1Water in the Mogao Grottoes,China:where it comes from and how it is driven显示文摘The Dunhuang Mogao Grottoes in China was designated as a world heritage site by UNESCO in 1987 and is famous for its cultural relics. Water is the most active factor that harms the relics in the caves as it damages the grotto murals and painted sculptures. Thus, determining the water sources and driving forces of water movement is a key issue for protecting these cultural relics. These issues have troubled relics protectors for a long time. In this study, the authors chose a representative cave in the Mogao Grottoes and, by completely sealing the cave to make a closed system, measured the water vapor from the surrounding rock. This was accomplished by installing a condensation–dehumidification temperature–humidity control system for the collection of water vapor. The results show that there is continuous evaporation from the deep surrounding rock into the cave. The daily evaporation capacity is determined to be 1.02 g/(d?m2). The water sources and driving forces of water movement were further analyzed according to the character of the water evaporation and by monitoring the temperature and humidity of the surrounding rock. It was found that the water vapor in the cave derives from phreatic water. Moreover, the yearly fluctuation of temperature in the surrounding rock and geothermal forces are the basic powers responsible for driving phreatic evaporation. Under the action of the yearly temperature fluctuations, decomposition and combination of bound water acts as a 'pump' that drives phreatic water migration and evaporation. When the temperature rises, bound water decomposes and evaporates; and when it falls, the rock absorbs moisture. This causes the phreatic water to move from deep regions to shallow ones. 2The First Crested Duck Genome Reveals Clues to Genetic Compensation and Crest Cushion Formation显示文摘The Chinese crested(CC)duck is a unique indigenous waterfowl breed,which has a crest cushion that affects its survival rate.Therefore,the CC duck is an ideal model to investigate the genetic compensation response to maintain genetic stability.In the present study,we first generated a chromosome-level genome of CC ducks.Comparative genomics revealed that genes related to tissue repair,immune function,and tumors were under strong positive selection,indicating that these adaptive changes might enhance cancer resistance and immune response to maintain the genetic stability of CC ducks.We also assembled a Chinese spot-billed(Csp-b)duck genome,and detected the structural variations(SVs)in the genome assemblies of three ducks(i.e.,CC duck,Csp-b duck,and Peking duck).Functional analysis revealed that several SVs were related to the immune system of CC ducks,further strongly suggesting that genetic compensation in the anti-tumor and immune systems supports the survival of CC ducks.Moreover,we confirmed that the CC duck originated from the mallard ducks.Finally,we revealed the physiological and genetic basis of crest traits and identified a causative mutation in TAS2R40 that leads to crest formation.Overall,the findings of this study provide new insights into the role of genetic compensation in adaptive evolution.Guobin Chang Xiaoya Yuan Qixin Guo Hao Bai Xiaofang Cao Meng Liu Zhixiu Wang Bichun Li Shasha Wang Yong Jiang Zhiquan Wang Yang Zhang Qi Xu Qianqian Song Rui Pan Lingling Qiu Tiantian Gu Xinsheng Wu Yulin Bi Zhengfeng Cao Yu Zhang Yang Chen Hong Li Jianfeng Liu Wangcheng Dai Guohong Chen 2023Genomics, Proteomics & Bioinformatics2023,21,3:0
3Cysteine dioxygenase and taurine are essential for embryo implantation by involving in E2-ERαand P_(4)-PR signaling in mouse显示文摘Background Taurine performs multiple physiological functions,and the maintenance of taurine level for most mammals relies on active uptake from diet and endogenous taurine synthesis through its synthesis enzymes,including cysteine dioxygenase(CDO).In addition,uterus tissue and uterus fluid are rich in to urine,and to urine synthesis is regulated by estrogen(E2)and progesterone(P_(4)),the key hormones priming embryo-uterine crosstalk during embryo implantation,but the functional interactions and mechanisms among which are largely unknown.The present study was thus proposed to identify the effects of CDO and taurine on embryo implantation and related mechanisms by using Cdo knockout(KO)and ovariectomy(OVX)mouse models.Results The uterine CDO expression was assayed from the first day of plugging(d 1)to d 8 and the results showed that CDO expression level increased from d 1 to d 4,followed by a significant decline on d 5 and persisted to d 8,which was highly correlated with serum and uterine taurine levels,and serum P_(4) concentration.Next,Cdo KO mouse was established by CRISPER/Cas9.It was showed that Cdo deletion sharply decreased the taurine levels both in serum and uterus tissue,causing implantation defects and severe subfertility.However,the implantation defects in Cdo KO mice were partly rescued by the taurine supplementation.In addition,Cdo deletion led to a sharp decrease in the expressions of P_(4)receptor(PR)and its responsive genes Ihh,Hoxa10 and Hand2.Although the expression of uterine estrogen receptor(ERa)had no significant change,the levels of ERa induced genes(Muc1,Ltf)during the implantation window were upregulated after Cdo deletion.These accompanied by the suppression of stroma cell proliferation.Meanwhile,E2inhibited CDO expression through ERa and P_(4)upregulated CDO expression through PR.Conclusion The present study firstly demonstrates that taurine and CDO play prominent roles in uterine receptivity and embryo implantation by involving in E2-ERa and P_(4)-PR signaling.These are crucial for our understanding the mechanism of embryo implantation,and infer that taurine is a potential agent for improving reproductive efficiency of livestock industry and reproductive medicine.Di Zhang Zhijuan Wang Xuan Luo Hongzhou Guo Guobin Qiu Yuneng Gong Hongxu Gao Sheng Cui 2023Journal of Animal Science and Biotechnology2023,14,3:0
