1Targeting p53 pathways:mechanisms,structures,and advances in therapy显示文摘The TP53 tumor suppressor is the most frequently altered gene in human cancers,and has been a major focus of oncology research.The p53 protein is a transcription factor that can activate the expression of multiple target genes and plays critical roles in regulating cell cycle,apoptosis,and genomic stability,and is widely regarded as the“guardian of the genome”.Accumulating evidence has shown that p53 also regulates cell metabolism,ferroptosis,tumor microenvironment,autophagy and so on,all of which contribute to tumor suppression.Mutations in TP53 not only impair its tumor suppressor function,but also confer oncogenic properties to p53 mutants.Since p53 is mutated and inactivated in most malignant tumors,it has been a very attractive target for developing new anti-cancer drugs.However,until recently,p53 was considered an“undruggable”target and little progress has been made with p53-targeted therapies.Here,we provide a systematic review of the diverse molecular mechanisms of the p53 signaling pathway and how TP53 mutations impact tumor progression.We also discuss key structural features of the p53 protein and its inactivation by oncogenic mutations.In addition,we review the efforts that have been made in p53-targeted therapies,and discuss the challenges that have been encountered in clinical development.Haolan Wang Ming Guo Hudie Wei Yongheng Chen 2023Signal Transduction and Targeted Therapy2023,8,4:4

6Recent strategies for constructing efficient interfacial solar evaporation systems显示文摘Interfacial solar evaporation(ISE)is a promising technology to relieve worldwide freshwater shortages owing to its high energy conversion efficiency and environmentally sustainable potential.So far,many innovative materials and evaporators have been proposed and applied in ISE to enable highly controllable and efficient solar-to-thermal energy conversion.With rational design,solar evaporators can achieve excellent energy management for lowering energy loss,harvesting extra energy,and efficiently utilizing energy in the system to improve freshwater production.Beyond that,a strategy of reducing water vaporization enthalpy by introducing molecular engineering for water-state regulation has also been demonstrated as an effective approach to boost ISE.Based on these,this article discusses the energy nexus in two-dimensional(2D)and three-dimensional(3D)evaporators separately and reviews the strategies for design and fabrication of highly efficient ISE systems.The summarized work offers significant perspectives for guiding the future design of ISE systems with efficient energy management,which pave pathways for practical applications.Yida Wang Junqing Hu Li Yu Xuan Wu Yingying Zhang Haolan Xu 2023Nano Research Energy2023,2,2:1

12Fabrication of UV responsive micelles-containing multilayers and their influence on cell adhesion显示文摘Multilayers incorporated with stimuli-responsive substances by means of layer-by-layer(LbL) self-assembly are much attractive due to their advantages of stimuli-responsiveness and potential applications in different fields. In this study, pyrenemethyl acrylate(PA) was synthesized, and was copolymerized with acrylic acid(AA) to obtain the amphiphilic and photodegradable P(PA-co-AA)polymers with a PA:AA molar ratio of 1.3:3, and an average molecular weight of 6.9 kDa and polydispersity index of 1.04. They formed micelles spontaneously when dispersed in aqueous solution with a size of 27.5 nm in a dry state and 136.6 nm in a wet state. The micelles were readily decomposed to form aggregates as a result of the cleavage of the pyrenemethyl ester bonds under UV-irradiation. UV-responsive micelles-containing multilayers were prepared by LbL self-assembly of the UV-responsive micelles and polyallylamine hydrochloride(PAH). UV-irradiation of the multilayers resulted in the decomposition of micelles, leading to larger surface roughness, and enhanced swelling ratio and wettability of the multilayers. In vitro culture of A549, HepG2 and endothelial cells showed significantly better adhesion at 4 h on the UV-illuminated multilayers, whereas the cell proliferation was not affected significantly until 5 d.Haolan Zhang Danyu Wang Xue Lin Nikolaos Politakos Jimena S. 14Boosting extraction of Pb in contaminated soil via interfacial solar evaporation of multifunctional sponge显示文摘Interfacial solar water evaporation is a reliable way to accelerate water evaporation and contaminant remediation.Embracing the recent advance in photothermal technology,a functional sponge was prepared by coating a sodium alginate(SA)impregnated sponge with a surface layer of reduced graphene oxide(rGO)to act as a photothermal conversion medium and then subsequently evaluated for its ability