| 1 | YTH Domain: A Family of N^6-methyladenosine (m^6A) Readers显示文摘Like protein and DNA, different types of RNA molecules undergo various modifications. Accumulating evidence suggests that these RNA modifications serve as sophisticated codes to mediate RNA behaviors and many important biological functions. N^6-methyladenosine(m^6 A)is the most abundant internal RNA modification found in a variety of eukaryotic RNAs, including but not limited to m RNAs, t RNAs, r RNAs, and long non-coding RNAs(lnc RNAs). In mammalian cells, m^6 A can be incorporated by a methyltransferase complex and removed by demethylases, which ensures that the m^6 A modification is reversible and dynamic. Moreover, m^6 A is recognized by the YT521-B homology(YTH) domain-containing proteins, which subsequently direct different complexes to regulate RNA signaling pathways, such as RNA metabolism, RNA splicing, RNA folding, and protein translation. | 2 | Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. | 3 | Engineering an enthesis-like graft for rotator cuff repair:An approach to fabricate highly biomimetic scaffold capable of zone-specifically releasing stem cell differentiation inducers显示文摘Rotator cuff(RC)attaches to humerus across a triphasic yet continuous tissue zones(bone-fibrocartilage-tendon),termed“enthesis”.Regrettably,rapid and functional enthesis regeneration is challenging after RC tear.The existing grafts bioengineered for RC repair are insufficient,as they were engineered by a scaffold that did not mimic normal enthesis in morphology,composition,and tensile property,meanwhile cannot simultaneously stimulate the formation of bone-fibrocartilage-tendon tissues.Herein,an optimized decellularization approach based on a vacuum aspiration device(VAD)was developed to fabricate a book-shaped decellularized enthesis matrix(O-BDEM).Then,three recombinant growth factors(CBP-GFs)capable of binding collagen were synthesized by fusing a collagen-binding peptide(CBP)into the N-terminal of BMP-2,TGF-β3,or GDF-7,and zone-specifically tethered to the collagen of O-BDEM to fabricate a novel scaffold(CBP-GFs/O-BDEM)satisfying the above-mentioned requirements.After ensuring the low immunogenicity of CBP-GFs/O-BDEM by a novel single-cell mass cytometry in a mouse model,we interleaved urine-derived stem cell-sheets into this CBP-GFs/O-BDEM to bioengineer an enthesis-like graft.Its high-performance on regenerating enthesis was determined in a canine model.These findings indicate this CBP-GFs/O-BDEM may be an excellent scaffold for constructing enthesis-like graft to patch large/massive RC tears,and provide breakthroughs in fabricating graded interfacial tissue. | Can Chen Qiang Shi Muzhi Li Yang Chen Tao Zhang Yan Xu Yunjie Liao Shulin Ding Zhanwen Wang Xing Li Chunfeng Zhao Lunquan Sun Jianzhong Hu Hongbin Lu | 2022 | Bioactive Materials2022,7,10: | 4 |

| 13 | Investigation on Strengthening Mechanism of China Low-Activation Ferrite Steel upon Thermo-Mechanical Treatment显示文摘The objective of this study was to investigate the influence of strengthening mechanisms on the high-temperature mechanical properties of China low-activation ferrite(CLF-1)steel,which underwent thermodynamic design and thermo-mechanical treatment(TMT).The microstructure characterization in the normalized and tempered condition and the TMT condition was carried out using optical microscopy,X-ray diffractometer,and scanning electron microscopy with electron backscatter diffraction.High-resolution transmission electron microscopy was employed to determine the crystallographic structures of precipitated phases.The results indicated that the addition of Ti led to an increase in the allocation of C in MC phase and an enhancement in the content of MC phase.Compared to CLF-P steel in the normalized and tempered condition,a 1.5-fold increase in dislocation density and an order of magnitude improvement in MX phase density were achieved after TMT.The formation of high-density nano-scale MC phases during TMT played a significant role in precipitation strengthening due to their favorable coherent relationship with the matrix and low interfacial free energy.The excellent high-temperature mechanical properties observed in CLF-P steel after TMT can be attributed to the combined effects of precipitation strengthening,dislocation strengthening,and lath strengthening. | Dongtian Yang Liangyin Xiong Hongbin Liao Guoping Yang Xiaoyu Wang Shi Liu | 2024 | Acta Metallurgica Sinica(English Letters)2024,37,2: | 0 |

| 15 | Effect of nozzle inlet parameters on the dry granulation atomization process of Si_(3)N_(4) ceramic bearing balls显示文摘In this paper,the atomization characteristics of Si3N4 ceramic dry granulation affect the performance of Si3N4 ceramic bearing balls.In order to improve the dry granulation characteristics and the comprehensive performance of Si3N4 ceramic bearing balls,the atomization mechanism of the spinning nozzle used for Si3N4 dry granulation was studied in detail.The interaction between air and binder in the pressure-swirl nozzle is analyzed based on VOF method,the modified realizable k−εturbulence model is used to simulate the flow field inside and outside the pressure-swirl nozzle,the effects of nozzle inlet parameters including the number of tangential inlets and the deflection angle of tangential inlets on the binder volume fraction,velocity distribution and pressure distribution are analyzed.The results show that when the number of tangential inlets increases from 1 to 4,the swirl strength of gas–liquid two-phase in the nozzle increases,the mean diameter of air core increases from 1.51 mm to 2.01 mm,and the spray cone angle increases from 18.5◦to 26.4◦.Besides,when the deflection angle of tangential inlet increases from 0◦to 15◦,the swirl strength of gas–liquid two-phase in the nozzle with the deflection angle of tangential inlet of 10◦is the largest,and the mean diameter of air core and spray cone angle is 3.04 mm and 30.7◦,respectively.Based on the atomization experiment platform of the electric control fuel system,the mean diameter of air core and spray cone angle are measured,the micromorphology of Si3N4 particles is observed,which verifies the correctness of numerical simulation.When the Si3N4 particles are prepared by dry granulation,taking the atomization performance of nozzle into consideration,the pressure-swirl nozzle with 4 tangential inlets and 10◦deflection angle should be selected. | Dongling Yu Xiaohui Zhang Hongbin Luo Dahai Liao Nanxing Wu | 2021 | International Journal of Modeling, Simulation, and Scientific Computing2021,12,6: | 0 |
