1Splicing factor PRP-19 regulates mitochondrial stress response显示文摘Animals respond to mitochondrial perturbation by activating the mitochondrial unfolded protein response(UPR^(mt))to induce the transcription of mitochondrial stress response genes.In Caenorhabditis elegans,activation of UPR^(mt) allows the animals to maintain organismal homeostasis,activate the innate immune response,and promote lifespan extension.Here,we show that splicing factors such as Precursor RNA processing 19(PRP-19)are required for the induction of UPR^(mt) in C.elegans.PRP-19 also modulates mitochondrial perturbation-induced innate immune response and lifespan extension.Knockdown of PRP-19 in mammalian cells suppresses UPR^(mt) activation and disrupts the mitochondrial network.These findings reveal an evolutionarily conserved mechanism that maintains mitochondrial homeostasis and controls innate immunity and lifespan through splicing factors.Peixue Xia Liankui Zhou Jialiang Guan Wanqiu Ding Ying Liu 2022Life Metabolism2022,1,1:0
2Synthesis of ultrafine dual-phase structure in CrFeCoNiAl_(0.6)high entropy alloy via solid-state phase transformation during sub-rapid solidification显示文摘High entropy alloys(HEAs)with superb mechanical properties have been traditionally produced by solid-ification and subsequent heat treatment,In this paper,we demonstrate a new route via one-step process using sub-rapid cooling.Under proper cooling rates,the CrFeCoNiAl^(0.6) HEA could form ultrafineFCC+BCC dual-phase structure.By varying cooling rate,we can control the fraction of the BCC phaseand refinement of FCC microstructures that have tunable mechanical properties in yield strength and hardness ranging from~580 to~1460 MPa and~260 to~550 Hv.We show that the structure-property-processing relation originates from the sideplate microstructures formed during fast cooling that havespecific crystallographic orientation relationship between the FCC and BCC phases and chemical segrega tion.This work provides a new setting for better understanding of the solid-state phase transformation in HEAs under sub-rapid solidification conditions as well as a novel method for development of high-performance HEAs.Chen Chen Yanzhou Fan Wei Wang Hang Zhang Jialiang Hou Ran Wei Tao Zhang Tan Wang Mo Li Shaokang Guan Fushan Li 2022Journal of Materials Science & Technology2022,,18:0
3A novel N^(6)-Deoxyadenine methyltransferase METL-9 modulates C.elegans immunity via dichotomous mechanisms显示文摘N_(6)-Methyldeoxyadenine(6mA)has been rediscovered as a DNA modification with potential biological function in metazoans.However,the physiological function and regulatory mechanisms regarding the establishment,maintenance and removal of 6mA in eukaryotes are still poorly understood.Here we show that genomic 6mA levels change in response to pathogenic infection in Caenorhabditis elegans(C.elegans).We further identify METL-9 as the methyltransferase that catalyzes DNA 6mA modifications upon pathogen infection.Deficiency of METL-9 impairs the induction of innate immune response genes and renders the animals more susceptible to pathogen infection.Interestingly,METL-9 functions through both 6mA-dependent and-independent mechanisms to transcriptionally regulate innate immunity.Our findings reveal that 6mA is a functional DNA modification in immunomodulation in C.elegans.Chengchuan Ma Tingling Xue Qi Peng Jie Zhang Jialiang Guan Wanqiu Ding Yi Li Peixue Xia Liankui Zhou Tianyu Zhao Sheng Wang Li Quan Chuan-Yun Li Ying Liu 2023Cell Research2023,33,8:0
