| 1 | Pathways of flower infection and pollen-mediated dispersion of Pseudomonas syringae pv. actinidiae, the causal agent of kiwifruit bacterial canker显示文摘Flowers can provide a protected and nutrient-rich environment to the epiphytic microflora,thus representing a sensible entry point for pathogens such as Pseudomonas syringae pv.actinidiae(Psa).This bacterium can colonize both male and female Actinidia flowers,causing flower browning and fall,and systemic invasion of the host plant,eventually leading to its death.However,the process of flower colonization and penetration into the host tissues has not yet been fully elucidated.In addition,the presence of Psa in the pollen from infected flowers,and the role of pollination in the spread of Psa requires confirmation.The present study employed a Psa strain constitutively expressing the fluorescent GFPuv protein,to visualize in vivo flower colonization.Microscopy observations were performed by means of confocal laser scanning and wide-field fluorescent microscopy,and were coupled with the study of Psa population dynamics by quantitative PCR(q-PCR).The pathogen was shown to colonize stigmata,move along the stylar furrow,and penetrate the receptacles via the style or nectarhodes.Once the receptacle was invaded,the pathogen migrated along the flower pedicel and became systemic.Psa was also able to colonize the anthers epiphytically and endophytically.Infected male flowers produced contaminated pollen,which could transmit Psa to healthy plants.Finally,pollinators(Apis mellifera and Bombus terrestris)were studied in natural conditions,showing that,although they can be contaminated with Psa,the pathogen’s transmission via pollinators is contrasted by its short survival in the hive. | Irene Donati Antonio Cellini Giampaolo Buriani Sofia Mauri Callum Kay Gianni Tacconi Francesco Spinelli | 2018 | Horticulture Research2018,5,1: | 3 |
| 2 | Thermal conductivity of glasses: first-principles theory and applications显示文摘Predicting the thermal conductivity of glasses from first principles has hitherto been a very complex problem.The established Allen-Feldman and Green-Kubo approaches employ approximations with limited validity—the former neglects anharmonicity,the latter misses the quantum Bose-Einstein statistics of vibrations—and require atomistic models that are very challenging for first-principles methods.Here,we present a protocol to determine from first principles the thermal conductivityκ(T)of glasses above the plateau(i.e.,above the temperature-independent region appearing almost without exceptions in theκ(T)of all glasses at cryogenic temperatures).The protocol combines the Wigner formulation of thermal transport with convergence-acceleration techniques,and accounts comprehensively for the effects of structural disorder,anharmonicity,and Bose-Einstein statistics.We validate this approach in vitreous silica,showing that models containing less than 200 atoms can already reproduceκ(T)in the macroscopic limit.We discuss the effects of anharmonicity and the mechanisms determining the trend ofκ(T)at high temperature,reproducing experiments at temperatures where radiative effects remain negligible. | Michele Simoncelli Francesco Mauri Nicola Marzari | 2023 | npj Computational Materials2023,,1: | 1 |

| 4 | Vulnerabilities in mIDH2 AML confer sensitivity to APL-like targeted combination therapy显示文摘Although targeted therapies have proven effective and even curative in human leukaemia, resistance often ensues. IDH enzymes are mutated in ~20% of human AML, with targeted therapies under clinical evaluation. We here characterize leukaemia evolution from mutant IDH2 (mIDH2)-dependence to independence identifying key targetable vulnerabilities of mIDH2 leukaemia that are retained during evolution and progression from early to late stages. Mechanistically, we find that mIDH2 leukaemia are metastable and vulnerable at two distinct levels. On the one hand, they are characterized by oxidative and genotoxic stress, in spite of increased 1-carbon metabolism and glutathione levels. On the other hand, mIDH2 leukaemia display inhibition of LSD1 and a resulting transcriptional signature of all-trans retinoic acid (ATRA) sensitization, in spite of a state of suppressed ATRA signalling due to increased levels of PIN1. We further identify GSH/ROS and PIN1/LSD1 as critical nodes for leukaemia maintenance and the combination of ATRA and arsenic trioxide (ATO) as a key therapeutic modality to target these vulnerabilities. Strikingly, we demonstrate that the combination of ATRA and ATO proves to be a powerfully synergistic and effective therapy in a number of mouse and human mIDH1/2 leukemic models. | 7 | Relevant incidental findings at abdominal multi-detector contrast-enhanced computed tomography:A collateral screening?显示文摘AIM: To investigate the prevalence of relevant incidental findings(RIFs) detected during routine abdominal contrast-enhanced computed tomography(Ce CT).METHODS: We retrospectively evaluated the reports of a consecutive series of abdominal Ce CT studies performed between January and May 2013. For each report, patients' age and sex, admission as inpatient or outpatient, clinical suspicion as indicated by the requesting physician, availability of a previous abdominal examination, and name of the reporting radiologist were recorded. Based on the clinical suspicion, the presence and features of any RIFs(if needing additional workup) was noted.RESULTS: One thousand forty abdominal Ce CT were performed in 949 patients(528 males, mean age 66 ±14 years). No significant difference was found between inpatients and outpatients age and sex distribution(P > 0.472). RIFs were found in 195/1040(18.8%) Ce CT [inpatients = 108/470(23.0%); outpatients = 87/570(15.2%); P = 0.002]. RIFs were found in 30/440(6.8%) Ce CT with a previous exam and in 165/600(27.5%) without a previous exam(P < 0.001). Radiologists' distribution between inpatients or outpatients was significantly different(P < 0.001). RIFs prevalence increased with aging, except for a peak in 40-49 year group. Most involved organs were kidneys, gallbladder, and lungs.CONCLUSION: A RIF is detected in 1/5 patients undergoing abdominal Ce CT.
