2Expenditure and financial burden for the diagnosis and treatment of colorectal cancer in China:a hospital.based,multicenter,cross-sectional survey显示文摘Background: The increasing prevalence of colorectal cancer(CRC) in China and the paucity of information about relevant expenditure highlight the necessity of better understanding the financial burden and effect of CRC diagnosis and treatment. We performed a survey to quantify the direct medical and non-medical expenditure as well as the resulting financial burden of CRC patients in China.Methods: We conducted a multicenter, cross-sectional survey in 37 tertiary hospitals in 13 provinces across China between 2012 and 2014. Each enrolled patient was interviewed using a structured questionnaire. All expenditure data were inflated to the 2014 Chinese Yuan(CNY; 1 CNY = 0.163 USD). We quantified the overall expenditure and financial burden and by subgroup(hospital type, age at diagnosis, sex, education, occupation, insurance type, household income, clinical stage, pathologic type, and therapeutic regimen). We then performed generalized linear modeling to determine the factors associated with overall expenditure.Results: A total of 2356 patients with a mean age of 57.4 years were included, 57.1 % of whom were men; 13.9% of patients had stage I cancer; and the average previous-year household income was 54,525 CNY.The overall average direct expenditure per patient was estimated to be 67,408 CNY, and the expenditures for stage Ⅰ, Ⅱ, Ⅲ, and Ⅳ disease were 56,099 CNY, 59,952 CNY, 67,292 CNY, and 82,729 CNY, respectively. Non-medical expenditure accounted for 8.3%of the overall expenditure. The 1-year out-of-pocket expenditure of a newly diagnosed patient was 32,649 CNY, which accounted for 59.9% of their previous-year household income and caused 75.0% of families to suffer an unmanageable financial burden. Univariate analysis showed that financial burden and overall expenditure differed in almost all subgroups(P < 0.05), except for sex. Multivariate analysis showed that patients who were treated in specialized hospitals and those who were diagnosed with adenocarcinoma or diagnosed at a later stage were likely to spend more,whereas those with a lower household income and those who underwent surgery spent less(all P < 0.05).Conclusions: For patients in China, direct expenditure for the diagnosis and treatment of CRC seemed catastrophic,and non-medical expenditure was non-ignorable. 4MiR-125a-5p Regulates Vitamin D Receptor Expression in a Mouse Model of Experimental Autoimmune Encephalomyelitis显示文摘Multiple sclerosis(MS)is a chronic and incurable autoimmune neurodegenerative disease of the central nervous system.Although the symptoms of MS can be managed by vitamin D3 treatment alone,this condition cannot be completely eradicated.Thus,there might be unknown factors capable of regulating the vitamin D receptor(VDR).Genome-wide analysis showed that miRNAs were associated with VDRs.We sought to determine the role and mechanism of action of miRNA-125a-5p and VDRs in a model of MS,mice with experimental autoimmune encephalomyelitis(EAE),which was induced by myelin oligodendrocyte glycoprotein 35–55 peptides.EAE mice showed decreased mean body weight but increased mean clinical scores compared with vehicle or control mice.And inflammatory infiltration was found in the lumbosacral spinal cord of EAE mice.In addition,VDR expression was significantly lower while the expression of miR-125a-5p was markedly higher in the spinal ventral horn of EAE mice than in vehicle or control mice.Importantly,activation of VDRs by paricalcitol or inhibition of miR-125a-5p by its antagomir markedly decreased the mean clinical scores in EAE mice.Interestingly,VDR and miR-125a-5p were co-localized in the same neurons of the ventral horn.More importantly,inhibition of miR-125a-5p remarkably blocked the decrease of VDRs in EAE mice.These results support a critical role for miR-125a-5p in modulating VDR activity in EAE and suggest potential novel therapeutic interventions.Han-Chun Long Rui Wu Chun-Feng Liu Fei-Long Xiong Zu Xu Dian He Yi-Fan Zhang Bing Shao Ping-An Zhang Guang-Yin Xu Lan Chu 2020Neuroscience Bulletin2020,36,2:6
