2Probiotic Leuconostoc mesenteroides ssp. cremoris and Streptococcus thermophilus induce IL-12 and IFN-γ production显示文摘AIM: To investigate the capacity of potentially probiotic strains from six bacterial genera to induce cytokine production alone or in combinations in order to identify potential enhancing or synergistic effects in order to select probiotic bacteria for in vivo purposes. METHODS: Cytokine production in human peripheral blood mononuclear cells (PBMC) in response to stimulation with eleven different potentially probiotic bacterial strains from Streptococcus , Lactobacillus , Bifidobacterium , Lactococcus , Leuconostoc and Propionibacterium genera was analysed. Production and mRNA expression of TNF-α, IL-12, IFN-γ and IL-10 were determined by ELISA and Northern blotting, respectively. RESULTS: All tested bacteria induced TNF-α production. The best inducers of Th1 type cytokines IL-12 and IFN-γ were Streptococcus and Leuconostoc strains. All Bifidobacterium and Propionibacterium strains induced higher IL-10 production than other studied bacteria. Stimulation of PBMC with any bacterial combinations did not result in enhanced cytokine production suggesting that different bacteria whether gram-positive or gram- negative compete with each other during host cell interactions.CONCLUSION: The probiotic S. thermophilus and Leuconostoc strains are more potent inducers of Th1 type cytokines IL-12 and IFN-γ than the probiotic Lactobacillus strains. 3Potentially probiotic bacteria induce efficient maturation but differential cytokine production in human monocyte-derived dendritic cells显示文摘AIM: To analyze the ability of nine different potentially probiotic bacteria to induce maturation and cytokine production in human monocyte-derived dendritic cells (moDCs). METHODS: Cytokine production and maturation of moDCs in response to bacterial stimulation was analyzed with enzyme-linked immunosorbent assay (ELISA) and flow cytometric analysis (FACS), respectively. The kinetics of mRNA expression of cytokine genes was determined by Northern blotting. The involvement of different signaling pathways in cytokine gene expression was studied using specific pharmacological signaling inhibitors. RESULTS: All studied bacteria induced the maturation of moDCs in a dose-dependent manner. More detailed analysis with S. thermophilus THS, B. breve Bb99, and L. lactis subsp. cremoris ARH74 indicated that these bacteria induced the expression of moDC maturationmarkers HLA class Ⅱ and CD86 as efficiently as pathogenic bacteria. However, these bacteria differed in their ability to induce moDC cytokine gene expression. S. thermophilus induced the expression of pro-inflammatory (TNF-α, IL-12, IL-6, and CCL20) and Th1 type (IL-12 and IFN-γ) cytokines, while B. breve and L. lactis were also potent inducers of anti- inflammatory IL-10. Mitogen-activated protein kinase (MAPK) p38, phosphatidylinositol 3 (PI3) kinase, and nuclear factor-kappa B (NF-κB) signaling pathways were shown to be involved in bacteria-induced cytokine production. 4Probiotic intervention has strain-specific anti-inflammatory effects in healthy adults显示文摘AIM: To evaluate the effects of three potentially anti- inflammatory probiotic bacteria from three different genera on immune variables in healthy adults in a clinical setting based on previous in vitro characterization of cytokine responses. METHODS: A total of 62 volunteers participated in this randomized, double-blind and placebo-controlled parallel group intervention study. The volunteers were randomized to receive a milk-based drink containing either Lactobacillus rhamnosus GG (LGG), Bifidobacterium animalis ssp. lactis Bb12 (Bb12), or Propionibacterium freudenreichii ssp. shermanii JS (PJS) or a placebo drink for 3 wk. Venous blood and saliva samples were taken at baseline and on d 1, 7 and 21. Fecal samples were collected at baseline and at the end of intervention. RESULTS: The serum hsCRP expressed as the median AUC0-21 (minus baseline) was 0.018 mg/L in the placebo group, -0.240 mg/L in the LGG group, 0.090 mg/L in the Bb12 group and -0.085 mg/L in the PJS group (P = 0.014). In vitro production of TNF-α from in vitro cultured peripheral blood mononuclear cells (PBMC) was significantly lower in subjects receiving LGG vs placebo. IL-2 production from PBMC in the Bb12 group was significantly lower compared with the other groups. 