1Near-diffraction-limited linearly polarized narrow-linewidth random fiber laser with record kilowatt output显示文摘In this paper, we propose and experimentally investigate a linearly polarized narrow-linewidth random fiber laser(RFL) operating at 1080 nm and boost the output power to kilowatt level with near-diffraction-limited beam quality using a master oscillation power amplifier. The RFL based on a half-opened cavity, which is composed of a linearly polarized narrow-linewidth fiber Bragg grating and a 500 m piece of polarization-maintained Ge-doped fiber, generates a 0.71 W seed laser with an 88 pm full width at half-maximum(FWHM) linewidth and a 22.5 dB polarization extinction ratio(PER) for power scaling. A two-stage fiber amplifier enhances the seed laser to the maximal 1.01 k W with a PER value of 17 dB and a beam quality of M_x^2=1.15 and M_y^2=1.13. No stimulated Brillouin scattering effect is observed at the ultimate power level, and the FWHM linewidth of the amplified random laser broadens linearly as a function of the output power with a coefficient of about 0.1237 pm∕W.To the best of our knowledge, this is the first demonstration of a linearly polarized narrow-linewidth RFL with even kilowatt-level near-diffraction-limited output, and further performance scaling is ongoing.JIANGMING XU LONG HUANG MAN JIANG JUN YE PENGFEI MA JINYONG LENG JIAN WU HANWEI ZHANG PU ZHOU 2017Photonics Research2017,5,4:20

3Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 4The effects of microRNA-34a regulating Notch-1/NF-κB signaling pathway on lipopolysaccharide-induced human umbilical vein endothelial cells显示文摘BACKGROUND: Notch-1/NF-κB signaling plays a key role in the cecal ligation and puncture(CLP)-induced sepsis. This study aims to investigate the intervention effects of microRNA-34a(miR-34a) lentivirus regulating Notch-1/NF-κB signaling pathway on lipopolysaccharide(LPS)-induced human umbilical vein endothelial cells(HUVEC).METHODS: HUVEC were divided into four groups as the following: they were infected with negative control lentivirus(NC group) or miR-34a lentivirus(OE group); LPS(1 g/mL) was added on the third day on the basis of NC group and OE group for 24 hours(NC+LPS group or OE+LPS group). The levels of TNF-α, IL-1β, IL-6, and IL-10 in the cell supernatants, and the mRNA and protein expression of Notch-1 and NF-κB in the HUVEC were evaluated.RESULTS: After 24 hours, the levels of TNF-α, IL-1β, IL-6 in the cell supernatants and the protein expression of NF-κB from NC+LPS group were significantly higher than those of NC group, but IL-10 level and the protein expression of Notch-1 in NC+LPS group were the opposite. After intervention of miR-34a lentivirus, the cell supernatants TNF-α and the protein expression of NF-κB in OE+LPS group after 24 hours markedly decreased compared to NC+LPS group. 9A Crucial Role of GA-Regulated Flavonol Biosynthesis in Root Growth of Arabidopsis显示文摘Flavonols have been demonstrated to play many important roles in plant growth, development, and communication with other organisms. Flavonol biosynthesis is spatiotemporally regulated by the subgroup 7 R2R3-MYB (SG7 MYB) transcription factors including MYB11/MYB12/MYB111. However, whether SG7- MYB activity is subject to post-translational regulation remains unclear. Here, we show that gibberellic acid (GA) inhibits flavonol biosynthesis via DELLA proteins in Arabidopsis. Protein-protein interaction analyses revealed that DELLAs (RGA and GAI) interacted with SG7 MYBs (MYB12 and MYB111) both in vitro and in vivo, leading to enhanced affinity of MYB binding to the promoter regions of key genes for flavonol biosynthesis and thus increasing their transcriptional levels. We observed that the level of auxin in the root tip was negatively correlated with root flavonol content. Furthermore, genetic assays showed that loss-of-function mutations in MYB 12, which is predominantly expressed in roots, partially rescued the short-root phenotype of the GA-deficient mutant ga1-3 by increasing root meristem size and mature cell size. Consistent with these observations, exogenous application of the flavonol quercetin restored the root meristem size of myb12 ga1-3 to that of ga1-3. 10kW-level,narrow-linewidth linearly polarized fiber laser with excellent beam quality through compact one-stage amplification scheme显示文摘In this manuscript, we demonstrate high-power, narrow-linewidth linearly polarized fiber laser with excellent beam quality through compact one-stage amplification scheme. By employing a single-mode–multimode–single-mode structure seed laser, a linearly polarized Yb-doped fiber laser with narrow linewidth and high output power is achieved.This laser, when used as a master oscillator, can be capable of suppressing the ASE in the process of power amplification.Thus, only one-stage amplification structure is used to scale up the laser power, and linearly polarized output with a polarization extinction ration of 14 d B, a narrow linewidth of 0.3 nm and an output power of 1018 W are achieved.Moreover, due to the good beam quality of seed laser and the well-designed amplifier stage, the beam quality of the output laser is near-diffraction-limited with M_x^2 ~1.18 and M_y^2 ~ 1.24 at the maximum power, and without mode instability occurring.Man Jiang Pengfei Ma Long Huang Jiangming Xu Pu Zhou Xijia Gu 2017High Power Laser Science and Engineering2017,5,4:7
11Improving the stability and bioavailability of tea polyphenols by encapsulations:a review显示文摘Tea polyphenols(TPPs)have attracted significant research interest due to their health benefits.However,TPPs are sensitive to certain environmental and gastrointestinal conditions and their oral bioavailability was found to be very low.Delivery systems made of food-grade materials have been reported to improve the shelf-life,bioavailability and bioefficacy of TPPs.This review discusses the chemistry of TPPs;the setbacks of TPPs for application;and the strategies to counteract application limitations by rationally designing delivery systems.An overview of different formulations used to encapsulate TPPs is provided in this study,such as emulsion-based systems(liposome,nanoemulsion,double emulsion,and Pickering emulsion)and nano/microparticles-based systems(protein-based,carbohydrate-based,and bi-polymer based).In addition,the stability,bioavailability and bioactivities of encapsulated TPPs are evaluated by various in vitro and in vivo models.The current findings provide scientific insights in encapsulation approaches for the delivery of TPPs,which can be of great value to TPPs-fortified food products.Further explorations are needed for the encapsulated TPPs in terms of their applications in the real food industry as well as their biological fate and functional pathways in vivo.Zhiya Yin Ting Zheng Chi-Tang Ho Qingrong Huang Qingli Wu Man Zhang 2022Food Science and Human Wellness2022,11,3:7

14IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE.Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu 2021Cellular & Molecular Immunology2021,18,7:6

16Mucus production stimulated by IFN-AhR signaling triggers hypoxia of COVID-19显示文摘Silent hypoxia has emerged as a unique feature of coronavirus disease 2019(COVID-19).In this study,we show that mucins are accumulated in the bronchoalveolar lavage fluid(BALF)of COVID-19 patients and are upregulated in the lungs of severe respiratory syndrome coronavirus 2(SARS-CoV-2)-infected mice and macaques.We find that induction of either interferon(IFN)-P or IFN-y upon SARS-CoV-2 infection results in activation of aryl hydrocarbon receptor(AhR)signaling through an IDO-Kyn-dependent pathway,leading to transcriptional upregulation of the expression of mucins,both the secreted and membrane-bound,in alveolar epithelial cells.Consequently,accumulated alveolar mucus affects the blood-gas barrier,thus inducing hypoxia and diminishing lung capacity,which can be reversed by blocking AhR activity.These findings potentially explain the silent hypoxia formation in COVID-19 patients,and suggest a possible intervention strategy by targeting the AhR pathway.Yuying Liu Jiadi Lv Jiangning Liu Man Li Jing Xie Qi Lv Wei Deng Nannan Zhou Yabo Zhou Jiangping Song Peng Wang Chuan Qin Wei-Min Tong Bo Huang 2020Cell Research2020,30,12:5

18High N-doped hierarchical porous carbon networks with expanded interlayers for efficient sodium storage显示文摘Sodium-ion batteries (SIBs) have been attracting considerable attention as a promising candidate for large-scale energy storage because of the abundance and low-cost of sodium resources. However, lack of appropriate anode materials impedes further applications. Herein, a novel self-template strategy is designed to synthesize uniform flowerlike N-doped hierarchical porous carbon networks (NHPCN) with high content of N (15.31 at.%) assembled by ultrathin nanosheets via a self-synthesized single precursor and subsequent thermal annealing. Relying on the synergetic coordination of benzimidazole and 2-methylimidazole with metal ions to produce a flowerlike network, a self-formed single precursor can be harvested. Due to the structural and compositional advantages, including the high N doping, the expanded interlayer spacing, the ultrathin two-dimensional nano-sized subunits, and the three-dimensional porous network structure, these unique NHPCN flowers deliver ultrahigh reversible capacities of 453.7 mAh·g^−1 at 0.1 A·g^−1 and 242.5 mAh·g^−1 at 1 A·g^−1 for 2,500 cycles with exceptional rate capability of 5 A·g^−1 with reversible capacities of 201.2 mAh·g^−1. 19Lichtenstein inguinal hernia repairs with porcine small intestine submucosa: a 5- year follow-up. a prospective randomized controlled study显示文摘Porcine small intestine submucosa(SIS)biologic patch has been used in inguinal hernia repair.However,there are lttle data available to assess the long-term effect after repair.This study aimed to explore the long-term effect of SIS patch in open inguinal hernia repair.Sevent-six patients with unilateral inguinal hernia were treated with Lichtenstein tension-free hernia repair using SIS patch(Beijing Datsing Bio-Tech Co,Ltd.)and Surgisis patch(COOK,USA)in Tianjn Union Medical Center and China-Japan Friendship Hospital.In the trial,the long-term efficacy of the treatment group and the control group were compared.A total of 66 patients in both groups received long-term follow-up(>5years)after surgery,with a followup rate of 86.8%.During the follow-up period,there was one case of recurrence,one case of chronic pain in the control group.There was no statistically significant difference(P>0.05)in terms of recurrence,chronic pain,foreign body sensation and infection between the two groups of patients.After long-term observations,it has been found that the porcine small intestinal submucosa(SIS)biological patch is safe and effective for inguinal hernia Lichtenstein repair,and has a low recurrence rate and complication rate.Baoshan Li Xin Zhang Yi Man Jiadong Xie Wei Hu Huang Huang Yinlong Wang Hongguang Ma 2021Regenerative Biomaterials2021,8,1:5
