1Apoptosis and Proinflammatory Cytokine Responses of Primary Mouse Microglia and Astrocytes Induced by Human H1N1 and Avian H5N1 Influenza Viruses显示文摘Patients with an influenza virus infection can be complicated by acute encephalopathy and encephalitis. To investigate the immune reactions involved in the neurocomplication,mouse microglia and astrocytes were isolated,infected with human H1N1 and avian H5N1 influenza viruses,and examined for their immune responses. We observed homogeneously distributed viral receptors,sialic acid (SA)-α2,3-Galactose (Gal) and SA-α2,6-Gal,on microglia and astrocytes. Both viruses were replicative and productive in microglia and astrocytes. Virus-induced apoptosis and cytopathy in infected cells were observed at 24 h post-infection (p.i.). Expression of IL-1β,IL-6 and TNF-α mRNA examined at 6 h and 24 h p.i. was up-regulated,and their expression levels were considerably higher in H5N1 infection. The amounts of secreted proinflammatory IL-1β,IL-6 and TNF-α at 6 h and 24 h p.i. were also induced,with greater induction by H5N1 infection. This study is the first demonstration that both human H1N1 and avian H5N1 influenza viruses can infect mouse microglia and astrocytes and induce apoptosis,cytopathy,and proinflammatory cytokine production in them in vitro. 2Microfluidic Electrospray Niacin Metal-Organic Frameworks Encapsulated Microcapsules for Wound Healing显示文摘Niacin metal-organic frameworks(MOFs)encapsulated microcapsules with alginate shells and copper-/zinc-niacin framework cores were in situ synthesized by using a microfuidic electrospray approach for wound healing.As the alginate shells were bacteriaresponsively degradable,the niacin MOFs encapsulated microcapsules could intelligently,controllably,and programmably release calcium,copper,and zinc ions,depending on the degree of infections.Te released ions could not only kill microbes by destroying their membrane and inducing the outfow of nutrient substance,but also activate copper/zinc superoxide dismutase(Cu/ZnSOD)to eliminate oxygen free radicals and rescue the cells from oxidative stress injury.Furthermore,the simultaneously released niacin could promote hemangiectasis and absorption of functional metal ions.Tus,the niacin MOFs encapsulated microcapsules were imparted with outstanding antibacterial,antioxidant,and angiogenesis properties.Based on an in vivo study,we have also demonstrated that the chronic wound healing process of an infected full-thickness skin defect model could be signifcantly enhanced by using the niacin MOFs encapsulated microcapsules as therapeutic agent.Terefore,the microfuidic electrospray niacin MOFs encapsulated microcapsules are potential for clinical applications.Guopu Chen Yunru Yu Xiuwen Wu Gefei Wang Guosheng Gu Feng Wang Jianan Ren Huidan Zhang Yuanjin Zhao 2019Research2019,,1:21
3Wave load computation in direct strength analysis of semi-submersible platform structures显示文摘A wave load computation approach in direct strength analysis of semi-submersible platform structures was presented in this paper. Considering the differences in shape of pontoon, column and beam, the combination of accumulative chord length cubic parameter spline theory and analytic method was adopted for generating the wet surface mesh of platform. The hydrodynamic coefficients of platform were calculated by the three-dimensional potential flow theory of the linear hydrodynamic problem for platform with low forward speed. The equation of platform motions was established and solved in frequency domain, and the responses of wave-induced loads on the platform can be obtained. With the interpolation method being utilized, the pressure loads on shell elements for finite element analysis (FEA) were converted from those on the hydrodynamic computation mesh, which pave the basis for FEA with commercial software.A computer program based on this method has been developed, and a calculation example of semi-submersible platform was illustrated.Analysis results show that this method is a satisfying approach of wave loads computation for this kind of platform.ZHANGHai-bin RENHui-long DAIYang-shan GEFei 2004Journal of Marine Science and Application2004,3,1:7
4Species-specific traits determine shrub-annual interactions during a growing season显示文摘Stress gradient hypothesis predicted that facilitative interactions usually increase in intensity and are importance with abiotic stress.By contrast,facilitation may be lost in time,when it involves the growth of benefactors or beneficiaries.Less is known about which response pattern is more common in arid desert.We present an empirical study to explore shrub-annual interactions at the community and individual level along the course of a single growing season in a desert steppe in northwest China.Here the severity of drought stress may increase in time due to uneven precipitation during plant growing season.We assessed growth responses of annuals in understory where two dominant shrubs were removed.Annuals responses showed a switch from weakly positive to more strongly positive beneath Calligonum mongolicum,whereas from positive to negative beneath Nitraria sphaerocarpa during the growing season.Additionally,annual species with contrasting functional traits showed distinct growth responses to canopies removal.There was evidence of an increase in soil moisture below the canopy of shrubs,but a decrease in potential evaporation rate and photosynthetically active radiation,which can partly explain these species-specific responses.We conclude that the balance between competitive and facilitative effects in shrub-annual interactions is not only governed by the severity of stress but also determined by plant traits,such as canopy structure of shrubs and functional traits of their understory annuals.GeFei ZHANG WenZhi ZHAO 2015Journal of Arid Land2015,7,3:4
