Characterization and Mapping of a Salt-Sensitive Mutant in Rice (Oryza sativa L.)显示文摘A salt-sensitive mutant designated rice salt sensitive 2 (rss2) was isolated from the M2 generation of the rice cultivar Nipponbare (Oryza sativa L. ssp. japonica) mutagenized with ethyl methanesulfonate (EMS). This mutant exhibited a greater decrease in salt tolerance with a significant increase in Na+ content in its shoots. Genetic analysis indicated that the increase in Na+ in rss2 was controlled by a single recessive gene. Further genome-wide analysis of the linkage map constructed from the F2 population of rss2/Zhaiyeqing 8 (ZYQ8) showed that two quantitative trait loci (QTLs) on chromosomes 1 and 6 were responsible for the Na+ concentration in shoots, which explained 14.5% and 53.3%, respectively, of the phenotypic variance. The locus on chromosome 1, but not that on chromosome 6, was also detected in the F2 population of Nipponbare/ZYQ8, suggesting that the QTL on chromosome 6 was responsible for the salt sensitivity in rss2. By analyzing the recombination events in 220 mutant individuals of an enlarged mapping population of rss2/ZYQ8, the rss2 locus was precisely mapped to an interval of 605.3 kb between insertion/deletion (InDel) markers IM21962 and IM22567. This finding will facilitate the cloning of the rss2 locus and provide insight into the physiological mechanisms of salt sensitivity in rice. | Jingwei Zhou Fuzheng Wang Ping Deng Wen Jing Wenhua Zhang | 2013 | Journal of Integrative Plant Biology2013,55,6: | 5
Cancer metabolism and tumor microenvironment:fostering each other?显示文摘The changes associated with malignancy are not only in cancer cells but also in environment in which cancer cells live.Metabolic reprogramming supports tumor cells’high demand of biogenesis for their rapid proliferation, and helps tumor cells to survive under certain genetic or environmental stresses.Emerging evidence suggests that metabolic alteration is ultimately and tightly associated with genetic changes,in particular the dysregulation of key oncogenic and tumor suppressive signaling pathways.Cancer cells activate HIF signaling even in the presence of oxygen and in the absence of growth factor stimulation. This cancer metabolic phenotype, described firstly by German physiologist Otto Warburg, ensures enhanced glycolytic metabolism for the biosynthesis of macromolecules. The conception of metabolite signaling,i.e.,metabolites are regulators of cell signaling, provides novel insights into how reactive oxygen species(ROS)and other metabolites deregulation may regulate redox homeostasis, epigenetics, and proliferation of cancer cells. Moreover, the unveiling of noncanonical functions of metabolic enzymes, such as the moonlighting functions of phosphoglycerate kinase 1(PGK1),reassures the importance of metabolism in cancer development. The metabolic, microRNAs,and ncRNAs alterations in cancer cells can be sorted and delivered either to intercellular matrix or to cancer adjacent cells to shape cancer microenvironment via media such as exosome.Among them, cancer microenvironmental cells are immune cells which exert profound effects on cancer cells.Understanding of all these processes is a prerequisite for the development of a more effective strategy to contain cancers. | Yiyuan Yuan Huimin Li Wang Pu Leilei Chen Dong Guo Hongfei Jiang Bo He Siyuan Qin Kui Wang Na Li Jingwei Feng Jing Wen Shipeng Cheng Yaguang Zhang Weiwei Yang Dan Ye Zhimin Lu Canhua Huang Jun Mei Hua-Feng Zhang Ping Gao Peng Jiang Shicheng Su Bing Sun Shi-Min Zhao | 2022 | Science China(Life Sciences)2022,65,2: | 5
Design and baseline data of a populationbased metabonomics study of eye diseases in eastern China: the Yueqing Ocular Diseases Investigation显示文摘Background:China is undergoing a massive transition toward an urban and industrial economy. These changes will restructure the demographics and economy which will eventually influence the future patterns of disease. The risk factors of vision-impairing eye diseases remain ambiguous and poorly understood.Metabolomics is an ideal tool to understand and shed light on the ocular disease mechanisms for earlier treatment. This article aims to describe the design, methodology and baseline data of the Yueqing Ocular Diseases Investigation(YODI),a developed county population-based study to determine the prevalence and primary causes of visual impairment;also with metabonomics analysis we aimed to identify, predict and suggest some preventive biomarkers that cause blindness. Methods:A population-based,cross-sectional study.Randomized clustering sampling was used to identify adults aged 50 years and older in Xiangyang Town,Yueqing county-level City. The interviews covered demographic, behavioral, ocular risk factors and mental health state. The ocular examination included visual acuity, autorefraction, intraocular pressure, anterior and posterior segment examinations, fundus photography, retinal tomography and angiography, and visual field testing.Anthropometric measurements included height and weight, waist and hip circumference, blood pressure, pulse rate, electrocardiogram, and abdominal ultrasound scan.A venous blood sample was collected for laboratory tests and metabonomics studies. Results:Of the 5319 individuals