| 2 | Overexpression of p27^(KIP1)induced cell cycle arrest in G_1 phase and subsequent apoptosis in HCC-9204 cell line显示文摘AIM We have previously reported that inducibleover-expression of Bak may prolong cell cycle inG1 phase and lead to apoptosis in HCC-9204 cells.This study is to investigate whether p27KIP1playsan important role in this process.METHODS In order to elucidate the exactfunction of p27KIP1in this process,a zinc induciblep27KIP1stable transfectant and transient p27KIP1-GFP fusion transfectant were constructed.Theeffects of inducible,p27KIP1on cell growth,cellcycle arrest and apoptosis were examined in themock,control pMD vector,and pMD-KIP1transfected HCC-9204 cells.RESULTS This p27KIP1-GFP transfectant maytransiently express the fusion gene.The cellgrowth was reduced by 35% at 48 h of p27KIP1induction with zinc treatment as determined bytrypan blue exclusion assay.These differencesremained the same after 72 h of p27KIP1expression,p27KIP1caused cell cycle arrest after24 h of induction,with 40% increase in G1population.Prolonged p27KIP1expression in thiscell line induced apoptotic cell death reflected byTUNEL assay.Fourty-eight h and 72 h of p27KIP1expression showed a characteristic DNA ladder onagarose gel electrophoresis. | 5 | Diagnostic value of FIB-4, aspartate aminotransferaseto-platelet ratio index and liver stiffness measurement in hepatitis B virus-infected patients with persistently normal alanine aminotransferase显示文摘AIM To assess the diagnostic value of FIB-4, aspartate aminotransferase-to-platelet ratio index(APRI), and liver stiffness measurement(LSM) in patients with hepatitis B virus infection who have persistently normal alanine transaminase(PNALT).METHODS We enrolled 245 patients with chronic hepatitis B: 95 in PNALT group, 86 in intermittently elevated alanine transaminase(PIALT1) group [alanine transaminase(ALT) within 1-2 × upper limit of normal value(ULN)], and 64 in PIALT2 group(ALT > 2 × ULN). All the patients received a percutaneous liver biopsy guided by ultrasonography. LSM, biochemical tests, and complete blood cell counts were performed.RESULTS The pathological examination revealed moderate inflammatory necrosis ratios of 16.81%(16/95), 32.56%(28/86), and 45.31%(28/64), and moderate liverfibrosis of 24.2%(23/95), 33.72%(29/86), and 43.75%(28/64) in the PNALT, PIALT1, and PIALT2 groups, respectively. The degrees of inflammation and liver fibrosis were significantly higher in the PIALT groups than in the PNALT group(P < 0.05). No significant difference was found in the areas under the curve(AUCs) between APRI and FIB-4 in the PNALT group; however, significant differences were found between APRI and LSM, and between FIB-4 and LSM in the PNALT group(P < 0.05 for both). In the PIALT1 and PIALT2 groups, no significant difference(P > 0.05) was found in AUCs for all comparisons(P > 0.05 for all). In the overall patients, a significant difference in the AUCs was found only between LSM and APRI(P < 0.05).CONCLUSION APRI and FIB-4 are not the ideal noninvasive hepatic fibrosis markers for PNALT patients. | 6 | Treatment and outcomes of 1041 pediatric patients with neuroblastoma who received multidisciplinary care in China显示文摘Importance:Neuroblastoma is the most common extracranial malignant solid tumor in children.Multidisciplinary care is critical to improving the survival of pediatric patients with neuroblastoma.Objective:To systematically summarize the clinical characteristics of children with neuroblastoma and evaluate their prognosis with multidisciplinary care provided in a single center.Methods:This retrospective study analyzed the clinical data of 1041 patients with neuroblastoma who were diagnosed,treated,and followed-up in the Hematology-Oncology Center of Beijing Children’s Hospital from 2007 to 2019.Results:The median age at diagnosis was 34 months;80.8%of the patients were younger than 5 years of age.Notably,243 patients(23.3%)were classified as low-risk,249 patients(23.9%)were classified as intermediate-risk,and 549(52.7%)were classified as high-risk.Furthermore,956 patients underwent surgical resections;986(94.7%)patients received chemotherapy;and 176 patients with high-risk neuroblastoma received hematopoietic stem cell transplantation.The 5-year event-free survival(EFS)rate was 91.3%and 5-year overall survival(OS)rate was 97.5%in low-risk group;in the intermediate-risk group,these rates were 85.1%and 96.7%,respectively,while they were 37.7%and 48.9%in the high-risk group(P<0.001 for both).The 5-year EFS and OS rates were significantly higher in patients diagnosed between 2015 and 2019 than in patients diagnosed between 2007 and 2014(P<0.001).In total,278 patients(26.7%)exhibited tumor relapse or progression;the median interval until relapse or progression was 14 months.Of the 233 patients who died,83%died of relapse or progression of neuroblastoma and 4.3%died of therapy-related complications.Interpretation:The 5-year OS rate was low in high-risk patients,compared with low-and intermediate-risk patients.Multidisciplinary care is critical for improvement of survival in pediatric patients with neuroblastoma.Additional treatment strategies should be sought to improve the prognosis of patients with high-risk neuroblastoma. | Yan Su Hong Qin Chenghao Chen Shengcai Wang Shihan Zhang Dawei Zhang Mei Jin Yun Peng Lejian He Xiaoman Wang Suyun Qian Maoquan Qin Ming Ge Fuquan Zhang Qi Zeng Huanmin Wang Xiaoli Ma Xin Ni | 2020 | Pediatric Investigation2020,4,3: | 16 |