to enhance Pb extraction from contaminated soil driven by interfacial solar evaporation.The SA loaded sponge had a Pb adsorption capacity of 107.4 mg g^(-1).Coating the top surface of the SA sponge with rGO increased water evaporation performance to 1.81 kg m^(-2)h^(-1)in soil media under one sun illumination and with a wind velocity of 2 m s^(-1).Over 12 continuous days of indoor evaporation testing,the Pb extraction efficiency was increased by 22.0%under 1 sun illumination relative to that observed without illumination.Subsequently,Pb extraction was further improved by 48.9%under outdoor evaporation conditions compared to indoor conditions.Overall,this initial work shows the significant potential of interfacial solar evaporation technologies for Pb contaminated soil remediation,which should also be applicable to a variety of other environmental contaminants.Pan Wu Xuan Wu Yida Wang Jingyuan Zhao Haolan Xu Gary Owens 2023Green Energy & Environment2023,8,5:0
15Reactive oxygen species-scavenging nanoparticles coated with chondroitin sulfate protect cartilage against osteoarthritis in vivo显示文摘Osteoarthritis(OA)is a prevalent chronic inflammatory disease in joints.Current interventions confront systemic toxicity and insufficient bioavailability.The unbalanced microenvironment of OA joints mainly fosters over-expressed reactive oxygen species(ROS),extracellular matrix disintegration,and apoptosis of chondrocytes.In this study,a kind of ROS-scavenging,biodegradable and drug-free nanoparticles(PP NPs)were constructed by the crosslinking of poly(propylene fumarate)(PPF)and ROS-scavenging poly(thioketal)(PTK).The high content of PTK and high crosslinking density of PPF and PTK innovatively endowed the NPs with slow degradation and prolonged ROS-elimination ability.The NPs were further surface-modified with chondroitin sulfate(CS),one of the dietary supplements for osteoarthritis.The intrinsic properties of resultant PP-CS NPs were excavated in vitro and in vivo.The PP-CS NPs could desirably consume 1,10-diphenyl-2-picrylhydrazyl(DPPH)radicals without toxicity to RAW264.7 cells in vitro.With an average diameter of~300 nm,the PP-CS NPs could be intra-articular administrated in OA rats and showed prolonged joint retention time,allowing only one injection per month.Moreover,the PP-CS NPs possessed a prolonged ROS depletion and M2 macrophage induction effect,down-regulated inflammatory cytokines,and reduced glycosaminoglycans loss.Consequently,the PP-CS NPs protected articular surface erosion,inhibited uneven cartilage matrix,and attenuated OA progression.Zhaoyi Wang Hao Xiong Zihe Zhai Yuejun Yao Tong Zhou Haolan Zhang Cunyi Fan Changyou Gao 2023Nano Research2023,16,2:0
16Can Eurasia Experience a Cold Winter under a Third-Year La Nina in 2022/23?显示文摘The Northern Hemisphere(NH)often experiences frequent cold air outbreaks and heavy snowfalls during La Nina winters.In 2022,a third-year La Nina event has exceeded both the oceanic and atmospheric thresholds since spring and is predicted to reach its mature phase in December 2022.Under such a significant global climate signal,whether the Eurasian Continent will experience a tough cold winter should not be assumed,despite the direct influence of mid-to high-latitude,large-scale atmospheric circulations upon frequent Eurasian cold extremes,whose teleconnection physically operates by favoring Arctic air invasions into Eurasia as a consequence of the reduction of the meridional background temperature gradient in the NH.In the 2022/23 winter,as indicated by the seasonal predictions from various climate models and statistical approaches developed at the Institute of Atmospheric Physics,abnormal warming will very likely cover most parts of Europe under the control of the North Atlantic Oscillation and the anomalous anticyclone near the Ural Mountains,despite the cooling effects of La Nina.At the same time,the possibility of frequent cold conditions in mid-latitude Asia is also recognized for this upcoming winter,in accordance with the tendency for cold air invasions to be triggered by the synergistic effect of a warm Arctic and a cold tropical Pacific on the hemispheric scale.However,how the future climate will evolve in the 2022/23 winter is still subject to some uncertainty,mostly in terms of unpredictable internal atmospheric variability.Consequently,the status of the mid-to high-latitude atmospheric circulation should be timely updated by medium-term numerical weather forecasts and sub-seasonal-to-seasonal prediction for the necessary date information and early warnings.Fei ZHENG Bo WU Lin WANG Jingbei PENG Yao YAO Haifeng ZONG Qing BAO Jiehua MA Shuai HU Haolan REN Tingwei CAO Renping LIN Xianghui FANG Lingjiang TAO Tianjun ZHOU Jiang ZHU 2023Advances in Atmospheric Sciences2023,40,4:0
