1Establishment of a Core Collection for the Chinese annual wild soybean (Glycine Soja)显示文摘With more than 6000 accessions collected from across China, the Chinese National Crop Gene Bank (CNCGB) holds the largest collection of annual wild soybean (Glycine Soja) germplasm in the world. To facilitate the management and utilization of these germplasm collections, it is important to establish a Core Collection. This study compares five sampling strategies, namely random, constant, proportional, logarithmic and genetic diversity-based, to establish a Core Collection for the annual wild soybean germplasm. Among the strategies evaluated, the genetic di- versity-based was found to be the simplest and most efficient. Using hierarchical classification and cluster analysis, in the genetic diversity diversity-based strategy, 652 accessions, accounting for 10.65% of the total 6172 accessions, were se- lected out to represent the total accessions. The established Core Collection has the following features: (i) the 21 de- scriptors observed in the entire collection were compared by the Core Collection, all 18 quality characters of entire collec- tion were preserved by core collection, and coincidence rate of average was 98.4%; (ii) the variant of 13 descriptors of the two collections was very similar, with the coincidence index being 0.96; (iii) the coincidence rate of genetic diversity be- tween the two collections was 81.38% DNA alleles; (iv) the geographic distribution pattern of core collection was the same as the entire collection; (v) molecular marker analysis by 20 SSR primer pairs on 299 accessions showed that the Core Collection covered 83.64% of the entire collection. 2Progresses in studies of nuclear actin显示文摘Actin is a protein abundant in cells. Recently, it has been proved to be universally existent in the nuclei of many cell types. Actin and actin-binding proteins, as well as actin-related proteins, are necessary for the mediation of the conformation and function of nuclear actin, including the transformation of actin between unpolymerized and polym- erized, chromatin remodeling, regulation of gene expression and RNA processing as well as RNA transportation. 3Immunocytochemical identification of actin in mitochdria of Physarum polycephalum显示文摘Mitochondria isolated from the plasmodia of Physarum polycephalum Schw.are reacted with rabbit anti-actin antibody,and detected with FITC-conjugated sheep anti-rabbit IgG antibody.The results of indirect immunofluorescence show that actin exists in the mitochondria.Western blot analysis confirms the existence of actin in the protein preparation of the mitochondria.The indirect immunoelectron microscopic observation using the same antibodies verifies further that actin is the constituents of mitochondria,and it is dispersively distributed in the mitochondria of P.polycephalum.ZENGXianlu XINGMiao WANGXiaoguang SONGZhaoxia HAOShui 2003Progress in Natural Science:Materials International2003,13,1:0
4Impact of nuclear actin on the prophase chromosome construction of Physarum polycephalum显示文摘A cell free system efficiently promoting mitosis has been developed using the precise natural synchronous plasmodium of Physarum polycephalum . The content changes of nuclear cyclin B were exploited to represent the prophase process of Physarum polycephalum . The possible function of nuclear actin on chromosome construction was investigated by detecting the content changes of nuclear cyclin B in the late G 2 phase nuclei treated with cytochalasin B and incubated in the cell free system.