6The physical structures of snow and sea ice in the Arctic section of 150°-180°W during the summer of 2010显示文摘The physical structures of snow and sea ice in the Arctic section of 150°-180°W were observed on the basis of snow-pit, ice-core, and drill-hole measurements from late July to late August 2010. Almost all the investigated floes were first-year ice, except for one located north of Alaska, which was probably multi-year ice transported from north of the Canadian Arctic Archipelago during early summer. The snow covers over all the investigated floes were in the melting phase, with temperatures approaching 0℃ and densities of 295-398 kg/m3 . The snow covers can be divided into two to five layers of different textures, with most cases having a top layer of fresh snow, a round-grain layer in the middle, and slush and/or thin icing layers at the bottom. The first-year sea ice contained about 7%-17% granular ice at the top. There was no granular ice in the lower layers. The interior melting and desalination of sea ice introduced strong stratifications of temperature, salinity, density, and gas and brine volume fractions. The sea ice temperature exhibited linear cooling with depth, while the salinity and the density increased linearly with normalized depth from 0.2 to 0.9 and from 0 to 0.65, respectively. The top layer, especially the freeboard layer, had the lowest salinity and density, and consequently the largest gas content and the smallest brine content. Both the salinity and density in the ice basal layer were highly scattered due to large differences in ice porosity among the samples. The bulk average sea ice temperature, salinity, density, and gas and brine volume fractions were-0.8℃, 1.8, 837 kg/m3 , 9.3% and 10.4%, respectively. 7考虑目标光谱差异的机载离散激光雷达叶面积指数反演显示文摘利用间隙率模型反演LAI(Leaf Area Index),需要同时获取冠层间隙率和消光系数,后者与冠层叶倾角分布有关。基于点云数量构建激光雷达穿透指数LPI(LiDAR Penetration Index),用以代替冠层间隙率GF(Gap Fraction),并利用间隙率模型反演冠层LAI是利用LiDAR PCD(LiDAR Point Cloud Data)数据反演冠层LAI主要思路。冠层和背景的光谱差异是影响PCD数据中冠层和背景点云数量的重要因素,因此从LPI到GF的校正需要获取背景和冠层的后向散射系数比(μ=ρg/ρv)。本文基于PCD数据中点云强度进行μ值获取,用以实现LPI到GF的校正;在假设区域内叶倾角满足椭球形叶倾角分布的基础上,利用样地尺度下的多角度GF,采用有约束的非线性最优化方法获取椭球形叶倾角分布参数χ,实现冠层消光系数的获取;最后利用间隙率模型实现基于PCD数据的LAI反演。本文探讨了基于PCD数据进行冠层LAI反演时,样地尺度Rxy_Tile、样方尺度Rxy_Plot以及进行背景和冠层分割的高度阈值Ht对模型的影响。结果显示,由于区域内地衣植被广泛覆盖,基于点云强度的μ值接近1,符合区域特点;经过μ值校正后的GF对冠层间隙率具有较好的反映能力(R2=0.78,RMSE=0.09);对于优势种明显的区域,基于样地尺度内多角度GF的χ值反演受样地内冠间大间隙的影响,选择合适的样地尺度能够减小LAI反演过程中的系统性误差;结合地面参考数据,确定的最优Rxy_Tile、Rxy_Plot和Ht分别为950 m、10 m和2.6 m,在此基础上反演的LAI与地面测量数据具有高度的一致性(R2=0.84,RMSE=0.51);与Rxy_Plot相比,基于间隙率模型的LAI反演对Ht的选择更为敏感。田罗 屈永华 Korhonen Lauri Korpela Ilkka Heiskanen Janne 2020遥感学报2020,24,12:5
8Underground space planning in Helsinki显示文摘This paper gives insight into the use of underground space in Helsinki,Finland.The city has an underground master plan(UMP) for its whole municipal area,not only for certain parts of the city.Further,the decision-making history of the UMP is described step-by-step.Some examples of underground space use in other cities are also given.The focus of this paper is on the sustainability issues related to urban underground space use,including its contribution to an environmentally sustainable and aesthetically acceptable landscape,anticipated structural longevity and maintaining the opportunity for urban development by future generations.Underground planning enhances overall safety and economy efficiency.The need for underground space use in city areas has grown rapidly since the 21 st century;at the same time,the necessity to control construction work has also increased.The UMP of Helsinki reserves designated space for public and private utilities in various underground areas of bedrock over the long term.The plan also provides the framework for managing and controlling the city's underground construction work and allows suitable locations to be allocated for underground facilities.Tampere,the third most populated city in Finland and the biggest inland city in the Nordic countries,is also a good example of a city that is taking steps to utilise underground resources.Oulu,the capital city of northern Finland,has also started to ‘go underground'.An example of the possibility to combine two cities by an 80-km subsea tunnel is also discussed.A new fixed link would generate huge potential for the capital areas of Finland and Estonia to become a real Helsinki-Tallinn twin city.Ilkka Vhaho 2014Journal of Rock Mechanics and Geotechnical Engineering2014,6,5:4
9Mission impossible completed:unlocking the nomenclature of the largest and most complicated subgenus of Cortinarius,Telamonia显示文摘So far approximately 144,000 species of fungi have been named but sequences of the majority of them do not exist in the public databases.Therefore,the quality and coverage of public barcode databases is a bottleneck that hinders the study of fungi.Cortinarius is the largest genus of Agaricales with thousands of species world-wide.The most diverse subgenus in Cortinarius is Telamonia and its species have been considered one of the most taxonomically challenging in the Agaricales.Its high diversity combined with convergent,similar appearing taxa have earned it a reputation of being an impossible group to study.In this study a total of 746 specimens,including 482 type specimens representing 184 species were sequenced.Also,a significant number of old types were successfully sequenced,105 type specimens were over 50 years old and 18 type specimens over 100 years old.Altogether,20 epi-or neotypes are proposed for recently commonly used older names.Our study doubles the number of reliable DNA-barcodes of species of C.subgenus Telamonia in the public sequence data-bases.This is also the first extensive phylogenetic study of the subgenus.A majority of the sections and species are shown in a phylogenetic context for the first time.Our study shows that nomenclatural problems,even in difficult groups like C.subgenus Telamonia,can be solved and consequently identification of species based on ITS barcodes becomes an easy task even for non-experts of the genus.Kare Liimatainen Tuula Niskanen Bálint Dima Joseph F.Ammirati Paul M.Kirk Ilkka Kytövuori 2020Fungal Diversity2020,,5:4

