1Dynamic immune profiling identifies the stronger graft-versus-leukemia (GVL) effects with haploidentical allografts compared to HLA-matched stem cell transplantation显示文摘Haploidentical stem cell transplantation(haplo-SCT)achieves superior or at least comparable clinical outcomes to HLA-matched sibling donor transplantation(MSDT)in treating hematological malignancies.To define the underlying regulatory dynamics,we analyzed time courses of leukemia burden and immune abundance of haplo-SCT or MSDT from multiple dimension.First,we employed two nonirradiated leukemia mouse models which carried human AML-ETO or MLL-AF9 fusion gene to establish haplo-identical and major histocompatibility(MHC)-matched transplantation models and investigated the immune cell dynamic response during leukemia development in vivo.We found that haplo-matching the MHCs of leukemia cells with recipient mouse T cells prolonged leukemic mice survival and reduced leukemia burden.The stronger graft-versus-leukemia activity in haplo-SCT group mainly induced by decreased apoptosis and increased cytotoxic cytokine secretion including tumor necrosis factor–α,interferon-γ,pore-forming proteins and CD107a secreted by T cells or natural killer cells.Furthermore,we conducted a prospective clinical trial which enrolled 135 patients with t(8;21)acute myeloid leukemia that displayed minimal residual disease before transplantation and underwent either haplo-SCT or MSDT.The results showed that the haplo-SCT slowed the kinetics of the leukemia burden in vivo and reduced the cumulative incidence of relapse compared with MSDT.Ex vivo experiments showed that,1 year after transplantation,cytotoxic T lymphocytes from the haplo-SCT group had higher cytotoxicity than those from the MSDT group during the same period.Our results unraveled the role of immune cells in superior antileukemia effects of haplo-SCT compared with MSDT.Huidong Guo Ying-Jun Chang Yan Hong Lan-Ping Xu Yu Wang Xiao-Hui Zhang Ming Wang Huan Chen Yu-Hong Chen Feng-Rong Wang Wei-Han Yu-Qian Sun Chen-Hua Yan Fei-Fei Tang Xiao-Dong Mo Kai-Yan Liu Xiao-Jun Huang 2021Cellular & Molecular Immunology2021,18,5:11
2Selection of Chelated Fe(Ⅲ)/Fe(Ⅱ) Catalytic Oxidation Agents for Desulfurization Based on Iron Complexation Method显示文摘Optimization of factors influencing the experiments on reactions involving 8 different chelating agents and soluble Fe(III)/Fe(II) salts was carried out to yield chelated iron complexes. A combination of optimized influencing factors has resulted in a Fe chelating capacity of the iron-based desulfurization solution to be equal to 6.83—13.56 g/L at a redox potential of 0.185—0.3. The desulfurization performance of Fe(III)/Fe(II) chelating agents was investigated on a simulated sulfur-containing industrial gas composed of H2 S and N2 in a cross-flow rotating packed bed. Test results have revealed that the proposed iron-based desulfurization solution showed a sulfur removal efficiency of over 99% along with a Fe chelating capacity exceeding 1.35 g/L. 3Synthesis and assembly with mesoporous silica of platinum (Ⅱ) porphyrin complexes bearing carbazyl groups: Luminescent and oxygen sensing properties显示文摘A series of platinum meso-tetrakis [3-methoxy-4-(N-carbazyl)n-alkyloxyphenyl]porphyrin (Pt-4Cn-TPP, n = 4, 6 and 8) are synthesized. Pt-4C4-TPP, Pt-4C6-TPP and Pt-4C8-TPP exhibit similar luminescent properties in solution and solid state. Three protonated platinum (II) porphyrins are assembled with mesoporous silica MCM-48, respec- tively, resulting in assembly materials Pt-4Cn-TPP4+/ MCM-48 (n = 4, 6 and 8). The luminescent intensity of Pt-4Cn-TPP4+/MCM-48 can be extremely quenched by molecular oxygen with high sensitivity (I0/I100 > 9). The Stern-Volmer plots of these assembly materials display considerable linearity within a wide range of oxygen concentration (0 to 100%). 4Experimental study and simulation of a three-phase flow stirred bioreactor显示文摘In order to obtain the reasonable operating conditions and minimize the power consumption in the stirred bioreactor, the hydrodynamic experiments in the stirred bioreactor have been taken to obtain the basic data. Subsequently, an Eulerian model for the gas–liquid–solid three phase flow in the stirred bioreactor has been proposed and the CFD simulation has been conducted. By comparing the results of experiment and simulation, it can be concluded that the simulation results were consistent with the experimental data. The inner relationship between operating variables and indicators could be obtained by comparing the results of just suspension speed, gas holdup, power consumption and operational maps, further the reasonable operating conditions could be also determined under the minimum power consumption. 