1Hepatitis B and C virus-induced hepatitis: Apoptosis, autophagy, and unfolded protein response显示文摘AIM: To investigate the co-incidence of apoptosis, autophagy, and unfolded protein response(UPR) in hepatitis B(HBV) and C(HCV) infected hepatocytes.METHODS: We performed immunofluorescence confocal microscopy on 10 liver biopsies from HBV and HCV patients and tissue microarrays of HBV positive liver samples. We used specific antibodies for LC3β, cleaved caspase-3, BIP(GRP78), and XBP1 to detect autophagy, apoptosis and UPR, respectively. AntiHCV NS3 and anti-HBs antibodies were also used to confirm infection. We performed triple blind counting of events to determine the co-incidence of autophagy(LC3β punctuate), apoptosis(cleaved caspase-3), and unfolded protein response(GRP78) with HBV and HCV infection in hepatocytes. All statistical analyses were performed using SPSS software for Windows(Version 16 SPSS Inc, Chicago, IL, United States). P-values < 0.05 were considered statistically significant. Statistical analyses were performed with Mann-Whitney test to compare incidence rates for autophagy, apoptosis, and UPR in HBV- and HCV-infected cells and adjacent noninfected cells.RESULTS: Our results showed that infection of hepatocytes with either HBV and HCV induces significant increase(P < 0.001) in apoptosis(cleavage of caspase-3), autophagy(LC3β punctate), and UPR(increase in GRP78 expression) in the HCV- and HBVinfected cells, as compared to non-infected cells of the same biopsy sections. Our tissue microarray immunohistochemical expression analysis of LC3β in HBV^(Neg) and HBV^(Pos) revealed that majority of HBVinfected hepatocytes display strong positive stainingfor LC3β. Interestingly, although XBP splicing in HBVinfected cells was significantly higher(P < 0.05), our analyses show a slight increase of XBP splicing was in HCV-infected cells(P > 0.05). Furthermore, our evaluation of patients with HBV and HCV infection based on stage and grade of the liver diseases revealed no correlation between these pathological findings and induction of apoptosis, autophagy, and UPR.CONCLUSION: The results of this study indicate that HCV and HBV infection activates apoptosis, autophagy and UPR, but slightly differently by each virus. 2Preoperative clinical and paraclinical predictors of choledocholithiasis显示文摘BACKGROUND:Choledocholithiasis is the most common cause of obstructive jaundice and cholangitis and occurs in about 10%of patients with symptomatic gallstone.The aim of this study was to find preoperative and non-invasive tests for predicting common bile duct stones(CBDs). METHODS:Findings of clinical examination,history laboratory tests and ultrasound(US)in 60 patients with CBDs were compared retrospectively,with 60 patients with gallstones.The data were collected from medical charts The sensitivity,specificity and positive predictive value (PPV)were determined. RESULTS:The comparison between the two groups showed significant differences in anorexia,itching,dark urine subicterus(slightly elevated serum bilirubin without clinical evidence of jaundice)and jaundice appearance increased bilirubin level,aspartate aminotransferase(AST) and alanine aminotranferase(ALT),CBD diameter>6 mm and stone observation under US(P<0.05).The highest specificity(100%)and PPV(100%)were found in dark urine and pale colored stool,history of cholangitis,and icterus.Among paraclinical tests,alkaline phosphatase (ALP)had the highest specificity(72.1%)and PPV(12.5%) Under US,stone observation in CBD was the most important factor with a sensitivity of 94.5%,a specificity of 100%and a PPV of 100%. 5甲醇-水二元混合物中Co(bpb)与叠氮离子反应的动力学研究(英文)显示文摘Co(bpb) [bpbH2is N,N′-o-phenylenebis(pyridine-2-carboxamide) ,C18H12N4O2] complex has active axial sites like a porphyrin complex. We studied the coordination of azide ion(N3-) to Co(bpb) in binary methanol-water mixtures by spectrophotometric method at the temperature range of 283-303 K. From the temperature dependence of the rate constant,activation parameters(Ea,ΔH,ΔS,and ΔG) were obtained. An isokinetic temperature at about 302 K was observed at which the formation rate of Co(bpb) -N3-was more or less independent of the solvent composition. The resulting ΔH against TΔS plot showed a good linear correlation,indicating the existence of enthalpy-entropy compensation in azide complexation process. Under optimum conditions and based on the absorbance of Co(bpb) -N3-produced through complex formation,a spectrophotometric method for the determination of N 3-in solution was developed. A linear relationship between the absorbance and N3-concentration was obtained in the range of(0.85-5.00) × 10-4 mol·L-1(3.6-21.0 μg·mL-1) . The detection limit was 2.5×10-5 mol·L-1(1.0 μg·mL-1) . 6Does delayed gastric emptying shorten the H pylori eradication period? A double blind clinical trial显示文摘AIM: To evaluate the gastric emptying inhibitory effects of sugar and levodopa on H pylori eradication period. METHODS: A total of 139 consecutive patients were randomized into 6 groups. The participants with peptic ulcer disease or non-ulcer dyspepsia non-responding to other medications who were also H pylori-positive patients either with positive rapid urease test (RUT) or positive histology were included. All groups were pretreated with omeprazole for 2 d and then treated with quadruple therapy regimen (omeprazole, bismuth, tetracycline and metronidazole); all drugs were given twice daily. Groups 1 and 2 were treated for 3 d, groups 3, 4 and 5 for 7 d, and group 6 for 14 d. Groups 1 to 4 received sugar in the form of 10% sucrose syrup. Levodopa was prescribed for groups 1 and 3. Patients in groups 2 and 4 were given placebo for levodopa and groups 5 and 6 received placebos for both sugar and levodopa. Upper endoscopy and biopsies were carried out before treatment and two months after treatment. Eradication of H pylori was assessed by RUT and histology 8 wk later.RESULTS: Thirty patients were excluded. Per-protocol analysis showed successful eradication in 53% in group 1, 56% in group 2, 58% in group 3, 33.3% in group 4, 28% in group 5, and 53% in group 6. Eradication rate, patient compliance and satisfaction were not significantly different between the groups.
