1Promotion of the articular cartilage proteoglycan degradation by T-2 toxin and selenium protective effect显示文摘Objective: To identify the relationship between T-2 toxin and Kashin-Beck disease (KBD),the effects of T-2 toxin on aggrecan metabolism in human chondrocytes and cartilage were investigated in vitro. Methods: Chondrocytes were isolated from human articular cartilage and cultured in vitro. Hyaluronic acid (HA),soluble CD44 (sCD44),IL-1β and TNF-α levels in super-natants were measured by enzyme-linked immunosorbent assay (ELISA). CD44 content in chondrocyte membrane was deter-mined by flow cytometry (FCM). CD44,hyaluronic acid synthetase-2 (HAS-2) and aggrecanases mRNA levels in chondrocytes were determined using reverse transcription polymerase chain reaction (RT-PCR). Immunocytochemical method was used to investigate expressions of BC-13,3-B-3(-) and 2-B-6 epitopes in the cartilage reconstructed in vitro. Results: T-2 toxin inhibited CD44,HAS-2,and aggrecan mRNA expressions,but promoted aggrecanase-2 mRNA expression. Meanwhile,CD44 expression was found to be the lowest in the chondrocytes cultured with T-2 toxin and the highest in control plus selenium group. In addition,ELISA results indicated that there were higher sCD44,IL-1β and TNF-α levels in T-2 toxin group. Similarly,higher HA levels were also observed in T-2 toxin group using radioimmunoprecipitation assay (RIPA). Furthermore,using monoclonal antibodies BC-13,3-B-3 and 2-B-6,strong positive immunostaining was found in the reconstructed cartilage cultured with T-2 toxin,whereas no positive staining or very weak staining was observed in the cartilage cultured without T-2 toxin. Selenium could partly inhibit the effects of T-2 toxin above. Conclusion: T-2 toxin could inhibit aggrecan synthesis,promote aggrecanases and pro-inflammatory cytokines production,and consequently induce aggrecan degradation in chondrocytes. 2Constituents of the anti-asthma herbal formula ASHMI^(TM) synergistically inhibit IL-4 and IL-5 secretion by murine Th2 memory cells,and eotaxin by human lung fibroblasts in vitro显示文摘OBJECTIVE:Anti-asthma herbal medicine intervention(ASHMI ),a combination of three traditional Chinese medicinal herbs developed in our laboratory,has demonstrated efficacy in both mouse models of allergic asthma,and a double-blind placebo-controlled clinical trial in patients with asthma.This study was designed to determine if the anti-inflammatory effects of individual herbal constituents of ASHMI? exhibited synergy. METHODS:Effects of ASHMI and its components aqueous extracts of Lingzhi(Ganoderma lucidum),Kushen(Sophora flavescens) and Gancao(Glycyrrhiza uralensis),on Th2 cytokine secretion by murine memory Th2 cells(D10.G4.1) and eotaxin-1 secretion by human lung fibroblast(HLF-1) cells were determined by measuring levels in culture supernatants by enzymelinked immunosorbent assay.Potential synergistic effects were determined by computing interaction indices from concentration-effect curve parameters. RESULTS:Individual Lingzhi,Kushen and Gancao extracts and ASHMI(the combination of individual extracts) inhibited production of interleukin(IL)-4 and IL-5 by murine memory Th2 cells and eotaxin-1 production by HLF-1 cells.The mean 25%-inhibitory-concentration(IC25) values(mg/mL) for ASHMI,Lingzhi,Kushen and Gancao for IL-4 production were 30.9,79.4, 123,and 64.6,respectively;for IL-5 production were 30.2,263,123.2 and 100,respectively; for eotaxin-1 were 13.2,16.2,30.2,and 25.1,respectively.The IC50 values(mg/mL) for ASHMI,Lingzhi,Kushen and Gancao for IL-4 production were 158.5,239.9,446.7,and 281.8, respectively;for eotaxin-1 were 38.1,33.1,100,and 158.5,respectively.The interaction indices of ASHMI constituents at IC25 were 0.35 for IL-4,0.21 for IL-5 and 0.59 for eotaxin-1.The interaction indices at IC50 values were 0.50 for IL-4 and 0.62 for eotaxin-1 inhibition.Inhibition of IL-5 did not reach IC50 values.All interaction indices were below 1 which indicated synergy. 3饮水微量元素与老年人群认知能力关系研究显示文摘在中国山东与河南两省农村研究了饮水中微量元素与老年人群认知能力 (CognitiveFunction)之间的关系。采用中国国标方法分析了饮水中氟、钙、硒、铝、铁、锌、镉、铅、砷微量元素和pH值 ;用老年性痴呆症 (Dementia)调查表 ,进行了≥ 6 5岁老年暴露人群认知能力的测定。调整年龄、性别和文化水平后 ,对微量元素与老年认知能力之间的关系进行直线模型、二次方模型和联合作用模型的统计分析 ,评价饮水中每种微量元素浓度对老年认知能力的影响。结果表明 ,单因素分析时发现几种微量元素对老年认知能力有明显作用。然而 ,在调整其它元素水平后 ,其中一些元素的作用消失。饮水中钙浓度对老年认知能力呈现一作用曲线 ,即随着饮水钙水平升高而认知能力增加 ;到某一点高峰后 ,又随钙水平增加而认知能力降低。而锌与镉之间存在一种相互作用 ,即在低饮水镉水平时 ,锌浓度与认知能力呈正相关 ,但在高饮水镉水平时 。梁超轲 吉荣娣 曹静祥 姜玉亭 余波 马凤 吴云鹏 应波 张妍 孙淑庄 Li Yiming Christine L Emsley Gao Sujuan Kathleen S Hall Hugh C Hendrie 2003卫生研究2003,32,5:12
