3Effect of Linguizhugan decoction on hyperlipidemia rats with intermittent fasting显示文摘OBJECTIVE:To explore the effect of Linguizhugan decoction on hyperlipidemia rats with caloric restriction.METHODS:The hyperlipidemia model of rat was induced by high fat diet for 8 weeks.After the model was established,26 rats were randomly divided into 4 groups:the control group(n=6),the model group(n=6),the intermittent fasting(IF) group(n= 8),and the IF and herbal medicine(IFH) group(n= 6).IF group was applied intermittent fasting every other day.The IFH group was given Linguizhugan decoction every day and intermittent fasting every other day.Blood samples were taken at the end of 16 weeks,and serum ghrelin and lipid was tested.RESULTS:Serum ghrelin in the IF group significantly increased(P<0.01).Serum ghrelin in IFH group was lower than the IF group(P<0.05),but higher than the model group(P<0.01).CONCLUSION:Linguizhugan decoction may play a part in regulation of energy and appetite in hyperlipidemia rats with IF.Yubin Yang Jian Qin Bin Ke Junjie Zhang Lanying Shi Qiong Li 2013Journal of Traditional Chinese Medicine2013,33,2:6
4Diffusion mechanism simulation of cloud manufacturing complex network based on cooperative game theory显示文摘Cloud manufacturing is a specific implementation form of the 'Internet + manufacturing' strategy. Why and how to develop cloud manufacturing platform(CMP), however, remains the key concern of both platform operators and users. A microscopic model is proposed to investigate advantages and diffusion forces of CMP through exploration of its diffusion process and mechanism. Specifically, a three-stage basic evolution process of CMP is innovatively proposed. Then, based on this basic process, a more complex CMP evolution model has been established in virtue of complex network theory, with five diffusion forces identified. Thereafter, simulations on CMP diffusion have been conducted. The results indicate that, CMP possesses better resource utilization,user satisfaction, and enterprise utility. Results of simulation on impacts of different diffusion forces show that both the time required for CMP to reach an equilibrium state and the final network size are affected simultaneously by the five diffusion forces. 5CRISPR-Cas9-based genome-wide screening identified novel targets for treating sorafenib-resistant hepatocellular carcinoma:a cross-talk between FGF21 and the NRF2 pathway显示文摘The treatment of hepatocellular carcinoma(HCC)has been dominated by multikinase inhibitors for more than a decade.However,drug resistance can severely restrict the efficacy of these drugs.Using CRISPR/CAS9 genome library screening,we evaluated Kelch-like ECH-associated protein 1(KEAP1)as a key regulator of sorafenib’s susceptibility in HCC.We also investigated whether KEAP1’s knockdown can stabilize nuclear factor(erythroid-derived 2)-like 2(NRF2)protein levels that led to sorafenib’s resistance,including an NRF2 inhibitor that can synergize with sorafenib to abolish HCC’s growth in vitro and in vivo.Furthermore,we clarified that fibroblast growth factor 21(FGF21)is an important downstream regulator of NRF2 in HCC.Intriguingly,we observed that FGF21 bound to NRF2 through the C-terminus of FGF21,thereby stabilizing NRF2 by reducing its ubiquitination and generating a positive feedback loop in sorafenib-resistant HCC.These findings,therefore,propose that targeting FGF21 is a promising strategy to combat HCC sorafenib’s resistance.Jiang Chen Shi Jiang Huijiang Shao Bixia Li Tong Ji Daniel Staiculescu Jiayan He Jie Zhao Liuxin Cai Xiao Liang Junjie Xu Xiujun Cai 2022Science China(Life Sciences)2022,65,10:4
6Monitoring of atmospheric radionuclides from the Fukushima nuclear accident and assessing their impact on Xi'an, China显示文摘Aerosol radionuclides (131I, 134Cs, 137Cs) and gaseous radioactive xenon (133Xe) were monitored at Xi'an, China following the accident at the Fukushima nuclear power plant in March 2011. The additional annual effective dose attributable to the Fukushima emissions was much lower than the public annual effective dose from natural radiation, according to Chinese national standards. The monitoring results were compared with data from other countries as well as with the radionuclide concentrations observed in Xi'an after the Chernobyl nuclear accident in 1986. Possible transport pathways of the released radionuclides from Fukushima to Xi'an were investigated. 