| 1 | Effect of ginseng polysaccharides and dendritic cells on the balance of Th1/Th2 T helper cells in patients with non-small cell lung cancer显示文摘OBJECTIVE: To investigate the effect of thorascopic administration of ginseng polysaccharides(GPS)plus dendritic cells(DC) on T helper cell type 1/T helper cell type 2(Th1/Th2) balance in patients with non-small cell lung cancer(NSCLC).METHODS: A total of 96 NSCLC patients were divided evenly into two groups. The control group was treated with DCs alone and the treatment group was treated with DCs plus GPS. After DCs and GPS were administered thoracoscopically, once a week,4 times for 30 days, the patients' quality of life was measured with the Functional Assessment of Can-cer Treatment-Lung(FACT-L) questionnaire before and after treatment. Serum interferon-γ(INF-γ), interleukin-4(IL-4), IL-2 and IL-5 were examined before and after treatments.RESULTS: The level of Th1 cytokines(INF-γ, IL-2)and the ratio of Th1/Th2 cytokines(INF-γ/IL-4, IL-2/IL-5) increased in both treatment groups, while Th2cytokines(IL-4, IL-5) and FACT-L scores decreased(P<0.01). Furthermore, after treatment Th1 cytokines(INF-γ, IL-2) and the ratio of Th1/Th2 cytokines(INF-γ/IL-4, IL-2/IL-5) were higher in the DCs +GPS group than in the control group(P<0.05). Conversely, FACT-L scores and Th2 cytokines(IL-4, IL-5)were higher in the control group than in the DCs +GPS group(P<0.05).CONCLUSION: The treatment regime of DCs plus GPS had a greater effect on NSCLC patients' immune function as compared with DCs alone. | 3 | miR-30a-5p promotes glomerular podocyte apoptosis via DNMT1-mediated hypermethylation under hyperhomocysteinemia显示文摘Abnormal elevation of homocysteine(Hcy)level is closely related to the development and progression of chronic kidney disease(CKD),with the molecular mech2anisms that are not fully elucidated.Given the demonstration that miR-30a-5p is specifically expressed in glomerular podocytes,in the present study we aimed to investigate the role and potential underlying mechanism of miR-30a-5p in glomerular podocyte apoptosis induced by Hcy.We found that elevated Hcy downregulates miR-30a-5p expression in the Cbs^(+/-)-mice and Hcy-treated podocytes,and miR-30a-5p directly targets the 3′-untranslated region(3′-UTR)of the forkhead box A1(FOXA1)and overexpression of miR-30a-5p inhibits FOXA1 expression.By nMS-PCR and MassARRAY quantitative methylation analysis,we showed the increased DNA methylation level of miR-30a-5p promoter both in vivo and in vitro.Meanwhile,dual-luciferase reporter assay showed that the region between--1400 and--921 bp of miR-30a-5p promoter is a possible regulatory element for its transcription.Mechanistic studies indicated that DNA methyltransferase enzyme 1(DNMT1)is the key regulator of miR-30a-5p,which in turn enhances miR-30a-5p promoter methylation level and thereby inhibits its expression.Taken together,our results revealed that epigenetic modification of miR-30a-5p is involved in glomerular podocyte injury induced by Hcy,providing a diagnostic marker candidate and novel therapeutic target in CKD induced by Hcy. | Ning Ding Lin Xie Fei Ma Shengchao Ma Jiantuan Xiong Guanjun Lu Huiping Zhang Yideng Jiang | 2022 | Acta Biochimica et Biophysica Sinica2022，54，1： | 4 |
| 4 | Application of isotopic and hydro-geochemical methods in identifying sources of mine inrushing water显示文摘Isotopic and hydro-geochemical surveys were carried out to identify the source of mine inrushing water at the #73003 face in the Laohutai Mine. Based on the analysis of isotopes and hydro-chemical features of surface water, groundwater from different levels and the inrushing water, a special relationship between water at the #73003 face and cretaceous water has been found. The results show that the isotopic and hydro-chemical features of the inrushing water are completely different from those of other groundwater bodies, except for the cretaceous water. The isotopic and hydrochemical characteristics of cretaceous water are similar to the inrushing water of the #73003 face, which aided with obtaining the evidence for the possible source of the inrushing water at the #73003 face. The isotope calculations show that the inrushing water at the #73003 face is a mixture of cretaceous water and Quaternary water. Water from the cretaceous conglomerate is the main source, accounting for 67% of the inrushing water, while the Quaternary water accounts for 33%. | 5 | Ataxia,acute mountain sickness,and high altitude cerebral edema显示文摘Previous investigations suggest that ataxia is common and often one of the most reliable warning signs of high altitude cerebral edema(HACE). The aim of this study was to investigate the diagnostic role of ataxia in acute mountain sickness(AMS)and HACE among mountain rescuers on the quake areas,and in approaching the relation between AMS and HACE. After the earthquake on April 14,2010,approximately 24 080 lowland rescuers were rapidly transported from sea level or lowlands to the mountainous rescue sites at 3 750 ~ 4 568 m,and extremely hardly worked for an emergency treatment after arrival. Assessments of acute altitude illness on the quake areas were using the Lake Louise Scoring System. 