1Beclin 1 cleavage by caspase-3 inactivates autophagy and promotes apoptosis显示文摘Autophagy and apoptosis are both highly regulated biological processes that play essential roles in tissue homeostasis,development and diseases.Autophagy is also described as a mechanism of death pathways,however,the precise mechanism of how autophagy links to cell death remains to be fully understood.Beclin 1 is a dual regulator for both autophagy and apoptosis.In this study we found that Beclin 1 was a substrate of caspase-3 with two cleavage sites at positions 124 and 149,respectively.Furthermore,the autophagosome formation occurred,followed by the appearance of morphological hallmarks of apoptosis after staurosporine treatment.The cleavage products of Beclin 1 reduced autophagy and promoted apoptosis in HeLa cells and the cells in which Beclin 1 was stably knocked down by specific shRNA.In addition,the cleavage of Beclin 1 resulted in abrogating the interaction between Bcl-2 with Beclin 1,which could be blocked by z-VAD-fmk.Thus,our results suggest that the cleavage of Beclin 1 by caspase-3 may contribute to inactivate autophagy leading towards augmented apoptosis.Yushan Zhu Lixia Zhao Lei Liu Ping Gao Weili Tian Xiaohui Wang Haijing Jin Haidong Xu Quan Chen 2010Protein & Cell2010,1,5:17
2Paleoclimate Modeling in China: A Review显示文摘This paper provides a review of paleoclimate modeling activities in China. Rather than attempt to cover all topics, we have chosen a few climatic intervals and events judged to be particularly informative to the international community. In historical climate simulations, changes in solar radiation and volcanic activity explain most parts of reconstructions over the last millennium prior to the industrial era, while atmospheric greenhouse gas concentrations play the most important role in the20 th century warming over China. There is a considerable model–data mismatch in the annual and boreal winter temperature change over China during the mid-Holocene [6000 years before present(ka BP)], while coupled models with an interactive ocean generally perform better than atmospheric models. For the Last Glacial Maximum(21 ka BP), climate models successfully reproduce the surface cooling trend over China but fail to reproduce its magnitude, with a better performance for coupled models. At that time, reconstructed vegetation and western Pacific sea surface temperatures could have significantly affected the East Asian climate, and environmental conditions on the Qinghai–Tibetan Plateau were most likely very different to the present day. During the late Marine Isotope Stage 3(30–40 ka BP), orbital forcing and Northern Hemisphere glaciation, as well as vegetation change in China, were likely responsible for East Asian climate change. 3Effects of 1-MCP on proline,polyamine,and nitric oxide metabolism in postharvest peach fruit under chilling stress显示文摘Peach(Prunus persica Batsch‘Yuhualu’)fruit are sensitive to chilling injury(CI).Proline,polyamine(PA),and nitric oxide(NO)are important small regulators of various metabolic pathways under chilling stress that mitigate CI.Ethylene is known to promote senescence and CI,while 1-methylcyclopropene(1-MCP)is an antagonist that inhibits the effects of ethylene.However,how1-MCP and ethylene affect proline,PA,and NO levels under chilling stress remains unclear.To address these questions,1-MCP(1μL·L^(−1))and ethylene(1μL·L^(−1))treatments were applied to peach fruit.Fruit were stored at 4°C for 28 d,then moved to 25°C for 3 d immediately after cold storage.Peach fruit exhibited CI symptoms after 7 d of cold storage with enhanced electrolyte leakage and malondialdehyde contents.The 1-MCP treatment significantly(P<0.05)restrained peach CI,and fruit did not exhibit CI symptoms until 14 d of cold storage.Proline and PAs in peach under chilling stress weremostly synthesized from glutamate and arginine,which were catalyzed by1-pyrroline-5-carboxylate synthetase and arginine decarboxylase,respectively.1-MCPtreated fruit exhibited higher proline and PA contents and enhanced chilling tolerance compared to the control,while ethylene-treated fruit had lower proline and PA contents and reduced chilling tolerance.Ethylene-treated fruit,which exhibited more severe CI symptoms compared to the control,had significantly(P<0.05)lower NO contents and NO synthase activities.However,NOmay not be a direct acting factor in 1-MCPinduced chilling tolerance,as 1-MCP-treated fruit had lower NO contents and NO synthase activities compared to the control.In conclusion,proline and PA clearly played direct and important roles in 1-MCP-induced peach chilling tolerance,while NO may not be actively involved.Chunlu Qian Zhengjie Ji Qin Zhu Xiaohua Qi Qianqian Li Jiandong Yin Jun Liu Juan Kan Man Zhang Changhai Jin Lixia Xiao 2021Horticultural Plant Journal2021,7,3:4
