1Rapid,specific and sensitive detection of Vibrio vulnificus by loop-mediated isothermal amplification targeted to vvhA gene显示文摘Vibrio vulnificus is an estuarine bacterial pathogen for human.The rapid,specific and sensitive detection of V.vulnificus is urgently needed for early disease diagnosis and timely treatment of V.vulnificus infection.In the study,a loop-mediated isothermal amplification(LAMP) technique was developed for V.vulnificus detection with a set of primers,composed of two out primers and two inner primers targeted to vvh A gene.The optimal amplification temperature was 63°C and the reaction only took 35 min.The amplification products could not only be detected by agarose gel electrophoresis with ladder-like pattern bands,but also could be visualized using calcein with naked eye directly.Forty-five strains were tested for the specificity of LAMP assay,and all the V.vulnificus strains were identified correctly while other strains were negative results.The sensitive of the new LAMP assay was 100-fold more sensitive than the conventional PCR.Meanwhile,all the V.vulnificus strains were detected correctly in spiked,clinical and environmental samples by the new LAMP assay.Compared with other well-known techniques,the new LAMP assay targeted to vvh A gene was extremely rapid,simple,sensitive and specific for V.vulnificus identification.ZHANG Lina WANG Mingyi CONG Dianxia DING Shuyan CONG Rinan YUE Jinyong GENG Jianli HU Chengjin 2018Acta Oceanologica Sinica2018,37,4:1
2Data-driven Reliability Assessment of an Electric Vehicle Penetrated Grid Utilizing the Diffusion Estimator and Slice Sampling显示文摘Due to the stochasticity of charging behaviors of electric vehicles(EVs),it is difficult to anticipate when charging load demand will be densely concentrated.If massive charging loads and the system peak profile appear at the same time,it may pose a risk to the reliable operation of power grids.For a system integrated with renewable energies,this risk can be much higher because of their unsteady power output.With load measurements more widely collected,this paper presents a data-driven framework to assess the reliability of a power grid considering charging EVs.Specifically,the diffusion estimator is firstly applied to estimate the probability density function of EV charging loads,which possesses both regional adaptivity and good boundary estimation performance.Then,charging load samples are produced through slice sampling.It is capable of sampling from irregularly-shaped distributions with high accuracy.The proposed approach is verified by the numerical results from the simulations on a modified IEEE 30-bus test system based on real measurement data.Songyu Huang Chengjin Ye Si Liu Wei Zhang Yi Ding Ruoyun Hu Jianbai Li 2023CSEE Journal of Power and Energy Systems2023,9,5:0
3Extension and application of a database for the rapid identification of Vibrio using MALDI-TOF MS显示文摘Rapid and accurate identification of Vibrio species has been problematic because phenotypic characteristics are variable within species and biochemical identification requires two or more days to complete.Matrix-assisted laser desorption ionization–time of flight mass spectrometry(MALDI-TOF MS)has become a powerful tool for rapidly distinguishing between related bacterial species.However,its accuracy depends on the number of strains in a database.In the current study,we extend and apply the Vibrio database based on MALDI-TOF MS.A total of 74 strains of Vibrio representing 28 species were identified and included in new database.A phylogenetic tree based on rpoB sequence and dendrograms were constructed.We analyzed 30 clinical Vibrio of three species to evaluate the new database and carried out PCA dendrogram analyses for differences of strains.We created a new database that offered fast and accurate Vibrio identification.MSP and PCA dendrogram analyses provided technical support to track sources and incidences of Vibrio infection.In addition,the discovery of characteristic and differential peaks is useful for the future identification of Vibrio.This represents a powerful tool for the rapid and accurate classification and identification of Vibrio and closely related species.Jing Wu Yuexia Zhou Xiaofei Liu Yuan Cao Chengjin Hu Yingjian Chen 2020Acta Oceanologica Sinica2020,39,10:0
4人组织型激肽释放酶6在卵巢上皮性肿瘤组织中的表达及其与临床病理特征和预后的关系(英文)显示文摘Objective: The aim of this study was to approach the relationship between the expression of human kallikrein 6 (hK6) in ovarian neoplasm and the clinicopathologic variables and prognosis for finding a new tumor marker for ovarian cancer. Methods: Through immunohistochemistry to examine the expression of hK6 in 19 cases with benign, 11 cases with borderline and 45 cases with malignant ovarian neoplasms and statistically analyzed whether the expression of hK6 correlated with the clinicopathologic variables and prognosis in patients with ovarian cancer. Results: The positive rate of hK6 in ovarian cancer tissues (60.0%) was significantly higher than that in the benign (15.8%) and borderline (27.3%) ovarian neoplasm tissues (P < 0.01). The expression of hK6 in low-grade ovarian cancer tissues was higher than that in high-grade (68.4% vs 14.3%; P < 0.05); hK6 in late–stage (stage III) was more frequently expressed than that in early-stage (stage I or II) (76.7% vs 26.7%; P < 0.01); and it significantly higher in ovarian carcinomas with lymph node metastases than those without lymph node metastases (77.8% vs 33.3%; P < 0.01 ); moreover, the expression of hK6 in the cancer tissues that the patients died or their pathogenetic condition recurred or their tumor metastasized within 3 years after surgery was higher (75.0%) than that in the cancer tissues that the pathogenetic condition of the patients was stable (42.9%; P < 0.05). Conclusion: The expression of hK6 in ovarian cancer tissues was higher than that in the benign and borderline ovarian neoplasm tissues, and high hK6 expression correlated with late–stage, low-grade, node metastasis and poor prognosis of patients. hK6 could potentially be a novel tumor marker for ovarian cancer that can predict the prognosis of the patients.Chengjin Hu Fang Zhang Kuixiang Chen Yingjian Chen 2010The Chinese-German Journal of Clinical Oncology2010,9,1:0
