| 1 | 人类皮下和网膜脂肪组织中脂联素基因的表达和调控的研究显示文摘目标:探讨脂联素在人类皮下和网膜脂肪组织中mRNA表达水平,以及糖皮质激素和PPARγ激动剂对脂联素的调控作用。方法:应用real-time摇RT-PCR方法观察了16名成人腹部皮下和网膜脂肪组织中脂联素mRNA表达水平以及与肥胖的关系。通过体外培养,观察地塞米松(dexamethasone,DEX)和/或PPARγ激动剂(troglitazone,TGZ)对不同部位脂肪组织脂联素的表达和分泌的调控作用。结果:皮下和网膜脂肪组织脂联素mRNA表达丰富但无显著部位差异,网膜脂肪组织脂联素mRNA水平与BMI呈显著负相关(r=0.56,P<0.05)。体外培养24h后,TGZ处理组脂肪组织脂联素基因的表达明显增加(皮下:70%;网膜:102%),且网膜脂肪组织脂联素的分泌明显高于对照组(P<0.05);DEX则下调网膜脂肪组织脂联素mRNA的表达水平36%,TGZ可逆转这种抑制作用。结论:网膜脂肪组织中脂联素mRNA的表达随BMI增加而下降,并受到糖皮质激素及PPARγ激动剂调控。脂联素的部位特异性表达和调控可能是内脏型肥胖连接胰岛素抵抗的重要分子机制。 | 李晓南 陈荣华 潘亚 Tommy Olsson Olle Hernell | 2006 | 南京医科大学学报(自然科学版)2006,26,1: | 12 |
| 2 | 人类皮下和网膜脂肪组织脂肪细胞因子的表达显示文摘目的探讨脂肪细胞因子在成人皮下和网膜脂肪组织中的表达水平。方法应用即刻定量PCR方法测定16例成人皮下和网膜脂肪组织中瘦素、脂联素、肿瘤坏死因子α(TNF-α)和抵抗素的表达水平。结果正常体重组和超重组皮下脂肪组织瘦素mRNA水平均明显高于网膜组织(P<0.01);正常组网膜脂肪组织脂联素mRNA水平较皮下组织有升高趋势(P=0.07);TNF-α和抵抗素mRNA表达水平低下且无显著部位差异。无论皮下或网膜脂肪组织瘦素表达水平均与体重指数(BMI)成正相关(皮下:r=0.68,P<0.01;网膜:r=0.71,P<0.01)),而网膜脂肪组织脂联素表达与BMI呈明显负相关(r=-0.60,P<0.05)。结论基因编码的一些细胞因子在脂肪组织中的表达呈部位特征,肥胖时皮下和网膜脂肪组织瘦素过度表达,而内脏脂肪组织脂联素表达下调,这可能是肥胖相关的胰岛素抵抗发生的潜在危险因素。 | 李晓南 陈荣华 Tommy Olsson Olle Hernell | 2005 | 江苏医药2005,31,12: | 5 |
| 3 | 儿童皮下和网膜脂肪组织瘦素表达水平及其临床意义显示文摘【目的】 对不同年龄儿童皮下和网膜脂肪组织中瘦素 (leptin)mRNA表达水平进行调查。 【方法】 配对的皮下和网膜脂肪组织从腹部择期手术患儿 ( 3 7例 ,3月~ 12岁 )和成人 ( 9名 )获取。应用real timeRT PCR方法测定不同部位脂肪组织中leptinmRNA表达水平 ,并探讨其与体块指数 (BMI)、腰围、臀围及腰臀围比 (WHR )、年龄、性别及肥胖的相互关系。 【结果】 与成人相一致 ,儿童leptin的表达亦有明显的部位性差异 ,皮下脂肪组织中leptinmRNA水平显著高于网膜 (P <0 .0 0 1)。皮下脂肪组织leptinmRNA表达水平与BMI呈显著正相关 (r =0 .69,P <0 .0 0 1) ,而网膜leptin与BMI呈较弱的正相关 (r =0 .3 2 ,P <0 .0 5 ) ;皮下和网膜leptin与WHR的均无显著相关 ,但网膜leptin与腰围呈较弱的正相关 (r =0 .3 3 ,P <0 .0 5 )。不同年龄组中脂肪组织的leptin表达水平差异有显著性(P <0 .0 1) ,6~ 12岁组皮下和网膜脂肪组织leptinmRNA表达均高于 1~ 5岁组 (P <0 .0 5 ) ,与成人水平相比 ,则差异无显著性 (P >0 .0 5 )。过重和肥胖儿童脂肪组织leptinmRNA表达水平显著高于非肥胖儿童。 【结论】 儿童不同部位脂肪组织leptinmRNA表达水平在一定程度上反映了脂肪组织的生长和分布。肥胖儿童脂肪组织leptin的? | 李晓南 陈荣华 Tommy Olsson Olle Hernel | 2004 | 中国儿童保健杂志2004,12,3: | 2 |
