1气相色谱-串联质谱检测蔬菜中氟吡菌酰胺及其代谢物残留显示文摘采用气相色谱-串联质谱(GC-MS/MS)技术建立了同时检测7种类型蔬菜中氟吡菌酰胺及其代谢物残留的分析方法。样品采用乙腈提取,无水硫酸镁和氯化钠盐析后,经分散固相萃取(PSA)净化,GC-MS/MS检测。结果表明:在0.01、0.1和1 mg/kg 3个添加水平下,氟吡菌酰胺及其代谢物2-(三氟甲基)苯甲酰胺(BZM)的平均回收率在80%-108%之间,相对标准偏差(RSD)在1.5%-8.4%之间。氟吡菌酰胺及其代谢物在7种蔬菜中的检出限(LOD)(S/N=3)分别为2.17-3.13μg/kg和1.22-2.04μg/kg,两者的定量限(LOQ)均为0.01 mg/kg。该方法操作简便、稳定和快速,可以满足大部分蔬菜中氟吡菌酰胺及其代谢物的定性与定量分析要求。李文卓 钱圆 MATSUMOTO Haruna 刘晓玉 秦丽 王蒙岑 朱国念 2016农药学学报2016,18,6:14
2五氟磺草胺在稻田中的消解动态及残留特性显示文摘建立了超高效液相色谱-质谱联用检测五氟磺草胺在水稻植株、稻田土壤、田水和糙米中残留的分析方法,结合田间试验研究了五氟磺草胺在稻田环境中的消解及残留特性,并对稻米中五氟磺草胺残留的膳食暴露进行了初步评估。结果表明:在0.005~0.5 mg/L范围内,五氟磺草胺的质量浓度与对应的峰面积间呈良好线性关系,检出限(LOD)为0.001~0.002 mg/kg,定量限(LOQ)为0.003~0.005 mg/kg。在0.005~0.5 mg/kg添加水平下,五氟磺草胺在水稻植株、稻田土壤、田水和糙米中的平均回收率在89%~106%之间,相对标准偏差在2.8%~8.5%之间。浙江、福建和黑龙江2年3地的田间试验表明:0.025%五氟磺草胺颗粒剂在水稻植株、稻田土壤和田水中的消解半衰期分别为1.5~3.3,3.0~4.7和1.6-3.0 d,说明该药剂在稻田环境中消解速率较快。以五氟磺草胺有效成分含量37.5和56.3 g/hm^2分别施药1次,于水稻成熟期采样检测,发现其在糙米中的残留量低于0.005 mg/kg,表明其膳食摄入风险很低,该研究结果可为五氟磺草胺风险评估提供一定参考。李文卓 钱圆 MATSUMOTO Haruna 杨斌 王蒙岑 朱国念 2017农药学学报2017,19,1:6
3Human PBMC-transferred murine MHC class I/II-deficient NOG mice enable long-term evaluation of human immune responses显示文摘Immunodeficient mice engrafted with human peripheral blood cells are promising tools for in vivo analysis of human patient individual immune responses.However,when human peripheral blood mononuclear cells(PBMCs)are transferred into NOG(NOD/Shi-scid,IL-2rg null)mice,severe graft versus host disease(GVHD)hinders long term detailed analysis.Administration of human PBMCs into newly developed murine MHC class I-and class II-deficient NOG(NOG-dKO;NOG-Iab,B2m-double-knockout)mice showed sufficient engraftment of human immune cells with little sign of GVHD.Immunization with influenza vaccine resulted in an increase in influenza-specific human IgG Ab,indicating induction of antigen-specific B cells in the NOG-dKO mice.Immunization with human dendritic cells pulsed with HLA-A2 restricted cytomegalovirus peptide induced specific cytotoxic T cells,indicating the induction of antigen-specific T cells in the NOG-dKO mice.Adoptive cell therapies(ACTs)using melanoma antigen recognized by T cells(MART-1)-specific TCR-transduced activated T cells showed strong tumor growth inhibition in NOG-dKO mice without any sign of GVHD accompanied by preferential expansion of the transferred MART-1-specific T cells.ACTs using cultured human melanoma infiltrating T cells also showed anti-tumor effects against autologous melanoma cells in NOG-dKO mice,in which changes in human cancer phenotypes by immune intervention,such as increased CD271 expression,could be evaluated.Therefore,NOG-dKO mice are useful tools for more detailed analysis of both the induction and effector phases of T-cell and B-cell responses for a longer period than regular NOG mice.Tomonori Yaguchi Asuka Kobayashi Takashi Inozume Kenji Morii Haruna Nagumo Hiroshi Nishio Takashi Iwata Yuyo Ka Ikumi Katano Ryoji Ito Mamoru Ito Yutaka Kawakami 2018Cellular & Molecular Immunology2018,15,11:6

