| 3 | Effects of aminoguanidine on nitric oxide production induced by inflammatory cytokines and endotoxin in cultured rat hepatocytes显示文摘AIM To study the effects of aminoguanidine(AG) and two L-arginine analogues Nω-nitro-L-arginine methyl ester (L-NAME) and Nω-nitro-L-arginine (L-NNA) on nitric oxide (NO) productioninduced by cytokines (TNF-α, IL-11β, and IFN-γ)and bacterial lipopolysaccharide (LPS) mixture(CM) in the cultured rat hepatocytes, andexamine their mechanisms action.METHODS Rat hepatocytes were incubatedwith AG, L-NAME, L-NNA, Actinomycin D (ActD)and dexamethasene in a medium containing CM(LPS plus TNF-α, IL-1β, and IFN-γ) for 24 h. NOproduction in the cultured supernatant wasmeasured with the Griese reaction. IntracellularcGMP level was detected with radioimmunoasey.RESULTS NO production was markedlyblocked by AG and L-NAME in a dose-dependentmanner under inflammatory stimuli conditiontriggered by CM in vitro. The rate of themaximum inhibitory effects of L-NAME (38.9%)was less potent than that obtained with AG(53.7%, P<0.05). There was no significantdifference between the inhibitory effects of AGand two L-arginine analogues on intracellularcGMP accumulation in rat cultured hepatocytes.Non-specific NOS expression inhibitordexamethasone ( DEX ) and iNOS mRNAtranscriptional inhibitor ActD also significantlyinhibited CM-induced NO production. | 5 | Fabrication of homogeneously dispersed graphene/Al composites by solution mixing and powder metallurgy显示文摘Graphene-reinforced aluminum(Al) matrix composites were successfully prepared via solution mixing and powder metallurgy in this study. The mechanical properties of the composites were studied using microhardness and tensile tests. Compared to the pure Al alloy, the graphene/Al composites showed increased strength and hardness. A tensile strength of 255 MPa was achieved for the graphene/Al composite with only 0.3 wt% graphene, which has a 25% increase over the tensile strength of the pure Al matrix. Raman spectroscopy, Fourier transform infrared spectroscopy, scanning electron microscopy, and transmission electron microscopy were used to investigate the morphologies, chemical compositions, and microstructures of the graphene and the graphene/Al composites. | 6 | Timing of IUI Treatment after hCG Administration 1-48h Affecting Pregnancy Rate显示文摘Objective To compare the different time of administration of hCG affecting pregnancy rate of IUI. Methods A total of 189 infertile couples underwent 331 cycles of IUI with husband's sperm. They were separated into 3 groups according to the time of hCG administration in IUI: hCG 1-23 h (group A); hCG 24-36 h (group B); hCG 37-48 h (group C). Results There were no statistical differences among 3 groups. None of the other relative factors, such as the female age, the different methods of ovulation and the cause of infertility, showed differences in pregnancy rate among 3 groups. | 8 | Baihui(DU20)-penetrating-Qubin(GB7) acupuncture inhibits apoptosis in the perihemorrhagic penumbra显示文摘Baihui(DU20)-penetrating-Qubin(GB7) acupuncture can inhibit inflammatory reactions and activate signaling pathways related to proliferation after intracerebral hemorrhage.However,there is no research showing the relationship between this treatment and cell apoptosis.Rat models of intracerebral hemorrhage were established by injecting 60 μL of autologous blood into the right side of the caudate-putamen.Six hours later,the needle traveled subcutaneously from the Baihui acupoint to Qubin acupoint.The needle was alternately rotated(180 ± 10 turns/min) manually along clockwise and counter-clockwise directions.Stimulation lasted for 7 days,and was performed three times each for 6 minutes with 6-minute intervals between stimulations.Rats intraperitoneally receiving Sonic hedgehog pathway activator,purmorphamine(1 mg/kg per day),served as positive controls.Motor and sensory function were assessed using the Ludmila Belayev test.Extent of pathological changes were measured in the perihemorrhagic penumbra using hematoxylin-eosin staining.Apoptosis was examined by terminal deoxynucleotidyl transferase(Td T)-mediated d UTP nick end labeling assay.Expression of smoothened(Smo) and glioma-associated homolog 1(Gli1) was determined by western blot assay.Our results showed that Baihui-penetrating-Qubin acupuncture promoted recovery of motor and sensory function,reduced the apoptotic cell percentage in the perihemorrhagic penumbra,and up-regulated Smo and Gli1 expression.We conclude that Baihui-penetrating-Qubin acupuncture can mitigate hemorrhage and promote functional recovery of the brain in a rat model of intracerebral hemorrhage,possibly by activating the Sonic hedgehog pathway. | Beng Zhang Xiao-Hong Dai Xue-Ping Yu Wei Zou Wei Teng Xiao-Wei Sun Wei-Wei Yu Hao Liu Hui Wang Meng-Juan Sun Meng Li | 2018 | Neural Regeneration Research2018,13,9: | 9 |

