| 1 | Expression of trefoil factors and TWIST1 in colorectal cancer and their correlation with metastatic potential and prognosis显示文摘AIM To detect the expression of trefoil factors(TFFs)and TWIST1 in colorectal cancer(CRC)and analyze their correlation with metastasis and survival.METHODS This study examined the expression of TFF1,TFF3 and TWIST1 in a total of 75 tumor samples,47 matched normal samples(15 cm from the lesion margin),30metastatic lymph nodes,and 10 liver metastatic cancer samples from patients with CRC.The relationship was then analyzed between the protein expression and different clinical records.TFF1,TFF3,TWIST1,E-cadherin,vimentin andβ-catenin m RNA and protein expression levels were measured in colon cancer cell lines with different metastatic potentials(HIEC,HT29,SW620,and Lo Vo cells),and the correlation of the expression levels with epithelial-mesenchymal transition(EMT)was discussed.RESULTS It was found that 66.7%(50/75),78.7%(59/75)and54.7%(41/75)of tumor tissue samples exhibited positive staining for TFF1,TFF3 and TWIST1 and so did 27.3%(13/47),100%(47/47)and 17%(8/47)of adjacent normal colorectal tissues.Compared with adjacent normal tissues,significant differences were found in the expression of all three proteins in different cancerous tissues(P<0.05).Higher expression of TFF3 and TWIST1 was significantly correlated with lymph node metastasis(P=0.034,P=0.000),advanced stage(P=0.031,P=0.003),and poorer survival(P=0.042 for the TFF3 group,P=0.003for the TWIST1 group).The expression of TFF3 and TWIST1 in cancer cell lines was higher than that in HIEC(a normal human intestinal epithelial cell line)(P<0.05),and the expression intensity demonstrated a tendency to rise with increased metastatic potential both at the protein and m RNA levels.However,TFF1expression demonstrated the opposite tendency.It was also observed that the expression of E-cadherin andβ-catenin tended to decrease while that of vimentin,TWIST1 and Snail tended to rise with the increase in metastatic potential.CONCLUSION The expression of TFF3 and TWIST1 might be associated with the survival of patients with CRC after curative resection and might be pivotal predictors of disease progression.TFF3 may be correlated to the invasiveness of CRC. | Akram Yusup Bailikezi Huji Cheng Fang Fei Wang Tuerxunjiang Dadihan Hai-Jiang Wang Halmurat Upur | 2017 | World Journal of Gastroenterology2017,23,1: | 7 |
| 2 | An magnetic resonance-based plasma metabonomic investigation on abnormal Savda in different complicated diseases显示文摘OBJECTIVE: To provide potential evidence for the existence of abnormal Savda, we assessed host metabonomic responses and dynamic changes occurring in various diseases using1H nuclear magnetic resonance(NMR)-based metabonomics.METHODS: Plasma samples taken from patients with complicated diseases with abnormal Savda(n=140, including 35 cases each of diabetes, asthma,breast cancer, and cervical carcinoma) and from healthy controls(n=35) were analyzed by1H NMR(600 MHz), and the spectral profiles were analyzed by multivariate analysis using orthogonal projection to latent structure with discriminant analysis.RESULTS: Supervised modeling of the data provided very good discrimination between patients and healthy controls. Compared with the healthy controls, the patient groups with different disease conditions displayed similar metabolic changes, characterized by lower creatine, creatinine, lactate, and amino acid levels(including isoleucine, leucine, valine, alanine, and 1-methylhistidine) and higher lipid levels(very low-density lipoproteins and unsaturated lipids). | 8 | Expression of airway mucus-associated proteins in rats with chronic obstructive pulmonary disease with a cold-dryness symptom pattern显示文摘OBJECTIVE: To reveal the effects on expression of airway mucus-associated proteins in rats with chronic obstructive pulmonary disease(COPD) and a cold-dryness symptom pattern induced by elastase and smoking.METHODS: The COPD model was established with an elastase dose into the trachea combined with exposure to smoking; the COPD model cold-dryness symptom pattern was further developed by exposure to a cold, dry environment. After 90 days,pathologic lung sections, inflammatory cytokine levels(measured by enzyme linked immunosorbent assay), m RNA and protein expression of mucus-associated proteins and aquaporins(measured by real-time polymerase chain reaction and western blots) were examined.RESULTS: Cytokines interleukin-6(IL-6), interleukin-8(IL-8), and tumor necrosis factor-α(TNF-α) in the COPD and the cold-dryness symptom pattern COPD groups were all significantly higher than in controls(each P < 0.01). IL-6 and IL-8 levels were higher in the cold-dryness symptom pattern COPD group than in the COPD group(each P < 0.05). The AQP5 m RNA expression in the cold-dryness symptom pattern COPD and COPD groups was lower than in the control group(P < 0.01), and that in the cold-dryness symptom pattern COPD group was lower than the COPD group(P < 0.05). The expression of MUC5 AC and MUC5 B m RNAs in the cold-dryness symptom pattern COPD group and COPD group was higher than in the control group(each P < 0.01), and that in the cold-dryness symptom pattern COPD group was higher than the COPD group(P < 0.01, and P < 0.05, respectively).The ratio of MUC5 AC m RNA/MUC5 B m RNA was COPD group < the cold-dryness symptom pattern COPD group < the control group. AQP4 and AQP5 