5Reprogramming of phytopathogen transcriptome by a non-bactericidal pesticide residue alleviates its virulence in rice显示文摘Bacteria equipped with virulence systems based on highly bioactive small molecules can circumvent their host's defense mechanisms.Pathogens employing this strategy are currently threatening global rice production.In the present study,variations in the virulence of the highly destructive Barkholderiaplantarii were observed in different rice-producing regions.The environment-linked variation was not attributable to any known host-related or external factors.Co-occurrence analyses indicated a connection between reduced virulence and 5-Amino-l,3,4-thiadiazole-2-thiol(ATT),a non-bactericidal organic compound.ATT,which accumulates in rice plants during metabolization of specific agrochemicals,was found to reduce virulence factor secretion by B.plantarii up to 88.8%and inhibit pathogen virulence by hijacking an upstream signaling cascade.Detailed assessment of the newly discovered virulence inhibitor resulted in mechanistic insights into positive effects of ATT accumulation in plant tissues.Mechanisms of virulence alleviation were deciphered by integrating high-throughput data,gene knockout mutants,and molecular interaction assays.TroK,a histidine protein kinase in a two-component system that regulates virulence factor secretion,is likely the molecular target antagonized by ATT.Our findings provide novel insights into virulence modulation in an important plant-pathogen system that relies on the host's metabolic activity and subsequent signaling interference.Haruna Matsumoto Yuan Qian Xiaoyan Fan Sunlu Chen Yanxia Nie Kun Qiao Dandan Xiang Xinzhong Zhang Meng Li Bo Guo Peilin Shen Qiangwei Wang Yunlong Yu Tomislav Cemava Mengcen Wang 2022Fundamental Research2022,2,2:2
6尿素中添加腐植酸、黄腐酸、重过磷酸钙和氯化钾能抑制氨的挥发显示文摘该试验比较了不同混合物,尿素-重过磷酸钙-氯化钾、尿素-重过磷酸钙-氯化钾-腐植酸、尿素-重过磷酸钙-氯化钾-黄腐酸、尿素-重过磷酸钙-氯化钾-酸性混合物(黄腐酸+腐植酸)在对氨损失,土壤pH值,铵态氮和可利用硝态氮积累等方面的影响,以尿素单独施用作为对照。方法:在实验室条件下,利用一个封闭式动态空气流量系统来评估是否混有重过磷酸钙、氯化钾、腐植酸、黄腐酸的情况下尿素的有效性。氨损失、土壤pH值、土壤中铵态氮以及可利用硝态氮是由标准程序测定的。结果:与对照组(单独施用尿素)相比,处理尿素-重过磷酸钙-氯化钾,尿素-重过磷酸钙-氯化钾-腐植酸,尿素-重过磷酸钙-氯化钾-黄腐酸,尿素-重过磷酸钙-氯化钾-酸性混合物(黄腐酸+腐植酸)中氨损失明显地减少,减少率由12.92%、20.12%、25.94%达到100%,土壤中的铵态氮也有类似的变化。从所有的处理来看,只有尿素-重过磷酸钙-氯化钾-黄腐酸,尿素-重过磷酸钙-氯化钾-酸性混合物(腐植酸+黄腐酸)明显地积累了土壤中可利用硝态氮,这一结果与本研究中pH值的结果一致。结论:尿素、重过磷酸钙、氯化钾中混合腐植酸或者酸性的腐植酸和黄腐酸混合物能够显著减少氨损失。这项研究结果有助于提高尿素中N,P,K使用的有效性,同时减少环境污染。Shamsuddin Rosliza Osumanu Haruna Ahmed Nik Muhamad Ab.Majid 张丽娜 宋丽艳 2010腐植酸2010,,2:2
7有机氮肥对马来西亚沙捞越州酸性土壤玉米干物质产量及铵态氮和硝态氮含量的影响显示文摘腐殖质中的腐植酸和黄腐酸可以提高土壤中可交换性铵(NH4+)的回收率。这两种酸固定和保持NH4+的能力已经在许多研究中得到了证实,二者通过提高植株的光合作用速率、促进根系发育影响植物的生长和养分吸收。因此,本研究调查了这两种酸(液态)对玉米干物质产量、土壤铵态氮和硝态氮(NO3-)含量的影响。根据液态有机氮肥的组成,采用标准方法分离提纯腐植酸。有机氮肥在玉米种植后第10天、第28天分别施入盆栽土壤,然后在种植后第54天或孕穗期采集植株和土壤样品。土壤样品分析pH、铵态氮和硝态氮含量,植株样品测定干物质产量。结果表明,在酸性条件下,液态有机氮肥(黄腐酸、腐植酸或二者同时施用)的施用可以提高土壤中NH4+的累积量。随土壤活性碳储量的增加,腐植酸分子上羧基官能团对土壤NH4+的吸附能力增强。然而,酸含量较低时对干物质产量的影响明显减小。各处理间硝态氮的有效性没有达到统计上的差异显著水平。较低的pH值能抑制土壤硝化作用进程,同时降低土壤NO3-含量。这表明液态腐植酸和/或黄腐酸在提高尿素利用效率方面发挥着重要作用。然而,与腐植酸分子特性有关的作用机理还需要进一步详细研究。本研究为液态和叶面有机肥料的发展提供一定的依据。Susilawati Kasim Osumanu Haruna Ahmed Nik Muhamad Ab.Majid Mohd Khanif Yusop Mohamadu Boyie Jalloh 王艳群 孙志梅 2011腐植酸2011,,2:2
