3Four Closely-related RING-type E3 Ligases,APD1-4,are Involved in Pollen Mitosis II Regulation in Arabidopsis显示文摘Ubiquitination of proteins is one of the critical regulatory mechanisms in eukaryotes.In higher plants,protein ubiquitination plays an essential role in many biological processes,including hormone signaling,photomorphogenesis,and pathogen defense.However,the roles of protein ubiquitination in the repro-ductive process are not clear.In this study,we identified four plant-specific RING-finger genes designated Aberrant Pollen Development 1 (APD1) to APD4,as regulators of pollen mitosis Ⅱ (PMⅡ) in Arabidopsis thaliana (L.).The apd1 apd2 double mutant showed a significantly increased percentage of bicellular-like pollen at the mature pollen stage.Further downregulation of the APD3 and APD4 transcripts in apd1 apd2 by RNA interference (RNAi) resulted in more severe abnormal bicellular-like pollen phenotypes than in apd1 apd2,suggesting that cell division was defective in male gametogenesis.All of the four genes were expressed in multiple stages at different levels during male gametophyte development.Confocal analysis using green florescence fusion proteins (GFP) GFP-APD1 and GFP-APD2 showed that APDs are associated with intracellular membranes.Furthermore,APD2 had E2-dependent E3 ligase activity in vitro,and five APD2-interacting proteins were identified.Our results suggest that these four genes may be involved,redundantly,in regulating the PMⅡ process during male gametogenesis.Guo Luo Hongya Gu Jingjing Liu Li-Jia Qu 2012Journal of Integrative Plant Biology2012,54,10:8
4Magnetic properties and large magnetocaloric effects in amorphous Gd-Al-Fe alloys for magnetic refrigeration显示文摘A series of the amorphous Gd75-55Al25-5Fe0-40 alloy ribbons were prepared by melt spinning.The structure,magnetic properties and magnetocaloric effect(MCE) of these alloys were investigated.The prepared samples have shown the characteristics of a second-order phase transition with zero hysteresis loss and the Tc can be tuned by changing the Fe contents.For the different compositions,the magnetic entropy change(--Sm) for a field change of 0-5 T reached a maximum value of 7.14 J kg-1 K-1 in the Gd70Al20Fe10 alloy near its Curie temperature(Tc) of 149 K.The non-linear composition dependence of(-ΔSm) could be caused by the competitions between Fe-Fe,Fe-Gd and Gd-Gd interactions.The refrigeration capacity(RC) values of these alloys are about 532-780 J/kg under a magnetic field change of 0-5 T.The results indicate that amorphous GdFeAl alloys can be considered as ideal candidates for a magnetic refrigerant in the temperature range of 104-222 K.ZHENG ZhiGang ZHONG XiChun SU KunPeng YU HongYa LIU ZhongWu ZENG DeCheng 2011Science China(Physics,Mechanics & Astronomy)2011,54,7:8
5The Arabidopsis Anaphase-Promoting Complex/Cyclosome Subunit 1 is Critical for Both Female Gametogenesis and Embryogenesis显示文摘Anaphase-promoting complex/cyclosome (APC/C), a multisubunit E3 ligase, plays a critical role in cell cycle control, but the functional characterization of each subunit has not yet been completed. To investigate the function of APC1 in Arabidopsis, we analyzed four mutant alleles of APC1, and found that mutation in APC1 resulted in significantly reduced plant fertility, accumulation of cyclin B, and disrupted auxin distribution in embryos. The three mutant alleles apc1-1, apc1-2 and apc1-3 shared variable defects in female gametogenesis including degradation, abnormal nuclear number, and disrupted polarity of nuclei in the embryo sac as well as in embryogenesis, in which embryos were arrested at multiple stages. All of these defects are similar to those previously identified in apc4. The mutant apc1-4, in which the T-DNA was inserted after the transmembrane domain at the C-terminus, showed much more severe phenotypes; that is, most of the ovules were arrested at the one-nucleate female gametophyte stage (stage FG1). In the apc1 apc4 double mutants, the fertility was further reduced by one-third in apc1-1/+ apc4-1/+, and in some cases no ovules even survived in siliques of apc1-4/+ apc4-1/+. 