1Expression,deleton and mnutation of ρ16 gene in human gastric cancer显示文摘AIM To investigate the relationship between the expression of p16 gene and the gastric carcinogenesis,depth of invasion and lymph node metastases, and to evaluate the deletion and mutation of exon 2 in p16 gene in gastric carcinoma.METHODS The expression of P16 protein was examined by streptavidin-peroxidase conjugated method (S-P); the deletion and mutation of p16 gene were respectively examined by polymerase chain reaction (PCR) and polymerase chain reaction single-strand conformation polymorphism analysis (PCR-SSCP) in gastric carcinoma.RESULTS Expression of P16 protein was detected in 96.25% (77/80) of the normal gastric mucosa, in 92.00% (45/50) of the dysplastic gastric mucosa and in 47.54% (58/122) of the gastric carcinoma. The positive rate of P16 protein expression in gastric carcinoma was significantly lower than that in normal gastric mucosa and dysplastic gastric mucosa (P<0.05). The positive rate of P16 protein expression in mucoid carcinoma 10.00% (1/ 10) was significantly lower than that in poorly differentiated carcinoma 51.22% ( 21/ 41 ),undifferentiated carcinoma 57.69% (15/26) and signet ring cell carcinoma 62.50% (10/ 16) (P<0.05). The positive rate of p16 protein in 30 cases paired primary and lymph node metastatic gastric carcinoma: There was 46.67% (14/30) in primary gastric carcinoma, 16.67% (5/30) in lymph node metastatic gastric carcinoma. The positive rate of lymph node metastatic carcinoma was significantly lower than that of primary carcinoma (P<0.05). There was of p16 gene mutation in exon 2, but 5 cases displayed deletion of p16 gene in exon 2 in the 25 primary gastric carcinomas.CONCLUSIONS The expression loss of P16 protein related to the gastric carcinogenesis, gastric carcinoma histopathological subtypes and lymph metastasis. The mutation of p16 gene in exon 2 may not be involved in gastric carcinogenesis. 3Biological characteristics of an improved triploid crucian carp显示文摘An improved triploid crucian carp(ITCC) was produced by crossing improved tetraploids(G1×AT,) with improved red crucian carp(IRCC,),which were obtained by distant crossing and gynogenesis.The biological characteristics of ITCC,including the number and karyotype of chromosomes,gonadad and pituitary structures,phenotype,and growth rate are reported.ITCC possessed 150 chromosomes with the karyotype 33m+51sm+33st+33t.In the breeding season,both ovary-like and testis-like gonads of ITCC were unable to produce normal mature gametes.The ultrastructure of the pituitary of ITCC showed that most of the endocrine granules in gonadotrophic hormone(GTH) cells had not been released,providing endocrinological evidence for the sterility of ITCC.Compared with triploid crucian carp(TCC) produced by mating Japanese crucian carp with allotetraploid hybrids,ITCC not only retained the excellent traits of fast growth rate and sterility,but also acquired improved morphological characteristics,including higher body,shorter tail and smaller head.CHEN Song,WANG Jing,LIU ShaoJun,QIN QinBo,XIAO Jun,DUAN Wei,LUO KaiKun,LIU JinHui & LIU Yun Key Laboratory of Protein Chemistry and Developmental Biology of State Education Ministry of China,College of Life Sciences,Hunan Normal University,Changsha 410081,China 2009Science China(Life Sciences)2009,52,8:14
4The cloning of Dmc1 cDNAs and a comparative study of its expression in different ploidy cyprinid fishes显示文摘Dmc1 (disrupted meiotic cDNA) is a functionally specific gene, which was firstly discovered in yeast and then found to encode a protein required for homologous chromosome synapsis during the process of meiosis. In this investigation, we cloned the partial cDNAs of Dmc1 of diploid red crucian carp, Japanese crucian carp, common carp, triploid crucian carp and allotetraploid hybrids by using a pair of degenerate primers based on the conservative sequence of amino acids of the DMC1 protein in yeast, mouse and human. The full length cDNAs were then obtained by rapid amplification of cDNA ends (RACE). Our data showed that the full length cDNAs of Dmc1 in the three diploid fishes are all 1375 bp long, while it is 1383 bp long in triploids and 1379 bp long in allotetraploids. And despite of the variation in length, all the cDNAs encode a protein of 342 amino acids. A high homology of 97.3% of the DMC1 protein can be drawn by comparing the amino acid sequences in the three diploids, which is also of 86%, 86% and 95% similarity to human, mouse and zebrafish, respectively. A comparative study of the expression pattern of Dmc1 was carried out by RT-PCR using specific primers against the same se-quences of coding regions in different ploidy cyprinid fishes, from which it was showed that Dmc1 was expressed only in gonads of these five kinds of fishes. The expression pattern of Dmc1 in both ovaries and testes from different ploidy fishes within breeding season was also studied by Real-time PCR, and the results showed that the expression of this gene was greatly different among the three different ploidy fishes, which was the highest of triploid and lowest of allotetraploids. The histological sections data showed matured gonads of both diploid red crucian carp and allotetraploids in breeding season, although the latter demonstrated a higher maturation, and no gonadal maturation could be observed in triploids. In conclusion, we