4Serum pepsinogen Ⅱ is a better diagnostic marker in gastric cancer显示文摘AIM:To investigate screening makers for gastric cancer,we assessed the association between gastric cancer and serum pepsinogens(PGs).METHODS:The subjects comprised 450 patients with gastric cancer,111 individuals with gastric atrophy,and 961 healthy controls.Serum anti-Helicobacter pylori(H.pylori) immunoglobulin G(IgG),PGⅠand PG Ⅱ were detected by enzyme-linked immunosorbent assay.Gastric atrophy and gastric cancer were diagnosed by endoscopy and histopathological examinations.Odds ratios and 95%CIs were calculated using multivariate logistic regression.RESULTS:Rates of H.pylori infection remained high in Northeastern China.Rates of H.pylori IgG positivity were greater in the gastric cancer and gastric atrophy groups compared to the control group(69.1% and 75.7% vs 49.7%,P < 0.001).Higher levels of PG Ⅱ(15.9 μg/L and 13.9 μg/L vs 11.5 μg/L,P < 0.001) and lower PGⅠ/PG Ⅱ ratio(5.4 and 4.6 vs 8.4,P < 0.001) were found in patients with gastric cancer or gastric atrophy compared to healthy controls,whereas no correlation was found between the plasma PGⅠconcentration and risk of gastric cancer(P = 0.537).In addition,multivariate logistic analysis indicated that H.pylori infection and atrophic gastritis were independent risk factors for gastric cancer.Lower plasma PGⅠ/PG Ⅱ ratio was associated with higher risks of atrophy and gastric cancer.Furthermore,plasma PG Ⅱ?level?significantly?correlated?with?H.pyloriinfected gastric cancer.CONCLUSION:Serum PG Ⅱ concentration and PG Ⅰ/PG Ⅱ ratio are potential biomarkers for H.pyloriinfected gastric disease.PG Ⅱ is independently associated with risk of gastric cancer.Xue-Yuan Cao Zhi-Fang Jia Mei-Shan Jin Dong-Hui Cao Fei Kong Jian Suo Jing Jiang 2012World Journal of Gastroenterology2012,18,48:27
5Expression of miR-125b in the new,highly invasive glioma stem cell and progenitor cell line SU3显示文摘MicroRNA (miR)-125b has been shown to play a potential role in the development of glioma stem cells.However,the relationship between miRNA and glioma stem cells is still elusive.This study was designed to elucidate this potential relationship.We established a highly invasive glioma stem cell and progenitor (GSCP) cell line SU3.SU3 cell suspensions were injected into nude mice brains in situ,and the invasiveness of graft tumors was analyzed using hematoxylin and eosin staining as well as immunohistochemistry.Real-time polymerase chain reaction (PCR) was used to measure the expression levels of miR-125b in SU3 and other cells.In vitro,SU3 cells expressed CD133 and nestin as well as differentiation markers glial fibrillary acidic protein (GFAP) and β-tubulin III,which were consistent with the characteristics of glioma stem cells.Scratch assays indicated that the migration ability of SU3 cells was stronger than that of U251 stem cells (U251s).In vivo,SU3 cells invaded into each part of the mouse brain from the caudate nucleus in a diffuse pattern and highly expressed invasive and proliferative cell markers matrix metalloprotease 2 (MMP2),MMP9,and Ki-67.Real-time PCR results revealed that the levels of miR-125b and MMP9 were significantly higher in SU3 and SU2,also a highly invasive GSCP cell line we established before,than in U251s.High expression of miR-125b both in newly established GSCPs,SU3,and long-term cultured GSCPs,SU2 suggests that miR-125b exhibits oncogene-like behavior.This behavior should be considered in further studies of miR-125b in cancer stem cells.Furthermore,MMP9,which plays a role in cancer stem cell invasion,may be a target gene of miR-125b.Yi Wan Xi-Feng Fei Zhi-Min Wang Dong-Yi Jiang Han-Chun Chen Jian Yang Lei Shi Qiang Huang 2012Chinese Journal of Cancer2012,31,4:25

9APPLICATION OF WHOLE BODY DIFFUSION WEIGHTED MR IMAGING FOR DIAGNOSIS AND STAGING OF MALIGNANT LYMPHOMA显示文摘Objective To evaluate the clinical impact of whole body diffusion weighted imaging (WB-DWI) on diagnosis and staging of malignant lymphoma. Methods Thirty-one patients with suspected lymphadenopathy were enrolled. WB-DWI was performed by using short TI inversion recovery echo-planar imaging sequence with free breathing and built-in body coil. Axial T2- weighted imaging images of the same location were used as reference. The results of WB-DWI were compared with pathological results and other imaging modalities. The mean apparent diffusion coefficient (ADC) values of different kinds of lymph nodes were compared. Results WB-DWI was positive in all 18 cases with lymphoma, 5 cases with metastatic lymph nodes and 4 of 8 cases with benign lymphadenopathy. The mean ADC value of lymphomatous, metastatic and benign lymph nodes was (0.87 ± 0.17) × 10-3, (0.98 ± 0.09) × 10-3 and (1.20 ± 0.10) × 10-3 mm2/s. There was significant difference in ADC value between benign lymph nodes and other two groups (P < 0.01). The sensitivity, specificity and accuracy of WB-DWI in diagnosis of lymphoma were 100% (18/18), 30.8% (4/13) and 71.0% (22/31). When an ADC value of 1.08 × 10-3 mm2/s was used as the threshold value for differentiating malignant from benign lymph nodes, the best results