2MiR-214-3p promotes proliferation and inhibits estradiol synthesis in porcine granulosa cells显示文摘Background: Granulosa cells(GCs) proliferation and estradiol synthesis significantly affect follicular development.The miR-214-3p expression in the ovarian tissues of high-yielding sows is higher than that in low-yielding sows,indicating that miR-214-3p may be involved in sow fertility. However, the functions and mechanisms of miR-214-3p on GCs are unclear. This study focuses on miR-214-3p in terms of the effects on GCs proliferation and estradiol synthesis.Results: Our findings revealed that miR-214-3p promotes proliferation and inhibits estradiol synthesis in porcine GCs. MiR-214-3p can increase the percentage of S-phase cells, the number of EdU labeled positive cells, and cell viability. However, E2 concentration was reduced after miR-214-3p agomir treatment. We also found that miR-214-3p up-regulates the expression of cell cycle genes including cell cycle protein B(Cyclin B), cell cycle protein D(Cyclin D), cell cycle protein E(Cyclin E), and cyclin-dependent kinase 4(CDK4) at the transcription and translation levels, but down-regulates the mRNA and protein levels of cytochrome P450 family 11 subfamily A member 1(CYP11A1), cytochrome P450 family 19 subfamily A member 1(CYP19A1), and steroidogenic acute regulatory protein(StAR)(i.e., the key enzymes in estradiol synthesis). 3N-Doped Carbon Nanofibers Encapsulating CoO@Co_(9)S_(8) Nanoparticles:Preparation from S-Rich Co_(32) Coordination Cluster Precursors by Electrospinning and Application for Superior Li-ion Storage显示文摘Thiacalixarene-supported Co_(32) nanoclusters encapsulated in polyacrylonitrile nanofibers(Co_(32)@PAN-NFs)by electrospinning have been utilized as precursors to fabricate N-doped CoO@Co_(9)S_(8) carbon nanofibers(CoO@Co_(9)S_(8)@CNFs)for superior Li-ion storage.The S-rich Co_(32) clusters capped by organic sheets afforded the well dispersed cobalt oxide/sulfide nanoparticles embedded in carbon nanofiber composites by direct calcination.The N-doped CoO@Co_(9)S_(8)@CNFs nanocomposites have been utilized as anode materials for lithium ion battery with the reversible capabilities being of 1051.8,967.6,894.7,782.7,669.5 and 525.4 mA·h/g at 0.1,0.2,0.5,1,2 and 3 A/g,respectively.The CoO@Co_(9)S_(8)@CNFs also showed a relatively high stable capacity of 551.7 mA·h/g at the current density of 1 A/g after 200 cycles of rate experiments.The as-obtained N-doped CoO@Co_(9)S_(8)@CNFs nanocomposites exhibited superior reversible capacity,rate performance,Coulomb efficiency(74.5%vs.63.9%)and cyclic stability comparing with the CoO@Co_(9)S_(8)@C derived from simple annealing of Co_(32) templates.YANG Siran AI Feixue LI Ziping ZHAO Guiyan BI Yanfeng 2022Chemical Research in Chinese Universities2022,38,2:1

5消化道神经内分泌肿瘤临床病理特征及预后因素分析(英文)显示文摘Objective: The aim of the study was to analyze the clinicopathologic characteristics of gastroenteropancreatic neuroendocrine neoplasm(GEP-NEN) and to explore the prognostic factors for patients and differences of immunohistochemical markers between neuroendocrine tumor(NET) and neuroendocrine carcinoma(NEC). Methods: Retrospective reviews were conducted for the charts of 119 patients with GEP-NEN at the Affiliated Hospital of Qingdao University(China) from August 2003 to December 2013. Kaplan-Meier method was used to do the overall survivals analysis for the patients at different levels of predictive factors. Meanwhile, Cox proportional hazard model was used to select independent risk factors of survival. Analysis of variance was used to compare the expression of immunohistochemical markers among different pathological grades. Results: Among 119 patients, pancreas(45/119, 37.82%) and rectum(33/119, 27.73%) were mostly involved. The onset age of GEP-NEN in female group was younger than that of the male group. There were 13 deaths(10.92%) during 18.9(0.1–133.4) months follow-up period. Multivariate analysis indicated that neural invasion, gender and pathological grades of NET and NEC were independent risk factors. In neuroendocrine neoplasm(NEN), Syn expression in G2 was higher than G1 and G3, while Cg A showed no significant difference. All markers showed no significant differences between NET and NEC. Conclusion: GEP-NEN may occur at multiple sites of digestive system and lack specific clinical manifestations. Syn expression detected for the prognosis of G1, G2 and G3 tumors have clinical significance. Neural invasion, sex and pathological grades were independent prognostic factors for GEP-NEN patients. 