1Low-frequency HIFU induced cancer immunotherapy:tempting challenges and potential opportunities显示文摘Immunotherapy is playing an increasingly important role in the treatment of tumors.Different from the traditional direct killing or excision therapies,immunotherapy depends on autologous immunity to kill tumor cells and tissues by activating or enhancing the body's immune system.Large numbers of recent studies suggest that low-frequency HIFU can not only enhance the intensity of the body's anti-cancer immune response,but also improve the efficiency of immunotherapy drug delivery to strengthen the effects of tumor immunotherapy.The focused ultrasound(FUS)destructs the tumor and simultaneously generates tumor debris and tumor-associated antigens,which enhances the immunogenicity of the tumor and stimulates the immune cells,inducing the body's immune response.Microbubbles are clinically used as a contrast.As a matter of fact,the addition of microbubbles can reinforce the destructive effect of FUS on the tumor and activate a stronger immune response.The combined application of ultrasound and microbubbles can more effectively open the blood brain barrier(BBB),which is beneficial to improving the intake of immune cells or immunotherapy drugs and exerting a positive influence in the lesion area.Currently,microbubbles and nanoparticles are commonly used as gene and drug carriers.Using ultrasound,the immune-related gene or antigen delivery itself can enhance the immune response and improve the efficacy of the immunotherapy.Guilian Shi Mingchuan Zhong Fuli Ye Xiaoming Zhang 2019Cancer Biology & Medicine2019,16,4:3

5Application of biological optimization of hypofractionated radiotherapy post conservative surgery for breast cancer显示文摘Objective The aim of the study was to discuss the application of biological optimization and its difference from physical optimization in hypofractionated radiotherapy for breast cancer after conservative surgery.Methods This retrospective study enrolled 15 randomly chosen patients with left-sided breast cancer who received radiotherapy.The volumetric arc therapy(VMAT)technique was used to redesign treatment plans with physical functions(PF)group,biological-physical functions combined(BF+PF and PF+BF)groups,and biological functions(BF)group.The dosimetric differences based on the above four optimization methods were assessed by calculating and analyzing the corresponding dose-volume parameters.Results The target parameters of the four groups differed significantly(P<0.05)except for the conformity index(CI).The tumor control probability(TCP)values in the BF and BF+PF groups were higher than those in the PF and PF+BF groups.Moreover,the dose-volume parameters of the ipsilateral lung in the BF group were less than those of three other groups,while the monitor unit(MU)in the BF group was approximately 16%lower than those of the PF and PF+BF groups.Conclusion Biological functions were useful to increase the equivalent uniform dose(EUD)and TCP values of the target,decrease the dose-volume parameters of the organs-at-risk(OARs),and improve treatment efficiency.Ying Shao Yadi Wang Fuli Zhang Shi Wang 2020Oncology and Translational Medicine2020,6,3:1
6AMP-activated protein kinaseβ1 orβ2 deletion enhances colon cancer cell growth and tumorigenesis显示文摘Abnormal metabolism is a major hallmark of cancer and has been validated as a therapeutic target.Adenine monophosphate-activated protein kinase(AMPK),anαβγheterotrimer,performs essential functions in cancer progression due to its central role in maintaining the homeostasis of cellular energy.While the contributions of AMPKαand AMPKγsubunits to cancer development have been established,specific roles of AMPKβ1 and AMPKβ2 isoforms in cancer development are poorly understood.Here,we show the functions of AMPKβ1 and AMPKβ2 in colon cancer.Specifically,deletion of AMPKβ1 or AMPKβ2 leads to increased cell proliferation,colony formation,migration,and tumorigenesis in HCT116 and HT29 colon cancer cells.Interestingly,the AMPKβ1 and AMPKβ2 isoforms have slightly different effects on regulating cancer metabolism,as colon cancer cells with AMPKβ1 knockout showed decreased rates of glycolysis-related oxygen consumption,while AMPKβ2 deletion led to enhanced rates of oxygen consumption due to oxidative phosphorylation.These results demonstrate that functional AMPKβ1 and AMPKβ2 inhibit growth and tumorigenesis in colon cancer cells,suggesting their potential as effective targets for colon cancer therapy.Fuli Shi Zhimin Tang Shanshan Jiang Zhijuan Xiong Wansi Zhang Yuanjun Li Hui Lin Zhijun Luo Ying Ying 2022Acta Biochimica et Biophysica Sinica2022,54,8:0