| 10 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 11 | Isotopic composition of organic carbon and elemental carbon in PM2.5 in Hangzhou, China显示文摘In order to identify the sources of organic carbon (OC) and elemental carbon (EC) in airborne PM2.5 in Hangzhou, OC and EC were oxidized to CO2 offline in two thermal steps, and their carbon isotope compositions were measured by mass spectrometer. Results showed that the average δ13COC values were -52.8‰, -48.1‰, -50.0‰ and -52.5‰ in spring, summer, autumn, and winter, respectively, and the annual average value was -50.9‰ (-85.0‰ to -35.6‰), whereas δ13CEC values were -26.4‰, -26.9‰, -26.7‰ and -25.9‰ in the four seasons, and its annual value was -26.5‰ (-30.5‰ to -23.8‰). Large differences were found between isotope compositions of the two types of carbon. δ13COC values were much smaller than those of δ13CEC, and varied in a large range, and the ratios in both spring and winter were the smallest. δ13CEC varied in a small range, and its mean values in four seasons were nearly the same. | 12 | Association of genotypes of rs671 within ALDH2 with risk for gastric cardia adenocarcinoma in the Chinese Han population in high-and low-incidence areas显示文摘Objective: This study aimed to determine if gastric cardia adenocarcinoma(GCA) risk was associated with the lys(A or *2) allele at the rs671(glu504lys) polymorphism within the aldehyde dehydrogenase 2(ALDH2) gene in a Chinese Han population. We also aimed to investigate ALDH2 genotypic distributions between subjects from high- and low-incidence areas for both GCA and esophageal squamous cell carcinoma(ESCC).Methods: We designed a case-control study including 2,686 patients with GCA and 3,675 control subjects from high- and lowincidence areas for both GCA and ESCC in China. Taq Man allele discrimination assay was used to genotype the rs671 polymorphism. χ~2 test and binary logistic regression analysis were used to estimate the odds ratios for the development of GCA,and multivariate ordinal logistic regression was used to analyze ALDH2 genotypic distributions among different groups.Results: Compared with ALDH2*1/*1 homozygotes, ALDH2*1/*2 and ALDH2*2/*2 carriers did not increase the risk for GCA in the Chinese Han population(P>0.05). Interestingly, the ratio of homozygous or heterozygous ALDH2 *2 carriers in highincidence areas for both GCA and ESCC was lower than that in low-incidence areas(P<0.001).Conclusions: Genotypes of rs671 at ALDH2 may not increase GCA susceptibility in Chinese Han populations. In addition, the ALDH2 genotypic distribution differs between Chinese Han populations from high- and low-incidence areas for both GCA and ESCC. | 14 | Differentially expressed protein and gene analysis revealed the effects of temperature on changes in ascorbic acid metabolism in harvested tea leaves显示文摘Tea is an important non-alcoholic beverage worldwide.Tea quality is determined by numerous secondary metabolites in harvested tea leaves,including tea polyphenols,theanine,caffeine,and ascorbic acid(AsA).AsA metabolism in harvested tea leaves is affected by storage and transportation temperature.However,the molecular mechanisms underlying AsA metabolism in harvested tea leaves exposed to different storage and transportation temperature conditions remain unclear.Here we performed RP-HPLC to detect dynamic changes in AsA content in tea leaves subjected to high-(38°C),low-(4°C),or room-temperature(25°C)treatments.The AsA distribution and levels in the treated tea leaves were analyzed using cytological–anatomical characterization methods.The differentially expressed CsAPX1 and CsDHAR2 proteins,which are involved in the AsA recycling pathway,were identified from the corresponding proteomic data using iTRAQ.We also analyzed the expression profiles of 18 genes involved in AsA metabolism,including CsAPX1 and CsDHAR2.AsA was mainly distributed in tea leaf mesophyll cells.High-and low-temperature treatments upregulated the CsAPX1 and CsDHAR2 proteins and induced CsAPX and CsDHAR2 gene expression.These results indicated that the CsAPX1 and CsDHAR2 proteins might have critical roles in AsA recycling in tea leaves.Our results provide a foundation for the in-depth investigation of AsA metabolism in tea leaves during storage and transportation,and they will promote better tea flavor in tea production. | Hui Li Zhi-Wei Liu Zhi-Jun Wu Yong-Xin Wang Rui-Min Teng Jing Zhuang | 2018 | Horticulture Research2018,5,1: | 6 |