5Robust microwave absorption in silver-cobalt hollow microspheres with heterointerfaces and electric-magnetic synergism:Towards achieving lightweight and absorption-type microwave shielding composites显示文摘Lightweight and high-efficiency microwave shielding materials based on high absorption loss are the hotspot of current research.Herein,the silver-cobalt bimetallic hollow microspheres(Ag-Co-HM)with low reflection loss and broadband absorption efficiency were fabricated and then used to construct bimetallic hollow structures in polydimethylsiloxane/carbon nanotube composites(PDMS/CNT)to obtain the lightweight and absorption-type microwave shielding composites.The microwave absorption of Ag-Co-HM and the microwave shielding effectiveness(SE T)of the PDMS/CNT/Ag-Co-HM composites was evaluated and discussed.The Ag-Co-HM with a molar ratio(Co/Ag)of 2.07 exhibited a low reflection loss as-46.6 dB at a thickness of 2.6 mm,and an effective absorption bandwidth of 8.40 GHz.The SE T of the PDMS/CNT composites was greatly enhanced by the incorporation of the Ag-Co-HM.Furthermore,the absorption coefficient(A)was also improved by adding the Ag-Co-HM and was much higher than the reflection coefficient.The composites exhibited absorption-type microwave shielding composites.For example,the average microwave SE T of 59.4 dB and A value of 0.78 are achieved in the PDMS/CNT/Ag-Co-HM composites containing 5 wt%CNTs and 10 wt%Ag-Co-HMs,which are much higher than the cor-responding values of 33.7 dB and 0.63 in the PDMS/CNT composites with only 5 wt%CNTs.In addition,the hollow structure of Ag-Co-HM endows the PDMS/CNT/Ag-Co-HM composites to have a low density.Dian Yang Jun-Ru Tao Yi Yang Qian-Ming He Ming Wang 2023Journal of Materials Science & Technology2023,,7:4
6Facile preparation of pH-responsive PEGylated prodrugs for activated intracellular drug delivery显示文摘PEGylated prodrug,covalent attaching polyethylene glycol(PEG) polymer chains to therapeutic drugs,is one of the most promising techniques to improve the water-solubility,stability,and therapeutic effect of drugs.In this study,three PEGylated acid-sensitive prodrugs DOX-PEG-DOX with different molecular weights,were prepared via Schiff-base reaction between aldehyde-modified PEG and the amino groups of doxorubicin(DOX).This kind of amphiphilic polymeric prodrug could be self-assemble into nanoparticles in aqueous solution.The average particle size and morphologies of the prodrug nanoparticles under different pH conditions were observed by dynamic light scattering(DLS) and transmission electron microscopy(TEM),re s pectively.It turned out that the nanoparticles could be kept stable in the physiological environment,but degraded in acidic medium.Subsequently,we also investigated in vitro drug release behavior and found that the prodrug had acid-sensitive property.The cytotoxicity and intracellular uptake assays revealed that the prodrugs could rapidly internalized by HeLa or HepG2 cells to release DOX and effectively inhibited the proliferation of the tumor cells,which have the potential for use in cancer therapy.Yue Song Dian Li Jinlin He Mingzu Zhang Peihong Ni 2019Chinese Chemical Letters2019,30,12:3
7Synthesis and Crystal Structure of N-((3-(2-nitrophenyl) propanoyl)oxy)-N-phenylbenzamide显示文摘The title compound N-((3-(2-nitrophenyl) propanoyl)oxy)-N-phenylbenzamide(C22H18N2O5,Mr = 390.38) was synthesized with nitrobenzene as the starting materials and structurally characterized by 1H NMR,13 C NMR,IR,H RMS(E-SI) and single-crystal X-ray diffraction.The crystalline structure of the title compound was studied in detail.The single-crystal X-ray diffraction revealed that the compound crystallizes in monoclinic,space group P21/n,with a = 14.4481(7),b = 8.0089(5),c = 16.6808(9) ?,α = 90.00(5),β = 100.069(5),γ = 90.00(5)°,Mr = 390.38,V = 1900.46(18) ?3,Z = 4,Dc = 1.364 g/cm-3,μ = 0.098,F(000) = 816.0,the final R = 0.0530 and w R = 0.1141 for 2525 observed reflections(I > 2σ(I)),R(all data) = 0.0813,w R(all