8Rice metabolic regulatory network spanning the entire life cycle显示文摘As one of the most important crops in the world,rice(Oryza sativa)is a model plant for metabolome research.Although many studies have focused on the analysis of specific tissues,the changes in metab-olite abundance across the entire life cycle have not yet been determined.In this study,combining both tar-geted and nontargeted metabolite profiling methods,a total of 825 annotated metabolites were quantified in rice samples from different tissues covering the entire life cycle.The contents of metabolites in different tissues of rice were significantly different,with various metabolites accumulating in the plumule and radicle during seed germination.Combining these data with transcriptome data obtained from the same time period,we constructed the Rice Metabolic Regulation Network.The metabolites and co-expressed genes were further divided into 12 clusters according to their accumulation patterns,with members within each cluster displaying a uniform and clear pattern of abundance across development.Using this dataset,we established a comprehensive metabolic profile of the rice life cycle and used two independent strategies to identify novel transcription factors-namely the use of known regulatory genes as bait to screen for new networks underlying lignin metabolism and the unbiased identification of new glycerophospholipid metabolism regulators on the basis of tissue specificity.This study thus demonstrates how guilt-by-association analysis of metabolome and transcriptome data spanning the entire life cycle in cereal crops provides novel resources and tools to aid in understanding the mechanisms underlying important agro-nomic traits.Chenkun Yang Shuangqian Shen Shen Zhou Yufei Li Yuyuan Mao Junjie Zhou Yuheng Shi Longxu An Qianqian Zhou Wenju Peng Yuanyuan Lyu Xuemei Liu Wei Chen Shouchuang Wang Lianghuan Qu Xianqing Liu Alisdair R.Fernie Jie Luo 2022Molecular Plant2022,15,2:3
9A new cell death program regulated by toll-like receptor 9 through p38 mitogen-activated protein kinase signaling pathway in a neonatal rat model with sepsis associated encephalopathy显示文摘Background:Sepsis,a serious condition with high mortality,usually causes sepsis associated encephalopathy(SAE)that involves neuronal cell death.However,the cell death programs involved and their underlying mechanisms are not clear.This study aimed to explore the regulatory mechanisms of different cell death programs in SAE.Methods:A neonatal rat model of SAE was established by cecal ligation and perforation.Survival rate and vital signs(mean arterial pressure and heart rate)were monitored,nerve reflexes were evaluated,and cortical pathological changes were observed by hematoxylin and eosin staining.The expression of pyroptosis,apoptosis,and necroptosis(PANoptosis)-related proteins,mitogen-activated protein kinase(MAPK),and its upstream regulator toll-like receptor 9(TLR9)were detected.The expression of TLR9 in neurons was observed by immunofluorescence staining.The ultrastructure of neurons was observed by transmission electron microscope.Results:First,PANoptosis was found in cortical nerve cells of the SAE rats.Meanwhile,the subunits of MAPKs,p38 MAPK,Jun N-terminal kinase,and extracellular signal-regulated kinase(ERK)were activated.After pharmacologically inhibiting each of the subunits,only p38 MAPK was found to be associated with PANoptosis.Furthermore,blocking the p38 MAPK signaling pathway activated necroptosis but inhibited apoptosis and pyroptosis.When necroptosis was pharmacologically inhibited,apoptosis and pyroptosis were reactivated.Finally,we found that the expression of TLR9,a regulator of MAPKs,was significantly increased in this model.After down-regulation of TLR9,p38 MAPK,and ERK signaling pathways were inhibited,which led to the inhibition of PANoptosis.Further analysis found that down-regulation of TLR9 improved the survival rate and reduced the pathological changes in SAE rats.Conclusions:Our study showed that the programs comprising PANoptosis are activated simultaneously in SAE rats.TLR9 activated PANoptosis through the p38 MAPK signaling pathway.TLR9 may work as a potential target for SAE treatment.Ruixi Zhou Junjie Ying Xia Qiu Luting Yu Yan Yue Qian Liu Jing Shi Xihong Li Yi Qu Dezhi Mu 2022Chinese Medical Journal2022,,12:3
