1Big Grain3, encoding a purine permease, regulates grain size via modulating cytokinin transport in rice显示文摘Grain size is an important agronomic trait affecting grain yield,but the underlying molecular mechanisms remain to be elucidated.Here,we isolated a dominant mutant,big grainy(bg3-D),which exhibits a remarkable increase of grain size caused by activation of the PURINE PERMEASE gene,OsPUP4.BC3/OSPUP4 is predominantly expressed in vascular tissues and is specifically suppressed by exogenous cytokinin application.Hormone profiling revealed that the distribution of different cytokinin forms,in roots and shoots of the bg3-D mutant,is altered.Quantitative reverse transcription-PCR(qRT-PCR)analysis indicated that expression of rice cytokinin type-A RESPONSE REGULATOR(OsRR)genes is enhanced in the roots of the bgj-D mutant.These results suggest that OSPUP4 might contribute to the long-distance transport of cytokinin,by reinforcing cytokinin loading into vascular bundle cells.Furthermore,plants overexpressing OsPUP7,the closest homolog of OsPUP4,also exhibited a similar phenotype to the bg3-D mutant.Interestingly,subcellular localization demonstrated that OSPUP4 was localized on the plasma membrane,whereas OSPUP7 was localized to the endoplasmic reticulum.Based on these findings,we propose that OSPUP4 and OSPUP7 function in a linear pathway to direct cytokinin cell-to-cell transport,affecting both its long-distance movement and local allocation.Yunhua Xiao Dapu Liu Guoxia Zhang Shaopei Gao Linchuan Liu Fan Xu Ronghui Che Yiqin Wang Hongning Tong Chengcai Chu 2019Journal of Integrative Plant Biology2019,61,5:7

3A cryptic inhibitor of cytokinin phosphorelay controls rice grain size显示文摘Plant hormone cytokinin signals through histidine-aspartic acid(H-D)phosphorelay to regulate plant growth and development.While it is well known that the phosphorelay involves histidine kinases,histidine phosphotransfer proteins(HPs),and response regulators(RRs),how this process is regulated by external components remains unknown.Here we demonstrate that protein phosphatase with kelch-like domains(PPKL1),known as a signaling component of steroid hormone brassinosteroid,is actually a cryptic inhibitor of cytokinin phosphorelay in rice(Oryza sativa).Mutation at a specific amino acid D364 of PPKL1 activates cytokinin response and thus enlarges grain size in a semi-dominant mutant named s48.Overexpression of PPKL1 containing D364,either with the deletion of the phosphatase domain or not,rescues the s48 mutant phenotype.PPKL1 interacts with OsAHP2,one of authentic HPs,and D364 resides in a region resembling the receiver domain of RRs.Accordingly,PPKL1 can utilize D364 to suppress OsAHP2-to-RR phosphorelay,whereas mutation of D364 abolishes the effect.This function of PPKL1 is independent of the phosphatase domain that is required for brassinosteroid signaling.Importantly,editing of the D364-residential region produces a diversity of semi-dominant mutations associated with variously increased grain sizes.Further screening of the edited plants enables the identification of two genotypes that confer significantly improved grain yield.Collectively,our study uncovers a noncanonical cytokinin signaling suppressor and provides a robust tool for seed rational design.Dapu Liu He Zhao Yunhua Xiao Guoxia Zhang Shouyun Cao Wenchao Yin Yangwen Qian Yanhai Yin Jinsong Zhang Shouyi Chen Chengcai Chu Hongning Tong 2022Molecular Plant2022,15,2:3
4Pretreatment with low-frequency repetitive transcranial magnetic stimulation may influence neuronal Bcl-2 and Fas protein expression in the CA1 region of the hippocampus: A possible anti-epilepsy mechanism in a lithium-pilocarpine-induced epileptic rat mod显示文摘BACKGROUND: Bcl-2 and Fas proteins are well known as anti-apoptotic and pro-apoptotic factors, respectively. However, whether the anti-epileptic mechanism of low-frequency repetitive transcranial magnetic stimulation (rTMS) involves an anti-apoptotic effect via regulating Bcl-2 and Fas protein expression remains to be determined. OBJECTIVE: To verify the correlation between the anti-epileptic mechanism following pretreatment of low-frequency rTMS and anti-hippocampal apoptosis. DESIGN, TIME AND SETTING: A randomized controlled animal experiment was performed at Institute of Neurological Disorders, Affiliated Hospital of North Sichuan Medical College between September 2007 and March 2008. MATERIALS: Pilocarpine (053K13011) was provided by Sigma, USA; lithium was provided by Shanghai Biotechnology Co., Ltd., China; Dantec Maglite-r25 rTMS instrument was provided by