data) = 0.1335,completeness to theta of 26.020 is 99.8% and GOF = 1.085.The crystal structure contains three branched chains with its center placed at the midpoint of N.The phenyl ring(C(8)~C(13)) makes a dihedral angle of 83.5(1) ℃ with the phenyl ring(C(1)~C(6)),and 80.5(1) ℃with the nitro-substituted benzene ring(C(17)~C(22)),indicating that the three rings are not coplanar.Hydrogen bonds(C(10)–H(10)···O(3)),(C(15)–H(15B)···O(4) and C(16)–H(16B)···O(4)) together with π-π stacking interactions stabilize the structure of the molecule.The preliminary biological test shows that the title compound has good antitumor activity against A549 in vitro with the IC50 value of 3.17 μmol.LIU Xiao-Feng ZHANG Yang LI Jing ZHAO Jing-Si XI Na HE Dian 2015Chinese Journal of Structural Chemistry2015,34,5:2
8Synthesis of hydroxycinnamic acid derivatives as mitochondria-targeted antioxidants and cytotoxic agents显示文摘In order to develop agents with superior chemopreventive and chemotherapeutic properties against hepatocellular carcinomas, mitochondria-targeted hydroxycinnamic acids(Mito HCAs) were synthesized by conjugation with a triphenylphosphonium cation. These synthetic compounds were evaluated for their antioxidant activities in hepatic mitochondria, including against OH? àand ROO? àinduced lipid peroxidation. H_2O_2 production was decreased significantly by increasing glutathione peroxidase and catalase activities. In addition, cell proliferation data from three cell lines(HepG2, L02 and WI38) indicated that the Mito HCAs were selective for cancer cells. Interestingly, the Mito HCAs both with or without Ca^(2+)triggered mitochondrial dysfunction by inducing mitochondrial swelling, collapsing the mitochondrial membrane potential and causing cytochrome c release. 9Arabidopsis NIMIN1,2 mediate redox homeostasis and JA/ET pathways to modulate Botrytis cinerea resistance显示文摘Arabidopsis NIM1-INTERACTING1 (NIMIN1) and NIMIN2 are required for salicylic acid (SA)-dependent resistance against biotrophic pathogens. In this study, we have demonstrated that NIMIN1, 2 are also essential for plant defense response to necrotrophic pathogen Botrytis cinerea. The nimin1 and nimin2 mutants displayed a higher susceptibility against B. cinerea than the wild type, which correlated with a decrease in B. cinerea-induced PDF).2 expression. Mutation in NIMIN1 or NIMIN2 enhanced accumulation of hydrogen peroxide (H_2O_2) with reductions in the activities of three main antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) at the early time (24h) of upon B. cinerea infection. These mutations also resulted in a visible decrease in B.cinerea-induced Ethylene Responsive Factor 1 (ERF1), Octadecanoid-Responsive Arabidopsis AP2/ERF 59 (ORA59), Allene Oxide Cyclase (AOC3), Opda Reductase 3 (OPR3),ACC Synthesis 2 (ACS2) and ACS6 expression, but an advance in MYC2 expression,indicating that NIMIN1,2 are essential for B. cinerea-activated jasmonic acid (JA)/ethylene(ET) biosynthesis and signaling. However, mutation in NIMIN1 or NIMIN2 drastically suppressed JA-, but not ET-activated PDF1.2 expression. 12Comparative analysis of constitutes and metabolites for traditional Chinese medicine using IDA and SWATH data acquisition modes on LC-Q-TOF MS显示文摘Identification of components and metabolites of traditional Chinese medicines(TCMs)employing liquid chromatography-quadrupole time-of-flight mass spectrometry(LC-Q-TOF MS)techniques with information-dependent acquisition(IDA)approaches is increasingly frequent.A current drawback of IDA-MS is that the complexity of a sample might prevent important compounds from being triggered in IDA settings.Sequential window acquisition of all theoretical fragment-ion spectra(SWATH)is a dataindependent acquisition(DIA)method where the instrument deterministically fragments all precursor ions within the predefined m/z range in a systematic and unbiased fashion.Herein,the superiority of SWATH on the detection of TCMs’components was firstly investigated by comparing the detection efficiency of SWATH-MS and IDA-MS data acquisition modes,and sanguisorbin extract