1Biocompatibility studies of silk fibroin-based artificial nerve grafts in vitro and in vivo显示文摘Silk fibroin(SF)has been used extensively in the biomedical field including tissue engineering for the generation of artificial bones,skins or ligaments.We have previously reported on good in vitro biocompatibility of SF fibers with peripheral nerve tissues and cells.In the present study,we developed a novel design of the SF-based artificial nerve graft(SF graft)which was composed of a SF-nerve guidance conduit(NGC)inserted with SF fibers.MTT assay was performed to determine the cytotoxicity of the SF-NGC extract fluid on the cultured L929 cells derived from an immortalized mouse fibroblast cell line.In addition,this SF graft was implanted into adult rats for bridging a 10-mm long sciatic nerve defect.The following-up experiments at initial stage(1-4 week)of nerve regeneration including routine blood tests and histochemical investigation were conducted to evaluate the in vivo biocompatibility of the SF graft with peripheral nerves.The results demonstrated that the SF-NGC graft was biocompatible with the surrounding tissues and cells due to its low inflammatory potential with a grade 0 under the U.S.Pharmacopeia guidelines and it was generally suitable to a certain degree for bridging peripheral nerve defects in virtue of supporting Schwann cell adherence,expansion and migration.Therefore the SF graft is a promising alternative to classical autografts for peripheral nerve repair.Chen Xuemei Yang Yumin Wu Jian Zhao Yahong Ding Fei Gu Xiaosong 2007Progress in Natural Science:Materials International2007,17,9:5
2Nanoparticle mediated controlled delivery of dual growth factors显示文摘Peripheral nerve functional recovery after nerve injury generally requires multiple growth factors by synergistic effect.However,the optical combination of multiple synergistic growth factors for axonal regeneration has been scarcely considered up to now.Meanwhile,the use of growth factors in promoting nerve regeneration was limited by its short biological half-life in vivo,its vulnerability to structure disruption or hydrolyzation,leading to loss of bioactivity.Herein,a novel polymeric nanoparticle delivery system composed of heparin andε-poly-L-lysine(PL)was prepared for control release of nerve growth factor(NGF)and basic fibroblast growth factor(bFGF).The nanoparticles were synthesized by polyelectrolyte complexation in aqueous solution at room temperature,followed by cross-linking with biological genipin.The obtained nanoparticles had a spherical shape,with a mean diameter of about 246 nm,and high growth factors encapsulation efficiency as well as good stability.NGF and bFGF were encapsulated in the nanoparticles and showed a continuous and slow release behavior in vitro.The bioactivities of the released growth factors were evaluated,and exhibited the synergistic effect.The controlled release of the dual synergistic growth factors would improve the treatment of peripheral nerve injury to mimic the natural cellular microenvironments.ZHANG LuZhong ZHOU YouLang LI GuiCai ZHAO YaHong GU XiaoSong YANG YuMin 2014Science China(Life Sciences)2014,57,2:3
3Ubiquitin-specific protease 24 promotes EV71 infection by restricting K63-linked polyubiquitination of TBK1显示文摘TANK-binding kinase 1(TBK1)is an essential protein kinase for activation of interferon regulatory factor 3(IRF3)and induction of the type I interferons(IFN-I).Although the biochemical regulation of TBK1 activation has been studied,little is known about how enterovirus 71(EV71)employs the deubiquitinases(DUBs)to regulate TBK1 activation for viral immune evasion.Here,we found that EV71 infection upregulated the expression of ubiquitinspecific protease 24(USP24).Further studies revealed that USP24 physically interacted with TBK1,and can reduce K63-linked polyubiquitination of TBK1.Knockdown of USP24 upregulated TBK1 K63-linked polyubiquitination,promoted the phosphorylation and nuclear translocation of IRF3,and in turn improved IFN-I production during EV71 infection.As a consequence,USP24 knockdown dramatically inhibited EV71 infection.This study revealed USP24 as a novel regulator of TBK1 activation,which promotes the understanding of immune evasion mechanisms of EV71 and could provide a potential strategy for treatment of EV71 infection.Lichao Zang Jin Gu Xinyu Yang Yukang Yuan Hui Guo Wei Zhou Jinhong Ma Yan Chen Yumin Wu Hui Zheng Weifeng Shi 2023Virologica Sinica2023,38,1:2

5Graphene-like two-dimensional nanosheets-based anticorrosive coatings: A review显示文摘In recent years, graphene has been widely employed in the field of metal corrosion protection owing to its outstanding impermeability and chemical stability, with examples of such metal protection including pure graphene coatings and graphene-based composite coatings. But the conductive graphene could promote the electrochemical reaction at the interface and accelerate the corrosion of metal substrates. More emerging graphene-like 2D nanosheets are attracting research attention for the application of metal anticorrosion, because of their barrier properties and poor conductivity, mainly including boron nitride(BN),molybdenum disulfide(MoS_(2)), zirconium phosphate(ZrP), and titanium carbide(MXene). In this review,the application of these graphene-like 2D nanosheets to metal protection is comprehensively reviewed.First, the general preparation methods of 2D nanosheets are briefly introduced. Second, surface functionalization of 2D nanosheets, including covalent and non-covalent modification, is described in detail.Third, the anticorrosion performance and optimization measures of pure 2D nanosheets coatings are summarized. Next, the protection performance, anticorrosive mechanism, and optimizations of 2D nanosheets composite coatings are presented. 