4Four-year dynamic observation and study on standardized management of elderly patients with type 2 diabetes in Beijing Yongding Road Community显示文摘Purpose:This study aimed to understand the joint standard of blood glucose,blood li-pids,and blood pressure in elderly patients>60 years of age with type 2 diabetes in the Beijing Yongding Road Community.Methods:One hundred ninety-two elderly patients>60 years of age who were diagnosed with type 2 diabetes in Yongding Road Community served as the investigation subjects,under-went standard management,and the dynamic changes in blood glucose,blood lipids,and blood pressure were monitored for 48 months.Results:At the end of the observation period,the standard rates of fasting blood glucose and postprandial blood glucose were 72.55%and 80.00%,respectively,which were increased compared with 55.73%and 56.08%at baseline(P<0.01).The standard rate of HbA1c was 59.81%,which was increased compared with 53.44%at baseline(P>0.05).The standard rates of TG and LDL-C were 76.71%and 60.38%,respectively,which were increased compared with 54.69%and 34.74%at base-line,and the standard rate of HDL-C was 13.64%,which was decreased compared with 40.10%at baseline(P<0.01).The standard rate of BP was 58.33%,which was increased compared with 38.54%at baseline(P<0.01).The optimal control rate of blood glucose for 48 months(the standard times of the total measurement times in 48 months≥75%)was higher;the fasting blood glucose was 52.17%,the 2-h postprandial blood glucose was 60.22%,and the HbA1c was 46.45%.The optimal control rate of blood lipids was lower;LDL-C was 17.49%and HDL-C was 13.59%.The optimal control rate of BP was 9.13%.At the end of the observation period,the levels of fasting blood glucose and postprandial blood glucose were decreased by 0.7 mmol/L and 1.48 mmol/L,respectively,compared with the base-line(P<0.01).The level of HbA1c was decreased by 0.18%compared with the baseline(P<0.05).The levels of LDL-C and HDL-C were decreased by 0.4 mmol/L and 0.23 mmol/L,respectively,compared with the baseline(P<0.01).The levels of SBP and DBP were decreased by 4 mmHg compared with the baseline(P<0.01).At the end of the observation period,the joint standard rate of the three indices of HbA1c,LDL-C,and BP was 24.72%,which was increased compared with 6.25%at baseline(P<0.01).Conclusion:Standardized management of elderly patients with diabetes in the Community can improve the joint standard rate of blood glucose,blood lipids,and blood pressure.The op-timal control rate and joint standard rate are the important indices for evaluating the quality of diabetes management.Yue Li Dongyun Shen Wei Zhang Yimin Wang Hongmei Li Jie Geng Liying Yang Zhijun Xiao Yan Liu Zhigang Feng Linying Cao Yan Jin Lixia Shi 2014Family Medicine and Community Health2014,2,2:4
564 × 64 GM-APD array-based readout integrated circuit for 3D imaging applications显示文摘Using the high sensitivity of the avalanche photodiode(APD) detector operated in the Geigermode(GM), an array readout integrated circuit(ROIC) comprising a two-segment time-to-digital converter(TDC) is employed for wide-dynamic time interval measurement, where a 1-bit low-segment TDC is implemented by discriminating a single-phase clock period. The proposed 64 × 64 GM-APD array ROIC fabricated using Taiwan semiconductor manufacturing company(TSMC) 0.18-μm complementary metal oxide semiconductor(CMOS) technology can operate at a maximum frequency of 500 MHz provided by an external phase-locked loop clock. The time resolution is reduced to < 1 ns along with a maximum range of4 μs; the differential non-linearity(DNL) and integral non-linearity(INL) are restricted to approximately-0.15 to 0.15 least significant bit(LSB) and-0.3 to 0.32 LSB, respectively; and the power consumption is490 mW under a frame rate of 20 kH z. 6Altitudinal Variation in Digestive Tract Lengh in Feirana quadranus显示文摘Selective pressures favor variation in organ size in response to environmental changes and evolutionary process. In particular, changes in environmental temperature and rainfall at different altitudes often affect food resources, thereby mediating energy intake and allocation. The digestive tract provided a functional relationship between energy intake and allocation, of which gut morphology was associated with diet changes and food quality under different environments. Here we studied altitudinal variation in the digestive tract across four Feirana quadranus populations and tested the hypothesis that relative size of digestive tract should increase with increasing altitude. The results showed that although significant variation in length of the digestive tract was observed in females among populations, altitudinal variation in relative length of digestive tract or gut was non-significant. In addition, the digestive tract length was not correlated with temperature and precipitation across the four populations. 