73 % of the rescuers were found to be developed AMS. The incidence of high altitude pulmonary edema(HAPE)and HACE was 0.73 % and 0.26 %,respectively,on the second to third day at altitude. Ataxia sign was measured by simple tests of coordination including a modified Romberg test. The clinical features of 62 patients with HACE were analyzed. It was found that the most frequent,serious neurological symptoms and signs were altered mental status(50/62,80.6 %)and truncal ataxia(47/62,75.8 %). Mental status change was rated slightly higher than ataxia,but ataxia occurred earlier than mental status change and other symptoms. The earliest sign of ataxia was a vague unsteadiness of gait,which may be present alone in association with or without AMS. Advanced ataxia was correlated with the AMS scores,but mild ataxia did not correlate with AMS scores at altitudes of 3 750~4 568 m. Of them,14 patients were further examined by computerized tomographic scanning of the brain and cerebral magnetic resonance imagines were examined in another 15 cases. These imaging studies indicated that the presence of the cerebral edema was in 97 % of cases who were clinically diagnosed as HACE(28/29). | 7 | Investigation of ion-atom collision dynamics through imaging techniques显示文摘The principle and technique details of recoil ion momentum imaging are discussed and summarized. The recoil ion momentum spectroscopy built at the Institute of Modern Physics (Lanzhou) is presented. The first results obtained at the setup are analyzed. For 30 keV He2+ on He collision, it is found that the capture of single electron occurs dominantly into the first excited states, and the related scattering angle results show that the ground state capture occurs at large impact parameters, while the capture into excited states occurs at small impact parameters. The results manifest the collision dynamics for the sub-femto-second process can be studied through the techniques uniquely. | 8 | Tensile behavior of AA3104 aluminum sheets at low deformation degree显示文摘The texture of a rolled AA3104 aluminum sheet was measured by the X-ray transmission method. The Lankford values or r values (ratio of plastic strain) and yield strengths in directions of 0, 15, 30, 45, 60, 75, and 90° to RD (rolling direction) of the sheet were tested during tensile loading at a strain of 2%. r values were predicted by the Sachs model and the reaction stress model in con- sideration of the measured texture. The simulated results indicate that r values calculated by the Sachs model are more exactly ap- proaching with the experimental values on the whole than those predicted by the reaction stress model. | 10 | Studies on monitoring hemodynamics and oxygen dynamics of adult respiratory distress syndrome secondary to high altitude pulmonary edema显示文摘To study monitoring hemodynamics and oxygen dynamics of adult respiratory distress syndrome (ARDS) secondary to high altitude pulmonary edema (HAPE),we performed clinic and laboratory studies in 8 patients who preliminarily developed high altitude cerebral edema (HACE) and then ARDS occurred at an altitude of 4 500 m. After an initial emergency treatment on high mountains,all the patients were rapidly transported to a hospital at a lower altitude of 2 808 m. The right cardiac catheterizations were carried out within 5 h after hospitalized. The monitoring hemodynamics and oxygen dynamics were studied via a thermodilution Swan-Gaze catheter. The results showed that before treatments at the beginning of monitoring,there presented a significant pulmonary artery hypertension with a decreased cardiac function,and a lower oxygen metabolism in all the 8 patients. However,after some effective treatments,including mechanical ventilation and using dexamethasone,furosemide,etc,four days later the result of a repeated monitoring showed that their pulmonary artery pressure had been decreased with an improved cardiac function with all the oxygen metabolic indexes increased significantly. | 13 | Extracellular-superoxide dismutase DNA methylation promotes oxidative stress in homocysteine-induced atherosclerosis显示文摘In the present study,we investigate the effect of homocysteine(Hcy)on extracellular-superoxide dismutase(ECSOD)DNA methylation in the aorta of mice,and explore the underlying mechanism in macrophages,trying to identify the key targets of Hcy-induced EC-SOD methylation changes.ApoE--mice are fed different diets for 15 weeks,EC-SODand DNAmethyltransferase1(DNMT1)expression levels aredetected byRT-PCRandwestern blot analysis.EC-SOD methylation levels are assessed by ntMS-PCR.After EC-SOD overexpression or knockdown in macrophages,following the transfection of macrophages with pEGFP-N1-DNMT1,the methylation levels of ECSOD are detected.Our data show that the concentrations of Hcy and the area of atherogenic lesions are significantly increased in ApoE--mice fed with a high-methionine diet,and have a positive correlation with the levels of superoxide anions,which indicates that Hcy-activated superoxide anions enhance the development of atherogenic lesions.EC-SOD expression is suppressed by Hcy,and the content of superoxide anion is increased when EC-SOD is silenced by RNAi in macrophages,suggesting that EC-SOD plays a major part in oxidative stress induced by Hcy.Furthermore,the promoter activity of EC-SOD is increased following transfection with the-1/-110o fragment,and EC-SOD methylation level is significantly suppressed by Hcy,and more significantly decreased upon DNMT1 overexpression.In conclusion,Hcy may alter the DNA methylation status and DNMT1 acts as the essential enzyme in the methyl transfer process to disturb the status of EC-SOD DNA methylation,leading to decreased expression of Ec-sODandincreasedoxidativestressandatherosclerosis. | Shengchao Ma Guanjun Lu Qing Zhang Ning Ding Yuzhen Jie Hui Zhang Lingbo Xu Lin Xie Xiaoling Yang Huiping Zhang Yideng Jiang | 2022 | Acta Biochimica et Biophysica Sinica2022，54，9： | 1 |
