading frame, encoding 130 amino acids. The deduced peptide contains a putative signal peptide of 17 amino acids and mature peptide of 113 amino acids. The molecular mass of the deduced mature peptide is 12.3 ku. It is highly cationic with a theoretical isoelectric point of 8.5. The deduced amino acids sequence of this Crustin showed high homology with those of Penaeus (Litopenaeus) setferus. Northern blotting showed that the cloned Crustin gene was mainly expressed in haemocytes, gill, intestine, and RNA in situ hybridization indicated that the Crustin gene was constitutively expressed exclusively in haemocytes of these tissues. Capillary electrophoresis RT-PCR analysis showed that Crustin was up-regulated dramatically from 12 to 48 h after a brief decrease of mRNA during first 6 h in response to microbe infection. The level of Crustin mRNA began to restore at 72 h post-challenge. 4Clinical Study on the Anti-Aging Effect of a Cream Containing Ginseng Root Extract and L-Carnosine显示文摘The anti-aging effects of a cream containing L-carnosine and ginseng root extract to provide a reference for the evaluation and development of anti-aging cosmetics were tested. Sixty healthy females, aged 35-60,were grouped into test sample and placebo groups. Skin elasticity, gloss, and wrinkles of the participants were analyzed before use and after use for 8 weeks. Instrument measurement results showed that after 8 weeks,skin elasticity, gloss, and wrinkles in the members of the test sample group obviously improved relative to those of the placebo group. Clinical effect assessment results showed that subjects did not experience adverse skin reactions, and their skin gloss showed obvious improvement. Self-assessment results showed that 87%subjects are satisfied with the test sample. 5Acetylation of ezrin regulates membrane–cytoskeleton interaction underlying CCL18-elicited cell migration显示文摘Ezrin,a membrane–cytoskeleton linker protein,plays an essential role in cell polarity establishment,cell migration,and division.Recent studies show that ezrin phosphorylation regulates breast cancer metastasis by promoting cancer cell survivor and promotes intrahepatic metastasis via cell migration.However,it was less characterized whether there are additional post-translational modifications and/or post-translational crosstalks on ezrin underlying context-dependent breast cancer cell migration and invasion.Here we show that ezrin is acetylated by p300/CBP-associated factor(PCAF)in breast cancer cells in response to CCL18 stimulation.Ezrin physically interacts with PCAF and is a cognate substrate of PCAF.The acetylation site of ezrin was mapped by mass spectrometric analyses,and dynamic acetylation of ezrin is essential for CCL18-induced breast cancer cell migration and invasion.Mechanistically,the acetylation reduced the lipid-binding activity of ezrin to ensure a robust and dynamic cycling between the plasma membrane and cytosol in response to CCL18 stimulation.Biochemical analyses show that ezrin acetylation prevents the phosphorylation of Thr567.Using atomic force microscopic measurements,our study revealed that acetylation of ezrin induced its unfolding into a dominant structure,which prevents ezrin phosphorylation at Thr567.Thus,these results present a previously undefined mechanism by which CCL18-elicited crosstalks between the acetylation and phosphorylation on ezrin control breast cancer cell migration and invasion.This suggests that targeting PCAF signaling could be a potential therapeutic strategy for combating hyperactive ezrin-driven cancer progression.Xiaoyu Song Wanjuan Wang Haowei Wang Xiao Yuan Fengrui Yang Lingli Zhao McKay Mullen Shihao Du Najdat Zohbi Saravanakumar Muthusamy Yalei Cao Jiying Jiang Peng Xia Ping He Mingrui Ding Nerimah Emmett Mingming Ma Quan Wu Hadiyah-Nicole Green Xia Ding Dongmei Wang Fengsong Wang Xing Liu 2020Journal of Molecular Cell Biology2020,12,6:1