6A novel TIR-NBS-LRR gene regulates immune response to Phytophthora root rot in soybean显示文摘Phytophthora root rot(PRR),caused by Phytophthora sojae,is a devastating disease of soybean.The NBSLRR gene family is a class of plant genes involved in disease resistance.miRNA mediates plant response to biotic stresses by regulating the expression of target genes at the transcriptional or post-translational level.Glyma.16G135500,encoding an NBS-LRR-type protein,is a target of gma-miR1510 that responds to pathogen infections.We cloned and overexpressed Glyma.16G135500(naming it GmTNL16)and knocked down mi R1510 using short tandem target mimic technology to identify the roles of the GmTNL16/gma-mi R1510 pair in the interaction of soybean and the oomycete.By overexpressing GmTNL16 in transgenic hairy roots of soybean,we showed that biomass of P.sojae was lower in overexpressing hairy roots than in control roots.Thus,miR1510 expression was reduced upon P.sojae infection,reflecting the induced expression of GmTNL16 conferring resistance to P.sojae in soybean.Differentially expressed genes were enriched in plant-pathogen interaction,plant hormone signal transduction,and secondary metabolism by RNA sequencing analyze.In particular,jasmonate and salicylic acid pathway-associated genes,including JAZ,COI1,TGA,and PR,responded to P.sojae infection.All of these results indicate that the GmTNL16/gma-miR1510 pair participates in soybean defense response via the JA and SA pathways.Li Zhou Sushuang Deng Huidong Xuan Xingxing Fan Ruidong Sun Jinming Zhao Haitang Wang Na Guo Han Xing 2022The Crop Journal2022,10,6:1
7Effect of Forchlorfenuron (CPPU) and Ethychlozate on Fruit Development and Quality of Mango Cultivar 'Tainong 1'显示文摘The fruit swelling effect of forchlorfenuron( CPPU) and fruit thinning effect of ethychlozate on Tainong 1 mango( Mangifera indica L. cv.) were studied. Spraying 32 mg/L CPPU at 7 and 21 d post flowering for just once significantly improved the fruit size of mango,but had no significant impact on fruit quality and flavor. 8Target tracking based on frequency spectrum amplitude显示文摘The amplitude of frequency spectrum can be integrated with probabilistic data association (PDA) to distinguish the target with clutter echoes, especially in low SNR underwater environment. A new target-tracking algorithm is presented which adopts the amplitude of frequency spectrum to improve target tracking in clutter. The probabilistic density distribution of frequency spectrum amplitude is analyzed. By simulation, the results show that the algorithm is superior to PDA. 9Th2 polarization in target organs is involved in the alleviation of pathological damage mediated by transplanting granulocyte colony-stimulating factor-primed donor T cells显示文摘Acute graft-versus-host disease(a GVHD)is caused by allo-activated donor T cells infiltrating target organs.As a regulator of immune function,granulocyte colony-stimulating factor(G-CSF)has been demonstrated to relieve the a GVHD reaction.However,the role of G-CSF-primed donor Tcells in specific target organs is still unknown.In this study,we employed a classical MHC-mismatched transplantation mouse model(C57BL/6 into BALB/c)and found that recipient mice transplanted with GCSF-primed T cells exhibited prolonged survival compared with that of the PBS-treated group.This protective function against GVHD mediated by G-CSF-primed donor T cells was further confirmed by decreased clinical and pathological scores in this a GVHD mouse model,especially in the lung and gut.Moreover,we found that Tcells polarized towards Th2 cells and regulatory T cells were increased in specific target organs.In addition,G-CSF treatment inhibited inducible co-stimulator(ICOS)expression and increased the expression of tolerance-related genes in recipient mice.Our study provides new insight into the immune regulatory effects of G-CSF on T cell-mediated a GVHD,especially for its precise regulation in GVHD target organs.Yang Zhoui Leqing Cao Huidong Guo Yan Hong Ming Wang Ke Wang Xiaojun Huang Yingjun Chang 2021Science China(Life Sciences)2021,64,7:1

12中压全调节大功率电源系统在北斗三号卫星上的应用显示文摘China’s BeiDou Navigation Satellite System(BDS)construction has been completed and the system has been formally commissioned.Most of the Electric Power Systems(EPSs)for MEO satellites were developed by the Shanghai Institute of Space Power-sources.The 42 V medium-voltage fully-regulated high-power EPS has been adopted for the first time in medium Earth orbit,with an output power reaching about 3 kW.Compared with the 42 V medium-voltage semi-regulated bus power system used in the Regional Navigation BDS-2 satellite,the EPS of the BDS-3 MEO satellites has increased power by about 80%,adopting many newly developed products such as high-efficient triple junction GaAs solar cells,high-energy-density lithium ion batteries and a high-efficient autonomous power control unit(PCU).Based on the studies on the medium-voltage fully-regulated and high-power EPS technical principles,and the adaptability and reliability of various working modes,the test verifications for the EPS were conducted both on the ground and in orbit.Compared with other global navigation satellite systems such as GPS,Galileo and GLONASS,the EPS of the BDS-3 MEO satellite has a long design life time which is equivalent to that of the GPS and Galileo,but with a larger power supply capability and power ratio,distinguishing its advancement in the field of satellite power technology.LI Xuping LEI Hu XU Huidong LUO Guo 2020Aerospace China2020,21,4:0
