1PCR-based approaches for identification of multi-copy transgene integration sites in mouse genome显示文摘Generation of transgenic mice by DNA microinjection has become one of the most important technologies in studying gene function in vivo. Ran- dom integration of transgene often results in inser- tional mutation which may complicate phenotype analysis of transgenic mice and/or create a good opportunity to study the function of endogenous gene. However,the utilization of these potentially valuable resources is hampered due to lack of efficient ap- proaches for rapid identification of multi-copy trans- gene integration sites. Here we propose two PCR-based approaches to identifying trans- gene/genome junction sequences. The efficiency of these two strategies was tested in 9 independent transgenic mouse lines. Among them,the transgene in 8 mouse lines was clearly localized to certain chromosome regions. 7Advances in sequencing and key character analysis of mango (Mangifera indica L.)显示文摘Mango(Mangifera indica L.)is an important fruit crop in tropical and subtropical countries associated with many agronomic and horticultural problems,such as susceptibility to pathogens,including powdery mildew and anthracnose,poor yield and quality,and short shelf life.Conventional breeding techniques exhibit significant limitations in improving mango quality due to the characteristics of long ripening,self-incompatibility,and high genetic heterozygosity.In recent years,much emphasis has been placed on identification of key genes controlling a certain trait through genomic association analysis and directly breeding new varieties through transgene or genotype selection of offspring.This paper reviews the latest research progress on the genome and transcriptome sequencing of mango fruit.The rapid development of genome sequencing and bioinformatics provides effective strategies for identifying,labeling,cloning,and manipulating many genes related to economically important traits.Preliminary verification of the functions of mango genes has been conducted,including genes related to flowering regulation,fruit development,and polyphenol biosynthesis.Importantly,modern biotechnology can refine existing mango varieties to meet the market demand with high economic benefits.Miaoyu Song Haomiao Wang Zhiyi Fan Hantang Huang Huiqin Ma 2023Horticulture Research2023,10,2:0
8Hexyl substitution of pentathienoacene toward a significant improvement in charge transport显示文摘A novel semiconductor, dihexyl-substituted pentathienoacene(C6-PTA) is designed and synthesized in five steps with the total yield up to 25.2%. The introduction of hexyl chains endow the thin film semiconductor with about threefold increase in carrier mobility and one to three orders of magnitude improvement in current on/off ratio. Furthermore, single crystal FETs based on C6-PTA exhibited mobility up to 0.64 cm^2 V^(-1) s^(-1), which is over 50 times higher than the thin film counterpart.