7Thermal Growth and Nanomagnetism of the Quasi-one Dimensional Iron Oxide显示文摘Quasi-one dimensional iron oxide nanowires with flat needle shape were synthesized on the iron powders by a rather simple catalyst-free thermal oxidation process in ambient atmosphere. The characterization by field emission scanning electron microscopy (FE-SEM), X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD), Raman and high-resolution transmission electron microscopy (HRTEM) revealed that these nanostructures are single crystalline α-Fe2O3. The various dimensions with 40-170 nm in width and 1-8 μm in length were obtained by tuning the growth temperature from 280 to 480℃. A surface diffusion mechanism was proposed to account for the growth of quasi-one dimensional nanostructure. The typical α-Fe2O3 nanowires synthesized at 430℃ had a reduced Morin temperature TM of 131 K in comparison with their bulk counterpart. The coercivitis Hc of these nanowires are 321 and 65 Oe at 5 and 300 K, respectively. 8Construction of chimeric inducible promoters by elicitors of rice fungal blast pathogen and their expression in transgenic rice显示文摘The promoter fragments of wheat GstA1 and potato Gst1 have been amplified by PCR, cloned and fused respectively to the minimal promoter sequence of rice actin gene (Act1)) and its 5’ untranslated leader sequence together with GUS. The constructs with 2 chimeric promoters (WGA and PGA) have been transferred into rice in order to analyze their inducibility patterns in transgenic rice plants. 9OPEN ARCHITECTURE CNC SYSTEM HITCNC AND KEY TECHNOLOGY显示文摘Aiming at the characteristics of modularity and reconfigurable in open architecture com-puter numerical control (CNC) system,the open architecture CNC system,Harbin Institute of Tech-nology computer numerical control (HITCNC),is researched and manufactured based on the interface standards. The system’s external interfaces are coincident with the corresponding international stan-dards,and the internal interfaces follow the open modular architecture controller (OMAC) agreement. In the research and manufacturing process,object-oriented technology is used to ensure the openness of the HITCNC,and static programming is applied in the CNC system according to the idea of modu-larization disassembly. The HITCNC also actualizes real-time and unreal-time modules adopting real-time dynamical linked library (RTDLL) and component object model (COM). Finite state ma-chine (FSM) is adopted to do dynamically modeling of HITCNC. The complete separation between the software and the hardware is achieved in the HITCNC by applying the SoftSERCANS technique. The application of the above key techniques decreases the programming workload greatly,and uses software programs replacing hardware functions,which offers plenty technique ensures for the open-ness of HITCNC. Finally,based on the HITCNC,a three-dimensional milling system is established. On the system,series experiments are done to validate the expandability and interchangeability of HITCNC. 10Microstructure improvement related coercivity enhancement for sintered NdFeB magnets after optimized additional heat treatment显示文摘The micro structure, especially the Nd-rich phase and the grain boundary, in sintered NdFeB magnets plays an important role in magnetic reversal and coercivity mechanism. To better understand the effects of the microstructure on the coercivity, we investigated the microstructure and properties improvements of a commercial sintered NdFeB magnet after optimized additional heat treatment. The coercivity is enhanced from 1399 to 1560 kA/m. This enhancement has been explained in terms of the evolution of the grain boundary structure, and the formation of continuous thin layers of Nd-rich phase is important for high coercivity. The micromagnetic simulation together with the numerical analysis based on the nucleation model suggest that the reversed magnetic domains nucleate mainly at the interface of multijunctions of Nd_2 Fe_(14)B grains with high stray fields during the demagnetization process. Both improved anisotropy fields at grain boundaries and reduced stray fields at multi-junction Nd-rich phases contribute to the coercivity enhancement. 