7Derlin-1 and TER94/VCP/p97 are required for intestinal homeostasis显示文摘Adult stem cells are critical for the maintenance of residential tissue homeostasis and functions. However,the roles of cellular protein homeostasis maintenance in stem cell proliferation and tissue homeostasis are not fully understood. Here, we find that Derlin-1 and TER94/VCP/p97, components of the endoplasmic reticulum(ER)-associated degradation(ERAD) pathway, restrain intestinal stem cell proliferation to maintain intestinal homeostasis in adult Drosophila. Depleting any of them results in increased stem cell proliferation and midgut homeostasis disruption. Derlin-1 is specifically localized in the ER of progenitors, and its Cterminus is required for its function. Interestingly, we find that increased stem cell proliferation is resulted from elevated ROS levels and activated JNK signaling in Derlin-1-or TER94-deficient progenitors. Further removal of reactive oxygen species(ROS) or inhibition of JNK signaling almost completely suppresses increased stem cell proliferation. Together, these data demonstrate that the ERAD pathway is critical for stem cell proliferation and tissue homeostasis. 8Power and efficiency optimizations of Maisotsenko-Atkinson,Dual and Miller cycles and performance comparisons with corresponding traditional cycles显示文摘Maisotsenko cycle(M-cycle)has been combined with some cooling and power cycles,and behaves important thermodynamic advantage.Finite-time thermodynamics(FTT)is applied to establish three endoreversible models of M-Atkinson,M-Dual and M-Miller cycles.They are performed based on models of endoreversible Atkinson,Dual and Miller cycles by combing FTT model with M-cycle concept.Power output(POW)and thermal efficiency(TEF)of those M-cycles are studied and optimized by numerical calculations.The maximum power output(MPO)and the corresponding pressure ratio and TEF,the maximum TEF and the corresponding pressure ratio and POW,as well as optimal ranges of pressure ratio are obtained.Effects of mass flow rate of circulating water injection,initial cycle temperature and maximum cycle temperature on cycle POW,TEF and optimal pressure ratio range are analyzed.The optimal performances of the three M-cycles are compared with those of traditional Atkinson,Dual and Miller cycles under the same conditions.The results show that for the three M-cycles,end temperature of adiabatic expansion process of M-cycle is less than that of the corresponding traditional cycle,POW and TEF at arbitrary pressure ratio of M-cycle are much higher than those of the corresponding traditional cycle,and performance characteristics of M-cycles are superior to those of the corresponding traditional cycles.CHEN LinGen ZHU FuLi SHI ShuangShuang GE YanLin FENG HuiJun 2023Science China(Technological Sciences)2023,66,12:0
9Estimation of the effect of target and normal tissue sparing based on equivalent uniform dose-based optimization in hypofractionated radiotherapy for lung cancer显示文摘Objective This study aims to investigate the dosimetric differences among four planning methods of physical and biological optimization in hypofractionated radiation therapy for non-small cell lung cancer(NSCLC).Methods Ten NSCLC patients receiving radiation therapy were chosen for this retrospective study.Volumetric modulated arc treatment plans for each patient were remade with dose-volume(DV)functions,biological-physical functions,and biological functions,using the same constraint parameters during optimization.The dosimetric differences between the four types of plans were calculated and analyzed.Results For the target,equivalent uniform dose(EUD)of the EUD and EUD+DV groups was approximately 2.8%–3.6%and 3.2%–3.7%higher than those of the DV and DV+EUD groups,respectively.The average tumor control probability(TCP)of the EUD and EUD+DV groups was also significantly higher than those of the other two groups(P<0.05).The difference in heterogeneity index(HI)among the four groups was also statistically significant(P<0.05),while the difference of conformity index(CI)was not significant(P>0.05).For the organs at risk,the differences of EUD,V5,V10,V20,V30 of normal lung tissues were not statistically significant(P>0.05);however,the mean lung dose of the EUD and EUD+DV groups was slightly lower than those of the other two groups.Conclusion The biological optimization method has obvious advantages of improving EUD and TCP of the target,while decreasing the exposed dose of normal lung.This result is meaningful in choosing plan optimization methods in routine work.Ying Shao Fuli Zhang Shi Wang Weidong Xu Jing Jiang 2019Oncology and Translational Medicine2019,5,5:0
