| 1 | Oxidation state specific analysis of arsenic species in tissues of wild-type and arsenic(+3 oxidation state)methyltransferase-knockout mice显示文摘Arsenic methyltransferase(As3mt) catalyzes the conversion of inorganic arsenic(i As) to its methylated metabolites, including toxic methylarsonite(MAs~Ⅲ) and dimethylarsinite(DMAs~Ⅲ). Knockout(KO) of As3 mt was shown to reduce the capacity to methylate i As in mice. However, no data are available on the oxidation states of As species in tissues of these mice. Here, we compare the oxidation states of As species in tissues of male C57BL/6 As3mt-KO and wild-type(WT) mice exposed to arsenite(iA s~Ⅲ) in drinking water. WT mice were exposed to50 mg/L As and As3mt-KO mice that cannot tolerate 50 mg/L As were exposed to 0, 15, 20, 25 or30 mg/L As. iA s~Ⅲaccounted for 53% to 74% of total As in liver, pancreas, adipose, lung, heart, and kidney of As3mt-KO mice; tri- and pentavalent methylated arsenicals did not exceed 10% of total As. Tissues of WT mice retained iA s and methylated arsenicals: iA s~Ⅲ, MAs~Ⅲand DMAs~Ⅲ represented 55%‐68% of the total As in the liver, pancreas, and brain. High levels of methylated species, particularly MAs~Ⅲ, were found in the intestine of WT, but not As3mt-KO mice,suggesting that intestinal bacteria are not a major source of methylated As. Blood of WT mice contained significantly higher levels of As than blood of As3mt-KO mice. This study is the first to determine oxidation states of As species in tissues of As3mt-KO mice. | 5 | Mitochondrial transplantation strategies as potential therapeutics for central nervous system trauma显示文摘Mitochondria are essential cellular organelles critical for generating adenosine triphosphate for cellular homeostasis, as well as various mechanisms that can lead to both necrosis and apoptosis. The field of 'mitochondrial medicine' is emerging in which injury/disease states are targeted therapeutically at the level of the mitochondrion, including specific antioxidants, bioenergetic substrate additions, and membrane uncoupling agents. Consequently, novel mitochondrial transplantation strategies represent a potentially multifactorial therapy leading to increased adenosine triphosphate production, decreased oxidative stress, mitochondrial DNA replacement, improved bioenergetics and tissue sparing. | 6 | Metabolic syndrome does not affect sustained virologic response of direct-acting antivirals while hepatitis C clearance improves hemoglobin A1c显示文摘AIM To determine whether successful treatment with direc-tacting antivirals(DAA) is associated with improvements in hemoglobin A1 c(HbA1 c) and if type 2 diabetes mellitus(T2 DM) or metabolic syndrome affects sustained virologic response(SVR).METHODS We performed a retrospective analysis of all hepatitis C virus(HCV) patients at the VA Greater Los Angeles Healthcare System treated with varying DAA therapy between 2014-2016. Separate multivariable logistic regression was performed to determine predictors of HbA1 c decrease ≥ 0.5 after DAA treatment and predictors of SVR 12-wk post treatment(SVR12).RESULTS A total of 1068 patients were treated with DAA therapy between 2014-2016. The presence of T2 DM or metabolic syndrome did not adversely affect SVR12. 106 patients had both HCV and T2 DM. Within that cohort,patients who achieved SVR12 had lower mean HbA1 c pre treatment(7.35 vs 8.60,P = 0.02),and lower mean HbA1 c post-treatment compared to non-responders(6.55 vs 8.61,P = 0.01). The mean reduction in HbA1 c after treatment was greater for those who achieved SVR12 than for non-responders(0.79 vs 0.01,P = 0.03). In adjusted models,patients that achieved SVR12 were more likely to have a HbA1 c decrease of ≥ 0.5 than those that did not achieve SVR12(adjusted OR = 7.24,95%CI: 1.22-42.94). CONCLUSION In HCV patients with T2 DM,successful treatment with DAA was associated with a significant reduction in HbA1 c suggesting that DAA may have a role in improving insulin sensitivity. | 7 | Regulation of macrophage activation in the liver after acute injury: Role of the fibrinolytic