were obtained with sensitivity of 87.8% and specificity of 91.3%. Sixteen of eighteen cases (88.9%) of lymphoma were accurately staged in accordance with clinical staging. Conclusions WB-DWI is a sensitive, but less specific technique for diagnosis of lymphoma. It is difficult to differentiate lymphomatous from metastatic lymph nodes using WB-DWI. 11Virulence and potential pathogenicity of coccoid Helicobacter pylori induced by antibiotics显示文摘AIM To explore the virulence and the potential pathogenicity of coccoid Helicobacter pylori (H. pylori) transformed from spiral form by exposure to antibiotic.METHODS Three strains of H. pylori, isolated from gastric biopsy specimens of confirmed peptic ulcer, were converted from spiral into coccoid from by exposure to metronidazole.Both spiral and coccoid form of H. pylori were tested for the urease activity, the adherence to Hep-2 cells and the vacuolating cytotoxicity to Hela cells, and the differences of the protein were analysed by SDS-PAGE and Western blot,The mutation of the genes including ureA, ureB,hpaA; vacA and cagA, related with virulence,was detected by means of PCR and PCR-SSCP.RESULTS In the coccoid H. pylori, the urease activity, the adherence to Hep-2 cells and the vacuolating cytotoxicity to Hela cells alldecreased. In strain F44, the rate and index of adherence reduced from 70.0% ± 5.3% to 33% ±5.1% and from 2.6 ±0.4 to 0.96 ±0.3 (P<0.01),respectively. The invasion of coccoid H. pylori into Hep-2 cell could be seen under electronmicroscope. SDS-PAGE showed that the content of the protein with the molecular weight over Mr74 000 decreased, and the hybriditional signal in band Mr 125 000 weakened, while the band Mr 110000 and Mr63000 strengthened in coccoid H. pylori as shown in Western blot. 13NH_3-SCR denitration catalyst performance over vanadium–titanium with the addition of Ce and Sb显示文摘Selective catalytic reduction technology using NH3 as a reducing agent(NH3-SCR) is an effective control method to remove nitrogen oxides. TiO2-supported vanadium oxide catalysts with different levels of Ce and Sb modification were prepared by an impregnation method and were characterized by X-ray diffractometer(XRD), Brunauer–Emmett–Teller(BET), Transmission electron microscopy(TEM), Fourier transform infrared spectroscopy(FT-IR), UV–Vis diffuse reflectance spectroscopy(UV–Vis DRS), Raman and Hydrogen temperature-programmed reduction(H2-TPR). The catalytic activities of V5 CexS by/TiO2 catalysts for denitration were investigated in a fixed bed flow microreactor. The results showed that cerium, vanadium and antimony oxide as the active components were well dispersed on TiO2, and the catalysts exhibited a large number of d–d electronic transitions, which were helpful to strengthen SCR reactivity. The V5 CexS by/TiO2 catalysts exhibited a good low temperature NH3-SCR catalytic activity. In the temperature range of 210 to 400℃, the V5 CexS by/TiO2 catalysts gave NO conversion rates above 90%. For the best V5Ce35Sb2/TiO2 catalyst, at a reaction temperature of 210℃, the NO conversion rate had already reached 90%. The catalysts had different catalytic activity with different Ce loadings. 14Mature Archean continental crust in the Yangtze craton: Evidence from petrology, geochronology and geochemistry显示文摘Previous studies have shown that the Archean basement is widely distributed throughout the Yangtze craton. To date, however, Archean basement terrains have not been found, except for a few Kongling high-grade metamorphic terrains in the Huangling dome that have been confirmed to be of Archean age. To further understand the basement component and crustal evolution of the Yangtze craton, we carried out a petrological, geochronological and geochemical study of the Jinshan K-feldspar granite emplaced within the Yangpo Group, located in Huji Town, Zhongxiang City, Hubei Province. Results indicate that the zircon SHRIMP U-Pb age of the Jinshan granite is 2655±9 Ma, placing it within the middle Neoarchean. Chemically, this pluton yields abundant silica and alkalis and is depleted in Ca, Mg and Ti. Furthermore, it is enriched in Rb, Th, Ga, Y and Zr, depleted in Sr, Ba, Nb and Ta, and especially lacking in Eu. High ratios of FeO*/MgO (32.0 to 58.7) and 104 ×Ga/Al (3.19 to 3.41) were also found. The pluton exhibits characteristics typical of A-type granites with crustal source magmas. Moreover, the meta-sedimentary rock association of the Yangpo Group, into which the pluton intruded, clearly shows relatively stable depositional environments of a shallow shelf sequence. Therefore, before the middle Neoarchean, the Yangtze craton contained mature continental crust. 