1Biological activities of a neutral water-soluble agar polysaccharide prepared by agarase degradation显示文摘Depolymerization of agar was performed using agarase, which was extracted from the cell-free medium of a culture of marine bacterial Alteromonas sp. nov. SY 37-12. After ethanol fractionation and lyophilization, the water-soluble agar polysaccharide (WSAP3) was collected. The anti-tumor activity of the product was determined by using Sarcoma 180 tumor in mouse. The tumor inhibition rate of WSAP3 reached 48.7% at a dose of 64mg kg-1 after 15 days treatment. WSAP3 enhanced the activities of superoxide dismutase and catalase, which suggests that WSAP3 was effective in promoting the antioxidation ability and eliminating danger from free radicals. The result of flow cytometry showed that the WSAP3 had no activities of cell cycle inhibition or apoptosis-inducing activities. The anti-oxidation of WSAP3 was further confirmed by test in vitro, which might play an important role in anti-tumor activity. The immunological regulation of WSAP3, especially its effect on the phagocytosis ratio and phagocytosis index of macrophage was also assayed in test in vivo.王静雪 Mou Haijin Jiang Xiaolu Guan Huashi 2005High Technology Letters2005,11,4:24
2Promotive Effects of Alginate-Derived Oligosaccharides on the Inducing Drought Resistance of Tomato显示文摘In order to determine the role of alginate-derived oligosaccharides (ADO) in drought stress resistance of tomato (Ly-copersicon esculentum Miller) seedlings, the leaves were exposed to different concentrations of ADO (0.05%, 0.10%, 0.20%, 0.30% and 0.50%) after drought stress was simulated by exposing the roots to 0.6 molL-1 PEG-6000 solution for 6 h. Changes in biomass, electrolyte leakage and malondialdehyde (MDA), free proline, total soluble sugars (TSS) and abscisic acid (ABA), the enzyme activities of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD) and phenylalanine ammonia-lyase (PAL) were measured to investigate the effects of ADO treatment. The results showed that the treatment with an ADO concentration of 0.20% exhibited the highest performance of drought stress resistance in the tomato seedlings by decreasing the electrolyte leakage and the concentration of MDA, increasing the contents of free proline, TSS and ABA, and increasing the activities of CAT, SOD, POD and PAL after treatment with ADO. It is suggested that changes in electrolyte leakage, MDA, osmotic solutes, ABA, anti-oxidative enzyme and PAL activities were responsible for the increased drought stress resistance in tomato seedlings. 3The Antioxidant Effects of Complexes of Tilapia Fish Skin Collagen and Different Marine Oligosaccharides显示文摘An excess of reactive oxygen species (ROS) leads to a variety of chronic health problems.As potent antioxidants, marine bioactive extracts containing oligosaccharides and peptides have been extensively studied.Recently, there is a growing interest in protein-polysaccharide complexes because of their potential uses in pharmaceutical and food industries.However, only few studies are available on the antioxidant activities of such complexes, in terms of their ROS scavenging capability.In this study, we combined different marine oligosaccharides (isolated and purified) with collagen peptides derived from tilapia fish skin, and evaluated the an-tioxidant activity of the marine peptide-oligosaccharide complexes vis-à-vis the activity of their original component molecules.Bio-chemical and cellular assays were performed to measure the scavenging effects on 1, 1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl and superoxide radicals, and to evaluate the influences on the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and the level of malondialdehyde (MDA) in UV-induced photoaging models.The results indicated that the antioxidant activities of all the complexes were stronger than those of their individual components.Among the 11 complexes tested, two complexes, namely MA1000+CP and κ-ca3000+CP, turned out to be highly effective antioxidants.Although the detailed mechanisms of this improved scavenging ability are not fully understood, this work provides insights into the design of highly efficient peptide-oligosaccharide complexes for potential applications in pharmaceutical, cosmetics and food industries.REN Shuwen LI Jing GUAN Huashi 2010Journal of Ocean University of China2010,9,4:5
4全球海岸带环境问题现状和相互联系显示文摘本研究对全球海岸带中的人口分布、土地覆盖物分布、生物多样性热点和保护区等的现状进行了评估。海岸带系指海岸线100km范围以内的陆地。本研究试图回答以下这样一些问题:海岸带人口拥挤程度如何?海岸带土地覆盖物分布型式怎样?海岸带有多少地区被划作保护区?生物多样性热点现状如何?人与海岸环境之间有何联系?在研究中,我们采用了根据遥感和其它来源得来的综合性且彼此协调一致的地球空间数据集。采用地理信息系统分层(Layering)方法和彼此兼容的数据集,使我们能够对某些海岸带所存在的问题及其相互关系进行鉴别和量化。预计这类信息可为全球海岸带的管理提供依据,并且有助于国家政策和国际政策的制定。HuaShi AshbinduSingh 刘林群 2003AMBIO-人类环境杂志2003,32,2:4