recruited for the YODI,4769(89.7%)subjects were enrolled for analyses. The median age was 62.0 years, and 45.6%were male. The educational level of illiteracy or semi-illiteracy,primary,middle and high school or above was 29.8%,45.5%,20.1%,and 3.3%,respectively.Majority of the participants were female, younger, and less educated when compared with nonparticipants. The average body mass index and waist-hip ratios were 24.4±3.4 kg/m^(2) and 0.9±0.1 respectively.Blood sample collection reached a sample size of 1909(479 from subjects with selfreported diabetes and 1430 from one-third of the 4290 subjects without self-reported diabetes). Conclusions:The YODI provides population-based data with a high response rate(89.7%)on the prevalence and primary causes of major vision-impairing eye diseases in developed county areas in eastern China.Metabonomics analysis from YODI will provide further association of metabolic characteristics with the visual impairment eye diseases. The risk prediction model could be created and has the potential to be generalized to developed eastern areas in China for prevention. | Yuxuan Deng Yuanbo Liang Sigeng Lin Liang Wen Jin Li Yue Zhou Meixiao Shen Jingwei Zheng Kemi Feng Yanting Sun Kwapong Willaim Robert Jia Qu Fan Lu | 2020 | Eye and Vision2020,7,1: | 4

scNanoATAC-seq:a long-read single-cell ATAC sequencing method to detect chromatin accessibility and genetic variants simultaneously within an individual cell显示文摘Dear Editor,The accessible chromatin regions were enriched in regulatory elements in human genome and many genetic variations associated with human diseases were identified within them.1 Single-cell assay for transposase-accessible chromatin using sequencing(scATAC-seq)on the next-generation sequencing(NGS)platform is a well-established method to detect open chromatin regions within an individual cell.2 However, there remain challenges in detection of large-scale structural variations(SVs,including insertions, deletions, duplications, inversions and translocations)and haplotype phasing from scATAC-seq data, which can be well resolved by third-generation sequencing(TGS)platform-based single-molecule long-read sequencing. To integrate advantages of long-read sequencing into scATAC-seq,we developed single-cell assay for transposase‐accessible chromatin on Nanopore sequencing platform(scNanoATAC-seq),a platebased scATAC-seq method suitable for TGS platform(Supplementary information,Fig.S1). | Yuqiong Hu Zhenhuan Jiang Kexuan Chen Zhangxian Zhou Xin Zhou Yan Wang Jingwei Yang Bo Zhang Lu Wen Fuchou Tang | 2023 | Cell Research2023,33,1: | 1

Five new terpenoids from Viburnum odoratissimum var.sessiliflorum显示文摘Five new terpenoids, including two vibsane-type diterpenoids(1,2)and three iridoid allosides(3-5),together with eight known ones, were isolated from the leaves and twigs of Viburnum odoratissimum var.sessiliflorum.Their planar structures and relative configurations were determined by spectroscopic methods, especially 2D NMR techniques. The sugar moieties of the iridoids were confirmed asβ-D-allose by GC analysis after acid hydrolysis and acetylation. The absolute configurations of neovibsanin Q(1)and dehydrovibsanol B(2)were determined by quantum chemical calculation of their theoretical electronic circular dichroism(ECD)spectra and Rh2(OCOCF_(3))_(4)-induced ECD analysis. The anti-inflammatory activities of compounds 1,3,4,and 5 were evaluated using an LPS-induced RA W264.7 cell model.Compounds 3 suppressed the release of NO in a dose-dependent manner, with an IC_(50) value of 55.64μmol·L^(-1).The cytotoxicities of compounds 1-5 on HCT-116 cells were assessed and the results showed that compounds 2 and 3 exhibited moderate inhibitory activities with IC_(50) values of 13.8 and 12.3μmol·L^(-1),respectively. | LI Yang JIAN Yajiao XU Fan LUO Yongxin LI Zhixuan OU Yi WEN Yan JIN Jingwei ZHANG Chuarui GAN Lishe | 2023 | Chinese Journal of Natural Medicines2023,21,4: | 0

Optical experimental solution for the multiway number partitioning problem and its application to computing power scheduling显示文摘Quantum computing is an emerging technology that is expected to realize an exponential increase in computing power. Recently, its theoretical foundation and application scenarios have been extensively researched and explored. In this work, we propose efficient quantum algorithms suitable for solving computing power scheduling problems in the cloud-rendering domain, which can be viewed mathematically as a generalized form of a typical NP-complete problem, i.e., a multiway number partitioning problem. In our algorithm, the matching pattern between tasks and computing resources with the shortest completion time or optimal load balancing is encoded into the ground state of the Hamiltonian;it is then solved using the optical coherent Ising machine, a practical quantum computing device with at least 100 qubits. The experimental results show that the proposed quantum scheme can achieve significant acceleration and save 97% of the time required to solve combinatorial optimization problems compared with classical algorithms. This demonstrates the computational advantages of optical quantum devices in solving combinatorial optimization problems. Our algorithmic and experimental work will advance the utilization of quantum computers to solve specific NP problems and will broaden the range of possible applications. | Jingwei Wen Zhenming Wang Zhiguo Huang Dunbo Cai Bingjie Jia Chongyu Cao Yin Ma Hai Wei Kai Wen Ling Qian | 2023 | Science China(Physics,Mechanics & Astronomy)2023,66,9: | 0
