1Effects of basic fibroblast growth factor on angiogenin expression and cell proliferation in H7402 human hepatoma cells显示文摘Hepatocellular carcinoma (HCC) is one of the most common cancers worldwide. Basic fibroblast growth factor (bFGF), which is highly expressed in developing tissues and malignant cells, regulates cell growth, differentiation, and migration. Its expression is essential for the progression and metastasis of HCC. This study aims to investigate the effects of bFGF on the expression of angiogenin, another growth factor, which plays an important role in tumor angiogenesis, and on cell proliferation in H7402 human hepatoma cells. The bFGF sense cDNA or antisense cDNA was stably transfected into H7402 cells. Genomic DNA PCR analysis demonstrated that human bFGF sense cDNA or antisense cDNA was inserted into the genome. Furthermore, the expression of bFGF and angiogenin was examined by RT-PCR and Western blot assays. MTT and colony formation assays were employed to determine cell proliferation. Stable bFGF over-expressing and under-expressing transfectants were successfully established. Expression of angiogenin was decreased in the over-expressing bFGF cells (sense transfectants) and was increased in the under-expressing bFGF cells (antisense transfectants). Cell proliferation increased in the bFGF sense transfectants and decreased in the bFGF antisense transfectants. These results demonstrated that the endogenous bFGF may not only negatively regulate the angiogenin expression but also contribute to the overall cell proliferation in H7402 human hepatoma cells. 2Analysis of Mitochondrial DNA from the Ancient Tombs of Turfan显示文摘MtDNA was successfully extracted from ten individual bones (femurs) in the tombs of ancient Jushi in Turfan basin, dated back to the year about 3 000-2 500 years ago. By means of four overlapping primers, we got nucleotide sequence of the 218bp length. Ancient mtDNA was analyzed by the sequencing of hypervariable region Ⅰ of the mtDNA control region. The result shows that 9 haplotypes with 24 polymorphic sites were obtained. The phylogenetic analysis indicated that Mongolians and Altai are the population genetically closest to the Jushi groups and Jushi mtDNA pool being an admixture of eastern Asian and European lineages. 3Biosynthesis of Unsaturated Fatty Acids via Mortierella Isabellina Cultivated in a Medium Containing Butanol显示文摘Mortierella isabellina was found to accumulate large amounts of unsaturated fatty acids when it was grown in a medium containing butanol(5 g/L) and yeast extract(10 g/L) and cultivated at 25 ℃ for 5 d during which additional feeding butanol of 2 g/L was fed after being cultivated for 48 h . The resultant mycelial lipids accounted for 40 1% of the dry mycelia, while about 34% of butanol in the medium was converted. The mycelial lipids contained four kinds of unsaturated fatty acids, i.e ., palmitoleic(4 9%), oleic(54 1%), linoleic(10 4%) and linolenic(5 4%) acids. Those accounted for 74 8% of the total fatty acids. [CaHCO3]+ is a predominant compound species of Ca (II). Gd (III) can compete with Ca (II) for biological molecules. 6Efficacy and safety of blood pricking and cupping for treating nonspecific low back pain: a systematic review and Meta-analysis显示文摘OBJECTIVE: To systematically evaluate the efficacy and safety of blood pricking and cupping in the treatment of nonspecific low back pain(NLBP).METHODS:The China National Knowledge Infrastructure Database(CNKI), Wanfang Med Online Database(Wanfang), China Science and Technology Journal Database(VIP), Pub Med, Web of Science,EMBase, and Cochrane Library databases were searched from inception to October 31, 2021, for randomized controlled trials(RCTs) of the treatment of NLBP by blood pricking and cupping. The quality of each included study was evaluated according to the Cochrane Systematic Assessor’s Manual evaluation criteria, while the Meta-analysis was performed using RevMan 5.4software.RESULTS: A total of 13 thirteen RCTs included a total of 1088 subjects. The visual analogue scale scores indicated that blood pricking and cupping was superior to the other treatments at relieving NLBP pain [mean difference(MD) =-1.43;95% confidence interval(CI):-2.31 to 0.54;Z = 3.15;P = 0.002). The Oswestry Disability Index score of blood pricking and cupping was superior to that of the control group in terms of improving NLBP dysfunction(MD =-6.25;95%CI:-8.37 to-4.31);Z = 5.77;P < 0.000 01). 