7Oleic acid reduces steroidogenesis by changing the lipid type stored in lipid droplets of ovarian granulosa cells显示文摘Background:Oleic acid is an abundant free fatty acid present in livestock that are in a negative energy-balance state,and it may have detrimental effects on female reproduction and fertility.Oleic acid induces lipid accumulation in bovine granulosa cells,which leads to a foam cell-like morphology and reduced steroidogenesis.However,why oleic acid increases lipid accumulation but decreases steroidogenesis remains unclear.This study focused on oleic acid’s effects on lipid type and steroidogenesis.Results:Oleic acid increased the lipid accumulation in a concentration-dependent manner and mainly increased the triglyceride level and decreased the cholesterol ester level.Oleic acid also led to a decline in estradiol and progesterone production in porcine granulosa cells in vitro.In addition,oleic acid up-regulated the expression of CD36 and diacylglycerol acyltransferase 2,but down-regulated the expression of 3-hydroxy-3-methylglutarylcoenzyme A reductase,scavenger receptor class B member 1 and acetyl-Coenzyme A acetyltransferase 2,as well as steroidogenesis-related genes,including cytochrome P450 family 11 subfamily A member 1,cytochrome P450family 19 subfamily A member 1 and 3 as well as steroidogenic acute regulatory protein at the mRNA and protein levels.An oleic acid-rich diet also enhanced the triglyceride levels and reduced the cholesterol levels in ovarian tissues of female mice,which resulted in lower estradiol levels than in control-fed mice.Compared with the control,decreases in estrus days and the numbers of antral follicles and corpora lutea,as well as an increase in the numbers of the atretic follicles,were found in the oleic acid-fed female mice.Conclusions:Oleic acid changed the lipid type stored in lipid droplets of ovarian granulosa cells,and led to a decrease in steroidogenesis.These results improve our understanding of fertility decline in livestock that are in a negative energy-balance state.Xiaoge Zhou Zhaoyi Mo Yankun Li Liang Huang Sihai Yu Lan Ge Yamei Hu Shengjie Shi Lutong Zhang Liguang Wang Lei Gao Gongshe Yang Guiyan Chu 2022Journal of Animal Science and Biotechnology2022,13,3:1
8The novel peptide Nb PPI1 identified from Nicotiana benthamiana triggers immune responses and enhances resistance against Phytophthora pathogens显示文摘In plants, recognition of small secreted peptides, such as damage/danger-associated molecular patterns(DAMPs), regulates diverse processes,includingstressandimmune responses. Here, we identified an SGPS(Ser-Gly-Pro-Ser) motif-containing peptide, Nicotiana tabacum NtPROPPI, and its two homologs in Nicotiana benthamiana, NbPROPPI1 and NbPROPPI2. Phytophthora parasitica infection and salicylic acid(SA) treatment induced NbPROPPI1/2 expression. Moreover, SignalP predicted that the 89-amino acid NtPROPPI includes a 24-amino acid N-terminal signal peptide and NbPROPPI1/2-GFP fusion proteins were mainly localized to the periplasm. Transient expression of NbPROPPI1/2 inhibited P.parasiticacolonization,andNbPROPPI1/2 knockdown rendered plants more susceptible to P. parasitica. An eight-amino-acid segment in the NbPROPPI1 C-terminus was essential for its immune function and a synthetic 20-residue peptide, NbPPI1, derived from the C-terminus of NbPROPPI1 provoked significant immune responses in N. benthamiana. These responses led to enhanced accumulation of reactive oxygen species, activation of mitogen-activated protein kinases, and up-regulation of the defense genes Flg22-induced receptor-like kinase(FRK) and WRKY DNA-binding protein 33(WRKY33). The NbPPI1-induced defense responses require Brassinosteroid insensitive1-associated receptor kinase 1(BAK1). 9The chloroplast genome comparative characteristic of artificial breeding tree,a case about Broussonetia kazinoki×Broussonetia papyrifera显示文摘Broussonetia kazinoki×Broussonetia papyrifera(ZJGS)is a hybrid species in Moraceae family,which has a very complicated hybrid origin.The excellent characteristics of fast growth,strong soil and water conservation ability,high leaf protein content and stem fiber content in ZJGS make it both ecological benefits in the mining area and economically valuable.This study aims to further understand ZJGS and other Moraceae taxa through the ZJGS chloroplast(cp)genome structure and the comparison with 12 closely related Moraceae species.Among the 13 Moraceae species,the cp genome length of seven Broussonetia species(ranges from 160,239 bp to 162,594 bp)is larger than that of six Morus species(ranges from 158,459 bp to 159,265 bp).Among the 77 shared protein-coding genes(PCGs)in Moraceae species,the obvious positive selection of Ka/Ks ratios acted on petD and rpl16 genes of B.kazinoki and B.papyrifera,respectively.Phylogenetic analysis based on shared PCGs from 28 species shows that ZJGS is closely related to maternal B.kazinoki.These findings provide data support for the origin of ZJGS hybridization and provide genomic resources for future ZJGS resource development and molecular breeding.WAN ZHANG GUIYAN YANG YUNLIN ZHAO ZHENGGANG XU HUANG HUIMIN JIAKANG ZHOU 2022BIOCELL2022,46,3:0