4Isolation of bacteria from fermented food and grass carp intestine and their efficiencies in improving nutrient value of soybean meal in solid state fermentation显示文摘Background: Soybean meal is an excellent and cost-effective protein source; however, its usage is limited in the piglet due to the presence of anti-nutritional factors and the antigens glycinin and β-conglycinin. The objective of the current study was to screen and select for bacteria that can be efficiently adopted to ferment soybean meal in order to solve this problem.Results: Bacteria were isolated from fermented soy foods and the grass carp intestine, and strains selected for high protease, cellulase and amylase activities. The isolated bacteria were characterized as Bacillus cereus, Bacillus subtilis and Bacilus amyloliquefacien, respectively. Fermentation with food-derived Isolate-2 and fish-derived F-9 increased crude protein content by 5.32% and 8.27%, respectively; improved the amino acid profile by increasing certain essential amino acids, broke down larger soy protein to 35 k Da and under, eliminated antigenicity against glycinin and β-conglycinin, and removed raffinose and stachyose in the soybean meal following a 24-h fermentation.Conclusions: Our results suggest these two B. amyloliquefaciens bacteria can efficiently solid state ferment soybean meal and ultimately produce a more utilizable food source for growing healthy piglets.Samantha Medeiros Jingjing Xie Paul W.Dyce Hugh Y.Cai Kees De Lange Hongfu Zhang Julang Li 2018Journal of Animal Science and Biotechnology2018,9,3:12
5Identification of Sheep Ovary Genes Potentially Associated with Off-season Reproduction显示文摘Off-season reproduction is a favorable economic trait for sheep industry.Hu sheep,an indigenous Chinese sheep breed,demonstrates a higher productivity of lambs and displays year-around oestrous behavior under proper nutrition and environment.The genetic basis behind these traits,however,is not well understood.In order to identify genes associated with the off-season reproduction,we constructed a suppression subtractive hybridization(SSH) cDNA library using pooled ovary mRNAs of 6 oestrous Hu females as a tester and the pooled ovary mRNAs of 6 non-oestrous Chinese Merino females as a driver.A total of 382 resulting positive clones were obtained after the SSH.We identified 114 differentially up-regulated genes in oestrous Hu sheep by using subsequent screening and DNA sequencing,of which 8 were previously known,93 were reported for the first time in sheep,and 13 were novel with no significant homology to any sequence in the DNA databases.Functions of the genes identified are related to cell division,signal transduction,structure,metabolism,or cell defense.To validate the results of SSH,6 genes(Ntrk2,Ppap2b,Htra1,Nid1,Serpine2 and Foxola) were selected for conformational analysis using quantitative real-time PCR(qRT-PCR),and two of them(Htral and Foxola) were verified by Northern blot.All of the 6 genes were differentially up-regulated in the ovary of oestrous Hu.It is obvious that off-season reproduction is a complex trait involving multiple genes in multiple organs.This study helps to provide a foundation for the final identification of functional genes involved in the sheep ovary.Lei Chen Ka Liu Zongsheng Zhao Hugh T.Blair Peng Zhang Daquan Li Runlin Z.Ma 2012Journal of Genetics and Genomics2012,39,4:3