8Soil Hydraulic Properties: Influence of Tillage and Cover Crops显示文摘Understanding the effects of cover crops and tillage on soil physical properties is important for determining soil productivity. This study was conducted at Lincoln University's Freeman Center, USA to evaluate the effects of tillage and cover crop management on soil hydraulic properties. The field site included three replicate blocks in a randomized complete block design with each plot measuring 21.3 m in length and 12.2 m in width. Treatment factors were tillage at two levels(moldboard plow tillage vs. no tillage) and cover crop at two levels(cereal rye(Secale cereal) cover crop vs. no cover crop). Soil samples were collected in late spring/early summer from each treatment at 10-cm depth increments from the soil surface to a depth of 40 cm using cores(76.2-mm diameter and 76.2-mm length). Soil bulk density was 13% lower with tillage compared with no-tillage. Volumetric water content was significantly higher at 0.0 and -0.4 k Pa pressures with tillage compared with no tillage. Tillage increased the proportion of coarse mesopores by 32% compared with no tillage, resulting in 87% higher saturated hydraulic conductivity(K_(sat)). Cover crop increased the proportion of macropores by 24% compared with no cover crop; this can potentially increase water infiltration and reduce runoff. As a result of higher macroporosity, Ksat was higher under cover crop compared with no cover crop. This study demonstrated that tillage can benefit soil hydraulic properties in the short term, but these effects may not persist over time. 17A Deep Learning Approach to Mesh Segmentation显示文摘In the shape analysis community,decomposing a 3D shape intomeaningful parts has become a topic of interest.3D model segmentation is largely used in tasks such as shape deformation,shape partial matching,skeleton extraction,shape correspondence,shape annotation and texture mapping.Numerous approaches have attempted to provide better segmentation solutions;however,the majority of the previous techniques used handcrafted features,which are usually focused on a particular attribute of 3Dobjects and so are difficult to generalize.In this paper,we propose a three-stage approach for using Multi-view recurrent neural network to automatically segment a 3D shape into visually meaningful sub-meshes.The first stage involves normalizing and scaling a 3D model to fit within the unit sphere and rendering the object into different views.Contrasting viewpoints,on the other hand,might not have been associated,and a 3D region could correlate into totally distinct outcomes depending on the viewpoint.To address this,we ran each view through(shared weights)CNN and Bolster block in order to create a probability boundary map.The Bolster block simulates the area relationships between different views,which helps to improve and refine the data.In stage two,the feature maps generated in the previous step are correlated using a Recurrent Neural network to obtain compatible fine detail responses for each view.Finally,a layer that is fully connected is used to return coherent edges,which are then back project to 3D objects to produce the final segmentation.Experiments on the Princeton Segmentation Benchmark dataset show that our proposed method is effective for mesh segmentation tasks.Abubakar Sulaiman Gezawa Qicong Wang Haruna Chiroma Yunqi Lei 2023Computer Modeling in Engineering & Sciences2023,,5:1