5Effect of intraoperative fluid administration on perioperative outcomes in patients undergoing McKeown esophagogastrectomy显示文摘Objective: Fluid therapy is one of the key components of perioperative management. However, evidence of intraoperative fluid(IOF) administration affecting clinical outcomes following McKeown esophagogastrectomy remains limited. This study investigated the impact of IOF on clinical outcomes after McKeown esophagogastrectomy.Methods: Patients who underwent McKeown esophagogastrectomy between July 2013 and July 2016 were identified. Preoperative, intraoperative and postoperative variables for each eligible patient were retrospectively collected from our electronic medical records and anesthetic records. IOF rates were determined and their relationships to postoperative clinical outcomes were compared.Results: A total of 546 patients were enrolled in the analysis. The median IOF rate was 8.87 mL/kg/h. We divided the patients into two groups: a low fluid volume group(LFVG <8.87 mL/kg/h, n=273) and a high fluid volume group(HFVG ≥8.87 mL/kg/h, n=273). No statistically significant differences in postoperative clinical outcomes were found between LFVG and HFVG either before or after propensity score matching.Conclusions: No effect of IOF administration on clinical outcomes in patients undergoing McKeown esophagogastrectomy was identified. 7Effects of NS1 variants of H5N1 influenza virus on interferon induction, TNFa response and p53 activity显示文摘Non-structural protein 1(NS1)is an important virulence factor of the highly pathogenic H5N1 avian influenza virus.A five-amino-acid(5 aa)deletion at position 80–84 and an aspartic acid to glutamic acid substitution at position 92(D92E)are two major NS1 mutations that are highly correlated with enhanced virulence.To investigate the effect of these mutations in H5N1 virulence,three H5N1-NS1 variants were constructed:NS51(lacking 5 aa at position 80–84),NS51(I)(carrying a 5-aa insertion at position 80–84)and NS51(IM)(carrying both the 5-aa insertion and the D92E mutation).We examined the effects of these mutations on interferon(IFN)induction,tumor-necrosis factor(TNF)a response,p53 activity and apoptosis.We found that the D92E mutation eliminated NS1’s repressive effect on IFN induction,while the 5-aa deletion resulted in enhanced resistance to TNFa responses.We also observed that all three variants exhibited a similar suppressive effect on p53 transcriptional activity,although none of them significantly influenced apoptosis of host cells.Our findings shed new light on the role of NS1 in the pathogenicity of H5N1 virus.Weizhong Li Gefei Wang Heng Zhang Gang Xin Dangui Zhang Jun Zeng Xiaoxuan Chen Yanxuan Xu Youhong Cui Kangsheng Li 2010Cellular & Molecular Immunology2010,7,3:3
8Effects of sea level rise on storm surge and waves within the Yangtze River Estuary显示文摘Sea level rise (SLR) can cause water depth increase (WDI) and coastal inundation (CI).By applying the coupled FVCOM + SWAN model,this study investigates the potential impacts of WDI and CI,induced by a 1.0 m SLR,on storm surge and waves within the Yangtze River Estuary.A 1.0 m WDI decreases the maximum storm surge by 0.15 m and increases the maximum significant wave height by 0.35 m.The CI effect size is smaller when compared with WDI.CI decreases the maximum storm surge and significant wave height by 0.04 and 0.07 m,respectively.In the near-shore area,WDI significantly alters the local hydrodynamic environment,thereby stimulating changes in maximum storm surges and wave heights.Low-lying regions are negatively impacted by CI.Conversely,in deep-water areas,the relative change in water depth is minimal and the effect of CI is gradually enhanced.The combined effect of WDI and CI decreases the maximum surge by 0.31 m and increases the maximum significant wave height by 0.21 m.As a result,CI may be neglected when designing deep-water infrastructures.Nonetheless,the complex interactions between adoption and neglect of CI should be simulated to achieve the best seawall flood control standards and design parameters.Yongming SHEN Gefei DENG Zhihao XU Jun TANG 2019Frontiers of Earth Science2019,13,2:3

11Libration and end body swing stabilization of a parallel partial space elevator system显示文摘This paper studies the libration and stabilization of a parallel partial space elevator system in circular orbits. The system is made up of two paralleled partial space elevators, each of which consists of one main satellite, one end body and a climber moving along the tether between them.The libration characteristics of the elevator are studied through numerical analysis by a new dynamic model, and a novel control strategy is proposed to stabilize the swing of the end body by projecting the climber speeds only. Optimal control method is used to implement the new control strategy in the case where the climbers move in opposite direction.