protein expression in the cold-dryness symptom pattern COPD group was lower than that in the COPD group which was lower again than in the control group. MUC5 AC and MUC5 B expression in the cold-dryness symptom pattern COPD group was higher than in the COPD group and higher again than in the control group.CONCLUSION: Cold-dryness affects the expression of mucus-associated protein m RNA and its corresponding proteins, reducing the secretion of aquaporins and increasing the secretion of mucins. | 10 | Plasma amino acid profiling of cancer patients with abnormal Savda based on high-performance liquid chromatography显示文摘OBJECTIVE: To investigate metabolic signatures in plasma of cancer patients with abnormal Savda using plasma-free amino acid profiles, and to evaluate the diagnostic potential of these profiles for the detection and explanation of the mechanisms of different symptoms in traditional Uyghur medicine.METHODS: Plasma samples from cancer patients with abnormal Savda(n=85) or non-abnormal Savda(n=105) and a healthy control group(n=65)were analyzed using high-performance liquid chromatography(HPLC). Orthogonal projection to latent structures with discriminant analysis was used for the classification and prediction of abnormal Savda, and spectral profiles were subjected to Student's t-tests to assess statistical significance.RESULTS: Compared with the healthy group, the levels of aspartic acid, glutamate, glycine, histidine,arginine, threonine, alanine, proline, methionine,isoleucine, leucine and phenylalanine decreased significantly in plasma of cancer patients with abnormal Savda(all P<0.05). Serine, cystine, tyrosine,valine and lysine levels showed no significant differences(all P>0.05). Compared with non-abnormal Savda syndrome patients, abnormal Savda syndrome patients showed high concentrations of glutamate, serine, valine, isoleucine, leucine and phenylalanine(all P<0.05). The remaining plasma amino acids showed no significant differences(all P>0.05).CONCLUSION: Plasma-free amino acid profiling has the potential to assist in understanding and determining abnormal Savda. Metagenomic studies demonstrated the presence of significant differences in the structure of the intestinal microbiota in populations of Europeans and Asians. In this aspect,the Xinjiang Uygur Autonomous region of China is the most affected district,where substantially the indigenous Uighur,Kazakh and also Han populations is affected. There is an objective need to ascertain the microecological and metabolic status of persons of the indigenous population in Western China,which are in the risk zone. The test for microalbuminuria is reliable method of kidney failure primary stages diagnosis,reflecting the initial stages of vessel pathology and the development of MS. It invariably correlates with an increase of cardiovascular morbidity and mortality. We introduce this screening test for early detection of predisposition to the development of the MS in the example of Xinjiang population for use also in other regions of China.There is a need to carry out the search,selection and analysis of the new strains of probiotics in the Xinjiang from national milk products with lactic acid or other fermented products with the purpose of studying their clinical characteristics. | 13 | Effect of cold-dryness on pulmonary and immunologic function in chronic obstructive pulmonary disease model rats显示文摘OBJECTIVE: To study the effects of cold-dryness on pulmonary and immunologic function of peripheral T-lymphocytes in chronic obstructive pulmonary disease(COPD) model rats, and to provide references for the prevention and treatment of cold-dryness COPD in the Xinjiang region.METHODS: The COPD model was established with an elastase drip into the trachea combined with smoking. The cold-dryness COPD model was developed by stressing with a cold-dry environment.Success of the model was determined by observation of pathologic lung sections. Rats were sacrificed by exsanguination from the femoral artery and changes of peripheral blood CD4+, CD8+, and CD4+/CD8+were detected by flow cytometry. Data were analyzed with SAS 11.5 statistical software.RESULTS: On the ninetieth day after ending the experiment, Peak expiratory flow in the cold-dryness COPD group was lower than that in the COPD and normal control groups(P<0.01). The time of inspiration in the cold-dryness COPD group was higher than that in the COPD and normal groups(P<0.05).Time of expiration(Te) in the cold-dryness COPD group was higher than that in the COPD and normal groups(P<0.01). 50% tidal volume expiratory flow(EF50) in the cold-dryness COPD group was lower than that in the COPD and normal groups(P<0.01), and EF50 in the COPD group was lower than that in the normal group(P<0.05). CD4+ content of peripheral blood in the cold-dryness COPD group was lower than that in the COPD and the normal groups(P<0.05). CD8+ content in the cold-dryness COPD and COPD groups was higher than that in the normal control group(P<0.01), and CD8+ content in the cold-dryness COPD group was higher than that in the COPD group(P<0.01). CD4+/CD8+in the cold-dryness COPD group and the COPD group was lower than that in the normal control group(P<0.01), and CD4+/CD8+ in the cold-dryness COPD group was lower than that in the COPD group(P<0.05).CONCLUSION: In the cold-dryness COPD model,CD8+ increased and CD4+/CD8+ decreased. Moreover, cold-dryness may aggravate this state.