11Methods, units and quality requirements for the analysis of haemoglobin A_(1c) in diabetes mellitus显示文摘The formation of glycohemoglobin, especially the hemoglobin A1c(Hb_(A1c)) fraction, occurs when glucose becomes coupled with the amino acid valine in the β-chain of Hb; this reaction is dependent on the plasma concentration of glucose. Since the early 1970 s it has been known that diabetics display higher values of Hb_(A1c) because they have elevated blood glucose concentrations. Thus Hb_(A1c) has acquired a very important role in the treatment and diagnosis of diabetes mellitus. After the introduction of the first quantitative measurement of Hb_(A1c), numerous methods for glycohemoglobin have been introduced with different assay principles: From a simple minicolumn technique to the very accurate automated highpressure chromatography and lastly to many automated immunochemical or enzymatic assays. In early days, the results of the quality control reports for Hb_(A1c) varied extensively between laboratories, therefore in United States and Canada working groups(WG) of the Diabetes Controls and Complications Trial(DCCT) were set up to standardize the Hb_(A1c) assays against the DCCT/National Glycohemoglobin Standardization Program reference method based on liquid chromatography. In the 1990 s, the International Federation of Clinical Chemistry and Laboratory Medicine(IFCC) appointed a new WG to plan a reference preparation and method for the Hb_(A1c) measurement. When the reference procedureswere established, in 2004 IFCC recommended that all manufacturers for equipment used in Hb_(A1c) assays should calibrate their methods to their proposals. This led to an improvement in the coefficient of variation(CV%) associated with the assay. In this review, we describe the glycation of Hb, methods, standardization of the Hb_(A1c) assays, analytical problems, problems with the units in which Hb_(A1c) values are expressed, reference values, quality control aspects, target requirements for Hb_(A1c), and the relationship of the plasma glucose values to Hb_(A1c) concentrations. We also note that the acceptance of the mmol/mol system for Hb_(A1c) as recommended by IFCC, i.e., the new unit and reference ranges, are becoming only slowly accepted outside of Europe where it seems that expressing Hb_(A1c) values either only in per cent units or with parallel reporting of percent and mmol/mol will continue. We believe that these issues should be resolved in the future and that it would avoid confusion if mmol/mol unit for Hb_(A1c) were to gain worldwide acceptance.Ilkka Penttil? Karri Penttil? 12Experimental evidence on biodiversity impacts of variable retention forestry,prescribed burning,and deadwood manipulation in Fennoscandia显示文摘Intensive forest management has been applied in most Fennoscandian forests for a period of almost one felling rotation.This paradigm has produced even-aged and even-structured forests of different successional stages that cover about 90%of forest land.At the same time,wildfires have been nearly eliminated in most of the Fennoscandian nature.Consequently,hundreds of species are red-listed because of forest management.To support these species,forest management requires improvements.Variable retention forestry and habitat restoration have been suggested to mitigate negative effects of forest management on biodiversity,and these have been practiced to some extent during the past few decades.Here,we review experimental results on the effects of variable retention and two restoration measures(prescribed burning and artificial addition of coarse woody debris)on different species groups in Fennoscandia.Our key findings are as follows:(i)Many species respond positively to felling within a few years,apparently due to released and often ephemeral resources,such as fresh residue and stumps.Species associated with shady conditions are negatively impacted,but any retention supports many of these,and their species composition remains almost unaffected with 50-70%retention of the initial tree volume.(ii)These effects remain detectable for at least 10-30 years or,according to some studies,nearly 100 years,e.g.,in polypore fungi.(iii)Initial effects of prescribed burning on most species groups(apart from pyrophiles)are negative,but within 10-15 years post-fire sites begin to support many rare and threatened deadwood-dependent species.Epiphytic lichens,however,remain negatively affected.(iv)Artificial addition of deadwood(mostly high stumps)supports a wide spectrum of deadwood-dependent species,but the species composition differs from that of naturally died trees.(v)Moisture and micro-habitat variation are crucial for forest species at harvested sites,at least in forests dominated by Norway spruce.We conclude that felling method as such is of little importance for threatened forest species,although retention mitigates many negative effects.These species require microclimatic continuity,and maintenance and active increase of legacies,such as deadwood of different qualities(species,downed/standing,snag/log/stump,decay stage),very old trees,and tree species mixtures.Matti Koivula Ilkka Vanha-Majamaa 2020Ecological Processes2020,9,1:3

