| 1 | Mapping and validation of a dominant salt tolerance gene in the cultivated soybean(Glycine max) variety Tiefeng 8显示文摘Salt is an abiotic stress factor that strongly affects soybean growth and production. A single dominant gene has been shown to confer salt tolerance in the soybean cultivar Tiefeng 8.The objective of the present study was to genetically map the salt-tolerance gene in an F2:3population and a recombinant inbred line(RIL) population derived from a cross between two cultivated soybeans, Tiefeng 8(tolerant) and 85-140(sensitive). The F2:3families and RILs were treated with 200 mmol L-1Na Cl to evaluate salt tolerance. The F2:3population showed 1(42 tolerant): 2(132 segregating): 1(65 sensitive) segregation, indicating a single dominant gene for salt tolerance in Tiefeng 8. A sequence-characterized amplified region(SCAR) marker from a previously identified random amplified polymorphic DNA(RAPD)marker and four insertion/deletion polymorphism(In Del) markers were developed within the mapping region. Using these markers along with SSR markers, the salt-tolerance gene was mapped within 209 kb flanked by SCAR marker QS08064 and SSR marker Barcsoyssr_3_1301 on chromosome 3. Three markers that cosegregated with the salt tolerance gene and SCAR marker QS08064 were used to genotype 35 tolerant and 23 sensitive soybean accessions. These markers showed selection efficiencies of 76.2% to94.2%. The as-cast alloy was found to be composed of dendritic primaryα-Mg matrix, α-Mg +W(Mg_3Zn_3Gd_2) eutectic along grain boundaries, and icosahedral quasicrystalline I(Mg_3Zn_6Gd) phase within α-Mg matrix. During homogenization process, α-Mg +W(Mg_3Zn_3Gd_2) eutectic and I phase gradually dissolved into a-Mg matrix, while some rod-like rare earth hydrides(GdH2)formed within α-Mg matrix. Both the tensile yield strength and the elongation showed a similar tendency as a function of homogenization temperature and holding time. The optimized homogenization parameter was determined to be 505℃ for 16 h according to the microstructure evolution. [Methods] Phenol-sulfuric acid method was used to determine polysaccharide's content. Based on single-factor experiment and orthogonal test,the influences of solid-liquid ratio( A),ultrasonic extraction temperature( B),ultrasonic power( C) and ultrasonic time( D) on extraction rate of O. benghalensis polysaccharide were studied,and the optimal process of ultrasonic extraction was selected. [Results] The influence order of solid-liquid ratio( A),ultrasonic extraction temperature( B),ultrasonic power( C) and ultrasonic time( D) on extraction rate of O. benghalensis polysaccharide was A > D > B > C. | 14 | Rules of Changes in Soil Nutrients and Enzyme Activities of Larix principis-rupprechtii in Different Forest Ages显示文摘In this study,three different ages of Larix principis-rupprechtii forests in 5,10 and 20 years were selected as the research objects,and the changes in soil nutrient and soil enzyme activities in different growth stages were analyzed. The results showed that the contents of organic matter and available phosphorus in the soil of different growth stages showed a significant downward trend with the increase of soil depth.For different forest ages in the same soil layer,the soil available phosphorus content declined with the increase of the forest age. The organic matter content of 40-60 cm in 20 years of forest age was the lowest in July,which was 4. 17 g/kg,significantly lower than that in other soil layers. Besides,the soil available phosphorus content of 5 years of forest age reached the maximum in July,with an average of 4. 44 mg/kg,which was higher than the available phosphorus content in soil in May and September,but the difference between the three months was not significant. The changes in ammonium nitrogen and nitrate nitrogen content in soil with different forest ages were consistent with the changes in the soil depth,showing a downward trend. In the new leaf stage,the ammonium nitrogen content of the L. principis-rupprechtii forest land in5 years of age at 20-40 and 40-60 cm of the soil depth was 13. 47 and 9. 09 mg/kg,respectively,which was 46. 9% and 64. 2% lower than that at 0-20 cm( 25. 36 mg/kg) of the soil depth. The soil nitrate nitrogen content of 20 years of forest age was 19. 24 mg/kg,which was25. 8% lower than that of 10 years of forest age,showing significant difference( P < 0. 