1Host metabolism dysregulation and cell tropism identification in human airway and alveolar organoids upon SARS-CoV-2 infection显示文摘The coronavirus disease 2019(COVID-19)pandemic is caused by infection with the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),which is spread primary via respiratory droplets and infects the lungs.Currently widely used cell lines and animals are unable to accurately mimic human physiological conditions because of the abnormal status of cell lines(transformed or cancer cells)and species differences between animals and humans.Organoids are stem cell-derived selforganized three-dimensional culture in vitro and model the physiological conditions of natural organs.Here we showed that SARS-CoV-2 infected and extensively replicated in human embryonic stem cells(hESCs)-derived lung organoids,including airway and alveolar organoids which covered the complete infection and spread route for SARS-CoV-2 within lungs.The infected ceils were ciliated,club,and alveolar type 2(AT2)cells,which were sequentially located from the proximal to the distal airway and terminal alveoli,respectively.Additionally,RNA-seq revealed early cell response to virus infection including an unexpected downregulation of the metabolic processes,especially lipid metabolism,in addition to the well-known upregulation of immune response.Further,Remdesivir and a human neutralizing antibody potently inhibited SARS-CoV-2 replication in lung organoids.Therefore,human lung organoids can serve as a pathophysiological model to investigate the underlying mechanism of SARS-CoV-2 infection and to discover and test therapeutic drugs for COVID-19.Rongjuan Pei Jianqi Feng Yecheng Zhang Hao Sun Lian Li Xuejie Yang Jiangping He Shuqi Xiao Jin Xiong Ying Lin Kun Wen Hongwei Zhou Jiekai Chen Zhili Rong Xinwen Chen 2021Protein & Cell2021,12,9:7

3Vertical Distribution of Soil Carbon, Nitrogen, and Phosphorus in Typical Chinese Terrestrial Ecosystems显示文摘Characterization of the vertical distribution of soil organic carbon(C), nitrogen(N), and phosphorus(P) may improve our ability to accurately estimate soil C, N, and P storage. Based on a database of 21 354 records in 74 long-term monitoring plots from 2004 to 2013 in the Chinese Ecosystem Research Network(CERN), we built fitting functions to quantify the vertical distribution of soil C, N, and P(up to 100 cm depth) in the typical Chinese terrestrial ecosystems. The decrease of soil C, N, and P content with depth can be well fitted with various mathematical functions. The fitting functions differed greatly between artificial(agriculture) and natural(desert, forest, and grassland) ecosystems, and also differed with respect to soil C, N, and P content. In both the artificial and natural ecosystems, the best fitting functions were exponential functions for C, quadratic functions for N, and quadratic functions for P. Furthermore, the stoichiometric ratios of soil C, N, and P were ranked in descending order: grassland > forest > agriculture > desert, and were also associated with climate. This study is the first to build the fitting functions for the profile distribution of soil C, N, and P in China at a national scale. 4Multi-proxy evidence of environmental change related to collapse of the Liangzhu Culture in the Yangtze Delta,China显示文摘The Liangzhu Culture(5300–4300 cal.a BP)has been regarded as the culmination of the Neolithic Age in the Yangtze Delta and as an archaic state.However,the reason behind the collapse of the Liangzhu Culture remains controversial.Some scholars attributed it to social conflicts or foreign invasions,while others suggested that environmental deterioration,such as cold episodes,inundation and palaeo-typhoon events,disrupted the civilization.Notably,the yellowish silty sediments widely overlying the Liangzhu group sites have been presumed to be one of the vital clues to resolving these issues.The process and cause of their formation are key to understanding the decline in the Liangzhu Culture.In this study,two typical profiles composed of the Liangzhu cultural layer and yellowish silty sediments were collected in Liangzhu Ancient City.Multiple proxies,including pollen,phytoliths and diatoms,along with radiocarbon and optically stimulated luminescence dating and synthesized geochemical evidence from different sediments,were applied to reconstruct ecological and hydrological changes and discuss their linkage to cultural collapse.The regional ecology was typical freshwater wetlands during 4800–4400 cal.a BP,which facilitated rice farming and large-scale architectural engineering in Liangzhu Ancient City.Evidence from microfossils and dating of the yellowish silty sediments indicated that Liangzhu Ancient City was significantly affected by seawater after abandonment during 4300–3400 cal.a BP.Furthermore,geochemical evidence,including trace element and Sr-Nd isotopic compositions,suggested that the primary provenance of the yellowish silt was a mixture of sediments from the Qiantang River,East China Sea,Yangtze River and Yellow River,which had been transported to Liangzhu Ancient City by the Yellow Sea Coastal Current and a marine transgression.Therefore,based on evidence from archaeological sites and numerical simulation in the Yangtze Delta,it was supposed that the short-term accelerated relative sea-level rise at approximately 4400 cal.a BP caused a marine transgression around Taihu Lake,formed a large range of yellowish silty sediments,destroyed rice farming production,and ultimately led to the collapse of the Liangzhu Culture.Keyang HE Houyuan LU Guoping SUN Xiang JI Yonglei WANG Kaikai YAN Xinxin ZUO Jiangping ZHANG Bin LIU Ningyuan WANG 2021Science China Earth Sciences2021,64,6:3

15Polydopamine-Assisted Fabrication of Stable Silver Nanoparticles on Optical Fiber for Enhanced Plasmonic Sensing显示文摘We present a facile and effective method for fabrication of the localized surface plasmon resonance(LSPR)optical fiber sensor assisted by two polydopamine(PDA)layers with enhanced plasmonic sensing performance.The first PDA layer was self-polymerized onto the bare optical fiber to provide the catechol groups for the reduction from Ag^(+)to Ag^(o) through chelating and redox activity.As the reduction of Ag^(+)proceeds,Ag nanoparticles(NPs)were grown in-situ on the PDA layer with uniform distribution.The second PDA layer was applied to prevent Ag NPs from oxidating and achieve an improvement of LSPR signal.The PDA/Ag/PDA-based optical fiber sensor has an enhanced LSPR sensitivity of 961 nm/RIU and excellent oxidation resistance.The stable PDA/Ag/PDA-based LSPR sensor with high optical performance is very promising for future application in optical sensing field.Yiwen TANG Hui YUAN Jiangping CHEN Qiguo XING Rongxin SU Wei QI Zhimin HE 2020Photonic Sensors2020,10,2:0
