| 1 | Confocal-based fluorescence fluctuation spectroscopy with a SPAD array detector显示文摘The combination of confocal laser-scanning microscopy(CLSM)and fluorescence fluctuation spectroscopy(FFS)is a powerful tool in studying fast,sub-resolution biomolecular processes in living cells.A detector array can further enhance CLSM-based FFS techniques,as it allows the simultaneous acquisition of several samples-essentially images-of the CLSM detection volume.However,the detector arrays that have previously been proposed for this purpose require tedious data corrections and preclude the combination of FFS with single-photon techniques,such as fluorescence lifetime imaging.Here,we solve these limitations by integrating a novel singie-photon-avalanche-diode(SPAD)array detector in a CLSM system.We validate this new implementation on a series of FFS analyses:spot-variation fluorescence correlation spectroscopy,pair-correlation function analysis,and image-derived mean squared displacement analysis.We predict that the unique combination of spatial and temporal information provided by our detector will make the proposed architecture the method of choice for CLSM-based FFS. | Eli Slenders Marco Castello Mauro Buttafava Federica Villa Alberto Tosi Luca Lanzanó Sami Valtteri Koho Giuseppe Vicidomini | 2021 | Light(Science & Applications)2021,10,2: | 2 |

| 3 | Cytokinome profile evaluation in patients with hepatitis C virus infection显示文摘The‘‘omics sciences’’(genomics,transcriptomics,proteomics)are often used to study living organisms as a whole system by evaluating the complex expression patterns of genes,miRNA,proteins,and metabolites.This study aimed,through bioinformatics and systems biology,to decipher the cytokinome profile in the evolution of inflammatory processes leading to cancer.The cytokinome was defined as the totality of cytokines and their interactions in and around biological cells.The system biology approach would provide a better understanding of the complex interaction network of cytokines,especially in cancer patients.Acquired knowledge would enable health providers with tools to evaluate disease onset through progression as well as identifying innovative therapeutic strategies.Understanding the role each cytokine plays in the metabolic network is of great importance.This paper reviews our group’s‘‘omics’’work.In particular,it addresses the role cytokines play in liver disease in six different scenarios.The first is the role the cytokines play in chronic inflammatory diseases and cancers.The second is the significance of the cytokinome profile.The third is the role of liver cirrhosis as an inflammatory disease.The fourth is the comparison of cytokine levels evaluated in patients with chronic hepatitis C virus(HCV)or with HCV-related cirrhosis.The fifth is the comparison of cytokine levels evaluated in patients with HCV-related cirrhosis in the presence and absence of type 2 diabetes.And lastly,we present a comparison of cytokine levels evaluated in patients with HCV-related cirrhosis in the presence and absence of hepatocellular carcinoma. | Francesca Capone Eliana Guerriero Giovanni Colonna Patrizia Maio Alessandra Mangia Giuseppe Castello Susan Costantini | 2014 | World Journal of Gastroenterology2014,20,28: | 1 |

| 7 | Impaired diastolic function in nave untreated human immunodeficiency virus infected patients显示文摘AIM:To evaluate cardiac function and structure in un-treated human immunodeficiency virus(HIV) patients without clinical evidence of cardiovascular disease.METHODS:Fifty-three nave untreated HIV-infected patients and 56 healthy control subjects underwent clinical assessment,electrocardiography(ECG) and echocardiography,including tissue doppler imaging.Moreover,a set of laboratory parameters was obtained from all subjects,including HIV-RNA plasma levels,CD4 cell counts and tumor necrosis factor-α levels.RESULTS:The two groups showed normal ECG traces and no differences regarding systolic morphologic parameters.In contrast,a higher prevalence of left ventricular diastolic dysfunction(abnormal relaxation or pseudonormal filling pattern) was found in the HIV patients(36% vs 9% in patients and controls,respec-tively,P <0.001).CONCLUSION:Subclinical cardiac abnormalities ap-pear in an early stage of the HIV infection,indepen-dent of antiretroviral therapy.The data suggest that HIV per se plays a role in the genesis of diastolic dys-function. | Ugo Oliviero Giovanni Bonadies Giorgio Bosso Maria Foggia Valentina Apuzzi Mariarosaria Cotugno Antonio Valvano Enrico Leonardi Guglielmo Borgia Giuseppe Castello Raffaele Napoli Luigi Saccà | 2010 | World Journal of Cardiology2010,2,4: | 0 |