16Effects of warming and clipping on plant and soil properties of an alpine meadow in the Qinghai-Tibetan Plateau, China显示文摘Climate warming and livestock grazing are known to have great influences on alpine ecosystems like those of the Qinghai-Tibetan Plateau(QTP) in China. However, it is lacking of studies on the effects of warming and grazing on plant and soil properties in these alpine ecosystems. In this study, we reported the related research from manipulative experiment in 2010–2012 in the QTP. The aim of this study was to investigate the individual and combined effects of warming and clipping on plant and soil properties in the alpine meadow ecosystem. Infrared radiators were used to simulate climate warming starting in July 2010, while clipping was performed once in October 2011 to simulate the local livestock grazing. The experiment was designed as a randomized block consisting of five replications and four treatments: control(CK), warming(W), clipping(C) and warming+clipping combination(WC). The plant and soil properties were investigated in the growing season of the alpine meadow in 2012. The results showed that W and WC treatments significantly decreased relative humidity at 20-cm height above ground as well as significantly increases air temperature at the same height, surface temperature, and soil temperature at the depth of 0–30 cm. However, the C treatment did not significantly decrease soil moisture and soil temperature at the depth of 0–60 cm. Relative to CK, vegetation height and species number increased significantly in W and WC treatment, respectively, while vegetation aboveground biomass decreased significantly in C treatment in the early growing season. However, vegetation cover, species diversity, belowground biomass and soil properties at the depth of 0–30 cm did not differ significantly in W, C and WC treatments. Soil moisture increased at the depth of 40–100 cm in W and WC treatments, while belowground biomass, soil activated carbon, organic carbon and total nitrogen increased in the 30–50 cm soil layer in W, C and WC treatments. Although the initial responses of plant and soil properties to experimental warming and clipping were slow and weak, the drought induced by the downward shift of soil moisture in the upper soil layers may induce plant belowground biomass to transfer to the deeper soil layers. This movement would modify the distributions of soil activated carbon, organic carbon and total nitrogen. 18Clinical features and outcomes of patients with severe acute pancreatitis complicated with gangrenous cholecystitis显示文摘BACKGROUND: The effects of gangrenous cholecystitis (GC) and consequent surgical interventions on the clinical outcomes and prognosis of patients with severe acute pancreatitis are not clear. The present study was to characterize the clinical outcomes of patients with severe acute pancreatitis complicated with GC. METHODS: We retrospectively analyzed 253 consecutive patients hospitalized for acute pancreatitis in intensive care unit. Among them, 68 were diagnosed as having severe acute pancreatitis; 10 out of the 68 patients had GC. We compared these 10 patients with GC and 58 patients without GC. The indices analyzed included sepsis/septic shock, pancreatic encephalopathy, acute respiratory distress syndrome, acute renal failure, multiple organ dysfunction syndrome, and death. RESULTS: Specific CT images of GC in patients with severe acute pancreatitis included enlarged and high-tensioned gallbladder, wall thickening, lumenal emphysema, discontinuous and/or irregular enhancement of mucosa, and pericholecystic effusion. The rates of severe sepsis/septic shock (70.0% vs 24.1%, P<0.01), pancreatic encephalopathy (50.0% vs 17.2%, P<0.05), acute respiratory distress syndrome (90.0% vs 41.4%, P<0.01), multiple organ dysfunction syndrome (70.0% vs 24.1%, P<0.01), acute renal failure (40.0% vs 27.6%, P<0.05), and death (40.0% vs 13.8%, P<0.05) were significantly higher in patients with GC than in those without GC.CONCLUSION: CT scans can help to identify early GC in patients with severe acute pancreatitis; early diagnosis and intervention for patients with GC can reduce morbidity and mortality.Er-Zhen Chen Jie Huang Zhi-Wei Xu Jian Fei En-Qiang Mao Sheng-Dao Zhang 2013Hepatobiliary & Pancreatic Diseases International2013,12,3:13
19Seroepidemiological Investigation of Lyme Disease and Human Granulocytic Anaplasmosis among People Living in Forest Areas of Eight Provinces in China显示文摘Objective Lyme disease and Human granulocytic anaplasmosis are tick‐borne diseases caused by Borrelia burgdorferi and Anaplasma phagocytophilum respectively. We have investigated infection and co-infection of the two diseases in the population of forest areas of eight provinces in China by measuring seroprevalence of antibodies against B. burgdorferi and A. phagocytophilum. Methods Forest areas in 8 provinces were chosen for investigation using whole sampling and questionnaire survey methods. 3 669 serum samples from people in the forest areas were tested for the presence of antibodies by indirect immunofluorescent assay (IFA). Results Seroprevalence against B. burgdorferi was 3% to 15% and against A. phagocytophilum was 2% to 18% in the study sites in the 8 provinces in China. We also found co-infection of B. burgdorferi and A. phagocytophilum in 7 of the 8 provinces (the exception being the Miyun area in Beijing). The seroprevalence for both B. burgdorferi and A. phagocytophilum was significantly higher among people exposed to ticks than among people who were not exposed to ticks.