Dundee, Denmark. METHODS: A total of 21 adult male Wistar rats were randomly divided into control (n = 6), rTMS pretreatment (n = 9), and sham-stimulation (n = 6) groups. The rTMS pretreatment group was pretreated with low-frequency rTMS (0.5 Hz, 75% threshold intensity, 20 times/bundle, and 5 bundles/day), while the sham-stimulation group was sham-stimulated with a similar sound for 7 successive days to establish lithium-pilocarpine-induced epileptic state models. MAIN OUTCOME MEASURES: Epileptic stroke latency; neuronal morphology was observed using hematoxylin and eosin staining; mean positive-reactive cell number and mean absorbance of Bcl-2 and Fas protein in the hippocampal CA1 region was observed using immunohistochemistry. RESULTS: Epileptic latency in the rTMS pretreatment group was significantly enhanced (P < 0.01), and a number of degenerated neurons were observed to be apoptotic. Bcl-2 protein expression increased at each time point, but Fas protein expression decreased (P < 0.01). 5The U-box ubiquitin ligase TUD1 promotes brassinosteroid-induced GSK2 degradation in rice显示文摘Brassinosteroids(BRs)are a class of steroid hormones with great potential for use in crop improvement.De-repression is usually one of the key events in hormone signaling.However,how the stability of GSK2,the central negative regulator of BR signaling in rice(Oryza sativa),is regulated by BRs remains elusive.Here,we identify the U-box ubiquitin ligase TUD1 as a GSK2-interacting protein by yeast two-hybrid screening.We show that TUD1 is able to directly interact with GSK2 and ubiquitinate the protein.Phenotypes of the tud1 mutant are highly similar to those of plants with constitutively activated GSK2.Consistent with this finding,GSK2 protein accumulates in the tud1 mutant compared with the wild type.In addition,inhibition of BR synthesis promotes GSK2 accumulation and suppresses TUD1 stability.By contrast,BRs can induce GSK2 degradation but promote TUD1 accumulation.Furthermore,the GSK2 degradation process is largely impaired in tud1 in response to BR.In conclusion,our study demonstrates the role of TUD1 in BR-induced GSK2 degradation,thereby advancing our understanding of a critical step in the BR signaling pathway of rice.Dapu Liu Xiaoxing Zhang Qingliang Li Yunhua Xiao Guoxia Zhang Wenchao Yin Mei Niu Wenjing Meng Nana Dong Jihong Liu Yanzhao Yang Qi Xie Chengcai Chu Hongning Tong 2023Plant Communications2023,4,2:2

8Malaria from hyperendemicity to elimination along international borders in Yunnan, China during 2003‒2020: a case study显示文摘Background: Border malaria is one of the most intractable problems hindering malaria elimination worldwide.Movement of both the human population and anopheline mosquitoes infected with Plasmodium spp.can cause cross-border malaria transmission.The Yunnan border area was still hyperendemic for malaria in the early part of this century.The objective of this case study was to analyze the strategies,interventions and impacts of malaria control and elimination in the Yunnan border area.Main text: A total of 10,349 malaria cases and 17.1 per 10,000 person-years of annual parasite incidence(API)were reported in the border area in 2003.Based on natural village-based stratification,integrated interventions,including mass drug administration for radical cures and preventive treatment,clinically presumptive treatment of all febrile patients for malaria and indoor residual spraying or dipping bed nets with insecticides were successfully carried out from 2003 to 2013.The overall API was reduced to 0.6 per 10,000 person-years by 2013,while effective cross-border collaboration interventions dramatically reduced the malaria burden in the neighbouring border areas of Myanmar.From 2014 forward,the comprehensive strategy,including universal coverage of surveillance to detect malaria cases,a rapid response to possible malaria cases and effective border collaboration with neighbouring areas,successfully eliminated malaria and prevented reintroduction of malaria transmission in the Yunnan border area.Conclusions: In Yunnan malaria burden has successfully reduced by dynamically accurate stratification and comprehensive interventions;and then the region achieved elimination and prevented reintroduction of malaria transmission through intensive surveillance,rapid response and border collaboration.Other border areas should perform their own intervention trials to develop their own effective strategy.Hui Liu Yaowu Zhou Yan Deng Zurui Lin Canglin Zhang Qiyan Chen Chun Wei Kaixia Duan Peng Tian Hongning Zhou Jianwei Xu 2022Infectious Diseases of Poverty2022,11,3:1