17Developmental Temporal Patterns and Molecular Network Features in the Transcriptome of Rat Spinal Cord显示文摘The molecular network features of spinal cord development that are integral to tissue engineering remain poorly understood in placental mammals,especially in terms of their relationships with vital biological processes such as regeneration.Here,using a large-scale temporal transcriptomic analysis of rat spinal cord from the embryonic stage to adulthood,we show that fluctuating RNA expression levels reflect highly active transcriptional regulation,which may initiate spinal cord patterning.We also demonstrate that microRNAs(miRNAs)and transcriptional factors exhibit a mosaic profile based on their expression patterns,while differential alternative splicing events reveal that alternative splicing may be a driving force for the development of the node of Ranvier.Our study also supports the existence of a negative correlation between innate immunity and intrinsic growth capacity.Epigenetic modifications appear to perform their respective regulatory functions at different stages of development,while guanine nucleotidebinding protein(G protein)-coupled receptors(including olfactory receptors(ORs))may perform pleiotropic roles in axonal growth.This study provides a valuable resource for investigating spinal cord development and complements the increasing number of single-cell datasets.These findings also provide a genetic basis for the development of novel tissue engineering strategies.Jian Yang Lili Zhao Sheng Yi Fei Ding Yumin Yang Yan Liu Yongjun Wang Mei Liu Chengbin Xue Lian Xu Leilei Gong Xinghui Wang Yu Zhang Bin Yu Guo-li Ming Xiaosong Gu 2021Engineering2021,7,11:0
18Bionic peptide scaffold in situ polarization and recruitment of M2 macrophages to promote peripheral nerve regeneration显示文摘Tissue regeneration requires exogenous and endogenous signals,and there is increasing evidence that the exogenous microenvironment may play an even more dominant role in the complex process of coordinated multiple cells.The short-distance peripheral nerve showed a spontaneous regenerative phenomenon,which was initiated by the guiding role of macrophages.However,it cannot sufficiently restore long-distance nerve injury by itself.Based on this principle,we firstly constructed a proinflammatory model to prove that abnormal M2 expression reduce the guidance and repair effect of long-distance nerves.Furthermore,a bionic peptide hydrogel scaffold based on self-assembly was developed to envelop M2-derived regenerative cytokines and extracellular vesicles(EVs).The cytokines and EVs were quantified to mimic the guidance and regenerative microenvironment in a direct and mild manner.The bionic scaffold promoted M2 transformation in situ and led to proliferation and migration of Schwann cells,neuron growth and motor function recovery.Meanwhile,the peptide scaffold combined with CX3CL1 recruited more blood-derived M2 macrophages to promote long-distance nerve reconstruction.Overall,we systematically confirmed the important role of M2 in regulating and restoring the injury peripheral nerve.This bionic peptide hydrogel scaffold mimicked and remodeled the local environment for M2 transformation and recruitment,favoring long-distance peripheral nerve regeneration.It can help to explicate regulative effect of M2 may be a cause not just a consequence in nerve repair and tissue integration,which facilitating the development of pro-regenerative biomaterials.Pengxiang Yang Yong Peng Xiu Dai Jing Jie Deling Kong Xiaosong Gu Yumin Yang 2023Bioactive Materials2023,,12:0

20Effects of Different Sowing and Transplanting Time on Quality and Economic Benefit of Tobacco Variety Yunyan 105显示文摘In order to provide references for popularization and application of tobacco variety Yunyan 105,a field experiment was conducted to explore the effects of different sowing and transplanting time on agronomic traits,economic benefit and quality of Yunyan 105. Results showed that the agronomic characters,quality traits and economic benefit of the tobacco were relatively fine with sowing time February 4-14 and transplanting time April 1-10. Index scores of the physical properties of tobacco leaves were greater than 80 scores with sowing time February 14 and transplanting time April 10. The comprehensive index of conventional chemical compositions was 0. 39-0. 73. The harmony of conventional chemical compositions of lower leaves was the best with the comprehensive index of 0. 59 when the sowing time and transplanting time were February 4 and April 1 respectively. As for the middle and upper leaves the comprehensive indexes were respectively 0. 71 and 0. 73 with the sowing time February 14 and transplanting time April 10. The sensory evaluation of smoking quality and economic benefit were February 14 sowing and April 10 transplanting better,its taste of the tobacco leaves was pure and mild,the irritancy and the strength was less,the aftertaste was better,and the output value reached the highest 4 271. 17 yuan/667 m^2. The various sowing and transplanting time had no significant influence on appearance quality of tobacco leaves.