5Extraction,Isolation and Structural Characterization of Polysaccharides from a Red Alga Gloiopeltis furcata显示文摘Three kinds of polysaccharides:GFW,GFH and GFA,were sequentially extracted from a red alga Gloiopeltis furcatawith 25℃and 85℃water,and 60℃ 4%NaOHwater solution.Based on the defatted alga,the yields of the polysaccharide were57.9%,2.5%and 2.6%,respectively.Their monosaccharide compositions,average molecular weights and structural characters weredetermined by gas chromatography(GC),high performance liquid chromatography(HPLC),fourier transform infrared spectroscopy(FTIR)or13C-NMR spectroscopy.The results showed that GFW,GFH and GFA were all composed of D-galactose(Gal)and3,6-anhydro-L-galactose(AnG),and particularly GFA also contained xylose(Xyl).The average molecular weights of GFW,GFHand GFA were 22.6 kD,26.5 kD and 49.8 kD,respectively,with the respective sulfate content 31.2%,25.1%and 22.7%.The data ofFTIR and13C-NMR confirmed the sulfate ester location at C6 of galactose.It is concluded that all the three polysaccharides extractedfrom Gloiopeltis furcata were sulfated galactans,two being sulfated-agarose,and one being xylose-containing sulfated galactan.YU Guangli HU Yannan YANG Bo ZHAO Xia WANG Peipei JI Guoli WU Jiandong GUAN Huashi 2010Journal of Ocean University of China2010,9,2:3

7Bioassay-Guided Extraction of Crude Fucose-Containing Sulphated Polysaccharides from Sargassum fusiforme with Response Surface Methodology显示文摘The response surface methodology(RSM) combined with bioassays was employed to optimize the extraction process of crude fucose-containing sulphated polysaccharides(c FCSP) from Sargassum fusiforme. The central composite design(CCD) was used with four variables, five levels, and four responses. The four variables were p H value of hydrochloric acid solution, extraction temperature(℃), ratio of liquid to raw material(m L g^(-1)), and extraction time(h), respectively. Chemical and bioassay indices were used in combination as the response parameters, which included the yield of c FCSP, fucose content, proliferation rate of spleen cells, and lipopolysaccharide-induced proliferation of splenocytes. The experimental data were fitted to a second-order polynomial equation using multiple regression analysis, and examined using the appropriate statistical methods. The best extraction conditions were as follows: the p H value of hydrochloric acid solution was 3.50; the extraction temperature was 100℃; the ratio of liquid to raw material was 15.00 m L g^(-1) and the extraction time was 2.50 h. The experimental yield was close to the predicted from the model. The extract could promote spleen lymphocyte proliferation, especially the lipopolysaccharide-induced lymphocyte proliferation in vitro, which suggested that its immunomodulatory effect on B lymphocytes. 8Isolation of Protoplasts from Undaria pinnatifida by Alginate Lyase Digestion显示文摘The aim of this study is to isolate protoplasts from Undaria pinnatifida. Protoplasts of the alga were isolated enzymatically by using alginate lyase, which was prepared by fermenting culture of a strain Vibrio sp. 510. Monofacterial method was applied for optimizing digestion condition. The optimum condition for protoplast preparation is enzymatic digestion at 28 ℃ for 2 h using alginate lyase at the concentration of 213.36 U (8 mL) every 0.5 g fresh thalline with NaCl 50 and at the shaking speed of 150 r min -1 during digestion. The protoplast yield can reach 2.62±0.09 million per 0.5 g fresh leave under the optimum condition. 10Spatial and seasonal variations in bacterial communities of the Yellow Sea by T-RFLP analysis显示文摘Four typical coastal sites(rocky shore,sandy shore,mud flat shore,and artificial harbor)at the Yellow Sea were chosen to investigate the spatial and seasonal variations in bacterial communities.This was accomplished by using terminal restriction fragment length polymorphism(T-RFLP)analysis of PCR amplified 16S rDNA fragments.Two kinds of tetrameric restriction enzymes,HhaI and MspI,were used in the experiment to depict the bacterial community diversity in different marine environments.It was found that the community compositions digested by the two enzymes separately were different.However,the results of bacterial community diversity derived from them were similar.The MDA analysis results of T-RFLP profiles coming from HhaI and MspI both exhibited a significant seasonal community shift for bacteria and a relatively low spatial variation among the four locations.With HhaI as the sample,the pair wise Ttests also revealed that variations were minor between each pair of marine environments,with R ranging from 0.198 to 0.349.However,the bacterial community structure in the mud flat site depicted a larger difference than each of the other three sites(R ranging from 0.282 to 0.349).Hongyuan WANG Xiaolu JIANG Ya HE Huashi GUAN 2009Frontiers of Environmental Science & Engineering2009,3,2:2