12Simultaneous quantifi cation and evaluate the differences of two skeleton components in raw,salt and wine Angelicae Pubescentis Radix by UPLC-MS/MS in negative/positive modes coupled with chemometrics显示文摘The aim of this study was to establish an ultra-performance liquid chromatography coupled with triple-quadrupole tandem mass spectrometry method with positive and negative ion exchange scanning mode for simultaneous quantification of two different skeleton components to reveal the differences among raw and processed Angelicae Pubescentis Radix. The results of methodology showed that each compound had good linearity and recovery rates. And the chemometrics results indicate that there were differences not only in Angelicae Pubescentis Radix samples from different sources but also in raw and processed Angelicae Pubescentis Radix. The results of this study confirm that the method has been successfully applied in simultaneous determination and discovering the difference of two different skeleton components between raw and processed Angelicae Pubescentis Radix for the first time. 13Sympathetic skin response in patients with myasthenia gravis: A comparative analysis显示文摘BACKGROUND:The examination of sympathetic skin response is an important index for assessing the autonomic nerve function,and patients with myasthenia gravis are always accompanied by dysautonomia. Therefore,it will be important to know whether sympathetic skin response can be used as the index for the clinical evaluation of myasthenia gravis. OBJECTIVE:To investigate the diagnostic value of sympathetic skin response in the damage of autonomic nerve function of patients with myasthenia gravis. DESIGN:A case-controlled comparative observation. SETTING:Department of Neurology and Room of Nerve Electromyogram,the Affiliated Hospital of North Sichuan Medical College. PARTICIPANTS:Thirty outpatients or inpatients with myasthenia gravis were selected from the Department of Neurology,the Affiliated Hospital of North Sichuan Medical College from May 2006 to May 2007,including 9 males and 21 females,aged 8-72 years with a mean age of(28±5) years old. They were all accorded with the diagnostic standards of myasthenia gravis,accompanied by different severity of autonomic nerve symptoms,including poor skin nutrition,sweating of hands and feet,pyknocardia,persistent hypotension,abdominal pain,constipation,etc. They all had not taken any drug affecting the autonomic nerve function before the examination. Informed consents were obtained from all the patients. Meanwhile,30 healthy physical examinees were enrolled as the normal control group,including 10 males and 20 females,aged 10-75 years with a mean age of(31±5) years old. Approval was obtained from the hospital ethic committee. METHODS:After admission,the patients were examined with sympathetic skin response using DANTEC keypoint 2.0 electromyography evoked potential apparatus(Danmark). The changes of the latency and wave amplitude of sympathetic skin response were observed. The subjects in the normal control group were examined with the same methods at physical examination. Abnormality was judged by the disappearance of wave form,latency longer than that in the normal control group by Mean±2.5SD,or wave amplitude lower than the average value in the normal control group by 50%. MAIN OUTCOME MEASURES:The results of the latency and wave amplitude of sympathetic skin response were compared between the patients with myasthenia gravis and normal controls. RESULTS:All the 30 patients with myasthenia gravis and 30 healthy physical examinees were involved in the final analysis of results. There were no significant differences between the left and right upper and lower limbs in both the myasthenia gravis group and normal control group(P > 0.05). In the myasthenia gravis group,the abnormal rate of sympathetic skin response was 37%(11/30),the latency was prolonged and the wave amplitude was decreased as compared with those in the normal control group,and there were significant differences(P < 0.01). 