| 2 | Reducing Viral Inhibition of Host Cellular Apoptosis Strengthens the Immunogenicity and Protective Efficacy of an Attenuated HSV-1 Strain显示文摘Herpes simplex virus 1(HSV-1),a member of a herpesviruses,shows a high infectivity rate of 30%-60%in populations of various ages.Some herpes simplex(HSV)vaccine candidates evaluated during the past 20 years have not shown protective efficacy against viral infection.An improved understanding of the immune profile of infected individuals and the associated mechanism is needed.HSV uses an immune evasion strategy during viral replication,and various virus-encoded proteins,such as ICP47 and Vhs,participate in this process through limiting the ability of CD8+cytotoxic T lymphocytes to recognize target cells.Other proteins,e.g.,Us3 and Us5,also play a role in viral immune evasion via interfering with cellular apoptosis.In this work,to study the mechanism by which HSV-1 strain attenuation interferes with the viral immune evasion strategy,we constructed a mutant strain,M5,with deletions in the Us3 and Us5 genes.M5 was shown to induce higher neutralizing antibody titers and a stronger cellular immune response than our previously reported M3 strain,and to prevent virus infection more effectively than the M3 strain in an in vivo mouse challenge test. | Xingli Xu Yufeng He Shengtao Fan Min Feng Guorun Jiang Lichun Wang Ying Zhang Yun Liao Qihan Li | 2019 | Virologica Sinica2019,34,6: | 1 |

| 4 | Pathogenic analysis of coxsackievirus A10 in rhesus macaques显示文摘Coxsackievirus A10(CV-A10)is one of the etiological agents associated with hand,foot and mouth disease(HFMD)and also causes a variety of illnesses in humans,including pneumonia,and myocarditis.Different people,particularly young children,may have different immunological responses to infection.Current CV-A10 infection animal models provide only a rudimentary understanding of the pathogenesis and effects of this virus.The characteristics of CV-A10 infection,replication,and shedding in humans remain unknown.In this study,rhesus macaques were infected by CV-A10 via respiratory or digestive route to mimic the HFMD in humans.The clinical symptoms,viral shedding,inflammatory response and pathologic changes were investigated in acute infection(1–11 day post infection)and recovery period(12–180 day post infection).All infected rhesus macaques during acute infection showed obvious viremia and clinical symptoms which were comparable to those observed in humans.Substantial inflammatory pathological damages were observed in multi-organs,including the lung,heart,liver,and kidney.During the acute period,all rhesus macaques displayed clinical signs,viral shedding,normalization of serum cytokines,and increased serum neutralizing antibodies,whereas inflammatory factors caused some animals to develop severe hyperglycemia during the recovery period.In addition,there were no significant differences between respiratory and digestive tract infected animals.Overall,all data presented suggest that the rhesus macaques provide the first non-human primate animal model for investigating CV-A10 pathophysiology and assessing the development of potential human therapies. | Suqin Duan Fengmei Yang Yanyan Li Yuan Zhao Li Shi Meng Qin Quan Liu Weihua Jin Junbin Wang Lixiong Chen Wei Zhang Yongjie Li Ying Zhang Jingjing Zhang Shaohui Ma Zhanlong He Qihan Li | 2022 | Virologica Sinica2022,37,4: | 1 |
| 5 | Targeting LncRNA LLNLR-299G3.1 with antisense oligonucleotide inhibits malignancy of esophageal squamous cell carcinoma cells in vitro and in vivo显示文摘Accumulating evidence has indicated that long non-coding RNAs(lncRNAs)play critical roles in the development and progression of cancers,including esophageal squamous cell carcinoma(ESCC).However,the mechanisms of lncRNAs in ESCC are still incompletely understood and therapeutic attempts for in vivo targeting cancer-associated lncRNA remain a challenge.By RNA-sequencing analysis,we identified that LLNLR-299G3.1 was a novel ESCC-associated lncRNA.LLNLR-299G3.1 was up-regulated in ESCC tissues and cells and promoted ESCC cell proliferation and invasion.Silencing of LLNLR-299G3.1 with ASO(antisense oligonucleotide)resulted in opposite effects.Mechanistically,LLNLR-299G3.1 bound to cancerassociated RNA binding proteins and regulated the expression of cancer-related genes,including OSM,TNFRSF4,HRH3,and SSTR3.ChIRP-seq(chromatin isolation by RNA purification and sequencing)revealed that these genes contained enriched chromatin binding sites for LLNLR-299G3.1.Rescue experiments confirmed that the effects of LLNLR-299G3.1 on ESCC cell proliferation were dependent on interaction with HRH3 and TNFRSF4.Therapeutically,intravenous delivery of placental chondroitin sulfate A binding peptide-coated nanoparticles containing antisense oligonucleotide(pICSA-BP-ANPs)strongly inhibited ESCC tumor growth and significantly improved animal survival in vivo.Overall,our results suggest that LLNLR-299G3.1 promotes ESCC malignancy through regulating gene-chromatin interactions and targeting ESCC by pICSA-BP-ANPs may be an effective strategy for the treatment of lncRNA-associated ESCC. | LI TIAN YONGYI HUANG BAOZHEN ZHANG YI SONG LIN YANG QIANQIAN CHEN ZHENG WANG YILING WANG QIHAN HE WENHAN YANG SHUYONG YU TIANYU LU ZICHEN LIU KAIPING GAO XIUJUN FAN JIAN SONG RIHONG ZHAI | 2023 | Oncology Research2023,31,4: | 0 |
