| 1 | Effect of Sirtuin 1 inhibition on matrix metalloproteinase 2 and Forkhead box O3a expression in breast cancer cells显示文摘Breast cancer is the most common invasive cancer in women worldwide.Sirtuin 1(SIRT1)has recently been shown to have implications in regulating cancer cell growth and apoptosis.SIRT1 regulates Forkhead box O3a(FOXO3a)by both inhibiting FOXO3-induced apoptosis and potentiating the ability of FOXO3a to resist oxidative stress.Matrix metalloproteinase 2(MMP2)participates in tumor invasion and metastasis by degrading extracellular matrix.SIRT1 up regulates MMP2 expression by its deacetylation activity.This study aimed to investigate the expression of SIRT1,FOXO3a and MMP2 in breast tissues of women with breast cancer.In addition,the effect of SIRT1 inhibition on both FOXO3a and MMP2 expression in breast cancer(MCF-7)cells was assessed.The expression levels of SIRT1,FOXO3a and MMP2 in the breast tissues were determined by real-time PCR in 60 patients with malignant tumor and in 24 patients with benign tumors.After SIRT1 inhibition,protein levels of SIRT1 and FOXO3a were assessed by Western Blot and levels of MMP2 by ELISA in MCF-7 cells.The expression levels of SIRT1,FOXO3a and MMP2 were significantly higher in breast cancer tissues compared to in benign breast tumor and adjacent normal tissues.SIRT1,MMP2 and FOXO3a expression were associated directly with each other.SIRT1 inhibition suppresses MMP2 and FOXO3a expression compared to control MCF7.Sirtinol(SIRT1 inhibitor)effectively induced inhibition of MMP2 and FOXO3a expression in MCF-7 cells,indicating the promising therapeutic strategy of targeting SIRT1 for breast cancer. | Hanan Abdelmawgoud Rehab Refaat El Awady | 2017 | Genes & Diseases2017,4,4: | 3 |
| 2 | Neutron-gamma discrimination method based on blind source separation and machine learning显示文摘The discrimination of neutrons from gamma rays in a mixed radiation field is crucial in neutron detection tasks.Several approaches have been proposed to enhance the performance and accuracy of neutron-gamma discrimination.However,their performances are often associated with certain factors,such as experimental requirements and resulting mixed signals.The main purpose of this study is to achieve fast and accurate neutron-gamma discrimination without a priori information on the signal to be analyzed,as well as the experimental setup.Here,a novel method is proposed based on two concepts.The first method exploits the power of nonnegative tensor factorization(NTF)as a blind source separation method to extract the original components from the mixture signals recorded at the output of the stilbene scintillator detector.The second one is based on the principles of support vector machine(SVM)to identify and discriminate these components.In addition to these two main methods,we adopted the Mexican-hat function as a continuous wavelet transform to characterize the components extracted using the NTF model.The resulting scalograms are processed as colored images,which are segmented into two distinct classes using the Otsu thresholding method to extract the features of interest of the neutrons and gamma-ray components from the background noise.We subsequently used principal component analysis to select the most significant of these features wich are used in the training and testing datasets for SVM.Bias-variance analysis is used to optimize the SVM model by finding the optimal level of model complexity with the highest possible generalization performance.In this framework,the obtained results have verified a suitable bias–variance trade-off value.We achieved an operational SVM prediction model for neutron-gamma classification with a high true-positive rate.The accuracy and performance of the SVM based on the NTF was evaluated and validated by comparing it to the charge comparison method via figure of merit.The results indicate that the proposed approach has a superior discrimination quality(figure of merit of 2.20). | Hanan Arahmane El-Mehdi Hamzaoui Yann Ben Maissa Rajaa Cherkaoui El Moursli | 2021 | Nuclear Science and Techniques2021,32,2: | 2 |

| 4 | Adherence to medication: A nation-wide study from the Children's Cancer Hospital, Egypt显示文摘AIM: To investigate adherence to medical regimen and predictors for non-adherence among children with cancer in Egypt. METHODS: We administered two study specific questionnaires to 304 parents of children diagnosed with cancer at the Children's Cancer Hospital in Cairo, Egypt, one before the first chemotherapy treatment and the other before the third. The questionnaires were translated to colloquial Egyptian Arabic, and due, to the high illiteracy level in Egypt an interviewer read thequestions in Arabic to each parent and registered the answers. Both questionnaires consisted of almost 90 questions each. In addition, a Case Report Form was filled in from the child's medical journal. The study period consisted of 7 mo(February until September 2008) and we had a participation rate of 97%. Descriptive statistics are presented and Fisher's exact test was used to check for possible differences between the adherent and non-adherent groups. A P-value below 0.05 was considered significant. Software used was SAS version 9.3 for Windows(SAS Institute Inc., Cary, NC, United States).RESULTS: Two hundred and eighty-one(90%) parents answered the second questionnaire, regarding their child's adherence behaviour. Approximately two thirds of the children admitted to their third chemotherapy treatment had received medical recommendations upon discharge from the first or second chemotherapy treatment(181/281, 64%). Sixty-eight percent(123/181) of the parents who were given medical recommendations reported that their child did not follow the recommendations. Two main predictors were found for non-adherence: child resistance(111/123, 90%) and inadequate information(100/123, 81%). In the adherent group, 20% of the parents(n = 12/58) reported trust in their child's doctor while 14 percent 8/58 reported trust in the other health-care professionals. Corresponding numbers for the non-adherent group are 8/123(7%) for both their child's doctor and other health-care professionals.