7Carotenoids in Aquaculture:An Overview显示文摘Recent studies on the role of carotenoids in aquaculture have investigated their function in regulating pigmentation,growth,survival,and immune activity of aquatic animals.These carotenoids include astaxanthin,canthaxanthin,lutein,zeaxanthin,and tunaxanthin.Astaxanthin and canthaxanthin are the main synthetic carotenoids used world-wide in the aquaculture industry.They play an important role in protecting cells and organisms against photosensitized oxidations.Direct evidence of carotenoids functioning as antioxidants in vivo has been reported in animal models.In addition to their established biological effects as antioxidants,carotenoids appear to have a great variety of cellular actions that make them remarkable physiological modulators.Consequently,recent attention has been focused on improving our understanding of carotenoids on the physiology and metabolism in aquatic animals with a view to enhance the efficacy of astaxanthin as a carotenoid pigment.This review aims to summarize the function and research progress of carotenoids in aquaculture.We discuss the methods used to study the absorption,transport,metabolic transformation and deposition of carotenoids,and their spatial and temporal expression.Jin Niu Lixia Tian Heizhao Lin Yongjian Liu2 2011Journal of Animal Science and Biotechnology2011,2,1:2

9Paclitaxel formulation with stable sustained-release behavior and its biological safety evaluation显示文摘Biological safety and stable sustained-release of the drug are two crucial issues involved in the formulation of paclitaxel.Focusing on these issues, by using the FDA approved polylactide as carrier material, soybean lecithin as surfactant and maltodextrin as thickener, paclitaxel loaded PLA microspheres were simply prepared by solvent evaporation, thus guaranteeing the biological safety. The introduction of maltodextrin as a thickener aided to a stable sustained-release of paclitaxel. Surface morphology, particle size, drug loading rate, encapsulation efficiency and in vitro drug release behavior were investigated.Biological safety evaluations such as acute toxicity, allergies, hemolysis, skin stimulation and genotoxicity test were also carried out. 11Oxidative coupling of cinnamic acid derivatives and their radical-scavenging activities显示文摘4-Hydroxycinnamic acid derivatives,including ferulic acid (FA) (1),sinapic acid (SA) (2) and caffeic acid (CA) (3),are widely distributed in the plant kingdom,and undergo oxidative cross-coupling leading to the corresponding dehydrodimers,trimers and even higher oligomers in plants. In order to evaluate the antioxidative ability of these 4-hydroxycinnamic acid derivatives and their oligomers,we synthesized 8-8'-bis-lactone-dimers (8-8'-DiFA (4),8-8'-DiSA (5) and 8-8'-DiCA (6)),as well as a new trimer (7),by the reaction of the corresponding monomers (1-3) with Ag2O in methanol,and assayed their free radical-scavenging activity by the reaction with 2,2-diphenyl-1-picrylhydrazyl radical (DPPH·). 16Ethyl acetate fraction in ethanol extract from root of 'Dai-Bai-Jie' (Marsdenia tenacissima):anti-tumor activity in A549cancer cells显示文摘OBJECTIVE: To evaluate the anti-tumor activity of ethyl acetate fraction(EFA), extracted with ethanol from the root of 'Dai-Bai-Jie' in A549 cancer cells and its underlying mechanism.METHODS: 'Dai-Bai-Jie' was extracted with 95%ethanol-aqueous(DBJ-1), 50% ethanol-aqueous(DBJ-2), and water(DBJ-3) by reflux method. 95%ethanol-aqueous extract was separated byethyl acetate(EFA) and n-butyl alcohol(DBJ-5), consecutively. The SRB method was used to evaluate the cytotoxic activity. Annexin V-FITC staining was applied to observe the apoptosis and analyze the cell cycle activated by EFA in A549 tumor cell. Western blot was used to detect the apoptosis/related pro-teins expressions. A549 tumor cellsbearing nude mice model was employed to measure the tumor volume, mice weight, and tumor inhibition ratio in order to verify the antitumor activity in vivo.RESULTS: DBJ-1 and EFA showed better cytotoxic activity on A549 tumor cells with IC5025 and 3.5 μg/mL, respectively. EFA can exhibit the proliferation,arrest cell cycle at G0/G1 phase, and induce apoptosis in A549 tumor cells in vitro. The mechanisms of apoptosis induced by EFA may be associated with decreasing Bcl-2 protein expression and increasing p53, Bax, Caspase-3, and Caspase-8 proteins expression. EFA also possessed significant anti-tumor efficacy in nude mice, and little toxicity was observed in the host.CONCLUSION: EAF could induce A549 tumor cells apoptosis and G0/G1 cell cycle arrest.