system显示文摘The liver functions,in part,to prevent exposure of the body to potentially harmful substances ingested in the diet.While it is highly efficient at accomplishing this,it is frequently prone to liver injury due to the biotransformation of xenobiotics into toxic metabolites.To counter this injury,the liver has evolved a unique capacity to rapidly and efficiently repair itself.Successful resolution of acute liver injury relies on hepatic macrophage populations that orchestrate the reparative response.After injury,Kupffer cells,the resident macrophages of the liver,become activated and secrete proinflammatory cytokines.These cytokines recruit other immune cells,including monocyte-derived macrophages,to the liver where they contribute to the repair process.Monocyte-derived macrophages traffic into the necrotic foci where they rapidly phagocytose dead cell debris.Simultaneous with this process,these cells change phenotype from a proinflammatory macrophage to a prorestorative macrophage that produce pro-mitogenic growth factors and antiinflammatory cytokines.Ultimately this process triggers resolution of inflammation,and along with proliferation of other hepatic cells,restores the liver architecture and function.While the mechanisms regulating specific macrophage functions during repair remain to be elucidated,recent studies indicate a key role for the fibrinolytic system in coordinating macrophage function during repair.In this review,we will highlight the function and role of hepatic macrophages in repair after acute liver injury,and will discuss the role of the fibrinolytic enzyme,plasmin,in regulation of these various processes. | Katherine Roth Jenna Strickland Bryan L Copple | 2020 | World Journal of Gastroenterology2020,26,16: | 3 |
| 8 | Biopolymer-nanotube nerve guidance conduit drug delivery for peripheral nerve regeneration:In vivo structural and functional assessment显示文摘Peripheral nerve injuries account for roughly 3%of all trauma patients with over 900,000 repair procedures annually in the US.Of all extremity peripheral nerve injuries,51%require nerve repair with a transected gap.The current gold-standard treatment for peripheral nerve injuries,autograft repair,has several shortcomings.Engineered constructs are currently only suitable for short gaps or small diameter nerves.Here,we investigate novel nerve guidance conduits with aligned microchannel porosity that deliver sustained-release of neurogenic 4-aminopyridine(4-AP)for peripheral nerve regeneration in a critical-size(15 mm)rat sciatic nerve transection model.The results of functional walking track analysis,morphometric evaluations of myelin development,and histological assessments of various markers confirmed the equivalency of our drug-conduit with autograft controls.Repaired nerves showed formation of thick myelin,presence of S100 and neurofilament markers,and promising functional recovery.The conduit’s aligned microchannel architecture may play a vital role in physically guiding axons for distal target reinnervation,while the sustained release of 4-AP may increase nerve conduction,and in turn synaptic neurotransmitter release and upregulation of critical Schwann cell neurotrophic factors.Overall,our nerve construct design facilitates efficient and efficacious peripheral nerve regeneration via a drug delivery system that is feasible for clinical applications. | Ohan S.Manoukian Swetha Rudraiah Michael R.Arul Jenna M.Bartley Jiana T.Baker Xiaojun Yu Sangamesh G.Kumbar | 2021 | Bioactive Materials2021,6,9: | 2 |
| 9 | 甘肃齐家文化中仪式性开颅手术初探(英文)显示文摘作为脑外科手术的一种,开颅术在世界多地史前遗址中均有发现。大量发现表明开颅术在颅骨上的尺寸、位置及手术原因千差万别。开颅术在中国境内亦有发现;但在齐家文化(2300-1500BC)遗址中十分少见。位于甘肃省甘南藏族自治区临潭县陈旗乡的磨沟遗址是一处重要的齐家文化墓葬遗址。本文将重点讨论一例磨沟出土的开颅个体,该成年男性个体(M179:R2)颅骨上有愈合程度较高的开颅术的痕迹;同时,将其与其他同时期(3000~0BC)中国出土的开颅术个体进行比较分析,从而论证阐述开颅术实行的原因及过程。该个体左侧顶骨冠状缝后侧位置处有一圆形穿孔,其小孔边缘不甚规则,且有明显愈合痕迹。穿孔切口的特征显示该穿孔由刮削法完成。但由于该穿孔高度愈合,我们无法准确判断完成穿孔所使用的工具。该个体上开颅术的特征以及相关考古学资料使得作者们认为M179:R2进行开颅术的原因或与巫术仪式有关。同时,磨沟出土的其他带有开颅术个体(大多数是男性,且颅骨开颅处愈合程度较高)也支持这一观点。 | Jenna M DITTMAR 詹小雅 Elizabeth BERGER 毛瑞林 王辉 赵永生 叶惠媛 | 2019 | 人类学学报2019,38,3: | 2 |