| 8 | The effects of microRNA-34a regulating Notch-1/NF-κB signaling pathway on lipopolysaccharide-induced human umbilical vein endothelial cells显示文摘BACKGROUND: Notch-1/NF-κB signaling plays a key role in the cecal ligation and puncture(CLP)-induced sepsis. This study aims to investigate the intervention effects of microRNA-34a(miR-34a) lentivirus regulating Notch-1/NF-κB signaling pathway on lipopolysaccharide(LPS)-induced human umbilical vein endothelial cells(HUVEC).METHODS: HUVEC were divided into four groups as the following: they were infected with negative control lentivirus(NC group) or miR-34a lentivirus(OE group); LPS(1 g/mL) was added on the third day on the basis of NC group and OE group for 24 hours(NC+LPS group or OE+LPS group). The levels of TNF-α, IL-1β, IL-6, and IL-10 in the cell supernatants, and the mRNA and protein expression of Notch-1 and NF-κB in the HUVEC were evaluated.RESULTS: After 24 hours, the levels of TNF-α, IL-1β, IL-6 in the cell supernatants and the protein expression of NF-κB from NC+LPS group were significantly higher than those of NC group, but IL-10 level and the protein expression of Notch-1 in NC+LPS group were the opposite. After intervention of miR-34a lentivirus, the cell supernatants TNF-α and the protein expression of NF-κB in OE+LPS group after 24 hours markedly decreased compared to NC+LPS group. A new method of electrostatic precipitation of the corona discharges with spraying water(CDSW) was introduced. A new electrode configuration and the circulation spraying of water were employed in the method. The efficient electrostatic precipitation for adhesive and fine dusts can be accomplished without any drain water during a long operating period. The fundamental structure, discharge characteristics, mechanism of spraying and precipitation principle of the electrostatic precipitation using CDSW were described and analyzed. The V I characteristics, spraying state, supplying water quantity, influence of temperature and clean of the electrodes were researched in series experiments. The treating effects of circulating spraying using the corona plasma at the same time of electrostatic precipitation were investigated. | 11 | Primary site and regional lymph nodeinvolvement are independent prognosticfactors for early?stage extranodal nasal?typenatural killer/T cell lymphoma显示文摘Background:Nasal-type extranodal natural killer/T-cell lymphoma(ENKTCL) originates primarily in the nasal cavity or extra-nasal sites within the upper aerodigestive tract.However,it is unclear whether the primary site can serve as an independent prognostic factor or whether the varying clinical outcomes observed with different primary sites can be attributed merely to their propensities of regional lymph node involvement.The aim of this study was to investigate the prognostic implications of the primary site and regional lymph node involvement in patients with early-stage nasal-type ENKTCL.Methods:To develop a nomogram,we reviewed the clinical data of 215 consecutively diagnosed patients with early-stage nasal-type ENKTCL who were treated in Sun Yat-sen University Cancer Center with chemotherapy and radiotherapy between 2000 and 2011.The predictive accuracy and discriminative ability of the nomogram were determined using a concordance index(C-index) and calibration curve.Results:The 5-year overall survival(OS) and progression-free survival(PFS) rates of patients with nasal ENKTCL were higher than those of patients with extra-nasal ENKTCL(OS:68.2%vs.46.0%,P = 0.030;PFS:53.4%vs.26.6%,P = 0.010).The 5-year OS and PFS rates of patients with Ann Arbor stage IE ENKTCL were higher than those of patients with Ann Arbor stage HE ENKTCL(OS:66.3%vs.59.2%,P = 0.003;PFS:51.4%vs.40.3%,P = 0.009).Multivariate analysis showed that age >60 years,ECOG performance status score >2,elevated lactate dehydrogenase(LDH) level,extranasal primary site,and regional lymph node involvement were significantly associated with lower 5-year OS rate;age >60 years,elevated LDH level,extra-nasal primary site,and regional lymph node involvement were significantly associated with lower 5-year PFS rate.The nomogram included the primary site and regional lymph node involvement based on multivariate analysis.The calibration curve showed good agreement between the predicted and actual 5-year OS and PFS rates,and the C-indexes of the nomogram for the OS and PFS rates were 0.697 and 0.634,respectively.Conclusions:The primary site and regional lymph node involvement are independent prognostic factors for earlystage ENKTCL treated with chemotherapy followed by definitive radiotherapy. | Shao‑Qing Niu Yong Yang Yi‑Yang Li Ge Wen Liang Wang Zhi‑Ming Li Han‑Yu Wang Lu‑Lu Zhang Yun‑Fei Xia Yu‑Jing Zhang | 2016 | Chinese Journal of Cancer2016,35,5: | 8 |