suggest that Dmc1 is specifically expressed in the period of meiosis in all the ploidy cyprinid fishes and directly related with the development of gonad in a manner of ploidy-independent way. 5Cell-SELEX-based aptamer-conjugated nanomaterials for enhanced targeting of cancer cells显示文摘Early detection and treatment of cancer depends on developing highly sensitive and specific methods for targeting cancer cells. To do this, aptamers, which are generated by a novel technique called cell-SELEX (systematic evolution of ligands by exponential enrichment), have been widely applied in cancer cell targeting based on such merits as high target affinity and specificity, small size, minimal immunogenicity, and ease of chemical modification. Furthermore, aptamers can gain more flexibility as cancer cell targeting tools when conjugated to nanomaterials, including metallic nanoparticles, quantum dots, silica nanoparticles, and carbon nanomaterials, among others. 7Formation and biological characterization of three new types of improved crucian carp显示文摘The improved tetraploids (G1×AT) were obtained by distant crossing and gynogenesis and the high-body individuals accounted for 2% among G1×AT. After mating with each other, the high-body in- dividuals produced three kinds of bisexual fertile diploid fishes: high-body red crucian carp, high-body fork-like-tails goldfish and gray common carp. The high-body red crucian carp mating with each other formed three types of improved crucian carp (ICC) including improved red crucian carp (IRCC), im- proved color crucian carp (ICCC) and improved gray crucian carp (IGCC). The phenotypes, chromo- some numbers, gonadal structure and fertility of the three kinds of ICC and their offspring were observed. All the three kinds of ICC possessed some improved phenotypes such as higher body, smaller head and shorter tail. The ratios of the body height to body length of IRCC, ICCC and IGCC were 0.54, 0.51 and 0.54, respectively. All of them were obviously higher than that of red crucian carp 0.41 (P<0.01). Three kinds of ICC had the same chromosome number as red crucian carp with 100 chromosomes. All the ICC possessed normal gonads producing mature eggs or sperm, which was important for the production of an improved diploid population. Compared with red crucian carp, all the ICC had stronger fertility such as higher gametes production, higher fertilization rate and higher hatchery rate. Three types of improved diploid fish population were generated from the three kinds of ICC by self-crossing, respectively. The ICC can serve as ornamental fish and edible fish. They are also ideal parents to produce triploids by mating with tetraploids. 11Analysis theory of random energy of train derailment in wind显示文摘Based on the analysis theory of random energy of train derailment, an analysis theory of random energy of train derailment in wind is suggested. Two methods are proposed -the time domain method and the frequency domain method of analysis theory of random energy of train derailment in wind. The curves of σ pw -v under various wind speeds are obtained through the computation. The original curve of σ p -v is expanded, which turns the analysis theory of random energy of train derailment into the all-weather theory. Train derailment condition has been established under wind action. The first and second criterions of train derailment have been proposed in light of wind action. The analysis of train derailment cases at home or abroad is made, in- cluding the first analysis of Xinjiang train derailment case encountered 13-level of gale, which explained the inevitability of train derailment. The analysis theory of random energy of train derailment in wind shows its validity and accuracy. The input energy σ pw of the transverse vibration of train-track(bridge)-wind system is linked to train speed. 13Effect of matrix cracking on the time delayed buckling of viscoelastic laminated circular cylindrical shells显示文摘The effect of matrix cracking on the bifurcation creep buckling of viscoelastic laminated circular cylindrical shells is investigated.The viscoelastic behavior of laminas is modeled by Schapery's integral constitutive equation with growing matrix cracks.The values of damage variables are correlated to non-dimensional density of matrix cracks relying on the formulas from mesomechanics approach,and the evolution equation predicting the growth rate of density of matrix cracks is assumed to follow a power type relation with transverse tensile stress.The governing equations for prebuckling creep deformation and bifurcation buckling of laminated circular cylindrical shells under axial compression are obtained on the basis of the Donnell type shallow shell theory and Kármán-Donnell geometrically nonlinear relationship.Corresponding solution strategy is constructed by integrating finite-difference technique,trigonometric series expansion method and Taylor's numerical recursive scheme for convolution integration.The bifurcation creep buckling of symmetrically laminated glass-epoxy circular cylindrical shells with matrix creep cracking coupled are examined for various geometrical parameters and parameters of damage evolution as well as boundary conditions.The numerical results show that matrix creep cracking remarkably shortens the critic time of bifurcation buckling and reduces the durable critic loads,and its effects become weak and finally vanish with the increase of the ratio of radius to thickness in the case of short laminated circular cylindrical shells,also the influence of the matrix creep cracking is mainly dependent on the boundary conditions at two ends for moderately long circular cylindrical shells.PENG Fan,FU YiMing & CHEN YaoJun College of Mechanics and Aerospace,Hunan University,Changsha 410082,China 2008Science China(Technological Sciences)2008,51,12:2