7 RCTs mentioned no adverse reactions, while one study reported mild syncope [7%(n = 3), all in the 17-30 years group] that was gradually relieved after rest. CONCLUSIONS: Blood pricking and cupping therapy can safely and effectively reduce pain and improve functional impairment in patients with NLBP. 8Impact of Interleukin-28B Polymorphism on Response to Treatment in Chinese Hepatitis C Patients显示文摘The candidate polymorphism rs12979860 of interleukin 28B(IL28B) gene was genotyped by means of polymerase chain reaction(PCR) amplification and direct PCR sequencing, and then the role of this single nucleotide polymorphism(SNP) in the response to the treatment of 172 Chinese patients with hepatitis C virus(HCV) chronic infection was analyzed. The results show that the frequencies of major homozygotes(CC) and heterozygotes(CT) of rs12979860 were 0.84 and 0.16, respectively, and the minor homozygotes(TT) wasn't found in this cohort. A highly significant association was found between the CC genotype and sustained virological response(SVR) in HCV geno type 1 infected patients(P<0.001) but not in HCV non-genotype 1 infected patients. In addition, a strong association was found between rs12979860 CC genotype and rapid virological response(RVR) in both HCV genotype 1 infected patients(P<0.001) and non-genotype 1 infected patients(P<0.001). Taken together, these results indicate that IL28B polymorphism plays a key role in response to hepatitis C therapy in Chinese patients. 9Two Ultraviolet Radiation Datasets that Cover China显示文摘Ultraviolet(UV) radiation has significant effects on ecosystems, environments, and human health, as well as atmospheric processes and climate change. Two ultraviolet radiation datasets are described in this paper. One contains hourly observations of UV radiation measured at 40 Chinese Ecosystem Research Network stations from 2005 to 2015. CUV3 broadband radiometers were used to observe the UV radiation, with an accuracy of 5%, which meets the World Meteorology Organization's measurement standards. The extremum method was used to control the quality of the measured datasets. The other dataset contains daily cumulative UV radiation estimates that were calculated using an all-sky estimation model combined with a hybrid model. The reconstructed daily UV radiation data span from 1961 to 2014. The mean absolute bias error and root-mean-square error are smaller than 30% at most stations, and most of the mean bias error values are negative, which indicates underestimation of the UV radiation intensity. These datasets can improve our basic knowledge of the spatial and temporal variations in UV radiation. 12Study of the Contact Force in Free-form Surfaces Compliant EDM Polishing by Robot显示文摘With an elastic negative pole being driven by ultra so nic vibration and being moved along the surface of work-piece compliantly by ro bot, a new kind of effective EDM, the compliant EDM, cuts the electrically condu ctive materials away and polishes work-piece of free-form surface. The study o f the contact force between the end of polishing tool and the surface of work-p iece is the key for the compliant EDM to study its cutting mechanism and to make better use of it. This paper makes a model for the contact force and verifies i t by experiments and simulation based on the theory of elastic body kinetics and dynamic stress concentration. 17Studies on the Factors Affecting on Somatic Embryogenesis in Soybean显示文摘The effects of the concentration of MS micro salts, 2,4-dichlorophenoxyacetic acid (2,4-D), 3-naphthalene acetic acid (NAA), proline and adenine on callus formation and somatic embryogenesis were investigated using orthogonal design with immature cotyledons of soybean. The results showed that the role of concentration of micro salts on frequency of callus formation and somatic embryogenesis were significant. MS medium supplemented with 2,4-D, NAA, proline and adenine could stimulate callus formation. The concentration of MS micro salts possessed obvious effects on somatic embryogenesis of soybean. 20The race to the nociceptor: mechanical versus temperature effects in thermal pain of dental neurons显示文摘The sensing of hot and cold stimuli by dental neurons differs in several fundamental ways. These sensations have been characterized quantitatively through the measured time course of neural discharge signals that result from hot or cold stimuli applied to the teeth of animal models. Although various hypotheses have been proposed to explain the underlying mechanism, the ability to test competing hypotheses against experimental recorded data using biophysical models has been hindered by limitations in our understanding of the specific ion channels involved in nociception of dental neurons. Here we apply recent advances in established biophysical models to test the competing hypotheses. We show that a sharp shooting pain sensa-tion experienced shortly following cold stimulation cannot be attributed to the activation of thermosensitive ion channels, thereby falsifying the so-called neuronal hypothesis,which states that rapidly transduced sensations of coldness are related to thermosensitive ion channels. Our results support a central role of mechanosensitive ion channels and the associated hydrodynamic hypothesis. In addition to the hydrodynamic hypothesis, we also demonstrate that the long time delay of dental neuron responses after hot stimulation could be attributed to the neuronal hypothesis—that a relatively long time is required for the temperature around nociceptors to reach some threshold.