12SNAIL1 is essential for female gametogenesis in Arabidopsis thaliana显示文摘Two yeast Brix family members Ssf1 and Ssf2,involved in large ribosomal subunit synthesis, are essential for yeast cell viability and mating efficiency. Their putative homologs exist in the Arabidopsis genome; however, their role in plant development is unknown. Here, we show that Arabidopsis thaliana SNAIL1(At SNAIL1), a protein sharing high sequence identity with yeast Ssf1 and Ssf2, is critical to mitosis progression of female gametophyte development.The snail1 homozygous mutant was nonviable and its heterozygous mutant was semi-sterile with shorter siliques.The mutation in SNAIL1 led to absence of female transmission and reduced male transmission. Further phenotypic analysis showed that the synchronic development of female gametophyte in the snail1 heterozygous mutant was greatly impaired and the snail1 pollen tube growth, in vivo, was also compromised. 13Magnetic properties and magnetocaloric effects in amorphous and crystalline Gd_(55)Co_(35)Ni_(10) ribbons显示文摘The magnetic properties and magnetocaloric effects of amorphous and crystalline Gd55Co35Ni10 ribbons are investigated.A main phase with a Ho 12 Co 7-type monoclinic structure(space group P21/c) and a minor phase with a Ho4Co3-type hexagonal structure(space group P63/m) are obtained for crystalline ribbon after annealing.The amorphous ribbons order ferromagnetically and undergo a second-order transition at 192 K.For crystalline Gd55Co35Ni10 ribbons,two magnetic phase transitions occur at 158 and 214 K,respectively.The peak value of-△SM under a field change of 0-5 T is 6.5 J/kg K at 192 K for amorphous Gd55Co35Ni10 ribbons.A relatively large magnetic entropy change(~5.0 J/kg K) under a field change of 0-5 T for the crystalline Gd55Co35Ni10 ribbons is obtained in the temperature interval range of 154-214 K.The large platform of magnetic entropy change and the negligible thermal/magnetic hysteresis loss mean the crystalline Gd55Co35Ni10 compound can satisfy the requirement of the Ericsson-type refrigerator working in the temperature range from 154K to 214K.ZHONG XiChun TANG PengFei GAO BeiBei MIN JiXiong LIU ZhongWu ZHENG ZhiGang ZENG DeChang YU HongYa QIU WanQi 2013Science China(Physics,Mechanics & Astronomy)2013,56,6:3
14Allogeneic hematopoietic stem cell transplantation for patients with acute leukemia显示文摘Objective: The purposes of this study were to assess the efficacy of allogeneic hematopoietic stem cell transplantation (HSCT) for acute leukemia (AL) and analyze the factors affecting the prognosis of these patients. Methods: The clinical and follow-up data of 93 AL patients (median age, 30 years) undergoing allogeneic HSCT in Xiangya Hospital over the past 12 years were collected, and the potential factors affecting the efficacy and prognosis of allogeneic HSCT patients were determined. Results: Hematopoietic reconstitution was achieved in 90 patients. At the last follow-up, the incidences of severe acute graft versus host disease (aGvHD) and extensive chronic GvHD (cGvHD) were 14.0% and 20.0%, the 3-year cumulative incidence of transplantation related mortality (TRM) and relapse rate were 16.8%±6.1% and 21.3%±6.7%, and the estimated 3-year overall survival (OS) and disease-free survival (DFS) of the patients were 64.6%±5.4% and 56.5%±5.5%, respectively. Univariate analysis indicated that age older than 40 years, HLA mismatch, and severe lung infection within the first 100 days after transplantation were risk factors for severe aGvHD, age older than 40 years, HLA mismatch, severe lung infection within the first 100 days after transplantation, and severe aGvHD were risk factors for TRM, high-risk AL and lack of cGvHD were risk factors for relapse (all P<0.05). Survival estimation showed that HLA mismatch, severe lung infection occurring within the first 100 days post-transplantation, high-risk AL severe aGvHD and lack of cGvHD were risk factors associated with poor prognosis (all P<0.05). Further multivariate analyses revealed that severe lung infection within the first 100 days post-transplantation, severe aGvHD and lack of cGvHD were independent risk factors for unfavorable outcomes (all P<0.05). 16Structure and magneto-electrical properties of Fe-C films prepared by magnetron sputtering显示文摘Fe-doped amorphous Fe x C 1 x granular films were prepared on n-Si(100) substrates by d.c.magnetron sputtering.The structural properties of Fe x C 1 x films were investigated by X-ray diffraction(XRD),atomic force microscope(AFM) and Raman spectroscopy.The results show that the iron and carbon of as-deposited films are in amorphous state,and the Fe x C 1 x films are diamond-like carbon(DLC) films.After doping iron into the DLC films,a smooth surface morphology of the Fe x C 1 x films has been obtained with the surface roughness Ra of about 0.231 nm for x=18at%.The Fe x C 1 x films have good soft magnetic properties with the coercivity of approximately 20 Oe.A high positive magnetoresistance(MR) up to 93% with x=1at% was observed in a Fe x C 1 x granular film at 300 K.The resistance characteristic of Fe-C films is changed at about 230 K and the positive MR effect can be understood by the p-n heterojunction theory.MA Lei LIU ZhongWu ZENG DeChang YU HongYa ZHONG XiaPing ZHANG XiaoZhong 2012Science China(Physics,Mechanics & Astronomy)2012,55,9:3