combinations are proposed where necessary to improve our traditional species concepts and provide updates on their classification.In addition to extensive morphological descriptions and appropriate asexual and sexual connections,DNA sequence data are also analysed from concatenated datasets(rDNA,TEF-a,RBP2 and b-Tubulin)to infer phylogenetic relationships and substantiate systematic position of taxa within appropriate ranks.Wherever new species or combinations are being proposed,we apply an integrative approach(morphological and molecular data as well as ecological features wherever applicable).Notes on 125 fungal taxa are compiled in this paper,including eight new genera,101 new species,two new combinations,one neotype,four reference specimens,new host or distribution records 16Analysis and prediction of 100 km-scale atmo-spheric duct interference in TD-LTE networks显示文摘Atmospheric ducts are horizontal layers that occur under certain weather conditions in the lower atmosphere.Radio signals guided in atmospheric ducts tend to experience less attenuation and spread much farther,i.e,hundreds of kilometers.In a large-scale deployed TD-LTE(Time Division Long Term Evolution)network,atmospheric ducts cause faraway downlink wireless signals to propagate beyond the designed protection distance and interfere with local uplink signals,thus resulting in a large outage probability.In this paper,we analyze the characteristics of ADI atmospheric duct interference(Atmospheric Duct Interference)by the use of real network-side big data from the current operated TD-LTE network owned by China Mobile.The analysis results yield the time varying and directional characteristics of ADI.In addition,we proposed an SVM(Support Vector Machine)-classi er based spacial prediction method of ADI by machine learning over combination of real network-side big data and real meteorological data.Furthermore,an implementation of ADMM(Alternating Direction Methods of Multipliers)framework is proposed to implement a distributed SVM prediction scheme,which reduces data exchange among di erent regions/cities,maintains similar prediction accuracy and is thus of a more practical use to operators.Ting Zhou Tianyu Sun Honglin Hu Hui Xu Yang Yang Ilkka Harjula Yevgeni Koucheryavy 2017Journal of Communications and Information Networks2017,2,1:2

20Variable retention forestry in European boreal forests in Russia显示文摘We explored whether,and to what extent,variable retention(VR)forestry has been applied in European boreal forests in northwestern Russia.Our survey revealed VR since 1910.Between 1910 and the 1960s,the statistics showing how much was retained are largely missing.However,for example,in the 1950s,on a large scale in the Republic of Karelia,up to 200-ha-sized harvesting areas,18–33%,were retention patches with a mean growing stock of 30–40m3 ha−1.In the fellings defined as“incomplete clear fellings,”which were the most common final felling type at that time,11–40%of the growing stock was left.Between the 1960s and the early 1990s,with more efficient harvesting and skidding techniques,conventional clear fellings with a much lower amount of retention were practiced.Concern about the regeneration of harvested areas gradually led to smaller(maximum 50 ha)harvesting areas and the increase of silvicultural activities.Until now,to ensure natural regeneration,patches of understory and 20–25 seed trees(i.e.,ca.15–25m3)per ha have been left permanently in harvesting areas.Landscape-scale retention for protecting ecosystem functions and biodiversity was legislated in 1978 by preserving key biotopes up to 1000 ha in size.Since 2001,promoted by forest certification,the key biotopes,such as paludified forest patches,buffers around water bodies,and habitats of red-listed species,have also been retained in harvesting areas,together with a dispersed retention of different elements.Quantitative estimates of the amount of key biotopes are largely missing.However,estimates of 1–13%in harvesting areas and 23%in whole managed landscapes have been given.VR applied during the last century has emulated natural disturbances and created diverse uneven-aged forest structures with high amounts of diverse coarse woody debris.We conclude that an analysis of past and current retention practices is essential for estimating the global role of Russian forestry.Further decisions on the general direction of Russian forestry and,specifically,retention practices are important to address the global challenges of biodiversity loss and climate change.Ekaterina Shorohova Sergey Sinkevich Aleksandr Kryshen Ilkka Vanha-Majamaa 2019Ecological Processes2019,8,1:1