05). | 15 | Smart light-responsive hierarchical metal organic frameworks constructed mixed matrix membranes for efficient gas separation显示文摘One type of new light-responsive hierarchical metal organic framework(MOF) has been successfully prepared using Co(NO_(3))_(3)·6H_(2)O as the metal salt and 4,4’-azobenzenedicarboxylic acid as the ligand by microwave method for the first time. It is found that MOF [Co(Az DC)] exhibits a light-responsive characteristic to SO_(2)adsorption due to the presence of azo group from the ligand. The light-responsive hierarchical MOFs are incorporated into Matrimid■ 5218(PI) matrix to prepare mixed matrix membranes(MMMs) for gas separation application. The morphology, crystallinity, chain mobility and thermal stability of MMMs are explored. Results show that Co(Az DC) may elevate both the CO_(2)(SO_(2)) permeability and CO_(2)(SO_(2))/N_(2)selectivity of the MMMs. In particular,the Co(Az DC) doped MMMs exhibit the significantly improved CO_(2)(SO_(2))/N_(2)selectivity from 33(123) for PI control membrane to 78(420) for MMMs, overcoming the 2008 Robeson upper bound for CO_(2)/N_(2)system. Sizesieving effect of Co(Az DC) with pore size 0.35 nm enhances the selectivity, while the –N=N– group from Co(Az DC) shows affinity to CO_(2)molecular rather than N_(2), also elevating selectivity of MMMs. In brief, enhanced selectivity of high-performance membrane is attributed to incorporation of Co(Az DC) particles, which displays synergistic effects both in size-sieving and CO_(2)-philic interaction for CO_(2)/N_(2)separation. Smart highly selective interface is constructed in MMMs by switching the configuration of MOFs from cis to trans. | 16 | Assessment and management of the performance risk of a pilot reclaimed water disinfection process显示文摘Chlorination disinfection has been widely used in reclaimed water treatment plants to ensure water quality. In order to assess the downstream quality risk of a running reclaimed water disinfection process, a set of dynamic equations was developed to simulate reactions in the disinfection process concerning variables of bacteria, chemical oxygen demand(COD), ammonia and monochloramine.The model was calibrated by the observations obtained from a pilot disinfection process which was designed to simulate the actual process in a reclaimed water treatment plant. A Monte Carlo algorithm was applied to calculate the predictive efuent quality distributions that were used in the established hierarchical assessment system for the downstream quality risk, and the key factors afecting the downstream quality risk were defined using the Regional Sensitivity Analysis method. | 17 | Dynamic profiles of DNA methylation and the interaction with histone acetylation during fiber cell initiation of Gossypium hirsutum显示文摘Background:Fiber,as the main product of cotton,provides main raw material for the textile industry.Many key factors have been revealed a significant role in fiber cell development including Myb proteins,phytohormones,fatty acid metabolites,and epigenetic modifications.DNA methylation is one of the important epigenetic modifications to regulate plant development and responses to abiotic or biotic stimuli.In general,DNA methylation consisting of 5mC and 6mA regulates the chromatin structure and gene transcription to affect plant development,however,the detailed role and underlying mechanism of DNA methylation in the fiber development of cotton are yet vague.Results:Here,systematical study of the 5mC and 6mA DNA methylation profiles during the fiber initiation period of Xu142 and its glabrous mutant Xu142fl represented a clear alteration of global DNA methylation associated with fiber cell initiation.Then,the genome-wide identification of genes responsible for methylation regulation at the fifth carbon of cytosine and the sixth carbon of adenine of DNA was operated in Gossypium hirsutum.As a result,13,10,6,and 17 genes were identified for 5mC methylation,5mC demethylation,6mA methylation,and 6mA demethylation,respectively.We then investigated the tissue expression pattern of all these genes,and some genes showed higher expression levels in fiber initiation,among which some displayed a significant change in transcription between Xu142 and Xu142fl.The possible interaction between histone acetylation and DNA methylation in fiber initiation through in vitro culture was studied by dot blot,and the results showed that repressed histone deacetylation by Trichostatin A(TSA)inhibited the global DNA methylation,and some causal genes(e.g.,GhDMT13,GhDAMT2,GhALKBH12,GhDM7)were also identified.Conclusions:In this study,all the findings indicated the interplay between histone acetylation and DNA methylation,supporting their important roles and providing precious clues for the epigenetic modifications associated with DNA methylation in the fiber development of cotton. | CHEN Guangyu LI Yonghui WEI Zhenzhen GAN Lei LIU Jisheng WANG Zhi | 2022 | Journal of Cotton Research2022,5,1: | 0 |