16Residual stress and radial stress gradient in polycrystalline diamond compacts显示文摘The differential thermal expansion of the polycrystalline diamond layer and the tungsten carbide substrate results in large residual stresses as PDC cutters cooling after sintering.The residual stresses on the top surface of the diamond layer of PDC were measured at five points along the radial direction of PDC using X-ray Diffraction Residual Stress Instrument,thus the stresses and their radial distribution were obtained.The results show that the stresses on the diamond surface are compressive,the biggest stress appears at the central point(about 1200 MPa),and that from the center to the edge of PDC,the magnitude of the stress decreases. 18Structural parameterization and functional prediction of antigenic polypeptome sequences with biological activity through quantitative sequence-activity models (QSAM) by molecular electronegativity edge-distance vector (VMED)显示文摘Only from the primary structures of peptides, a new set of descriptors called the molecular electro-negativity edge-distance vector (VMED) was proposed and applied to describing and characterizing the molecular structures of oligopeptides and polypeptides, based on the electronegativity of each atom or electronic charge index (ECI) of atomic clusters and the bonding distance between atom-pairs. Here, the molecular structures of antigenic polypeptides were well expressed in order to propose the auto-mated technique for the computerized identification of helper T lymphocyte (Th) epitopes. Furthermore, a modified MED vector was proposed from the primary structures of polypeptides, based on the ECI and the relative bonding distance of the fundamental skeleton groups. The side-chains of each amino acid were here treated as a pseudo-atom. The developed VMED was easy to calculate and able to work. Some quantitative model was established for 28 immunogenic or antigenic polypeptides (AGPP) with 14 (1― 14) Ad and 14 other restricted activities assigned as '1'(+) and '0'(-), respectively. The latter comprised 6 Ab(15-20), 3 Ak(21-23), 2 Ek(24-26), 2 H-2k(27 and 28) restricted sequences. Good results were obtained with 90% correct classification (only 2 wrong ones for 20 training samples) and 100% correct prediction(none wrong for 8 testing samples); while con-trastively 100% correct classification (none wrong for 20 training samples) and 88% correct classification (1 wrong for 8 testing samples). Both stochastic samplings and cross valida-tions were performed to demonstrate good performance. The described method may also be suitable for estimation and prediction of classes I and II for major histocompatibility an-tigen (MHC) epitope of human. It will be useful in immune identification and recognition of pro-teins and genes and in the design and devel-opment of subunit vaccines. Several quantitative structure activity relationship (QSAR) models were developed for various oligopeptides and polypeptides including 58 dipeptides and 31 pentapeptides with angiotensin converting enzyme (ACE) inhibition by multiple linear regression (MLR) method. In order to explain the ability to characterize molecular structure of polypeptides, a molecular modeling investigation on QSAR was performed for functional prediction of polypeptide sequences with anti-genic activity and heptapeptide sequences with tachykinin activity through quantitative se-quence-activity models (QSAMs) by the molecular electronegativity edge-distance vector (VMED). The results showed that VMED exhibited both excellent structural selectivity and good activity prediction. Moreover, the results showed that VMED behaved quite well for both QSAR and QSAM of poly-and oli-gopeptides, which exhibited both good estimation ability and prediction power, equal to or better than those reported in the previous references. Finally, a preliminary conclusion was drwan: both classical and modified MED vectors were very useful structural descriptors. 19Second-order calibration applied to quantification of two active components of Schisandra chinensis in complex matrix显示文摘The effectiveness of traditional Chinese medicine (TCM) against various diseases urges more low cost, speed and sensitive analytical methods for investigating the phamacology of TCM and providing a theoretical basis for clinical use. The potential of second-order calibration method was validated for the quantification of two effective ingredients of Schisandra chinensis in human plasma using spectrofluorimetry. The results obtained in the present study demonstrate the advantages of this strategy for multi-target determination in complex matrices. Although the spectra of the analytes are similar and a large number of interferences also exist, second-order calibration method could predict the accurate concentrations together with reasonable resolution of spectral profiles for analytes of interest owing to its 'second-order advantage'.