6IRE1-RACK1 axis orchestrates ER stress preconditioning-elicited cytoprotection from ischemia/reperfusion injury in liver显示文摘Endoplasmic reticulum(ER)stress is involved in ischemic preconditioning that protects various organs from ischemia/reperfusion(I/R)injury.We established an in vivo ER stress preconditioning model in which tunicamycin was injected into rats before hepatic I/R.The hepatic I/R injury,demonstrated by serum aminotransferase level and the ultra-structure of the liver,was alleviated by administration of tunicamycin,which induced ER stress in rat liver by activating inositol-requiring enzyme1(IRE1)andupregulating78 kDaglucose-regulated protein(GRP78).The proteomic identification for IRE1 binders revealed interaction and cooperation among receptor for activated C kinase 1(RACK1),phosphorylated AMPK,and IRE1 under ER stress conditions in a spatiotemporal manner.Furthermore,in vitro ER stress preconditioningwas induced by thapsigargin and tunicamycin in L02 and Hep G2 cells.Surprisingly,BCL2 was found to bephosphorylated by IRE1 under ER stress conditions to prevent apoptotic process by activation of autophagy.In conclusion,ER stress preconditioning protects against hepatic I/R injury,which is orchestrated by IRE1-RACK1 axis through the activation of BCL2.Our findings provide novel insights into the molecular pathways underlying ER stress preconditioning-elicited cytoprotective effect against hepatic I/R injury.Dong Liu Xing Liu Ti Zhou William Yao Jun Zhao Zhigang Zheng Wei Jiang Fengsong Wang Felix O.Aikhionbare Donald L.Hill Nerimah Emmett Zhen Guo Dongmei Wang Xuebiao Yao Yong Chen 2016Journal of Molecular Cell Biology2016,8,2:1

11Logic devices based on nucleic acid self-assembly显示文摘Nucleic acids are natural macromolecules with the ability to store and transmit information based on the strict base-pairing principle.Beyond the natural nucleic acid double helixes,various DNA/RNA nanostructures with customized geometries and functionalities have been fabricated.Featured with programmability and sequence-dependent responsiveness,DNA/RNA nanostructures have been employed for the rational design and construction of logic devices.When stimulated by internal molecular triggers and/or external stimuli,these logic gate devices can operate at nanoscale level in complex biological environments,performing logic operations and producing corresponding outputs.In this minireview,we summarize the recent advances of nucleic acid logic devices,which are responsive to various stimuli,including DNA/RNA strands,metal ions,small molecules,peptides,proteins,photo-irradiation,pH changes,and so forth.The applications of these devices in biosensing and biofunction regulation are also included.In the last part of the present study,we discuss the remaining challenges and perspectives of nucleic acid logic devices.Xuehui Xu Yingxu Shang Fengsong Liu Qiao Jiang Baoquan Ding 2021InfoMat2021,3,10:0
12Logic-Gated Plasmonic Nanodevices Based on DNA-Templated Assembly显示文摘Life has evolved numerous elegant molecular machines that recognize biological signals and affect mechanical changes precisely to achieve specific and versatile biofunctions.Inspired by nature,synthetic molecular machines could be designed rationally to realize nanomechanical operations and autonomous computing.We constructed logic-gated plasmonic nanodevices through coassembly of two gold nanorods(AuNRs)and computing elements on a tweezer-shaped DNA origami template.After recognition of various molecular inputs,such as DNA strands,glutathione,or adenosine,the geometry and plasmonic circular dichroism(CD)signals of the AuNR–origami nanodevices produced corresponding changes.Then we designed and characterized a set of modular Boolean logic-gated nanodevices(YES,NOT,AND,OR)and proceeded to construct a complicated 3-input circuit capable of performing Boolean OR-NOT-AND operations.Our plasmonic logic devices transduced external inputs into conformational changes and near-infrared(NIR)chiral outputs.This DNA-based self-assembly strategy holds great potential for applications in programmable optical modulators,molecular information processing,and bioanalysis.Fengsong Liu Qiao Jiang Qing Liu Na Li Zihong Han Chao Liu Zhaoran Wang Yunfei Jiao Jiashu Sun Baoquan Ding 2021CCS Chemistry2021,3,3:0