10Synergistic effects of multiple driving factors on the runoff variations in the Yellow River Basin,China显示文摘River runoff plays an important role in watershed ecosystems and human survival,and it is controlled by multiple environmental factors.However,the synergistic effects of various large-scale circulation factors and meteorological factors on the runoff on different time-frequency scales have rarely been explored.In light of this,the underlying mechanism of the synergistic effects of the different environmental factors on the runoff variations was investigated in the Yellow River Basin of China during the period 1950-2019 using the bivariate wavelet coherence(WTC)and multiple wavelet coherence(MWC)methods.First,the continuous wavelet transform(CWT)method was used to analyze the multiscale characteristics of the runoff.The results of the CWT indicate that the runoff exhibited significant continuous or discontinuous annual and semiannual oscillations during the study period.Scattered inter-annual time scales were also observed for the runoff in the Yellow River Basin.The meteorological factors better explained the runoff variations on seasonal and annual time scales.The average wavelet coherence(AWC)and the percent area of the significant coherence(PASC)between the runoff and individual meteorological factors were 0.454 and 19.89%,respectively.The circulation factors mainly regulated the runoff on the inter-annual and decadal time scales with more complicated phase relationships due to their indirect effects on the runoff.The AWC and PASC between the runoff and individual circulation factors were 0.359 and 7.31%,respectively.The MWC analysis revealed that the synergistic effects of multiple factors should be taken into consideration to explain the multiscale characteristic variations of the runoff.The AWC or MWC ranges were 0.320-0.560,0.617-0.755,and 0.819-0.884 for the combinations of one,two,and three circulation and meteorological factors,respectively.The PASC ranges were 3.53%-33.77%,12.93%-36.90%,and 20.67%-39.34%for the combinations one,two,and three driving factors,respectively.The combinations of precipitation,evapotranspiration(or the number of rainy days),and the Arctic Oscillation performed well in explaining the variability in the runoff on all time scales,and the average MWC and PASC were 0.847 and 28.79%,respectively.These findings are of great significance for improving our understanding of hydro-climate interactions and water resources prediction in the Yellow River Basin.WANG Junjie SHI Bing ZHAO Enjin CHEN Xuguang YANG Shaopeng 2021Journal of Arid Land2021,13,8:3
11Long-term clinical observation of patients with acute and chronic complete spinal cord injury after transplantation of Neuro Regen scaffold显示文摘Spinal cord injury(SCI)often results in an inhibitory environment at the injury site.In our previous studies,transplantation of a scaffold combined with stem cells was proven to induce neural regeneration in animal models of complete SCI.Based on these preclinical studies,collagen scaffolds loaded with the patients’own bone marrow mononuclear cells or human umbilical cord mesenchymal stem cells were transplanted into SCI patients.Fifteen patients with acute complete SCI and 51 patients with chronic complete SCI were enrolled and followed up for 2 to 5 years.No serious adverse events related to functional scaffold transplantation were observed.Among the patients with acute SCI,five patients achieved expansion of their sensory positions and six patients recovered sensation in the bowel or bladder.Additionally,four patients regained voluntary walking ability accompanied by reconnection of neural signal transduction.Among patients with chronic SCI,16 patients achieved expansion of their sensation level and 30 patients experienced enhanced reflexive defecation sensation or increased skin sweating below the injury site.Nearly half of the patients with chronic cervical SCI developed enhanced finger activity.These long-term follow-up results suggest that functional scaffold transplantation may represent a feasible treatment for patients with complete SCI.Fengwu Tang Jiaguang Tang Yannan Zhao Jiaojiao Zhang Zhifeng Xiao Bing Chen Guang Han Na Yin Xianfeng Jiang Changyu Zhao Shixiang Cheng Ziqiang Wang Yumei Chen Qiaoling Chen Keran Song Zhiwei Zhang Junjie Niu Lingjun Wang Qin Shi Liang Chen Huilin Yang Shuxun Hou Sai Zhang Jianwu Dai 2022Science China(Life Sciences)2022,65,5:3