| 12 | Endocrine Disruption Activity of 30-day Dietary Exposure to Decabromodiphenyl Ethane in Balb/C Mouse显示文摘Objective This study aimed to evaluate the hepatotoxicity,metabolic disturbance activity and endocrine disrupting activity of mice treated by Decabromodiphenyl ethane(DBDPE).Methods In this study,Balb/C mice were treated orally by gavage with various doses of DBDPE.After 30 days of treatment,mice were sacrificed; blood,livers and thyroid glands were obtained,and hepatic microsomes were isolated.Biochemical parameters including 8 clinical chemistry parameters,blood glucose and hormone levels including insulin and thyroid hormone were assayed.The effects of DBDPE on hepatic cytochrome P450(CYP) levels and activities and uridinediphosphate-glucuronosyltransferase(UDPGT) activities were investigated.Liver and thyroid glands were observed.Results There were no obvious signs of toxicity and no significant treatment effect on body weight,or liver-to-body weight ratios between treatment groups.The levels of ALT and AST of higher dose treatment groups were markedly increased.Blood glucose levels of treatment groups were higher than those of control group.There was also an induction in TSH,T3,and f T3.UDPGT,PROD,and EROD activities were found to have been increased significantly in the high dose group.Histopathologic liver changes were characterized by hepatocyte hypertrophy and cytoplasmic vacuolization.Our findings suggest that DBDPE can cause a certain degree of mouse liver damage and insufficiency.Conclusion DBDPE has the activity of endocrine disruptors in Bal/C mice,which may induce drug-metabolizing enzymes including CYPs and UDPGT,and interfere with thyroid hormone levels mediated by Ah R and CAR signaling pathways.Endocrine disrupting activity of DBDPE could also affect the glucose metabolism homeostasis. | SUN Ru Bao SHANG Shuai ZHANG Wei LIN Ben Cheng WANG Qiang SHI Yun XI Zhu Ge | 2018 | Biomedical and Environmental Sciences2018,31,1: | 7 |

| 15 | Dynamic model for sucking process of pneumatic cutting-type safflower harvest device显示文摘This study proposed a method using negative pressure sucking the petals adhered around cones.The structural parameters of the sucking device affect the flow-field distribution in the negative-pressure air chamber.In order to improve the harvesting efficiency and quality of the pneumatic cutting-type safflower harvest device,a dynamic model was established and the safflower petals upwind area were measured.According to the test,the size parameters of the thornless Yumin safflower were as follows:the average necking diameter was 6.30 mm,the average capitulum diameter was 20.89 mm and the average petal length was 22.39 mm.The measured maximum frontal area of the safflower petal was 11-40 mm2.Secondly,the required negative pressure power that resulted in the rise of safflower petals was calculated.In general,when the suction of the negative pressure reaches 9.8 m/s,the safflower petals can be sucked successfully.The simulation of the flow field in the suction mouth indicated that the streamlined suction mouth was beneficial in reducing resistance and the test results of high-speed photography showed that the aforementioned condition could improve the efficiency of the upright safflower.And the test verified that the rate of the unshaped petals in the cylindrical air tunnel was low and the efficiency of carding and shaping under negative pressure was considerably better in wet petals than in dry ones.Results of the upright safflower petal experiment were consistent with the theoretical analysis and simulation conclusion,and indicated the precision of the dynamic model and suction flower mouth orifice-shaped simulation analysis. | Ge Yun Zhang Lixin Qian Ying Jiao Xiaopan Chen Yuanbo | 2016 | International Journal of Agricultural and Biological Engineering2016,9,5: | 6 |

| 17 | Smoking and Its Relation to Metabolic Status among Chinese Adults: Analysis of a Nationwide Survey显示文摘Objective To update the estimated prevalence rates of smoking and examine major metabolic diseases associated with smoking status in Chinese adults. Methods Using a complex, multistage, probability sampling design, we recruited a nationally representative sample of 98,658 Chinese adults aged ≥18 years in 2010. Information on current, former, never, and passive smoking status was obtained using a standard questionnaire. All estimates were weighted to represent the overall Chinese adult population. Results The estimated proportion of current smokers was 28.3% for Chinese adults aged ≥18 years. The corresponding values of former and passive smokers were 5.1% and 21.4%, respectively. Additionally, former smokers were found to have a less favorable metabolic risk profile among all categories of smoking status in both men and women. The prevalence of metabolic diseases such as diabetes and hypertension also increased with a greater number of smoking pack-years in men.