6Increased expression of chondroitin sulphate proteoglycans in rat hepatocellular carcinoma tissues显示文摘AIM:To investigate the expression of chondroitin sulphate proteoglycans(CSPGs)in rat liver tissues of hepatocellular carcinoma(HCC).METHODS:Thirty male Sprague Dawley rats were randomly divided into two groups:control group(n=10) and HCC model group(n=20).Rats in the HCC model groups were intragastrically administrated with 0.2%(w/v)N-diethylnitrosamine(DEN)every 5 d for 16 wk,whereas 0.9%(w/v)normal saline was administered to rats in the control group.After 16 wk from the initiation of experiment,all rats were killed and livers were collected and fixed in 4%(w/v)paraformaldehyde.All tissues were embedded in paraffin and sectioned.Histological staining(hematoxylin and eosin and Toluidine blue)was performed to demonstrate the onset of HCC and the content of sulphated glycosaminoglycan(sGAG).Immunohistochemical staining was performed to investigate the expression of chondroitin sulphate(CS)/dermatan sulphate(DS)-GAG,heparan sulphate(HS)-GAG,keratan sulphate(KS)-GAG in liver tissues.Furthermore,expression and distribution of CSPG family members,including aggrecan,versican,biglycan and decorin in liver tissues,were also immunohistochemically determined.RESULTS:After 16 wk administration of DEN,malignant nodules were observed on the surface of livers from the HCC model group,and their hepatic lobule structures appeared largely disrupted under microscope.Toluidine blue staining demonstrated that there was an significant increase in sGAG content in HCC tissues when compared with that in the normal liver tissues from the control group[0.37±0.05 integrated optical density per stained area(IOD/area)and 0.21± 0.01 IOD/area,P<0.05].Immunohistochemical studies demonstrated that this increased sGAG in HCC tissues was induced by an elevated expression of CS/DS(0.28±0.02 IOD/area and 0.18±0.02 IOD/area,P< 0.05)and HS(0.30±0.03 IOD/area and 0.17±0.02 IOD/area,P<0.01)but not KS GAGs in HCC tissues.Further studies thereby were performed to investigate the expression and distribution of several CSPG components in HCC tissues,including aggrecan,versican,biglycan and decorin.Interestingly,there was a distinct distribution pattern for these CSPG components between HCC tissues and the normal tissues.Positive staining of aggrecan,biglycan and decorin was localized in hepatic membrane and/or pericellular matrix in normal liver tissues;however,their expression was mainly observed in the cytoplasm,cell membranes in hepatoma cells and/or pericellular matrix within HCC tissues.Semi-quantitative analysis indicated that there was a higher level of expression of aggrecan(0.43± 0.01 and 0.35±0.03,P<0.05),biglycan(0.32±0.01 and 0.25±0.01,P<0.001)and decorin(0.29±0.01 and 0.26±0.01,P<0.05)in HCC tissues compared with that in the normal liver tissues.Very weak versican positive staining was observed in hepatocytes near central vein in normal liver tissues;however there was an intensive versican distribution in fibrosis septa between the hepatoma nodules.Semi-quantitative analysis indicated that the positive rate of versican in hepatoma tissues from the HCC model group was much higher than that in the control group(33.61%and 21.28%,P <0.05).There was no positive staining in lumican and keratocan,two major KSPGs,in either normal or HCC liver tissues.CONCLUSION:CSPGs play important roles in the onset and progression of HCC,and may provide potential therapeutic targets and clinical biomarkers for this prevalent tumor in humans.Xiao-Li Jia Si-Yuan Li Shuang-Suo Dang Yan-An Cheng Xin Zhang Wen-Jun Wang Clare E Hughes Bruce Caterson 2012World Journal of Gastroenterology2012,18,30:2
7Occurrence and influence of residual gas released by crush methods on pore structure in Longmaxi shale in Yangtze Plate, Southern China显示文摘The composition of gas released under vacuum by crushing from the gas shale of Longmaxi Formation in Upper Yangtze Plate,Southern China was systematically investigated in this study.The effect of residual gas release on pore structures was checked using low-pressure nitrogen adsorption techniques.The influence of particle size on the determination of pore structure characteristics was considered.Using the Frenkel-Halsey-Hill method from low-pressure nitrogen adsorption data,the fractal dimensions were identified at relative pressures of 0‒0.5 and 0.5‒1 as D1 and D2,respectively,and the evolution of fractal features related to gas release was also discussed.The results showed that a variety component of residual gas was released from all shale samples,containing hydrocarbon gas of CH4(29.58%‒92.53%),C2H6(0.97%‒2.89%),C3H8(0.01%‒0.65%),and also some non-hydrocarbon gas such as CO2(3.54%‒67.09%)and