was used as a mode TCM.After optimizing the setting parameters of SWATH,rolling collision energy(CE)and variable Q1 isolation windows were found to be more efficient for sanguisorbin identification than the fixed CE and fixed Q1 isolation window.More importantly,the qualitative efficiency of SWATH-MS on sanguisorbins was found significantly higher than that of IDA-MS data acquisition.In IDA mode,18 kinds of sanguisorbins were detected in sanguisorbin extract.A total of 47 sanguisorbins were detected when SWATH-MS was used under rolling CE and flexible Q1 isolation window modes.Besides,26 metabolites of sanguisorbins were identified in rat plasma,and their metabolic pathways could be deduced as decarbonylation,oxidization,reduction,methylation,and glucuronidation according to their fragmental ions acquired in SWATH-MS mode.Thus,SWATH-MS data acquisition could provide more comprehensive information for the component and metabolite identification for TCMs than IDA-MS.Dian Kang Qingqing Ding Yangfan Xu Xiaoxi Yin Huimin Guo Tengjie Yu He Wang Wenshuo Xu Guangji Wang Yan Liang 2020Journal of Pharmaceutical Analysis2020,10,6:1

17Chrysophanol localizes in mitochondria to promote cell death through upregulation of mitochondrial cyclophilin D in HepG2 cells显示文摘Objective:Chrysophanol(Chry) displays potent anticancer activity in human cancer cells and animal models,but the cellular targets of Chry have not been fully defined.Herein,we speculated whether mitochondria were a target involved in Chry-induced cytotoxicity.Methods:Human liver cancer cell line HepG2 was incubated.The cytotoxicity was evaluated by MTT assay.Mitochondria localization was evaluated by a confocal microscopy.Mitochondrial membrane potential ΔΨm was detected by TMRE staining and determined by the flow cytometer.The levels of ATP,mitochondrial superoxide anions,and GSH/GSSG were determined according to the assay kits.The apoptosis were evaluated through Hoechst33342/PI and Annexin V/PI staining,respectively.The expression of cyclophilin D(CyPD) was determined by immunoblot method,and the interaction between CyPD and Chry was analyzed by molecule docking procedure.Results:Chry itself mainly localized in mitocho ndria to cause mitochondrial dysfunction and cell death in HepG2 cells.As regard to the mechanism,cyclosporin A as the inhibitor for the formation of mitochondrial permeability transition pore(mPTP) moderately suppressed cell death,indicating mPTP involved in the process of cell death.Further,Chry enhanced the protein expression of Cyclophilin D(CyPD) which is a molecular componentry and a modulator of mPTP,while antioxidant N-acetyl-L-cysteine inhibited the expression of CyPD.Molecule docking procedure disclosed two hydrogen-bonds existed in CyPD-Chry complex with-11.94 kal/mol of the binding affinity value.Besides,the mtDNA-deficient HepG_2-ρ0 cells were much resistant to Chry-induced cell death,indicating mtDNA at least partly participated in cell death.A combination of Chry and VP-16 produced the synergism effect toward cell viability andΔΨm,while Chry combined with Cis-Pt elicited the antagonism effect.Conclusion:Taken together,enrichment in mitochondria and actions on mPTP,CyPD and mtDNA provides an insight into the anticancer mechanism of Chry.The combination therapy for Chry with clinical drugs may deserve to further explore.Yu Xie Ling Zhang Yuan-yuan Li Dian He Li-fang Zheng 2021Chinese Herbal Medicines2021,13,2:1

20ON THE OPTIMIZATION OF VLSI ALLOCATION IN HIGH-LEVEL SYNTHESIS显示文摘Allocation is one of main tasks in the high-level synthesis. It includes module , functional unit allocation, storage allocation and interconnection allocation. This paper models the allocation problem as cluster analysis and applies a new algorithm, neighbor state transition (NST) algorithm, for cluster optimization. It is proved that the algorithm produces an asymptotically global optimal solution with the upper bound on the cost function (1 + O(1/n)2-ε)F*, When F' is the cost of the optimum solution, n is the problem size and e is a positive parameter arbitrarily close to zero.
