| 1 | Novel coumarin–benzimidazole derivatives as antioxidants and safer anti-inflammatory agents显示文摘Inspired from occurrence of anti-inflammatory activity of 3-substituted coumarins and antiulcer activity of various 2-substituted benzimidazoles,novel compounds have been designed by coupling coumarin derivatives at 3-position directly or through amide linkage with benzimidazole nucleus at 2-position.The resultant compounds are expected to exhibit both anti-inflammatory and antioxidant activities along with less gastric toxicity profile.Two series of coumarin–benzimidazole derivatives(4a–e and 5a–e)were synthesized and evaluated for anti-inflammatory activity and antioxidant activity.Compounds 4c,4d and 5a displayed good anti-inflammatory(45.45%,46.75%and 42.85%inhibition,respectively,versus 54.54% inhibition by indomethacin)and antioxidant(IC_(50) of 19.7,13.9 and 1.2 mmol/L,respectively,versus 23.4 mmol/L for butylatedhydroxytoluene)activities.Evaluation of ulcer index and in vivo biochemical estimations for oxidative stress revealed that compounds 4d and 5a remain safe on gastric mucosa and did not induce oxidative stress in tissues.Calculation of various molecular properties suggests the compounds to be sufficiently bioavailable. | Radha Krishan Arora Navneet Kaur Yogita Bansal Gulshan Bansal | 2014 | Acta Pharmaceutica Sinica B2014,4,5: | 6 |
| 2 | Absolute copy number differences of Y chromosomal genes between crossbred (Bos taurus×Bos indicus) and Indicine bulls显示文摘Background: The Y chromosome in mammal is paternally inherited and harbors genes related to male fertility and spermatogenesis. The unique intra-chromosomal recombination pattern of Y chromosome and morphological difference of this chromosome between Bos taurus and Bos indicus make it an ideal model for studying structural variation, especially in crossbred(Bos taurus × Bos indicus) bulls. Copy Number Variation(CNV) is a type of genomic structural variation that gives information complementary to SNP data. The purpose of this study was to find out copy number differences of four Y chromosomal spermatogenesis-related candidate genes in genomic DNA of crossbred and purebred Indicine bulls. Result: Four Y chromosomal candidate genes of spermatogenesis namely, sex determining gene on Y chromosome(SRY), DEAD box polypeptide 3-Y chromosome(DDX3Y), Ubiquitin specific peptidase 9, Y-linked(USP9Y), testis-specific protein on Y chromosome(TSPY) were evaluated. Absolute copy numbers of Y chromosomal genes were determined by standard curve-based quantitative real time PCR. Copy numbers of SRY and TSPY genes per unit amount of genomic DNA are higher in crossbred than Indicine bulls. However, no difference was observed in DDX3Y and USP9Y gene copy numbers between two groups. Conclusion: The present study demonstrates that the structural organization of Y chromosomes differs between crossbred and Indicine bulls which are reproductively healthy as observed from analysis of semen attributes. The absolute copy numbers of SRY and TSPY genes in unit mass of genomic DNA of crossbred bulls are significantly higher than Indicine bulls. No alteration in absolute copies of DDX3Y and USP9Y gene was found between the genome of crossbred and Indicine bulls. | 4 | Variation in Antioxidants, Bioactive Compounds and Antioxidant Capacity in Germinated and Ungerminated Grains of Ten Rice Cultivars显示文摘Present study was conducted to characterise ten rice(Oryza sativa L.) cultivars viz. IET-23466, Dhan-201, IET-23448, MAS-946, IET-23445, IET-23463, IET-23455, PR-123, PR-115 and IET-23449 based on antioxidants(total phenolics, flavonols and tannins), bioactive compounds(phytic acid, gamma amino butyric acid, tocopherol and reduced ascorbate) and antioxidant activity(1,1-diphenyl-2-picrylhydrazyl radical scavenging activity, hydroxyl radical scavenging activity, free radical antioxidant power and total reducing power) with an aim to identify cultivars containing higher health promoting components after germination. Three cultivars IET-23466, IET-23463 and PR-123 performed better as revealed by higher level of antioxidants, bioactive compounds and antioxidant activity before and after germination. Three cultivars MAS-946, IET-23445 and IET-23449 had moderate level of antioxidants, bioactive compounds and antioxidant activity. Four cultivars Dhan-201, IET-23448, IET-23455 and PR-115 performed intermediately. Strong positive correlation was observed among total phenolics and the antioxidant activity. Phytic acid was found to be negatively correlated to the antioxidant activity. | 13 | Four new degradation products of doxorubicin:An application of forced degradation study and hyphenated chromatographic techniques显示文摘Forced degradation study on doxorubicin(DOX) was carried out under hydrolytic condition in acidic,alkaline and neutral media at varied temperatures,as well as under peroxide,thermal and photolytic conditions in accordance with International Conference on Harmonization(ICH) guidelines Q1(R2).It was found extremely unstable to alkaline hydrolysis even at room temperature,unstable to acid hydrolysis at 80 ℃,and to oxidation at room temperature.It degraded to four products(O-I-O-IV) in oxidative condition,and to single product(A-I) in acid hydrolytic condition.These products were resolved on a C_8(150 mm × 4.6 mm,5 μm) column with isocratic elution using mobile phase consisting of HCOONH_4(10 mM,pH 2.5),acetonitrile and methanol(65:15:20,v/v/v).Liquid chromatography-photodiode array(LC-PDA) technique was used to ascertain the purity of the products noted in LC-UV chromatogram.For their characterization,a six stage mass fragmentation(MS^6) pattern of DOX was outlined through mass spectral studies in positive mode of electrospray ionization(+ ESI) as well as through accurate mass spectral data of DOX and the products generated through liquid chromatography-time of flight mass spectrometry(LC-MS-TOF) on degraded drug solutions.Based on it,O-l-O-IV were characterized as 3-hydroxy-9-desacetyldoxorubicin-9-hydroperoxide,l-hydroxy-9-desacetyldoxorubicin-9-hydroperoxide,9-desacetyldoxorubicin-9-hydroperoxide and 9-desacetyldoxorubicin,respectively,whereas A-I was characterized as deglucosaminyl doxorubicin.While A-I was found to be a pharmacopoeial impurity,all oxidative products were found to be new degradation impurities.The mechanisms and pathways of degradation of doxorubicin were outlined and discussed. | Dheeraj Kaushik Gulshan Bansal | 2015 | Journal of Pharmaceutical Analysis2015,5,5: | 1 |