10Insight into the characteristics of an important evolutionary model bird(Geospiza magnirostris)mitochondrial genome through comparison显示文摘Darwin’s finches are the most classic case of evolution.Early studies on the evolution of this species were mainly based on morphology.Until now,the mitochondrial genome of Geospiza magnirostris has been sequenced and the study explored the characteristics of the complete genome of G.magnirostris and verified the evolutionary position of it.The 13 PCGs initiated by ATN codons.The stop codons of three PCGs(ND2,COX3 and ND4)were incomplete,with only T-or TA-replacing complete form TAA or TAG.All the tRNA genes expressed a typical cloverleaf secondary structure,except for tRNA^(Ser1)(AGY),whose dihydrouridine(DHU)arm was lack and instead with a simple loop.In the sequence of the control region of G.magnirostris,we found six simple repeat tandem sequences with a total length of 42 bp.Two characteristic conserved overlapping junction(ATGCTAA)and(CAAGAAAG)were observed as reported for eight selected Passeriformes birds.A special conserved overlapping junction(ATCTTACC)involved in mitochondrial transcription termination was found between tRNA^(Tyr) and COX1 in G.magnirostris’s control region.Four most frequently used amino acids in G.magnirostris’s PCGs were Leu1(CUN),Ile,Thr,Ala.The codon usage of G.magnirostris was relatively average,and there was no particular bias.The ratio Ka/Ks results showed that G.magnirostris receives less natural selection pressure.The phylogenetic relationships and cluster analysis of relative codon usage showed that G.magnirostris and Thraupis episcopus clustered in one branch.The phylogenetic position of G.magnirostris was consistent with the traditional taxonomic of Thraupis.The results supported the conclusion that G.magnirostris belongs to the morphological classification of the family Thraupidae.ZHENGGANG XU LIANG WU JIAHAO CHEN YUNLIN ZHAO CHONGXUAN HAN TIAN HUANG GUIYAN YANG 2022BIOCELL2022,46,7:0
11Satellite observed shape-based overall rotation—A new aspect in eddy kinematics显示文摘With the development of satellite altimetry technology,the resolution of sea-level anomaly(SLA)datasets is constantly improving.Current spatial resolution levels can reach a grid size of(1/4)°×(1/4)°,with daily measurements that span from 1993 to 2018,allowing for the precise identification and tracking of individual eddies.In the current study,in addition to the internal circulation and migration of eddies,a new aspect in eddy kinematics is revealed and investigated for the first time:shape-based overall eddy rotation(SOER),based on the intrinsic elliptical shape of eddies identified from a high-resolution SLA dataset.We found that eddies can maintain an elliptical shape and a slow and stable SOER during their migration process.The SOER speed was observed to be negatively correlated to eddy lifetime,and exhibited a dependence on latitude,decreasing from low-and high-to mid-latitudes.The SOER direction tended to be consistent with the direction of internal circulation,particularly for long-lived eddies.In addition,we identified a negative relationship between internal circulation speed and SOER speed while the migration speed was positively related to SOER speed.These findings further expand and improve eddy kinematics,which is of great significance for the future study of eddy dynamics.Xueqing Yang Guiyan Han Chunyong Ma Chuanchuan Cao Jie Yang Ge Chen 2022Acta Oceanologica Sinica2022,41,5:0
12CLOCK inhibits the proliferation of porcine ovarian granulosa cells by targeting ASB9显示文摘Background Clock circadian regulator(CLOCK)is a core factor of the mammalian biological clock system in regulat-ing female fertility and ovarian physiology.However,CLOCK’s specific function and molecular mechanism in porcine granulosa cells(GCs)remain unclear.In this study,we focused on CLOCK’s effects on GC proliferation.Results CLOCK significantly inhibited cell proliferation in porcine GCs.CLOCK decreased the expression of cell cycle-related genes,including CCNB1,CCNE1,and CDK4 at the mRNA and protein levels.CDKN1A levels were upregulated by CLOCK.ASB9 is a newly-identified target of CLOCK that inhibits GC proliferation;CLOCK binds to the E-box element in the ASB9 promoter.Conclusions These findings suggest that CLOCK inhibits the proliferation of porcine ovarian GCs by increasing ASB9 level.Liang Huang Huan Yuan Shengjie Shi Xiangrong Song Lutong Zhang Xiaoge Zhou Lei Gao Weijun Pang Gongshe Yang Guiyan Chu 2023Journal of Animal Science and Biotechnology2023,14,5:0