N2(1.88%‒8.07%).The total yield of residual gas was in a range from 6.1μL/g to 17.0μL/g related to rock weight.The geochemical and mineralogical analysis suggested that the residual gas yield was positively correlated with quartz(R^2=0.5480)content.The residual gas released shale sample has a higher surface area of 17.20‒25.03 m^2/g and the nitrogen adsorption capacity in a range of 27.32‒40.86 ml/g that is relatively higher than the original samples(with 9.22‒16.30 m^2/g and 10.84‒17.55 ml/g).Clearer hysteresis loop was observed for the original shale sample in nitrogen adsorption-desorption isotherms than residual gas released sample.Pore structure analysis showed that the proportions of micro-,meso-and macropores were changed as micropores decreased while meso-and macropores increased.The fractal dimensions D1 were in range from 2.5466 to 2.6117 and D2 from 2.6998 to 2.7119 for the residual gas released shale,which is smaller than the original shale.This factor may indicate that the pore in residual gas released shale was more homogeneous than the original shale.The results indicated that both residual gas and their pore space have few contributions to shale gas production and effective reservoir evaluation.The larger fragments samples of granular rather than powdery smaller than 60 mesh fraction of shale seem to be better for performing effective pore structure analysis to the Longmaxi shale.Ming-liang Liang Zong-xiu Wang Guo-dong Zheng Hugh Christopher Greenwell Hui-jun Li Lin-yan Zhang Xing-qiang Feng Kai-xun Zhang 2020China Geology2020,3,4:2
8Inflammatory response to coronary artery bypass surgery: Does the heme-oxygenase-1 gene microsatellite polymorphism play a role?显示文摘Background Heme-oxygenase 1 (HO-1) is a rate-limiting enzyme in the degradation of heme to bilirubin, ferritin and carbon monoxide (CO) and may have significant anti-inflammatory function. The HO-1 gene promoter region shows microsatellite polymorphism with different (GT)n repeats, reported to differently induce gene expression, with the short allele associated with higher gene expression. We measured the acute inflammatory response using coronary artery bypass surgery (CABG) as a well-characterized and uniform stimulus and examined the correlation between levels of IL-6, C-reactive protein (CRP) and fibrinogen and their relationship to HO-1 genotype.Methods Two hundred and seventy-five consecutive patients undergoing CABG were genotyped for the HO-1 promoter polymorphism using PCR and automated DNA capillary sequencer. IL-6, CRP and fibrinogen were measured at baseline and 6, 24, 48, 72, 96 and 120 hours after CABG.Results Complete IL-6,CRP and fibrinogen measures were available in 220 patients. Before surgery IL-6 levels showed a strong correlation with CRP and fibrinogen ( r =0.48, P <0.0001; r =0.41, P <0.0001 respectively), with a significant correlation between CRP and fibrinogen ( r =0.61, P <0.0001). All three acute phase reactants showed a significant increase after CABG. After surgery, peak IL-6 was strongly correlated with peak CRP ( r =0.34, P =0.0009) but not with peak fibrinogen ( r =0.15, P =0.13), while peak CRP and peak fibrinogen were significantly correlated ( r =0.415, P <0.0001). HO-1 allelic repeats ranged from 22-42, with (GT) 25 and (GT) 32 being the two most common alleles, and subsequently divided into three groups according to previous published work: <30(GT)n were designated as S (short), 30-37(GT)n as M (middle) and long repeats with >37(GT)n as L (long); allele frequency 0.35, 0.58 and 0.07 respectively. Baseline CRP differed by genotype: those carrying at least one long allele having higher CRP than those with no long allele (3.76±0.79 vs. 2.07±0.17, P =0.013). Conversely, those carrying at least one short allele had higher fibrinogen levels than those with no short allele (3.83±0.79 vs. 3.51±0.88, P =0.006). Conclusions There is a strong correlation between the measured acute phase reactants both at baseline and after the inflammatory response to CABG in patients with coronary disease. There was an association between the HO-1 microsatellite polymorphism and CRP and fibrinogen levels at baseline but there was no similar association following CABG.