13Clinical Prediction Models for Hepatitis B Virus-related Acute-on-chronic Liver Failure:A Technical Report显示文摘Background and Aims:It is critical but challenging to predict the prognosis of hepatitis B virus-related acute-on-chronic liver failure(HBV-ACLF).This study systematically summarized and evaluated the quality and performance of available clinical prediction models(CPMs).Methods:A keyword search of articles on HBV-ACLF CPMs published in PubMed from January 1995 to April 2020 was performed.Both the quality and performance of the CPMs were assessed.Results:Fifty-two CPMs were identified,of which 31 were HBV-ACLF specific.The modeling data were mostly derived from retrospective(83.87%)and single-center(96.77%)cohorts,with sample sizes ranging from 46 to 1,202.Three-month mortality was the most common endpoint.The Asian Pacific Association for the Study of the Liver consensus(51.92%)and Chinese Medical Association liver failure guidelines(40.38%)were commonly used for HBV-ACLF diagnosis.Serum bilirubin(67.74%),the international normalized ratio(54.84%),and hepatic encephalopathy(51.61%)were the most frequent variables used in models.Model discrimination was commonly evaluated(88.46%),but model calibration was seldom performed.The model for end-stage liver disease score was the most widely used(84.62%);however,varying performance was reported among the studies.Conclusions:Substantial limitations lie in the quality of HBV-ACLF-specific CPMs.Disease severity of study populations may impact model performance.The clinical utility of CPMs in predicting short-term prognosis of HBV-ACLF remains to be undefined.Xia Yu Yi Lu Shanshan Sun Huilan Tu Xianbin Xu Kai Gong Junjie Yao Yu Shi Jifang Sheng 2021Journal of Clinical and Translational Hepatology2021,9,6:3

15Evolution of microstructures and intermetallic compounds at bonding interface in friction stir welding of dissimilar Al/Mg alloys with/without ultrasonic assistance显示文摘Complete understanding of the evolution behaviors of the microstructures and intermetallic compounds(IMCs)along the interface materials flow path in friction stir welding(FSW)of dissimilar Al to Mg alloys is of great significance.In this study,conventional FSW and ultrasonic vibration enhanced FSW(UVeFSW)experiments of Al/Mg alloys were performed,and the instantaneous evolution features of the interface materials around the tool were'frozen'by using the'sudden stop'and simultaneous cooling techniques.The microstructures and IMCs formation at different locations around the exit hole were observed and characterized by scanning electron microscope,energy dispersive spectrometer and transmission elec-tron microscope.It was found that before the materials started to deposit near the back of the tool,“IMC+Mg+IMC+Al”multilayer microstructure and simple IMC layer with(β+γ)sequentially emerged on the Al/Mg interface.With the application of ultrasonic vibration,the multi-layered interface structure only appeared at the middle stage of materials flow around the pin,and ultrasonic vibration just began to play a suppression role on the growth of two sub-layers IMC at a position where the materials deposit.With assistance of ultrasonic vibration in UVeFSW,the tool drove a larger volume of Mg alloy to move toward the retreating side,and the final IMCs thickness was thinner than that in FSW.Junjie Zhao ChuanSong Wu Lei Shi Hao Su 2023Journal of Materials Science & Technology2023,,8:2

18Exploring the energy transfer processes in Lu2(1-x)Y2xSiO5:Ce crystals显示文摘A systematical exploration of energy transfer processes in Lu_(2(1-x)Y_2 xSiO_5:Ce(LYSO) crystals under vacuum ultraviolet-ultraviolet(VUV-UV) excitation was implemented. The relationship between energy transfer and scintillation properties was established. It is revealed that there are mainly three energy transfer types in the crystal i.e. host → Ce1/Ce2/STEs, Ce1 →Ce2 and STEs → Ce1/Ce2. The influence of Y content of the LYSO crystals on the energy transfer efficiency of the above processes was carefully analyzed.