Prophylaxis of posterior capsule opacification through autophagy activation with indomethacin-eluting intraocular lens显示文摘Posterior capsule opacification(PCO)is the most common long-term postoperative complication of cataract surgery, leading to secondary vision loss.Optimized intraocular lens(IOL)structure and appropriate pharmacological intervention, which provides physical barriers and biological inhibition, respectively, can block the migration, proliferation, and epithelial-mesenchymal transition(EMT)of lens epithelial cells(LECs)for PCO prophylaxis.Herein,a novel indomethacin-eluting IOL(INDOM-IOL)with an optimized sharper edge and a sustained drug release behavior was developed for PCO prevention.Indomethacin(INDOM),an ophthalmic non-steroidal anti-inflammatory drug(NSAID)used for postoperative ocular inflammation, was demonstrated to not only be able to suppress cell migration and down-regulate the expression of cyclooxygenase-2(COX-2)and EMT markers, including alpha-smooth muscle actin(α-SMA)and cyclin D1,but also promote the autophagy activation in LECs.Additionally, autophagy was also verified to be a potential therapeutic target for the down-regulation of EMT in LECs.The novel IOL,serving as a drug delivery platform, could carry an adjustable dose of hydrophobic indomethacin with sustained drug release ability for more than 28 days.In the rabbit PCO model, the indomethacin-eluting IOL showed excellent anti-inflammatory and anti-PCO effects. In summary, indomethacin is an effective pharmacological intervention in PCO prophylaxis, and the novel IOL we developed prevented PCO in vivo under its sustained indomethacin release property, which provided a promising approach for PCO prophylaxis in clinical application. | Xiaobo Zhang Jing Wang Jingwei Xu Wen Xu Yin Zhang Chenqi Luo Shuang Ni Haijie Han Xingchao Shentu Juan Ye Jian Ji Ke Yao | 2023 | Bioactive Materials2023,,5: | 0
Under-ice ambient noise in the Arctic Ocean: observations at the long-term ice station显示文摘Under-ice ambient noise in the Arctic Ocean is studied using the data recorded by autonomous hydrophones at the long-term ice station during the 9th Chinese National Arctic Research Expedition.Time-frequency analysis of two 7-s-long ice-induced noise samples shows that both ice collision and ice breaking noise have many outliers in the time-domain(impulsive characteristic)and abundant frequency components in the frequency-domain.Ice collision noise lasts for several seconds while the duration of ice breaking noise is much shorter(i.e.,less than tens of milliseconds).Gaussian distribution and symmetric alpha stable(sαs)distribution are used in this paper to fit the impulsive under-ice noise. The sαs distribution can achieve better performance as it can track the heavy tails of impulsive noise while Gaussian distribution fails. This paper also analyzes the meteorological variables during the under-ice noise observation experiment and deduces that the impulsive ambient noise was caused by the combined force of high wind speed and increasing atmosphere temperature on the ice canopy. The Pearson correlation coefficients between long-term power spectral density variations of under-ice ambient noise and meteorological variables are also studied in this paper. | Xiao Han Jingwei Yin Yanming Yang Hongtao Wen Longxiang Guo | 2020 | Acta Oceanologica Sinica2020,39,9: | 0
Reconstructing unknown quantum states using variational layerwise method显示文摘In order to gain comprehensive knowledge of an arbitrary unknown quantum state, one feasible way is to reconstruct it,which can be realized by finding a series of quantum operations that can refactor the unitary evolution producing the unknown state. We design an adaptive framework that can reconstruct unknown quantum states at high fidelities, which utilizes SWAP test, parameterized quantum circuits(PQCs)and layerwise learning strategy. We conduct benchmarking on the framework using numerical simulations and reproduce states of up to six qubits at more than 96%overlaps with original states on average using PQCs trained by our framework, revealing its high applicability to quantum systems of different scales theoretically. Moreover,we perform experiments on a five-qubit IBM Quantum hardware to reconstruct random unknown single qubit states, illustrating the practical performance of our framework. For a certain reconstructing fidelity, our method can effectively construct a PQC of suitable length, avoiding barren plateaus of shadow circuits and overuse of quantum resources by deep circuits, which is of much significance when the scale of the target state is large and there is no a priori information on it.This advantage indicates that it can learn credible information of unknown states with limited quantum resources, giving a boost to quantum algorithms based on parameterized circuits on near-term quantum processors. | Junxiang Xiao Jingwei Wen Shijie Wei Guilu Long | 2022 | Frontiers of physics2022,17,5: | 0