17High-Q silica microdisk optical resonators with large wedge angles on a silicon chip显示文摘We experimentally demonstrate high optical quality factor silica microdisk resonators on a silicon chip with large wedge angles by reactive ion etching. For 2-μm-thick microresonators, we have achieved wedge angles of 59°, 63°,70°, and 79° with optical quality factors of 2.4 × 10~7, 8.1 × 10~6, 5.9 × 10~6, and 7.4 × 10~6, respectively, from ~80 μm diameter microresonators in the 1550 nm wavelength band. 18Structural and expressional analysis of a cDNA that expresses predominantly in rice stamens显示文摘The development of stamen is an important stage during the sexual reproduction in higher plants. Studying the genes that predominantly or even specifically expressed in stamens will aid in understanding the molecular mechanism of stamen development. The structure of a rice CHS-like gene (D5) which predominantly expressed in anthers has been analysized, showing high structural similarity with two other anther-specific genes (A1 and BA42) from Brassica napus. Northern analysis also reveals that the three genes share a similar expression pattern and that the expression of D5 may relate to the early development of pollens. 19An anther-specific chalcone synthase-like gene D5 related to rice pollen development显示文摘It was shown in a previous analysis that D5 gene from rice (Oryza sativa L.) was an anther-specific gene encoding a chalcone synthase-related protein. In this study, D5 gene was found specifically expressed in tapetum cells as well as in the peripheral cells of the vascular bundle of rice anthers by RNA in situ hybridization. In order to study its function, D5 was transformed into rice in both sense and antisense directions driven by a rice Actin 1 promoter. 20Rock crevices determine woody and herbaceous plant cover in the karst critical zone显示文摘The study of the critical zones(CZs) of the Earth link the composition and function of aboveground vegetation with the characteristics of the rock layers, providing a new way to study how the unique rock and soil conditions in karst regions affect the aboveground vegetation. Based on survey results of the rocks, soils and vegetation in the dolomite and limestone distribution areas in the karst area of central Guizhou, it was found that woody plant cover increases linearly with the number of cracks with a width of more than 1 mm, while the cover of herbaceous plants shows the opposite trend(p<0.01). The dolomite distribution area is characterized by undeveloped crevices, and the thickness of the soil layer is generally less than 20 cm, which is suitable for the distribution of herbaceous plants with shallow roots. Due to the development of crevices in the limestone distribution area, the soil is deeply distributed through the crevices for the deep roots of trees, which leads to a diversified species composition and a complicated structure in the aboveground vegetation. Based on moderate resolution imaging spectroradiometer(MODIS) remote sensing data from 2001 to 2010, the normalized differentiated vegetation index(NDVI) and annual net primary productivity(NPP) results for each phase of a 16-day interval further indicate that the NDVI of the limestone distribution area is significantly higher than that in the dolomite distribution area, but the average annual NPP is the opposite. The results of this paper indicate that in karst CZs, the lithology determines the structure and distribution of the soil, which further determines the cover of woody and herbaceous plants in the aboveground vegetation. Although the amount of soil in the limestone area may be less than that in the dolomite area, the developed crevice structure is more suitable for the growth of trees with deep roots, and the vegetation activity is strong.