| 15 | Effect of Spleen Lymphocytes on the Splenomegaly in Hepatocellular Carcinoma-bearing Mice显示文摘Objective To study the effect of spleen lymphocytes on the splenomegaly by hepatocellular carcinoma-bearing mouse model. Methods Cell counts,cell cycle distribution,the percentage of lymphocytes subsets and the levels of IL-2 were measured,and two-dimensional gel electrophoresis(2-DE) was used to investigate the relationship between spleen lymphocytes and splenomegaly in hepatocellular carcinoma-bearing mice. Results Compared with the normal group,the thymus was obviously atrophied and the spleen was significantly enlarged in the tumor-bearing group. Correlation study showed that the number of whole spleen cells was positively correlated with the splenic index. The cell diameter and cell-cycle phase distribution of splenocytes in the tumor-bearing group showed no significant difference compared to the normal group. The percentage of CD3+ T lymphocytes and CD8+ T lymphocytes in spleen and peripheral blood of tumor-bearing mice were substantially higher than that in the normal mice. Meanwhile,the IL-2 level was also higher in the tumor-bearing group than in the normal group. Furthermore,two dysregulated protein,β-actin and S100-A9 were identi?ed in spleen lymphocytes from H22-bearing mice,which were closely related to cellular motility. | 17 | Atorvastatin attenuates surgery-induced BBB disruption and cognitive impairment partly by suppressing NF-κB pathway and NLRP3 inflammasome activation in aged mice显示文摘In clinic,perioperative neurocognitive disorder is becoming a common complication of surgery in old patients.Neuroinflammation and blood-brain barrier(BBB)disruption are important contributors for cognitive impairment.Atorvastatin,as a strong HMG-CoA reductase inhibitor,has been widely used in clinic.However,it remains unclear whether atorvastatin could prevent anesthesia and surgery-induced BBB disruption and cognitive injury by its anti-inflammatory property.In this study,aged C57BL/6J mice were used to address this question.Initially,the mice were subject to atorvastatin treatment for 7 days(10 mg/kg).After a simple laparotomy under 1.5%isoflurane anesthesia,Morris water maze was performed to assess spatial learning and memory.Western blot analysis,immunohistochemistry,and enzyme-linked immunosorbent assay were used to examine the inflammatory response,BBB integrity,and cell apoptosis.Terminal-deoxynucleotidyl transferase mediated nick end labeling assay was used to assess cell apoptosis.The fluorescein sodium and transmission electron microscopy were used to detect the permeability and structure of BBB.The results showed that anesthesia and surgery significantly injured hippocampal-dependent learning and memory,which was ameliorated by atorvastatin.Atorvastatin could also reverse the surgery-induced increase of systemic and hippocampal cytokines,including IL-1β,TNF-α,and IL-6,accompanied by inhibiting the nuclear factor kappa-B(NF-κB)pathway and Nucleotide-Binding Oligomerization Domain,or Leucine Rich Repeat and Pyrin Domain Containing 3(NLRP3)inflammasome activation,as well as hippocampal neuronal apoptosis.In addition,surgery triggered an increase of BBB permeability,paralleled by a decrease of the ZO-1,occludin,and Claudin 5 proteins in the hippocampus.However,atorvastatin treatment could protect the BBB integrity from the impact of surgery,by up-regulating the expressions of ZO-1,occludin,and Claudin 5.These findings suggest that atorvastatin exhibits neuroprotective effects on cognition in aged mice undergoing surgery. | Pengfei Liu Quansheng Gao Lei Guan Yanting Hu Jingwen Jiang Teng Gao Weixuan Sheng Xinying Xue Hui Qiao Tianzuo Li | 2021 | Acta Biochimica et Biophysica Sinica2021,53,5: | 6 |