15Histological Assessment and Transcriptome Analysis Provide Insights into the Toxic Effects of Perfluorooctanoic Acid to Juvenile Half Smooth Tongue Sole Cynoglossus semilaevis显示文摘Perfluorooctanoic acid(PFOA)is a widespread synthetic persistent organic pollutant that may enrich along the food chain and affect the growth,development,reproduction,and lipid metabolism of aquatic organisms,particularly the benthic organisms.How-ever,the toxic effects of PFOA on the half-smooth tongue sole Cynoglossus semilaevis,a commercial benthic fish in China,have rarely been reported.Because juvenile fish are sensitive to environmental pollutants,in the present study,histological assessment and tran-scriptome sequencing were performed to determine the short-term impact of PFOA on juvenile half-smooth tongue soles.Histologi-cal analysis showed that PFOA exposure caused hepatocyte rupture,intestinal villi breakage,increased goblet cell count,and brain ab-normal.Transcriptome results showed that some interesting signaling pathways,such as glycolysis/gluconeogenesis,PPAR signaling pathway and GABAergic synapse signaling pathway,were enriched after PFOA exposure.In addition,some metabolic,immune and neural genes were differentially expressed,which including ependymin,hbb1-like and gad 1,and they were up-regulated after 14 days of exposure.Transcriptome results also indicated that half-smooth tongue sole might improve energy metabolism in response to PFOA toxicity after 7 days of exposure.These findings provide a basis for studying the ecological effects of PFOA on marine benthic fishes.ZHAN Min SHI Kunpeng ZHANG Xue FAN Qingxin XU Qian LIU Xinbao LI Zhujun LIU Hongning XIA Yanting SHA Zhenxia 2023Journal of Ocean University of China2023,22,6:0
16The divergence of brassinosteroid sensitivity between rice subspecies involves natural variation conferring altered internal auto-binding of OsBSK2显示文摘Japonica/geng and indica/xian are two major rice(Oryza sativa)subspecies with multiple divergent traits,but how these traits are related and interact within each subspecies remains elusive.Brassinosteroids(BRs)are a class of steroid phytohormones that modulate many important agronomic traits in rice.Here,using different physiological assays,we revealed that japonica rice exhibits an overall lower BR sensitivity than indica.Extensive screening of BR signaling genes led to the identification of a set of genes distributed throughout the primary BR signaling pathway with divergent polymorphisms.Among these,we demonstrate that the C38/T variant in BR Signaling Kinase2(OsBSK2),causing the amino acid change P13L,plays a central role in mediating differential BR signaling in japonica and indica rice.OsBSK2in indica plays a greater role in BR signaling than OsB SK2in japonica by affecting the auto-binding and protein accumulation of OsBSK2.Finally,we determined that OsBSK2 is involved in a number of divergent traits in japonica relative to indica rice,including grain shape,tiller number,cold adaptation,and nitrogen-use efficiency.Our study suggests that the natural variation in OsB SK2 plays a key role in the divergence of BR signaling,which underlies multiple divergent traits between japonica and indica.Wenchao Yin Lulu Li Zhikun Yu Fan Zhang Dapu Liu Hongkai Wu Mei Niu Wenjing Meng Xiaoxing Zhang Nana Dong Yanzhao Yang Jihong Liu Yongqiang Liu Guoxia Zhang Jianlong Xu Shimei Wang Chengcai Chu Qian Qian Hongning Tong 2022Journal of Integrative Plant Biology2022,64,8:0

18Identifying Game Processes Based on Private Working Sets显示文摘Fueled by the booming online games,there is an increasing demand for monitoring online games in various settings.One of the application scenarios is the monitor of computer games in school computer labs,for which an intelligent game recognition method is required.In this paper,a method to identify game processes in accordance with private working sets(i.e.,the amount of memory occupied by a process but cannot be shared among other processes)is introduced.Results of the W test showed that the memory sizes occupied by the legitimate processes(e.g.,the processes of common native windows applications)and game processes followed normal distribution.Using the T-test,a significant difference was identified between the legitimate processes and C/S-based computer games,in terms of the means and variances of their private working sets.Subsequently,we derived the density functions of the private working sets of the considered game processes and those of the legitimate processes.Given the private working set of a process and the derived probability density functions,the probability that the process is a legitimate process and the probability that the process is a game process can be determined.After comparing the two probabilities,we can easily determine whether the process is a game process or not.As revealed from the test results,the recognition accuracy of this method for C/S-based computer games was approximately 90%.Jinfeng Li Li Feng Longqing Zhang Hongning Dai Lei Yang Liwei Tian 2020Computers, Materials & Continua2020,,10:0
