| 1 | Developing DNA methylation-based diagnostic biomarkers显示文摘An emerging paradigm shift for disease diagnosis is to rely on molecular characterization beyond traditional clinical and symptom-based examinations. Although genetic alterations and transcription signature were first introduced as potential biomarkers, clinical implementations of these markers are limited due to low reproducibility and accuracy. Instead, epigenetic changes are considered as an alternative approach to disease diagnosis. Complex epigenetic regulation is required for normal biological functions and it has been shown that distinctive epigenetic disruptions could contribute to disease pathogenesis. Disease-specific epigenetic changes, especially DNA methylation, have been observed,suggesting its potential as disease biomarkers for diagnosis. In addition to specificity, the feasibility of detecting disease-associated methylation marks in the biological specimens collcted noninvasively,such as blood samples, has driven the clinical studies to validate disease-specific DNA methylation changes as a diagnostic biomarker. | 7 | The KDM4B–CCAR1–MED1 axis is a critical regulator of osteoclast differentiation and bone homeostasis显示文摘Bone undergoes a constant and continuous remodeling process that is tightly regulated by the coordinated and sequential actions of bone-resorbing osteoclasts and bone-forming osteoblasts.Recent studies have shown that histone demethylases are implicated in osteoblastogenesis;however,little is known about the role of histone demethylases in osteoclast formation.Here,we identified KDM4B as an epigenetic regulator of osteoclast differentiation.Knockdown of KDM4B significantly blocked the formation of tartrate-resistant acid phosphatase-positive multinucleated cells.Mice with myeloid-specific conditional knockout of KDM4B showed an osteopetrotic phenotype due to osteoclast deficiency.Biochemical analysis revealed that KDM4B physically and functionally associates with CCAR1 and MED1 in a complex.Using genome-wide chromatin immunoprecipitation(ChIP)-sequencing,we revealed that the KDM4B–CCAR1–MED1 complex is localized to the promoters of several osteoclast-related genes upon receptor activator of NF-κB ligand stimulation.We demonstrated that the KDM4B–CCAR1–MED1 signaling axis induces changes in chromatin structure(euchromatinization)near the promoters of osteoclast-related genes through H3K9 demethylation,leading to NF-κB p65 recruitment via a direct interaction between KDM4B and p65.Finally,small molecule inhibition of KDM4B activity impeded bone loss in an ovariectomized mouse model.Taken together,our findings establish KDM4B as a critical regulator of osteoclastogenesis,providing a potential therapeutic target for osteoporosis. | Sun-Ju Yi You-Jee Jang Hye-Jung Kim Kyubin Lee Hyerim Lee Yeojin Kim Junil Kim Seon Young Hwang Jin Sook Song Hitoshi Okada Jae-Il Park Kyuho Kang Kyunghwan Kim | 2021 | Bone Research2021,9,3: | 1 |

| 10 | Micro-Locational Fine Dust Prediction Utilizing Machine Learning and Deep Learning Models显示文摘Given the increasing number of countries reporting degraded air quality,effective air quality monitoring has become a critical issue in today’s world.However,the current air quality observatory systems are often prohibitively expensive,resulting in a lack of observatories in many regions within a country.Consequently,a significant problem arises where not every region receives the same level of air quality information.This disparity occurs because some locations have to rely on information from observatories located far away from their regions,even if they may be the closest available options.To address this challenge,a novel approach that leverages machine learning and deep learning techniques to forecast fine dust concentrations was proposed.Specifically,continuous location features in the form of latitude and longitude values were incorporated into our models.By utilizing a comprehensive dataset comprising weather conditions,air quality measurements,and location properties,various machine learning models,including Random Forest Regression,XGBoost Regression,AdaBoost Regression,and a deep learning model known as Long Short-Term Memory(LSTM)were trained.Our experimental results demonstrated that the LSTM model outperforms the other models,achieving the best score with a root mean squared error of 23.48 in predicting fine dust(PM10)concentrations on an hourly basis.Furthermore,the fact that incorporating location properties,such as longitude and latitude values,enhances the overall quality of the regression models was discovered.Additionally,the implications and contributions of our research were discussed.By implementing our approach,the cost associated with relying solely on existing observatories can be substantially reduced.This reduction in costs can pave the way for economically efficient fine dust observation systems,ensuring more widespread and accurate air quality monitoring across different regions. | Seoyun Kim Hyerim Yu Jeewoo Yoon Eunil Park | 2024 | Computer Systems Science & Engineering2024,48,2: | 0 |
