| 1 | Deletion of a Non-Catalytic Region Increases the Enzymatic Activity of a β-Agarase from Flammeovirga sp. MY04显示文摘A Glycoside hydrolase(GH) typically contains one catalytic module and varied non-catalytic regions(NCRs). However, effects of the NCRs to the catalytic modules remain mostly unclear except the carbohydrate-binding modules(CBMs). Aga G4 is a GH16 endo-β-agarase of the agarolytic marine bacterium Flammeovirga sp. MY04. The enzyme consists of an extra sugar-binding peptide within the catalytic module, with no predictable CBMs but function-unknown sequences in the NCR, which is a new characteristic of agarase sequences. In this study, we deleted the NCR sequence, a 140-amino acid peptide at the C-terminus and expressed the truncated gene, aga G4-T140, in Escherichia coli. After purification and refolding, the truncated agarase r Aga G4-T140 retained the same catalytic temperature and p H value as r Aga G4. Using combined fluorescent labeling, HPLC and MS/MS techniques, we identified the end-products of agarose degradation by r Aga G4-T140 as neoagarotetraose and neoagarohexaose, with a final molar ratio of 1.53:1 and a conversion ratio of approximately 70%, which were similar to those of r Aga G4. However, the truncated agarase r Aga G4-T140 markedly decreased in protein solubility by 15 times and increased in enzymatic activities by 35 times. The oligosaccharide production of r Aga G4-T140 was approximately 25 times the weight of that produced by equimolar r Aga G4. | 5 | Bandpass transmission spectra of a whisperinggallery microcavity coupled to an ultrathin fiber显示文摘Tapered fibers with diameters ranging from 1 to 4 μm are widely used to excite the whispering-gallery(WG)modes of microcavities. Typically, the transmission spectrum of a WG cavity coupled to a waveguide around a resonance assumes a Lorentzian dip morphology due to resonant absorption of the light within the cavity. In this paper, we demonstrate that the transmission spectra of a WG cavity coupled with an ultrathin fiber(500–700 nm)may exhibit both Lorentzian dips and peaks, depending on the gap between the fiber and the microcavity. | 10 | Dynamics and entanglement of a membrane-in-the-middle optomechanical system in the extremely-large-amplitude regime显示文摘The study of optomechanical systems has attracted much attention, most of which are concentrated in the physics in the smallamplitude regime. While in this article, we focus on optomechanics in the extremely-large-amplitude regime and consider both classical and quantum dynamics. Firstly, we study classical dynamics in a membrane-in-the-middle optomechanical system in which a partially reflecting and flexible membrane is suspended inside an optical cavity. We show that the membrane can present self-sustained oscillations with limit cycles in the shape of sawtooth-edged ellipses and exhibit dynamical multistability. Then, we study the dynamics of the quantum fluctuations around the classical orbits. By using the logarithmic negativity, we calculate the evolution of the quantum entanglement between the optical cavity mode and the membrane during the mechanical oscillation. | 11 | Very large group delay in VHF band using coupled high temperature superconducting resonators显示文摘Storing a very high frequency(VHF)band(30–300 MHz)electromagnetic wave has many potential applications,such as phase modulation,buffering,and radio frequency memory.It can be effectively achieved by applying coupled resonator-based electromagnetically induced transparency(EIT)due to its slow light effect.However,the wavelength in the VHF band is too long to design resonators,and the group delay is limited by the high resistive loss of metal.The practical application of EIT in the VHF band is still a big challenge.In this work,we propose and experimentally demonstrate EIT response in a high-temperature superconducting(HTS)microwave circuit with coupled-resonator-induced transparency.The chip size of the HTS circuit is only 34 mm×20 mm with a very low transparency frequency of 198.55 MHz.In addition,we implement very large group delay higher than 12.3μs and 16.2μs with working temperatures of 65 K and 50 K separately,which is much longer than the previous reported works on slow wave.The fabricated circuit is planar with working temperature about 65 K,and thus can be easily integrated into other microwave devices under the cryogenic conditions provided by a commercial portable Stirling cryocooler.Our proposed method paves a way for studying EIT in the microwave region due to the high quality factor of the HTS resonator,which has great potential use for radio-frequency memory in the future. | TIANNING ZHENG BIN WEI FUCHUAN LEI BISONG CAO | 2021 | Photonics Research2021,9,10: | 0 |

| 17 | Functional Cure of Chronic Hepatitis B with Antiviral Treatment in Children having High-level Viremia and Normal or Mildly Elevated Serum Aminotransferase显示文摘Background and Aims:There is a lack of data supporting the notion that antiviral treatments can beneft children with chronic hepatitis B(CHB)having high viremia and normal or mildly elevated serum alanine aminotransferase(ALT)levels.We aimed t analyze the efficacy of antiviral treatments in children with CHB and explore the factors associated with functional cure.Methods:Forty-eight children with CHB having high viremia and normal or mildly elevated serum ALT levels were screened in this real-world study.Thirty-two children received either interferon-alpha(IFN-α)monother-apy,IFN-a therapy with a nucleoside analog(NA)add-on,or IFN-α and NA combination therapy.The 16 children in the control group did not receive antiviral treatment.All 48 chil-dren were available for follow-up assessments for the entire 36-month study period.We identified a functional cure with respect to hepatitis B virus(HBV)DNA loss,loss/seroconver-sion of circulating hepatitis B e antigen(HBeAg),and loss of hepatitis B surface antigen(HBsAg)with or without serocon-version.Cox regression analysis was employed to evaluate the factors that may have influenced the functional cure.Re-sults:After 36 months,the cumulative functional cure rate was 56.25%(18/32)in the treated group and 0%(0/16)in the control group(p<0.001).In the treated group,the serum HBV DNA levels declined rapidly at the end of a 6-month visit and the cured children achieved a loss rate of 100%(18/18)within 16 months of beginning treatment,compared with 64.29%(9/14)of the uncured children(p<0.001).The rates of HBeAg seroconversion were significantly higher among the cured children than among the uncured children(p<0.001).All 16 children in the control group maintained high levels of serum HBV DNA and were positive for both serum HBeAg and HBsAg during the entire 36 months of the study period.Func tional cure was associated with younger ages(1-6 vs.7-14 years,p=0.013),CD8^(+)T lymphocyte counts(p=0.013),and B lymphocyte counts(p=0.003).No serious adverse events were observed.Conclusions:Antiviral treatment achieved a functional cure of CHB in a high proportion of children having high-level viremia and normal or mildly elevated ALT levels.Younger age and high peripheral lymphocyte counts were as sociated with this functional cure. | Jing Li Peiyao Fan Zhiqiang Xu Yi Dong Fuchuan Wang Weiguo Hong Jinfang Zhao Yinjie Gao Jianguo Yan Lili Cao Chao Zhang Shishu Zhu Fu-Sheng Wang Min Zhang | 2023 | Journal of Clinical and Translational Hepatology2023,11,5: | 0 |
